[Prediction and estimation on molar response values of alkane using molecular subgraph].

A new method based on topographical subgraph of alkanes is proposed and used for molar response values of alkanes. A novel coding, which derived from the numbers of molecular subgraph, is used to characterize molecular structure of alkanes. It showed that there is a very good correlation between the coding and molar response values. The results of multiple linear regression are as follows. Molar response values (SM) on FID: SM = 89.579 - 56.579x1 - 40.579x2 - 18.191x 3 - 2.677x4 + 11.818x5 + 25.690x6 + 40.421x7 + 54.421x8 + 13.926x9 + 15.524x10 + 13.923x11 + 13.857x12 + 30.079x13 + 30.403x14 + 32.677x15 + 29.030x16, N = 50, r = 0.9983, S = 1.9342; Molar response values (SM) on TCD: SM = 139.799 - 62.251x2 - 49.214x3 - 31.244x4 - 15.992x5 + 0x6 + 13.201x7 + 27.201x8 + 11.152x9 + 9.225x10 + 8.427x11 + 10.041x12 + 22.452x13 + 14.977x14 + 22.318x16, N = 32, r = 0.9963, S = 1.7668. Where r, S and N are regression coefficient, residual standard deviation and data number, respectively. Having tested the modeling by using back-propagation neural network (BPNN) and leave-one-out method, we got the correlation coefficient of cross validation, 0.989.

[Immune responses in mice vaccinated with recombinant plasmid pcDNA3 containing ROP1 gene from Toxoplasma gondii].

AIM: To observe the immune responses in BALB/c mice vaccinated with the constructed recombinant plasmid, pcDNA3, containing ROP1 gene from Toxoplasma gondii. METHODS: The plasmid DNA obtained by alkaline lysis were injected into the left leg of each mouse at a dosage of 100 micrograms. A booster vaccine was given using the same dosage two weeks later. Control groups were injected with the pcDNA3 blank plasmid and normal saline, respectively. 30, 50 and 70 days after the booster injection, the proliferation activity of T lymphocytes and the NK cell activity were determined using MTT assay, the number of CD4+/CD8+ T cells with indirect immunofluorescence assay and the titer of IgG antibody by ELISA. RESULTS: The spleens of mice enlarged obviously after immunization with pcDNA3-ROP1. The activity of proliferation of spleen T lymphocytes was higher in the immunization group than in the control groups; The NK cell activity in the immunization group was higher than those of control ones. The number of CD4+ T cells showed no obvious increase but CD8+ T cells obviously increased. The titer of IgG antibody in the immunization group showed no significant increase until 90 days later. CONCLUSION: Vaccination of mice with the recombinant plasmid, pcDNA3-ROP1, could elicit cellular and humoral immune response in mice.

[Cellular immune response in mice vaccinated with uv-attenuated Toxoplasma gondii ZS1 strain trophozoites].

AIM: To explore the protective effect and cellular immune response of uv-attenuated ZS1 strain trophozoites of Toxoplasma gondii in mice. METHODS: The ZS1 strain trophozoites of T. gondii were irradiated by uv-light with 2537 A wave length for 60 minutes. Mice were divided into 4 groups. Group 1 was vaccinated alone, group 2 was challenged with normal ZS1 trophozoites on d45 after vaccination, group 3 was infected alone, and group 4 was normal control. The changes in splenic T cell proliferation, level of CD4+ and CD8+ T cell, and NK cell activity were compared. RESULTS: Group 1 mice survived normally, no trophozoite, pseudo-cyst or cyst was detected in the tissues on d49 after vaccination. Group 2 mice survived longer than those of group 3. The T lymphocyte proliferation in response to soluble antigen of T. gondii was significantly enhanced in group 2, and suppressed in group 3. The level of CD4+ T cell in group 2 was decreased, resulting in a reverse of CD4+/CD8+ ratio. The NK cell activities in groups 1, 2 and 3 were all significantly increased. CONCLUSION: The uv-attenuated vaccine of T. gondii ZS1 strain could induce certain protective immunity against challenge infection, in which CD8+ T cell and NK cell might play an important role.

[A new adaptive filtering method for micro-particle image processing].

Based on the local gray level histogram of macro-particle image which is more special than other images, a new adaptive noise filtering method for macro-particle image processing is developed and described in this paper. It is made up of median filter and special order statistic (OS) filters (alpha-mean filter and inverse alpha-mean filter). This adaptive filter not only has satisfactory performance in both of noise-removal and edge-preservation of macro-particle image, but also is easy and quick to implement. An example is presented in this paper.

[Spectral studies on the mechanism of rare earth poly (vinyl chloride) thermal stabilizers].

Rare earth Poly (Vinyl Chloride) (PVC) thermal stabilizers are one kind of effective and non-toxic stabilizers developed recently. By photoacoustic (PA) spectra and FT-IR analysis, their excellent performance in PVC stabilization progress can be studied conveniently. Good color remaining ability of rare earth stabilizers can be shown clearly by PA spectra and compared with others easily. From the IR spectrum and process data, the extra stabilization ability of rare earth stabilizers may be attributed to their conformation transforming effect.

NMDA antagonist displays anticonvulsant effect via NO synthesis inhibition in penicillin-treated rat hippocampal slices.

The present study investigated the role of nitric oxide (NO) in epileptogenesis and whether this role correlated with ionotropic glutamate receptor (IGR). Using a self-constructed NO-sensitive microelectrode (SNM), we observed the effect of nitric oxide synthase (NOS) inhibitors, NMDA and non-NMDA selective antagonists on penicillin(PEN)-treated hippocampal slices by simultaneously recording evoked field potentials and nitric oxide release from CA1 pyramidal neurons. 7-nitroindazole (7-NI),Nomega-nitro-L-arginine (L-NNA) and DL-2-amino-phospho-novaleric acid (APV), but not 6,7-dinitroquinoxaline-2,3 (1h,4h)-dione(DNQX), depressed NO release and partly reversed PEN's epileptogenetic effect, while APV + 7-NI + L-NNA did not display a further inhibitory effect. These findings suggest both NOS inhibitor and NMDA antagonist involve as anticonvulsant factors in epileptogenesis, providing direct evidence for NO release in response to NMDA receptor activation. The anticonvulsant effect of NMDA antagonist may ascribe to its action on NO release.

Monocyte induction of IL-10 and down-regulation of IL-12 by iC3b deposited in ultraviolet-exposed human skin.

CD11b+ monocytic/macrophagic cells (Mo/Mph), which infiltrate into skin after UV irradiation, play an important role in UV-induced immunosuppression. Because in mice, blockade of CD11b (iC3b receptor) on monocytes and depletion of its ligand, iC3b, reverses UV-induced immunosuppression, we asked whether iC3b is deposited in human skin after UV, and whether iC3b can modulate the cytokine profile of Mo/Mph. Immunofluorescence studies revealed that iC3b was newly deposited in UV-exposed skin and was localized in apposition to infiltrating CD11b+ Mo/Mph. In addition, in situ hybridization studies showed that TNF-alpha mRNA was also induced in a similar microanatomic localization. To model the effects of these complex signals on infiltrating Mo/Mph following UV exposure, we then tested the effects of immobilized iC3b and TNF-alpha on resting blood monocytes. Both IL-10 mRNA synthesis and protein secretion were significantly induced by binding of iC3b in vitro and were synergistically increased by the presence of TNF-alpha. The effect was abrogated by a blocking Ab to CD11b, indicating CD11b-iC3b interaction. In contrast, iC3b binding resulted in suppression of IL-12 p40 mRNA and significantly inhibited the production of IL-12 p70 protein. Our studies thus define a novel mechanism for induction of tissue Mo/Mph into an IL-10high/IL-12low state via iC3b in combination with TNF-alpha.

Particle nucleation in the tropical boundary layer and its coupling to marine sulfur sources

New particle formation in a tropical marine boundary layer setting was characterized during NASA's Pacific Exploratory Mission-Tropics A program. It represents the clearest demonstration to date of aerosol nucleation and growth being linked to the natural marine sulfur cycle. This conclusion was based on real-time observations of dimethylsulfide, sulfur dioxide, sulfuric acid (gas), hydroxide, ozone, temperature, relative humidity, aerosol size and number distribution, and total aerosol surface area. Classic binary nucleation theory predicts no nucleation under the observed marine boundary layer conditions.

Posttransplant therapy using high-dose human immunoglobulin (intravenous gammaglobulin) to control acute humoral rejection in renal and cardiac allograft recipients and potential mechanism of action.

BACKGROUND: Intravenous gammaglobulin (i.v.IG) contains anti-idiotypic antibodies that are potent inhibitors of HLA-specific alloantibodies in vitro and in vivo. In addition, highly HLA-allosensitized patients awaiting transplantation can have HLA alloantibody levels reduced dramatically by i.v.IG infusions, and subsequent transplantation can be accomplished successfully with a crossmatch-negative, histoincompatible organ. METHODS: In this study, we investigated the possible use of i.v.IG to reduce donor-specific anti-HLA alloantibodies arising after transplantation and its efficacy in treating antibody-mediated allograft rejection (AR) episodes. We present data on 10 patients with severe allograft rejection, four of whom developed AR episodes associated with high levels of donor-specific anti-HLA alloantibodies. RESULTS: Most patients showed rapid improvements in AR episodes, with resolution noted within 2-5 days after i.v.IG infusions in all patients. i.v.IG treatment also rapidly reduced donor-specific anti-HLA alloantibody levels after i.v.IG infusion. All AR episodes were reversed. Freedom from recurrent rejection episodes was seen in 9 of 10 patients, some with up to 5 years of follow-up. Results of protein G column fractionation studies from two patients suggest that the potential mechanism by which i.v.IG induces in vivo suppression is a sequence of events leading from initial inhibition due to passive transfer of IgG to eventual active induction of an IgM or IgG blocking antibody in the recipient. CONCLUSION: I.v.IG appears to be an effective therapy to control posttransplant AR episodes in heart and kidney transplant recipients, including patients who have had no success with conventional therapies. Vascular rejection episodes associated with development of donor-specific cytotoxic antibodies appears to be particularly responsive to i.v.IG therapy.

Free radical intermediates of phenytoin and related teratogens. Prostaglandin H synthase-catalyzed bioactivation, electron paramagnetic resonance spectrometry, and photochemical product analysis.

Phenytoin and related xenobiotics can be bioactivated by embryonic prostaglandin H synthase (PHS) to a teratogenic free radical intermediate. The mechanism of free radical formation was evaluated using photolytic oxidation with sodium persulfate and by EPR spectrometry. Characterization of the products by mass spectrometry suggested that phenytoin photolyzes to a nitrogen-centered radical that rapidly undergoes ring opening to form a carbon-centered radical. PHS-1 was incubated with teratogen (phenytoin, mephenytoin, trimethadione, phenobarbital, and major metabolites) or its vehicle and the free radical spin trap alpha-phenyl-N-t-butylnitrone, and incubations were analyzed by EPR spectrometry. There was no alpha-phenyl-N-t-butylnitrone radical adduct in control incubations. For phenytoin, a putative unstable nitrogen-centered radical adduct and a stable carbon-centered radical adduct were detected. Free radical spin adducts also were detected for all other teratogens and metabolites except carbamazepine. The PHS inhibitor eicosatetraynoic acid abolished the free radical EPR signal. Incubation of 2'-deoxyguanosine with phenytoin and PHS-1 resulted in a 5-fold increase in its oxidation to 8-hydroxy-2'-deoxyguanosine. This is the first direct chemical evidence for PHS-catalyzed bioactivation of phenytoin and related teratogens to a free radical intermediate that initiates DNA oxidation, which may constitute a common molecular mechanism of teratologic initiation.

Interaction of Sulfate-Reducing Bacteria with Molybdenum Dissolved from Sputter-Deposited Molybdenum Thin Films and Pure Molybdenum Powder.

When sputter-deposited Mo thin films were exposed to sulfate-reducing bacterium Desulfovibrio desulfuricans, dissolved Mo markedly delayed the culture growth and reduced the rate of sulfate reduction. The interaction led to an orange coloration of the culture liquid. X-ray photoelectron spectroscopy of dried culture droplets revealed that Mo dissolution products existed mostly in pentavalent state, and a smaller amount of molybdate and molybdenum disulfide. In contrast, Mo dissolution in uninoculated medium was negligible. Subsequently, different concentrations of molybdate, ranging from 0.1 to 20 mM, were added to the growth medium and it was found that a low concentration of molybdate (1 mM) was able to reduce the culture growth rate and sulfate reduction by forming Mo(V)-S complexes. In order to study the dependence of the degree of interaction upon microbial activity and growth-dependent metabolic products, 1.0 g/L Mo powder was added to (a) the growth medium, (b) a 3-day-old culture, and, (c) the supernatants of 2 h to 5-day-old cultures. Ultraviolet-visible spectroscopy indicated that the Mo(V)-S complexes consisted of a Mo-S compound analogous to a binuclear dioxobridged Mo(V)-cysteine complex (314 nm) and Mo(V)-containing molybdenyl thiocyanate (468 nm). Dissolution of Mo was induced by H2S, a product of the bacterial sulfate reduction, and was further increased probably by sulfur-containing amino groups and proteins. Copyright 1998 Academic Press.

Resistance to adenovirally induced hyperleptinemia in rats. Comparison of ventromedial hypothalamic lesions and mutated leptin receptors.

Leptin regulates appetite and body weight via hypothalamic targets, but it can act directly on cultured pancreatic islets to regulate their fat metabolism. To obtain in vivo evidence that leptin may act peripherally as well as centrally, we compared the effect of adenovirally induced hyperleptinemia on food intake, body weight, and islet fat content in ventromedial hypothalamic-lesioned (VMHL) rats, sham-lesioned (SL) controls, and Zucker Diabetic Fatty (ZDF) rats in which the leptin receptor is mutated. Infusion with recombinant adenovirus containing the rat leptin cDNA increased plasma leptin by approximately 20 ng/ml in VMHL and ZDF rats but had no effect on their food intake, body weight, or fat tissue weight. Caloric matching of hyperphagic VMHL rats to SL controls did not reduce their resistance to hyperleptinemia. Whereas prediabetic ZDF rats had a fourfold elevation in islet fat, in VMHL rats islet fat was normal and none of them became diabetic. Isolated islets from ZDF rats were completely resistant to the lipopenic action of leptin, while VMHL islets exhibited 50% of the normal response; caloric matching of VMHL rats to SL controls increased leptin responsiveness of their islets to 92% of controls. We conclude that leptin regulation of adipocyte fat requires an intact VMH but that islet fat content is regulated independently of the VMH.

Ultrasonic shear wave velocities of MgSiO3 perovskite at 8 GPa and 800 K and lower mantle composition

Ultrasonic interferometric measurements of the shear elastic properties of MgSiO3 perovskite were conducted on three polycrystalline specimens at conditions up to pressures of 8 gigapascals and temperatures of 800 kelvin. The acoustic measurements produced the pressure (P) and temperature (T) derivatives of the shear modulus (G), namely ( partial differentialG/ partial differentialP)T = 1.8 +/- 0.4 and ( partial differentialG/ partial differentialT)P = -2.9 +/- 0.3 x 10(-2) gigapascals per kelvin. Combining these derivatives with the derivatives that were measured for the bulk modulus and thermal expansion of MgSiO3 perovskite provided data that suggest lower mantle compositions between pyrolite and C1 carbonaceous chondrite and a lower mantle potential temperature of 1500 +/- 200 kelvin.

Lithium stimulates gene expression through the AP-1 transcription factor pathway.

Lithium, a monovalent cation, is the mainstay in the treatment of manic-depressive (MDI) illness, but despite extensive research, its mechanism of action remains to be elucidated. Since lithium requires chronic administration for therapeutic efficacy, and because its beneficial effects last well beyond its discontinuation, it has been postulated that lithium may exert major effects at the genomic level. In the present study we found that lithium, at therapeutically relevant concentrations, increases AP-1 DNA binding activity in human SH-SY5Y cells and rat C6 glioma cells. Additionally, in both SY5Y and C6 cells transiently transfected with a reporter gene vector driven by an SV40 promoter, lithium increased the activity of the reporter gene in a time- and concentration-dependent manner. Furthermore, mutations in the AP-1 sites of the reporter gene promoter significantly attenuated lithium's effects. These data indicate that lithium stimulates gene expression through the AP-1 transcription factor pathway, effects which may play a role in its long-term mood-stabilizing effects.

Structural and enzymatic studies of a new analogue of coenzyme B12 with an alpha-adenosyl upper axial ligand.

A new analogue of coenzyme B12 (5'-deoxyadenosylcobalamin, AdoCbl), in which the configuration of the N-glycosidic bond in the Ado ligand is inverted [(alpha-ribo)AdoCbl], has been synthesized and its crystal structure determined by X-ray diffraction [MoKalpha, lambda = 0.71073 A, monoclinic P212121, a = 16.132(12) A, b = 21. 684(15) A, c = 27.30(3) A, 9611 independent reflections, R1 = 0. 0708]. As suggested by molecular mechanics modeling before the structure was known, the Ado ligand lies over the southern quadrant of the molecule, as is the case for AdoCbl. The most striking feature of the structure is disorder in the orientation of the adenine (Ade) moiety relative to the ribose of the Ado ligand. This was resolved with a two-state model in which in the major (0.57 occupancy) conformer the A16(O)-A11-A9(N)-A8 dihedral angle is 1.9 degrees and the Ade is virtually perpendicular to the corrin ring; in the minor conformer, the Ade is tilted down, and this dihedral is -48.7 degrees. The Co-C and axial Co-N bond lengths and the Co-C-C bond angle are quite similar to those in AdoCbl. The corrin ring is considerably flatter than that of AdoCbl, with a fold angle of 11.7 degrees. The molecule was successfully modeled by molecular mechanics (MM), and rotation of the Ado ligand relative to the corrin gave rise to four locally minimum structures with the Ado in the southern, eastern, northern, or western quadrant, with the southern conformation as the global minimum, as is the case with AdoCbl itself. Nuclear Overhauser effects (nOe's) observed by two-dimensional (2D) NMR were incorporated as restraints in molecular dynamics (MD) and simulated annealing (SA) calculations. A MD simulation at 300 K showed that only the southern conformation is populated with the Ado ligand confined to an arc from over C15 to over C12, while the Ade ring oscillates from perpendicular to parallel to the corrin ring. Twenty-seven structures were collected by MD-SA. Most of these annealed into the southern conformation, but examples of the other conformations were also found. The new analogue is a partially active coenzyme for the ribonucleotide reductase from Lactobacillus leichmanii with maximal activity that is 9.7% of that of AdoCbl itself, and a very high Km value (245 microM compared to 0.54 microM for AdoCbl). In addition, the rate constant for enzyme-induced carbon-cobalt bond cleavage of (alpha-ribo)AdoCbl is 160-fold smaller than that for AdoCbl, and only 1/3 as much cob(II)alamin is produced at the active site.
