Luteolytic prostaglandins. Synthesis and biological activity.

Analogues of PGF2 alpha with enhanced luteolytic activity were synthesized using the Corey synthesis. The luteolytic activity of the new prostaglandins was tested in the hamster. In addition the smooth muscle activity of the new compounds was compared with that of PGA2 on the longitudinal strip of rat stomach fundus. Structure-activity relationships in the new series of 17,18,19,20-tetranor-16-thienyl-oxy-PGF2 alpha are discussed.

Vasodilation and inhibition of platelet aggregation by prostacyclins with modified omega-side chain.

Prostacyclin analogs with modified omega-side chain were synthetized in search of therapeutically useful agents. To characterize the vasodilator and platelet-antiaggregating properties, prostacyclin analogs were tested on systemic blood pressure in anesthetized rats, relaxation of bovine coronary artery and inhibition of arachidonic acid induced human platelet aggregation. The sodium salt of prostacyclin induced a dose dependent decrease of blood pressure with an ED25 of 0.23 microgram/kg i.v., a marked relaxation of bovine coronary artery with an IC50 of 5.9 ng/ml and a strong inhibition of platelet aggregation with an ED50 of 3x10(-9) M. Similar results were obtained with prostacyclin-methylester. Replacement of the n-pentyl moiety attached to C-15 of prostacyclin by cyclohexyl, 2-(2-furyl)ethyl, 2-(3-thienyl)ethyl and especially by 3-thienyl-oxymethyl yielded analogs with comparable prostacyclin properties, while substitution by 1,1-dimethyloxaalkyl residues was followed by a marked loss of activity. The order of potency among the analogs of the sodium salt and methylester of prostacyclin with strong vasodepressor and antiaggregatory properties was identical in all three models used. The three test systems used for evaluation have demonstrated that suitable modifications of the omega-side chain of prostacyclin result in potent vasodilator and platelet-antiaggregating agents.

[A modified method for the quantitative determination of urinary oxalic acid].

The present paper describes a modified method of the quantitative determination of oxalic acid in the urine which is based on the reduction of oxalic acid to glycolic acid and on the transformation with chromotropic acid to a colorimetrically well measurable complex of colours. The method ascertains a good reproducibility and is simply to be performed in every clinico-chemical laboratory.

Left ventricular apical thin point.

The wall thickness of the apex of the left ventricle has been measured at its thinnest point in 60 adult hearts at necropsy. This measured 2 mm or less in 97% of them and 1 mm or less in 67%.

[Quantitative bacteriology of sputum and bronchial secretions in patients with chronic bronchitis (author's transl)].

The in vitro and in vivo reproducibility of the quantitative bacteriological analysis of sputum and bronchial secretions has been studied in patients with chronic bronchitis. Sputum produced during bronchial toilet showed a significant higher bacterial numeration than that produced by physiotherapy but the bacterial species identified were identical. No significant variations were observed between the qualitative and quantitative bacteriologic flora of bronchial secretions collected by bronchoscopy from different segments of the airways. Comparison of microbial counts on sputum specimens collected at different periods of the day and at weekly intervals over several weeks revealed that the sputum of clinically stable chronic bronchitic patients contain a relatively constant and potentially pathogenic bacterial flora. The emergence of a superinfection was emphasized by the parallel increase of the leukocytic and bacterial numeration of sputum.

Inhibition of viral multiplication by homologous methylated ribonucleic acids. V. Inhibition of myxoviruses in mouse.

Mice respond to intravenous injection of homologous methylated RNA by the production of a circulating interferon-like substance. The treatment with modified RNA induces a significant protection against viral infection. This effect is optimum when the treatment occurs a few hours before viral infection. The protective effect is a peculiar property of homologous methylated RNA as heterologous RNAs do not induce any resistance to the infection with influenza virus.

Impairment of enzyme induction by glucocorticoids in Zajdela hepatoma cells.

Whereas glucocorticoids induce TAT, TRP, GPT in liver and only TAT in HTC cells, no hormonal effect on the synthesis of these enzymes was found in Zajdela hepatoma cells grown in vivo as an ascitic tumor, or in vitro as layer cultures. Although these cells remain uninducible, the hormone penetrates normally, but a strong decrease of the specific binding of cytosol and nuclear proteins with the hormone was observed. The impairment at the level of the hormone receptors could account for the non-inducibility of enzyme synthesis in ZHC cells.

[Rheological and biochemical properties of sputum. Role of infection (author's tansl)].

Rheological and biochemical changes in sputum following repeated bronchial infections have been studied in six chronic bronchitics. Secretory IgA (IgAs) in the sputum decrease and serumalbumin increases as the number of acute bronchial infection episodes increases. Rheological properties of sputum follow a comparable course to that of secretory IgA. The decrease of the ratio-IgA-serum albumin and the loss of the viscoelastic properties of sputum reflect the non functional state of the bronchial mucosa. These changes may be considered as possible important factors altering mucus clearance and therfore favoring chronic bronchial obstruction.
