[Cloning and bioinformatics of human REV3 gene promoter region and its response to carcinogen N-methyl-N'-nitro-N-nitrosoguanidine].

OBJECTIVE: To understand the up regulatory mechanism of human REV3 gene induced by the chemical carcinogen N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). METHODS: Bioinformatic analysis of human REV3 gene promoter region was based on BLAST alignment, promoter prediction software and recognition of transcriptional factor binding sites. Cloning of human REV3 gene promoter region was performed by nested PCR. Response of human REV3 gene promoter to the chemical carcinogen MNNG was measured by transient transfection assay based on the dual luciferase reporter assay system. RESULT: Bioinformatic analysis showed that human REV3 gene promoter region was located on chromosome 6 PAC clone RP3-415N12, and that the hypothetical promoter region contained promoter sequences, rich CpG islands, and putative recognition sites for several transcriptional factors, including AP-1/c-Jun/c-Fos, AP-2, STAT, CREBP, and NF-kappaB. Reconstructed reporter plasmid pGL3- 2582 was established by inserting 2582 nucleotides from the promoter region into the luciferase reporter vector pGL3-Basic. Transient transfection assay showed the hypothetical REV3 promoter region had promoter function, and it responded to MNNG treatment (P<0.01). CONCLUSION: Human mutator REV3 gene promoter region has been successfully cloned. The response of REV3 promoter region to MNNG suggests that REV3 gene can be regulated at transcriptional level under conditions of genotoxic stress.

[Effect of emodin and apigenin on invasion of human ovarian carcinoma HO-8910PM cells in vitro].

BACKGROUND & OBJECTIVE: Emodin inhibited the activity of TPK and CK2 and the degradation of I-kappaB. Apigenin inhibited the activity of MAPK and PI(3)K. In this study the authors observed the effect of emodin and apigenin on the invasion of HO-8910PM cells in vitro. METHODS: Trypan blue dye exclusion assay was used to examine the cytotoxity of emodin and apigenin. Reconstituted basement membrane invasion assay was utilized to evaluate the invasive activity. Type IV collagenase production was analyzed by PAGE substrate zymography. RESULTS: Emodin had weaker cytotoxity on HO-8910PM cells than apigenin, their IC(50) after treatment with the chemicals for 48 hours were (35.30+/-3.50) micromol/L, and (28.92+/-2.60)micromol/L, while emodin significantly inhibited membrane invasion and adhesion and migration of HO-8910PM cells. Their inhibition rates after treated with the chemical of 40 micromol/L were (45.31+/-3.10)%,(25.42+/-1.70)%, and (41.59+/-1.90)%. Emodin inhibited the production but not activity of MMP-9. Apigenin inhibited migration and adhesion of HO-8910PM cells, their inhibition rates after treated with the chemical of 40 micromol/L were (29.04+/-1.70)% and (30.80+/-3.00)%, while weakly inhibited membrane invasion (the inhibition rate only was 12.1%) and inhibited neither production nor activity of MMP-9. CONCLUSION: Both emodin and apigenin had cytotoxicity on HO-8910PM cells. Emodin was a potential agent inhibiting tumor invasion and metastasis.

Microarray profile of differentially expressed genes in a monkey model of allergic asthma.

BACKGROUND: Inhalation of Ascaris suum antigen by allergic monkeys causes an immediate bronchoconstriction and delayed allergic reaction, including a pulmonary inflammatory infiltrate. To identify genes involved in this process, the gene-expression pattern of allergic monkey lungs was profiled by microarrays. Monkeys were challenged by inhalation of A. suum antigen or given interleukin-4 (IL-4) treatment; lung tissue was collected at 4, 18 or 24 h after antigen challenge or 24 h after IL-4. Each challenged monkey lung was compared to a pool of normal, unchallenged monkey lungs. RESULTS: Of the approximately 40,000 cDNAs represented on the microarray, expression levels of 169 changed by more than 2.5-fold in at least one of the pairwise probe comparisons; these cDNAs encoded 149 genes, of which two thirds are known genes. The largest number of regulated genes was observed 4 h after challenge. Confirmation of differential expression in the original tissue was obtained for 95% of a set of these genes using real-time PCR. Cluster analysis revealed at least five groups of genes with unique expression patterns. One cluster contained genes for several chemokine mediators including eotaxin, PARC, MCP-1 and MCP-3. Genes involved in tissue remodeling and antioxidant responses were also identified as regulated by antigen and IL-4 or by antigen only. CONCLUSION: This study provides a large-scale profile of gene expression in the primate lung following allergen or IL-4 challenge. It shows that microarrays, with real-time PCR, are a powerful tool for identifying and validating differentially expressed genes in a disease model.

[Comparison of Doppler examination and non-stress test in the prediction of intrauterine fetal hypoxia].

OBJECTIVE: To assess the predictive value of intrauterine fetal hypoxia by Doppler examination and non-stress test (NST). METHODS: The normal value of the peak-systolic and diastolic ratio (S/D), pulse index (PI), resistance index (RI), fast volume ratio (FVR) of different gestational age was created according to 4 326 normal pregnancy. S/D, PI, RI, FVR and NST was performed for 2 873 normal pregnancy at the last trimester. Fetal outcomes were recorded within 7 days of birth. RESULTS: (1) S/D, PI, and RI decreased with the gestational age advancing, but FVR didn't. When S/D, PI, RI, FVR or NST appeared abnormal, the ratio of intrauterine fetal hypoxia was 62.0%, 60.7%, 63.3%, 27.0% and 75.7% respectively, as these normal, the ratio of intrauterine fetal hypoxia was 12.8%, 13.9%, 13.0%, 15.7%, 7.5% respectively. There was significant difference (P < 0.01). (2) Logistic regression showed that the predictive value of RI and NST to intrauterine fetal hypoxia was significant (P < 0.05). CONCLUSIONS: The predictive value of NST and RI to intrauterine fetal hypoxia was stronger than other parameters. Multi-parameter may predict more accurately than any single index for intrauterine fetal hypoxia.

[The influence of high frequency partial liquid ventilation on the cardiopulmonary function in dogs with inhalation injury].

OBJECTIVE: To investigate the influence of high frequency partial liquid ventilation (HFJV) on the cardiopulmonary function in dogs with inhalation injury. METHODS: Sixteen mongrel dogs inflicted by hot steam inhalation were subjected to severe inhalation injury and were randomly divided into control (C) and treatment (T) groups. The dogs in both groups were all given HFJV. In addition, the dogs in T group were simultaneously supplied with perfluorocarbon liquid (3 ml/kg) into the lungs slowly via tracheal intubation for liquid ventilation. The blood gas analysis, pulmonary compliance, airway resistance and hemodynamic parameters were determined at 30, 60 and 90 minutes after ventilation. RESULTS: The PaO(2) in T group increased progressively, which was significantly higher than the post-injury value at all time points (P < 0.05). While the PaO(2) in C group exhibited no difference to the post-injury value at all time points. The PaCO(2) in T group increased obviously and was higher than the post-injury value at 60 and 90 post-ventilation minutes (P < 0.05). Furthermore, the PaO(2) in all the time points in T group was a little higher than that in C group (P > 0.05) and PaCO(2) in T group was much higher than that in C group at 90 min after ventilation (P < 0.05). But there was no difference between the two groups in terms of dynamic/static pulmonary compliance and airway resistance as well as the hemodynamics. CONCLUSION: Compared with simple HFJV, high frequency partial liquid ventilation seemed to be beneficial to the oxygenation after inhalation injury and to be no influence on the hemodynamics.

Upregulation of bax Gene Expression Promotes Paclitaxel-induced Apoptosis in Esophageal Carcinoma Cells.

An inducible mammalian expression vector of bax gene was constructed and the control ability of metallothionein II promoter in esophageal carcinoma cell line was systematically identified with luciferase report gene. After the transfection of it into human esophageal carcinoma cell line Eca109, Bax protein expression was analyzed by immunolcytochemical method. Paclitaxel-induced apoptosis was determined by TUNEL assay, DNA ladder assay and flow cytometry. Results showed that 140 &mgr;mol/L ZnSO(4) for 12 h is optimal for induction of bax gene expression. Under these conditions, a clonal transfectant X1097(#), expressing bax gene effectively, was obtained. It was found that, X1097(#) had higher apoptotic rate and was more sensible than Eca109. These results implied that Bax protein may play an important role in paclitaxel-induced apoptosis. Therefore, bax protein may be promising as an helping drug to improve therapeutic effects of paclitaxel.
