Fabrication of protein-conjugated silver sulfide nanorods in the bovine serum albumin solution.

Highly ordered silver sulfide nanorods conjugated with the Bovine Serum Albumin (BSA) protein have been successfully achieved at ambient temperature. Such a process is very simple and controllable, directly using silver nitrate and thioacetamide (TAA) as the reactants in the aqueous solution of BSA. The products have been characterized by XRD, HRTEM-SAED, SEM-EDS, TG-DTA, FT-IR, and CD spectroscopy. The results of the research show that the as-prepared Ag2S nanorods are monodispersed with sizes about 40 nm in diameter and 220 nm in length, and exhibit a high degree of crystallinity and good photoluminescence. Furthermore, an interesting mechanism is discussed for the formation of the Ag2S nanorods.

[The accuracy of diagnosing cervical intraepithelial neoplasia with colposcopically directed biopsy].

OBJECTIVE: To evaluate the accuracy of colposcopically directed biopsy in the diagnosis of cervical intraepithelial neoplasia (CIN). METHODS: The clinical data of 153 patients with CIN diagnosed by colposcopically directed biopsy and treated with cervical loop electrosurgical procedure (LEEP) shortly after were retrospectively studied. The consistency of pathological examination between colposcopically directed biopsy and LEEP was evaluated. The factors of missed diagnosis of cervical invasive carcinoma were analyzed. RESULTS: The diagnosis of 106 out of the 153 patients by colposcopically directed biopsy was consistent with that by LEEP as confirmed by pathologic diagnosis with a consistence rate of 69.3%, however, the diagnoses of 47 of the 153 patients by colposcopically directed biopsy were not consistent with those by LEEP, with an inconsistent rate of 30.7%, including 22 cases of overdiagnosis (14.4%, 22/153) and 25 cases of underdiagnosis (16.3%, 25/153). Eighteen patients were confirmed as with cervical invasive carcinoma by LEEP/hysterectomy at last with a missed diagnosis rate of 11.8%. The missed diagnosis rate of invasive carcinoma by colposcopically directed biopsy was significantly higher in the patients with cytologic diagnosis of with > or = HSIL than in the patients cytologically diagnosed as with < HSIL (chi(2) = 8.208, P < 0.05). CONCLUSION: The accuracy of diagnosing CIN with colposcopically directed biopsy is still unsatisfying. Attention should be paid on the patients with cytological diagnosis of > or = HSIL so as to avoid missed diagnosis of cervical carcinoma.

Altered expression of cellular membrane molecules of HLA-DR, HLA-G and CD99 in cervical intraepithelial neoplasias and invasive squamous cell carcinoma.

OBJECTIVE: The aim of this study was to investigate the role of HLA-DR, HLA-G and CD99 during cervical carcinogenesis and to examine the prognostic significance of these protein expressions in invasive squamous cell carcinoma (SCC). METHODS: Using specific antibodies for HLA-DR, HLA-G and CD99, we examined protein expressions in 19 normal cervix, 15 mild dysplasia (CIN I), 22 moderate dysplasia (CIN II), 23 severe dysplasia (CIN III), and 34 invasive squamous cell carcinoma by immunohistochemistry. And we detected the expression of Ki67 in the same specimens. RESULTS: None of normal cervix and CINs except three cases of CIN III expressed HLA-DR. HLA-DR expression increased progressively with the grade of the tumor, and significant differences could be observed between grade 1 and grade 2 (P<0.01) and between grade 1 and grade 3 (P<0.05). In all normal epithelial control samples, HLA-G expression was seen in ectocervical squamous and endocervical columnar epithelium and the staining was strong and uniform. Only a small proportion of CINs and SCCs showed reduced expression of HLA-G. Compared with the results in the control samples, CINs and SCCs showed significantly reduced expression of HLA-G (P<0.001). SCCs showed significantly increased expression of CD99 when compared with normal cervix and CINs (P<0.05). Ki67 was expressed in all specimens. Significant differences were observed between CINs and normal cervix (P<0.001) and SCCs and controls (P<0.001), but no significant differences could be observed between SCCs and CINs. None of the expressions of these proteins was associated with any of clinicopathological parameters. CONCLUSIONS: These results indicate that increased expression of HLA-DR and CD99 may be related to the evolution of cervical cancer. All protein expressions were not associated with clinicopathological parameters.

Presence of HBsAg, HBcAg, and HBVDNA in ovary and ovum of the patients with chronic hepatitis B virus infection.

OBJECTIVE: The purpose of this study was to investigate the presence and distribution of Hepatitis B virus antigen (HBVAg) and Hepatitis B virus deoxyribonucleic acid (HBVDNA) in ovary with chronic hepatitis B virus. STUDY DESIGN: The immunohistochemistry method and in situ hybridization method were used to detect the hepatitis B surface antigen (HBsAg), anti-hepatitis B core antibody (HBcAg), and HBVDNA in ovary of patients with chronic hepatitis B infection. RESULTS: HBsAg presented in the ovum-granular cell and interstitial cell of ovary. HBsAg located in the cytomembrane and cytoplasm. HBcAg presented in the ovum-granular cell-interstitial cell and endothelium cell of interstitial blood vessel of ovary, and the HBcAg may present in the ovum at different stage. HBcAg was situated at the cytomembrane, cytoplasm, and nucleus. HBVDNA was detected in the ovum at different stage as HBcAg and in granular cell, interstitial cell of ovary. In the same ovary tissue, HBVDNA was positive in some ova, but it was weakly positive or negative in other ova. HBVDNA distributed mainly in nucleus, but also can be detected in the cytoplasm. CONCLUSION: HBV could infect the ovum at different stage and replicate in it. This may be an important mechanism of HBV vertical transmission.

[Expression of human annexin V in different fetal tissues].

OBJECTIVE: To investigate the expression of human annexin-V (HA-V) in relation to HBV infection in different fetal tissues. METHODS: Immunohistochemistry was employed to detect the expression and distribution of HA-V in the liver, kidney, ovary, heart, fallopian tube, spleen, and thymus gland of human fetus. RESULTS: HA-V expression was detected in different tissues including the ovary, liver, intrahepatic bile duct, heart, kidney, lymphocytic cells in the thymus gland, epithelial cells of the fallopian, and cortical and medullary cells of the spleen. HA-V was distributed mainly in the cytoplasm of the cells. The liver tissues exhibited greater gray scale for HA-V expression than in the other tissues (P<0.05) and no significant difference was observed in the other tissues than the liver (P>0.05) in image analysis with Photoshop 7.0. CONCLUSION: HA-V is an inherent protein in fetal tissues with possible relation to HBV infection of different tissues as a HBV receptor. Greater amount of HA-V in the liver may account for the vulnerability of the liver to HBV infection.

[Evaluation of neoadjuvant chemotherapy for locally advanced cervical cancinoma].

OBJECTIVE: To investigate the effect of neoadjuvant chemotherapy on locally advanced cervical cancer, the factors affecting outcome of chemotherapy and the long-term survival rate in the patients. METHODS: A total of 64 patients with stageIb2-IIb of locally advanced cervical cancers treated with neoadjuvant chemotherapy between June 1999 and October 2004 in Women's Hospital, School of Medicine, Zhejiang University were retrospectively analysed. The effect of chemotherapy, as well as survival rate was evaluated. The related factors of effect, including age, histology, clinical stage, grade of differentiation, initial tumor size, chemotherapy regimens, number of courses were analysed. RESULTS: Overall effective rate of neoadjuvant chemotherapy was 80%. The effective rate was associated with histology. The patients with squamous cell cancer had significantly higher 5-year survival rate than those with adenocarcinoma (82% vs 6/9, P < 0.05). The rates of positive pelvic lymph node metastasis and parametrial invasion were significantly higher in complete or partial response patients than those in stable patients (P < 0.05). The overall 5-year survival rate was 89%. The 5-year survival rate in complete or partial response group was 100%, in stable disease group was 46% (P < 0.05). The 3, 5-year disease-free survival rate in complete or partial response group was 95% and 83%, respectively, while in stable disease group was 33% and 0, respectively. The disease-free survival rates in complete or partial response group were higher than those of stable disease group (P < 0.05). CONCLUSIONS: The clinical response to neoadjuvant chemotherapy is related to histology. The patients who have good response to chemotherapy should choose surgery. Chemotherapy can decrease the high risk factors of pathology in complete or partial response patients. Neoadjuvant chemotherapy for locally advanced cervical cancer patients seems to improve the long-term survival rate.

Effects of extract of dilong (pheretima) on the scalded skin in rats.

In order to investigate the effects of extract of Dilong (Pheretima) on the skin viability and lipid peroxidation after deep degree II scalds, the extract of Dilong (Pheretima) at two concentrations (5%, 10%) were respectively applied to the surface of wound. The oxygen consumption, the succinate dehydrogenase (SDH) activity, the Schiff's base content, and the healing time of wound surface were determined in Wistar rats at the 8th, 24th, 48th and 72th hour after scalds, with SD-Ag used as controls. The results indicated that the oxygen consumption and SDH activity decreased, and the schiff's base content increased after the scalds. After the application of the extract of Dilong, the oxygen consumption and SDH activity increased, and the schiff's base content decreased significantly as compared with the control group (P < 0.05 or P < 0.01), and the mean healing time of the wound surface was five days in advance in the two treatment groups as compared with the control group. It is suggested that the scalds are related with lipid peroxdation to a certain extent, and the extract of Dilong has actions of attenuating lipid peroxidation, and promoting healing of the wound.

[Deletion of OPCML gene and promoter methylation in ovarian epithelial carcinoma].

OBJECTIVE: To investigate the expression of OPCML gene in ovarian epithelial carcinoma and determine the relationship between mRNA expression and methylation of their promoters. METHOD: Twenty normal ovarian tissues and 89 ovarian epithelial tumor specimens (72 malignant, 17 benign), as well as 3 ovarian carcinoma cell lines (SKOV-3, CAOV3, and 3AO), were collected for detection of OPCML gene expression by reverse transcription-polymerase chain reaction and for detection of promoter methylation by restriction enzyme cut analysis from 7. 1999 to 7. 2003. RESULTS: Among ovarian epithelial carcinoma 19.4% expressed OPCML mRNA, while 85% of normal ovarian tissue and 76.5% of benign ovarian tumor. The ratio of expression of OPCML mRNA in ovarian epithelial carcinoma was significantly lower than those of normal (chi2 = 30.108, P = 0.0000) and benign tumors (chi2 = 21.162, P = 0.000). No OPCML mRNA expression was found in SKOV-3 and CAOV3, but was found in 3AO. Methylations were detected in 44.4% of cancer cells promoter, while 0% in normal ovarian tissue and benign ovarian tumors. The ratio of methylation of ovarian epithelial carcinoma was significantly higher than those of normal (chi2 = 13.630, P = 0.0000) and benign tumors (chi2 = 11.797, P = 0.000). Methylation was found in SKOV-3 and CAOV3, but not in 3AO. The relationship between gene expression and promoter methylation was correlated (r = 11.589, P = 0.002), especially at Hap I1 site (r = 11.640, P = 0.004). Methylation was also found in SKOV-3 and CAOV3 cell lines, but not in 3AO cell line. CONCLUSION: Deletion of OPCML gene exists in ovarian epithelial carcinoma cell. The gene promoter methylations, especially Hap II motif, may be one of pathways that contribute the inhibition of OPCML expression.

Fas-mediated pathway and apoptosis in normal cervix, cervical intraepithelial neoplasia and cervical squamous cancer.

The aim of this study was to examine the expression patterns of apoptosis-related antigens, such as Fas, FasL, and cFLIP, in cervical squamous cells, and investigate the role of Fas-mediated apoptosis in the pathogenesis of cervical neoplasia. Using specific antibodies for Fas, FasL, and cFLIP, we examined protein expression in 19 specimens of normal cervix, 15 mild dysplasia (CIN I), 22 moderate dysplasia (CIN II), 23 severe dysplasia (CIN III), and 34 invasive squamous cell carcinoma (SCC) by immunohistochemistry. We detected the apoptotic indices by TUNEL in the same specimens. Fas expression levels were quite similar in CIN I, CIN II and the normal cervix. Though Fas expression tended to increase in grade 2 cancer compared to grade 1 cancer and CIN III, and a slight decline was present in grade 3 compared with grade 2 cancer, these differences did not reach statistical significance. Almost all CINs did not express FasL, while FasL expression increased with the grade of the tumor. Statistically significant differences could be observed between grade 1 and grade 2 (p<0.01) and between grade 2 plus grade 3 and grade 1 (p<0.001). All cases of normal cervix and CIN, except two cases of CIN III, did not express cFLIP. cFLIP expression tended to increase with the grade of the tumor. Apoptosis was determined in all samples by TUNEL. There was a decreasing tendency of apoptotic cells from normal cervix to cancers. A negative correlation between cFLIP and apoptosis (r=-0.499 and p=0.000) was observed. Deregulated Fas/FasL system and constitutive expression of cFLIP in SCC may be an important mechanism by which SCCs escape apoptosis during the malignant transformation and progression of these tumors.

Expression of HBsAg and HBcAg in the ovaries and ova of patients with chronic hepatitis B.

AIM: To investigate the expression and distribution of HBV in the ovaries and ova. METHODS: The immunohistochemistry method was used to detect the HBsAg and HBcAg in the ovaries of patients with chronic hepatitis B. RESULTS: Expression of HBsAg in the ova, granular and interstitial cells of the ovaries was located in the cytomembrane and cytoplasm. Expression of HBcAg in the ova, granular, interstitial and endothelial cells of interstitial blood vessels of the ovaries was found in the cytomembrane, cytoplasm, and nuclei. CONCLUSION: HBV can infect the ova at different stages of development and replicate in it.

Expression of ST13 in colorectal cancer and adjacent normal tissues.

AIM: To investigate the in situ expression of suppression of tumorigenecity 13 (ST13) mRNA in both colorectal cancer and adjacent normal tissues. METHODS: Colorectal cancer cell lines SW1116, SW620 and CoLo205 were enrolled to confirm the feasibility of the in situ hybridization procedure. Seven colorectal cancer and adjacent normal tissues were included for RNA-RNA in situ hybridization. RESULTS: The expression of ST13 in the seven normal colon tissues was positive and the positive signals appeared in mucosal cells. Only three of the seven colorectal cancer tissues had positive hybridization signals that appeared in adenocarcinoma cells. CONCLUSION: The expression of ST13 decreases in colorectal cancer tissue compared with that in adjacent normal tissue. ST13 is mostly expressed in colorectal epithelia and adenocarcinoma cells.

Ovarian carcinoma cells effectively inhibit differentiation and maturation of dendritic cells derived from hematopoietic progenitor cells in vitro.

Loco-regional dissemination of ovarian carcinoma is associated with immunosuppression of the peritoneal cavity. One marked characteristic of the peritoneal immunity in this disease is the defective function of dendritic cells (DCs). In this study, the affect of ovarian carcinoma cells on DCs derived from hematopoetic progenitor cells was observed. The study demonstrated that the expansion, phenotype, and function of DCs generated from CD34+ precursors were significantly altered by the supernatant secreted by ovarian carcinoma cells, and this effect could be partly explained by tumoral overproduction of Vascular Endothelial Growth Factor (VEGF). The results indicated that a role of ovarian carcinoma cells in the differentiation and function of DCs could be associated with the immunosuppression and development of ovarian carcinoma.

Interactions of sympathetic nerves with capsaicin-sensitive sensory nerves: neurogenic mechanisms for phenol-induced hypertension in the rat.

BACKGROUND: Previous investigations have shown that norepinephrine is capable of inhibiting neurotransmission in capsaicin-sensitive sensory nerves via a prejunctional mechanism. The alteration in the activity of sympathetic or capsaicin-sensitive sensory nerves in the development of phenol-induced hypertension was observed separately in rats. METHODS: In the present study, we examined interactions of adrenergic nerves with capsaicin-sensitive sensory nerves in phenol-induced hypertensive rats. Blood pressure, the synthesis and release of calcitonin gene-related peptide (CGRP) and the content of nerve growth factor in (NGF) arteries were determined. RESULTS: Intrarenal injection of phenol caused a permanent elevation of blood pressure concomitantly with a decrease in the concentration of CGRP in plasma, the content of CGRP in dorsal root ganglia and the density of CGRP-containing nerves in the mesenteric artery, and vascular NGF content. Chronic treatment with prazosin (an alpha1-adrenoreceptor antagonist, 3 mg/kg per day) failed to alter the synthesis and release of CGRP and vascular NGF content, even though it completely normalized blood pressure. However, treatment with yohimbine (an alpha2-adrenoreceptor antagonist, 5 mg/kg per day) significantly increased CGRP level and vascular NGF content. Combined administration of prazosin and yohimbine not only significantly elevated the synthesis and release of CGRP and arterial NGF content, but also completely normalized blood pressure. CONCLUSION: These results indicate that the decreased production and release of CGRP and reduced vascular NGF content are attributed to the activation of alpha2-adrenoreceptors in phenol-induced hypertensive rats.

[Effect of high salt diet on expression of osteopontin mRNA in Sprague-Dawley rat kidney].

OBJECTIVE: To investigate the effect of high-salt diet on the expression of osteopontin (OPN) mRNA and its protein in Sprague-Dawley rat kidney. METHODS: Forty-eight male SD rats aged 10 weeks receiving normal salt diet were enrolled in study, and were divided into two groups and fed with high salt diet (4%NaCl) or normal salt diet (0.6%NaCl) for 12 weeks respectively, with 24 rats in each group. Tail systolic blood pressure and bodyweight were measured in all rats every week. At the end of 4th, 8th and 12th week, 6 rats in each group were sacrificed for detection of the expression of OPN mRNA and its protein in the kidney with quantitatine real-time (QRT)-PCR and immunohistochemistry, respectively. RESULTS: No significant difference was found in blood pressure, bodyweight and kidney weight/bodyweight between high-salt diet group and normal salt diet group (P>0.05). The expression of OPN mRNA (0.27+/-0.16 vs 0.15+/-0.13, P<0.05) and its protein (0.78+/-0.15 vs 0.61+/-0.11, P<0.01) in SD rat kidney with high-salt diet were up-regulated at the end of 12th week compared with the rats with normal salt diet. CONCLUSIONS: High-salt diet increase the expression of OPN mRNA and its protein in SD rat kidney. Renal injury induced by high salt intake might be independent of blood pressure change.

[Effects of high salt diet on blood pressure, renal morphology and osteopontin expression in Sprague-Dawley rat with Ang II-induced renal injury].

OBJECTIVE: To investigate the effect of high-salt diet on blood pressure, renal morphology and osteopontin (OPN) expression in Sprague-Dawley rat with Ang II-induced renal injury. METHODS: Thirty-six male SD rats aged 12 weeks receiving normal salt diet were rolled in the study, and received Ang II (100 ng.kg-1.min-1) for two weeks using subcutaneous minipump. Rats were then fed high sodium (n=18, 4% NaCl) or normal sodium (n=18, 0.6% NaCl) diet from third week respectively and lasted for 12 weeks. Eighteen rats in the control were sham-operated and fed normal sodium diet. Tail systolic blood pressure (SBP) was determined every two weeks. Expressions of TGF-beta1 OPN mRNA and its protein in SD rat kidney were measured with quantitative RT-PCR and immunohistochemistry respectively at the end of Ang II infusion and at the end of the experiment. Renal ultrastructure was observed by electronmicroscopy. RESULTS: SBP of the rats received Ang II increased significantly compared with controls (P?0.05). SBP returned to normal after Ang II infusion stopped. No significant difference existed in blood pressure between rats fed high salt diet and normal salt diet in the period of 12 weeks (P<0.05). Expressions of TGF-beta1, OPN mRNA and its protein in the kidney of the rats fed with high salt diet were up-regulated at the end of Ang II infusion (P>0.05) and at the end of the 12th week compared with the rats fed normal salt diet and controls (P<0.01). The kidney was damaged by Ang II, which was further aggravated by the infusion of high salt diet. CONCLUSIONS: High salt diet could aggravate the renal injury of the rats with Ang II-induced renal injury that may be independent of blood pressure change. Expressions of the TGF-beta1, OPN mRNA and its protein were up-regulated after renal injury.
