[Transduction efficiency of recombinant adeno-associated virus 2 in human bone marrow CD34+ hematopoietic stem/progenitor cells and mesenchyme stem cells].

OBJECTIVE: To investigate the transduction efficiency of recombinant adeno-associated virus 2 ( rAAV2) in human bone marrow CD34+ hematopoietic stem/progenitor cells and mesenchyme stem cells. METHODS: The rAAV2 containing green fluorescent protein genes (rAAV2/GFP) were constructed, packaged and purified. CD34+ hematopoietic stem/progenitor cells and mesenchyme stem cells were infected with the rAAV2/GFP. After transduction for 48 hours, the expression of GFP was detected under fluorescence microscope. Furthermore, the transduction efficiency of AAV transduced CD34+ with hydroxyurea (HU) pretreatment and that of untreated were compared. RESULTS: GFP genes were expressed in 5.3% +/- 1.7% CD34+ cells. After pretreatment with HU, the expression of the GFP gene in CD34+ cells increased to 13.2% +/- 2.8%, and 23% +/- 3.6% mesenchyme stem cells expressed the GFP gene. Conclusion The transduction efficiency of mesenchyme stem cells is higher than that of CD34+ hematopoietic stem/progenitor cells. HU pretreatment can obviously increase the transduction efficiency of CD34+ hematopoietic stem/progenitor cells.

[Association of cadherin CDH23 gene polymorphisms with noise induced hearing loss in Chinese workers].

OBJECTIVE: To investigate the association of cadherin 23 gene (CDH23) polymorphisms with the development of noise induced hearing loss (NIHL). METHODS: 194 workers exposed to occupational noise were drawn as the subjects in the cross-sectional epidemiology study. According to the result of audiometry, they were divided into two groups: the NIHL group and the normal group. The genotypes of 93 workers with NIHL and 101 normal workers were tested by polymerase chain reaction followed by restriction fragment polymorphism analysis (PCR-RFLP). RESULTS: There were no significant difference in the distribution of genotypes and alleles frequencies of the rs1227049 and rs1227051 positions between NIHL group and normal group (P > 0.05), there were significant differences between the two groups in the rs3802711 position and the terminal position of exon 7 (P < 0.01). After adjusted for age, sex, smoking, history of explosive noise exposure and cumulative noise exposure (CNE) with multiple logistic regression analysis, the risk of rs1227049 CC genotype was found significantly increased than that of the GG genotype, the OR value of which was 3.865 (95% CI 1.076 - 13.886), the risk of rs3802711 TT genotype was significantly higher than that of the CT genotype, the OR value of which was 6.088 (95% CI 2.485 - 14.917). GG genotype in the terminal position of exon 7 was also found a significantly higher risk than the AG genotype, the OR value of which was 5.769 (95% CI 2.745 - 12.121). CONCLUSION: It was suggested that genetic polymorphism in cadherin CDH23 gene might play an important role in the development of NIHL in Chinese workers. The individuals with the rs1227049 CC genotype, rs3802711 TT genotype and GG genotype in the terminal position of exon 7 might be more susceptible to NIHL.

[Coronary artery atherosclerotic plaque and stent visualizations by optical coherence tomography].

OBJECTIVE: To evaluate coronary artery atherosclerotic plaque characteristics and changes post coronary stenting by optical coherence tomography (OCT). METHODS: OCT images were obtained in 22 diseased coronary vessels after coronary angiography or percutaneous coronary interventions (PCI) in 20 patients and in 23 stents [7 sirolimus-eluting stents (SES) follow up at 4-29 months post stenting and 8 bare mental stents (BMS) at 4-35 months post stenting, 8 stents immediately after PCI]. RESULTS: All 22 vessels and 23 stents OCT images were successfully acquired. Two thromboses, 8 fibrous, 9 lipid-rich and 3 calcium plaques as well as 3 plaque ruptures were visualized by OCT. No significant neointimal proliferation and restenosis were found in SES stents and some struts were not covered with neointima even at 29 months post stenting. Significant neointimal proliferation on surfaces of stent struts were visualized in all 8 BMS stents and restenosis was detected in 3 BMS stents. OCT images obtained immediately after PCI showed that 3 stents were well positioned, tissue prolapse between coronary stent struts occurred in 4 stents and stent dissociation with vessel wall could be seen in 1 stent. CONCLUSIONS: OCT imaging can clearly visualize different types of atherosclerotic plaques. By providing detailed information on plaque characteristics, this technique might help cardiologists in choosing suitable stents and guiding preventive therapy for patients with coronary heart disease.

[Subretinal transplantation of human fetal lung fibroblasts expressed ciliary neurotrophic factor gene prevent photoreceptor degeneration in RCS rats].

OBJECTIVE: To investigate the efficacy of subretinal transplantation of CNTF gene transfected fibroblasts for preventing photoreceptor degeneration in RCS. METHODS: The human fetal lung fibroblasts with high level expression of CNTF were established by liposome mediated gene transfer and MTX selection. A 5 microl of cell suspension, containing 1 x 10(5) cells, was injected through pars plana of ciliary body into the subretinal space of the right eye at postnatal 4-5 weeks, the left eye was left without injection or injected with PBS as controls. The both eyes were enucleated for histopathological examinations at 2, 4, 6, 8, 10, 12 and 15 weeks following transplantation. RESULTS: The level of CNTF protein (91,046.15 pg/ml) expressed in the transfected cells was determined by sandwich enzyme-linked immunosorbent assay (ELISA). The four of seven eyes examined by light microscopy and the ten of 14 eyes examined by electro microscopy showed rescue effect. The prolonged photoreceptor survival, reduction of apoptotic cells and debris were observed in transplanted eyes in comparison with untreated or sham-injected eyes. CONCLUSION: This study provides the first indication that transplanted human fibroblasts with high level expression of CNTF are able to rescue photoreceptor degeneration in RCS dystrophic rat retina.

[Screening effective sequences of small interfering RNAs targeting MDR1 gene in human gastric cancer SGC7901/VCR cells].

OBJECTIVE: To screen effective sequences of small interfering RNA targeting MDR1 gene in human gastric cancer SGC7901/VCR cells. METHODS: Four siRNAs (MDR1si326, MDR1si1513, MDR1si2631 and MDR1si3071) targeting MDR1 gene were designed and synthesized by in vitro transcription. The siRNA duplexes were used to transfect into the human gastric cancer SGC7901/VCR cells. The expression level of MDR1 mRNA and P-gp were detected by RT-PCR and Western blotting, respectively. The accumulation of intracellular adriamycin (ADR) was examined by flow cytometry and the cell sensitivity to ADR was demonstrated by MTT. RESULTS: The SGC7901/VCR cells treated with 4 siRNAs led to reversal effect on multidrug resistance to different extents. Among the SGC7901/VCR cells treated by siRNAs for 48 h, the expression level of MDR1 mRNA in cells of MDR1si326 or MDR1si2631 group (0.42 +/- 0.07 or 0.49 +/- 0.02) was more decreased than that in cells of MDR1si1513 or MDR1si3071 group (P < 0.05). The accumulation of ADR in cells of MDR1si326 group was the most; in cells of MDR1si2631 group, more; in cells of MDR1si3071 group, lower and in cells of MDR1si1513 group, the lowest (P < 0.05). The relative reversal efficiency of cells of MDR1si2631 group to ADR was the highest and in cells of MDR1si326 group, higher (P < 0.05). There was no significant difference in the relative reversal efficiency between the cells of MDR1si1513 and MDR1si3071 groups (P > 0.05). The expression level of P-gp in cells of MDR1si326 group was the lowest among the SGC7901/VCR cells treated by siRNAs for 72 h. CONCLUSION: The MDR1si326 with most, MDR1si2631 with more, MDR1si3071 with less and MDR1si1513 with least reversal effects on MDR1 gene mediated multidrug resistance were found in the human gastric cancer SGC7901/VCR cells.

[Experimental study on the mechanism of oilfield wastewater treatment by using hydrolysis-acidification with aerobic biological processes].

Hydrolysis-acidification + aerobic biological processes were conducted experimentally to treat oilfield wastewater pretreated with physical and chemical treatment in Xinjiang oilfield. The results showed that when the COD concentration in influent was 190-220 mg x L(-1), that in effluent reduced to 65-75 mg x L(-1) under HRT of 10h in both hydrolysis-acidification process and aerobic biological process, reaching the strictest requirement of Effluent Standards for Wastewater from Petroleum Development Industry (GB3550-83). Using GC/MS technology, the relative content of various organic pollutants was analyzed to discover the transfer and degradation law in the oilfield wastewater in biological treatment process. The system of DNA extraction technique, PCR and DGGE reacting systems were practical to analyze the microbial community in the hydrolysis-acidification and aerobic biological processes. The predominant sequences of several 16S rDNA DGGE fragments were determined and confirmed in comparison in GeneBank (NCBI).

[Inhibition of hTERT antisense oligodeoxynucleotide on proliferation and telomerase activity in HL-60 cells].

This study was purposed to investigate the inhibition of hTERT antisense oligodeoxynucleotide (ASODN) on the proliferation and telomerase activity in HL-60 cells and to explore the relativity between the telomerase activity and the expression of hTERT gene in HL-60 cells. After treated by hTERT ASODN the expression of hTERT was detected by RT-PCR, the morphological changes of HL-60 cells was observed with inverted microscopy, the cell proliferation was measured by MTT method, and the telomerase activity was determined with TRAP-ELISA and TRAP-PAGE. The results showed that after sealing hTERT gene with ASODN for 72 hours, the expression of hTERT gene was significantly inhibited, the cell growth was repressed and the ability of proliferation decreased, and the effect was specific in sequence and dependent in dose and time. OD(450-690) values were 2.648 +/- 0.42, 1.504 +/- 0.47, 1.223 +/- 0.39, 0.944 +/- 0.16 respectively, as the cells were treated with 0, 10, 20, 30 micromol/L ASODN for 72 hours. The difference was significant as compared 10, 20, 30 micromol/L groups with 0 micromol/L ASODN group respectively (P < 0.05), but the difference was no significant when compared 20 micromol/L SODN group (2.376 +/- 0.65) with untreated group (2.648 +/- 0.42) (P > 0.05). TRAP-PAGE detection revealed that comparing ASODN groups with SODN groups the telomerase image bands were decreased and least was found in groups of 30 +/- mol/L. It is concluded that the hTERT ASODN may inhibit the proliferation and down-regulate the telomerase activity in HL-60 cells by sealing the expression of hTERT gene.

[The construction of recombinant adenovirus expressing bifunctional fusion protein sCAR-EGF and the detection of its activity].

To improve the targeting of adenovirus vector for gene therapy, a fusion gene sCAR-EGF, in which epidermal growth factor gene was fused to the 3' end of extracellular Coxsackie virus-adenovirus receptor gene, was constructed and cloned into shuttle plasmid pDC315 to obtain a recombinant plasmid pDC315-sCAR-EGF. With the AdMax system, AD-293 cells were co-transfected with pDC315-sCAR-EGF and adenovirus genomic plasmid pBHGloxdeltaE13cre. Through high efficiency site specific recombination, a replication-defective adenovirus Ad5-CMV-sCAR-EGF was constructed. The recombinant adenovirus was analyzed by PCR and Western blotting, the results indicated that Ad5-CMV-sCAR-EGF contained the fusion gene sCAR-EGF, and the adenovirus infected cells was induced to produce and secrete the fusion protein into the supernatant. We have demonstrated that the fusion protein sCAR-EGF is helpful for elevating the infection efficiency of Ad5-CMV-luc with the reporter gene in vitro, which providing a new approach to the gene therapy for tumors overexpressing EGFR.

[Expression and clinical significance of hypoxia-inducible factor-1alpha in gastric cancer].

BACKGROUND & OBJECTIVE: Hypoxia-inducible factor-1alpha (HIF-1alpha) is correlated to the genesis, progression, invasion, metastasis, and prognosis of some malignancies. This study was to evaluate the expression of HIF-1alpha in gastric cancer, and explore its correlation to clinical features of gastric cancer. METHODS: The expression of HIF-1alpha in 96 specimens of gastric cancer was detected by SP immunohistochemistry. The correlations of HIF-1alpha to clinicopathologic features and prognosis of gastric cancer were analyzed with SPSS10.0 software. RESULTS: Of the 96 specimens, 77 (80.2%) were HIF-1alpha-positive: 7 were grade (+), 29 were grade (++), 27 were grade (+++), and 14 were grade (++++). The positive rate of HIF-1alpha was significantly lower in stage I-II patients than in stage III-IV patients (66.7% vs. 87.3%, P=0.016), significantly lower in stage T1-T2 patients than in stage T3-T4 patients (57.9% vs. 88.0%, P=0.007), and slightly lower in the patients without distant metastasis than in the patients with distant metastasis (75.9% vs. 100%, P=0.055). The 5-year overall survival rate was significantly lower in HIF-1alpha-positive patients than in HIF-1alpha-negative patients (31.2% vs. 57.9%, P=0.027). However, HIF-1alpha was not an independent prognostic factor of gastric cancer in multivariate analysis. CONCLUSIONS: The expression of HIF-1alpha is correlated to patients' survival, tumor stage, invasion depth, and distant metastasis of gastric cancer, and may be considered as a reference criterion in evaluating the progression, metastasis, and prognosis of gastric cancer.

[Effect of hTERT ASODN on the oncogenicity and the inductive apoptosis of HL-60 cells].

OBJECTIVE: To investigate the effect of hTERT antisense oligodeoxynucleotide (ASODN) on the oncogenicity and the inductive apoptosis of HL-60 cells. METHODS: Apoptosis of HL-60 cells was detected by flow cytometry (FCM) and agarose gel electrophoresis. Both treated and untreated HL-60 cells were collected and transplanted into 5 BALB/c nude mice respectively, the formation of transplanted neoplasm and its morphologic change were observed. After the transplanted neoplasms were uniform with the ameliorated method in another 10 BALB/c nude mice, they were divided into 2 groups and injected ASODN and PBS into the neoplasm respectively. Seven days later, the tumor were measured, its morphology were observed, and the apoptotic cells were detected with a TUNEL kit. RESULTS: After 72 h treatment there were DNA ladders and early apoptosis peak in hTERT ASODN treated HL-60 cells but was none in SODN treated and blank control cells. In tumor formation experiment, neoplasms were formed in ASODN treated group at 16-17 d and untreated group at 12-13 d. Neoplasm was formed in 2 of 5 ASODN treated mice and 4 of 5 untreated mice respectively. In untreated mice tumor tissues were rich in blood vasa and stromal tissue compared with that in ASODN treated mice. In tumor therapy experiment, before treatment, there was no difference in the average neoplasm physical volume between ASODN treated group [(100.9 +/- 24.6) mm3] and PBS treated group [(98.4 +/- 23.1) mm3] (P > 0.05). After treatment, the neoplasm volume in ASODN treated group [(422.7 +/- 326.4) mm3] was smaller than that in PBS treated group [(786.4 +/- 357.6) mm3] (P < 0.05). Histologically, there were many apoptosis cells in ASODN treated group, but was seldom seen in PBS treated group. The TUNEL positive cells in ASODN treated group were much more than that in PBS treated group (P < 0.05). CONCLUSION: The hTERT ASODN induces apoptosis of HL-60 cells in vitro, reduces the tumor formation in BALB/c nude mice and inhibits the growth of the transplanted neoplasm.

[A new method of establishing experimental model of chronic myocardial infarction by incision below xiphoid process in black goats].

OBJECTIVE: To establish an effective model of myocardial infarction in black goat so as to provide a safe, convenient and credible model of myocardial infarction for treatment and research. METHODS: Sixteen black goats were made chronic myocardial infarction by ligation of far end of left anterior descending coronary artery through incision below xiphoid process. Electrocardiogram (ECG) and serum myocardial enzymes were investigated before and after occlusion. Echocardiographic measurements were performed, and left coronary artery angiography was performed with digital subtraction angiography (DSA) before infarction and 6 weeks after infarction. The myocardial ultrastructure were observed. RESULTS: All goats survived more than 6 weeks. ECG showed ambulatory change, ST-segment elevated half an hour after occlusion and pathologic Q waves 6 weeks after infarction, CK-MB significantly increased. Echocardiographic indexes showed significant decrease of maximal peak A, percent wall thickening(WHT) and ejecting fraction (EF), increase of end-systolic volume (ESV), end-diastolic volume (EDV), and dilation of left ventricle. DSA showed block or decrease of perfusion of far end of left anterior descending coronary artery. CONCLUSION: It is safe, convenient and credible to establish model of myocardial infarction by ligation of far end of left anterior descending coronary artery through incision below xiphoid process in black goat.

[Effects of bone marrow mesenchymal stem cells enriched by small intestinal submucosal films on cardiac function and compensatory circulation after myocardial infarction in goats].

OBJECTIVE: To investigate effects of the autologous bone mesenchymal stem cells (MSCs) enriched by the small intestinal submucosa (SIS) film implantation on the myocardial structure, cardiac function, and compensatory circulation after myocardial infarction in the goats. METHODS: Sixteen black goats were selected and divided randomly into the control group (n= 8)and the experimental group (n= 8). The chronic myocardial infarction models were made by the ligation of the far end of the left anterior descending coronary artery. At the same time, MSCs were aspired from the thigh bone of the goats in the experimental group. MSCs were isolated by the centrifugation through a percoll step gradient and purified by the plating culture and depletion of the non-adherent cells. Primary MSCs were cultured in the DMEM medium supplemented with the fetal bovine serum in vitro. After that, the cultures were labeled by 5- BrdU. The active cells were transplanted into the SIS film. Six weeks after the ligation, the MSCs-SIS film was implanted by its being sutured onto the infarction area; whereas, the control group underwent a sham-operation. In both groups, echocardiographic measurements were performed before infarction, 6 weeks after infarction and 6 weeks after the MSC-collagen implantation, respectively, to assess the myocardial structure and cardiac function. The left coronary artery angiography was performed with the digital subtraction angiography. RESULTS: In an assessment of the left ventricular function, at 6 weeks after operation, the stroke volume and the ejection fraction of the control group and the experimental group were 42.81+/-4.91, 37. 06+/-4.75 ml and 59.20%+/-5.41%, 44.56% +/-4.23%, respectively (P<0.05). The end-diastolic volume and the end-systolic volume of the control group and the experimental group were 72.55+/-8.13, 83.31+/-8.61 ml and 29.75+/-5. 98, 46.25+/-6.68 ml, respectively (P<0. 05). The maximal velocity of peak E of control group and experimental group were 54. 85+/-6.35 cm/s and 43.14+/-4.81cm/s (P<0. 01); and the maximal velocity of peak A of control group and experimental group were 52.33+/-6.65 cm/s and 56.91+/-6.34 cm/s (P>0. 05). Echocardiography showing a distinctly dilatation of left ventricle with the ventricular dyskinesia in control group, but without the ventricular dyskinesia in experimental group. The selective-coronary angiography revealed that the obvious compensatory circulation established between the anterior descending branch and the left circumflex branch in the experimental group. CONCLUSION: Implantation of the autologous MSCs enriched by the SIS film can prevent dilatation of the left ventricular chamber and can improve the contractile ability of the myocardium, cardiac function, and collateral perfusion.

FK962, a novel enhancer of somatostatin release, exerts cognitive-enhancing actions in rats.

FK962 (N-(1-acetylpiperidin-4-yl)-4-fluorobenzamide) is a derivative of FK960 (N-(4-acetyl-1-piperazinyl)-p-fluorobenzamide monohydrate), with putative anti-dementia properties. Here, we wanted to determine whether FK962 retained the ability of the parent compound to both facilitate somatostatinergic nerve activity in hippocampal neurons and to ameliorate cognitive dysfunction in rat models. FK962 (10(-9) - 10(- 6) M) significantly enhanced high K+-evoked somatostatin release from rat hippocampal slices. FK962 also significantly reduced somatostatin-induced inhibition of Ca2+ channels at 10(-9) - 10(-7) M in single rat hippocampal neurons using whole-cell patch-clamp. Furthermore, administration of FK962 (0.032-3.2 mg/kg, i.p.) significantly ameliorated memory deficits in passive avoidance task in animal models: scopolamine-treated rats, nucleus basalis magnocellularis-lesioned rats and aged rats. FK962 (0.01- 1) mg/kg, i.p.) significantly improved spatial memory deficits induced by nucleus basalis magnocellularis-lesion in water maze task. These results suggest that FK962 ameliorates cognitive impairment in rats via activation of the somatostatinergic nervous system in the hippocampus, indicating that FK962 could be a potent cognitive enhancer and therefore might be of therapeutic value for cognitive disorders such as Alzheimer's disease.
