P130 and its truncated form mediate p53-induced cell cycle arrest in Rb(-/-) Saos2 cells.

In the present study, we investigate the mechanism of how p53 induces growth arrest in Rb-defective Saos2 cells that express temperature-sensitive mutant p53 (ts p53). The activation of p53 at a permissive temperature (32.5 degrees C) induces the cell cycle arrest at both the G1 and G2 stages. The induction of several p53-responsive genes as well as a small form of p130 (S-p130) was detected upon p53 activation. S-p130 retained the functions as a pocket protein and was dominant over p130 at the protein level after 36 h at 32.5 degrees C. A canonical p53 binding site was identified in intron 4 of p130. Furthermore, a novel p53-inducible transcript containing a partial intron 4 sequence downstream of the p53 binding site and exon 5 of p130 was detected by RT-PCR, suggesting S-p130 is induced by p53 at transcriptional level. The results from gel shift assay and immunoprecipitation showed that S-p130 as well as p130 formed complexes with both E2F1 and E2F4 at a permissive temperature. Moreover, the transient expression of E1A (12S) and E2F1 effectively abrogated p53-induced cell cycle arrest. These results strongly suggested that p130 and its truncated form might substitute Rb in mediating p53-induced cell cycle arrest in Rb(-/-) Saos2 cells.

Cost-effective low timing jitter passively Q-switched diode-pumped solid-state laser with composite pumping pulses.

A novel scheme that combines gain switching with passive Q switching of a miniature diode-pumped solid-state laser is proposed and implemented. A composite pumping pulse, consisting of a long, low-intensity pulse and a following short, high-intensity pulse, is used to reduce the timing jitter. A greater-than-tenfold reduction in timing jitter is demonstrated.

Extreme mtDNA homogeneity in continental Asian populations.

Mitochondrial DNA (mtDNA) variation in continental Asia has not been well-studied. Here, we report mtDNA HV1 sequences for 84 Xi'an and 82 Changsha Han Chinese, 89 Honshu Japanese, and 35 Vietnamese. Comparison of these sequences with other Asian mtDNA sequences reveals high variability within populations, but extremely low differentiation among Asian populations. Correlations between genetic distance and geographic distance, based on mtDNA and Y chromosome variation, indicate a higher migration rate in females than in males. This may reflect patrilocality, as suggested previously, but another plausible hypothesis is that the demographic expansion associated with the spread of agriculture in Asia may be responsible for the extreme genetic homogeneity in Asia.

[Changes of element contents and their interrelationship in tibia of osteoporotic rats]

OBJECTIVE: To observe the changes of bone element contents in osteoporosis and their interrelationship. METHODS: Twelve female SD rats,10-month-old, were bilaterally ovariectomized (OVX group) and another ten rats were received sham-operation under anesthesia (SHAM group).The element contents in tibia, including Ca, P, Mg, Zn, Mn, Fe, Cu, Mo and Cr, were determined by atomic absorption spectrophotometer 7 month later. The data of contents of all elements were analyzed by simple regression. RESULTS: Compared with the SHAM group rats, the contents of Ca, P and Mg were decreased by 6.6 %(P<0.05), 6.3 %(P<0.05) and 14.9 %(P<0.01) respectively. The contents of Zn and Fe were reduced by 15.2 %(P<0.01) and 35.1 %(P<0.01) separately, Mo and Cr were decreased by 12.2 %(P>0.05) and 14.0 %(P>0.05), while the contents of Mn, Cu and Co were shown no change. There was a significant correlation among the contents of Mg, Mn, Zn, Ca and P. CONCLUSION: The contents of Ca, P, Mg, Zn and Fe were matkedly reduced in bone of osteoporotic rats induced by ovariectomy.

[Study of ABO blood group secretor type alpha(1,2)-fucosyltransferase gene polymorphism in Chinese].

Secretor gene (FUT2)-encoded secretor type alpha(1,2)-fucosyltransferase (Se enzyme) that regulates expression and secretion of ABO(H) antigens in epithelial cell of glands and body fluids. It has the extensive polymorphism and race specificity. In the present study, we investigated the distribution of the fusion gene of the FUT2 locus in 191 Manchu individuals from Liaoning Province and 208 Mongolian individuals from Inner Mongolia, and analyzed the polymorphism of the FUT2 gene in 90 unrelated Han Chinese from Shandong Province and 90 Mongolian from Inner Mongolia, respectively. The fusion gene was not found in the two investigated ethnic groups. The frequencies of G849A nonsense mutation in Shandong Han Chinese and Mongolian of Inner Mongolia individuals were the same, 0.0055.
