Posttranslational modifications of tau--role in human tauopathies and modeling in transgenic animals.

Alzheimer's disease (AD) is characterized histopathologically by beta-amyloid-containing plaques, neurofibrillary tangles (NFT), reduced synaptic density, and neuronal loss in selected brain areas. Plaques consist of aggregates of a small peptide termed Abeta which is derived by proteolysis of the larger amyloid precursor protein APP, whereas NFT are composed of hyperphosphorylated forms of the microtubule-associated protein tau. Tau pathology in the presence of scant or no beta-amyloid plaques characterizes additional neurodegenerative disorders collectively called tauopathies. In the course of plaque and NFT formation, the major proteinaceous components of these lesions undergo post-translational modifications. In the case of tau, these include phosphorylation of mainly serine and threonine, but also tyrosine residues. In addition, tau is subject to ubiquitination, nitration, truncation, prolyl isomerization, association with heparan sulfate proteoglycan, glycosylation, glycation and modification by advanced glycation end-products (AGEs). This review aims to provide insight into the complexity of tau modifications in human tauopathies such as AD and frontotemporal dementia with parkinsonism linked to chromosome 17 (FTDP-17). Selected aspects of the post-translational modification of tau have been reproduced in transgenic animal models. Most of this work has been done in mice, but insight has also been gained from studies in the sea lamprey, the nematode C. elegans and Drosophila. Attempts have been made to link specific post-translational modifications with tau aggregation and nerve cell dysfunction.

Expression of IFN-gamma and its receptor alpha in the peripheral blood of patients with chronic hepatitis C.

BACKGROUND: It has been known that intra-cellular immunity is important for defense against viral infections and this function lies with interferon gamma (INF-gamma). Here we evaluated the role of IFN-gamma system in the pathogenesis of chronic hepatitis C (CHC). METHODS: The levels of interferon gamma receptor alpha (IFNGR alpha) on the peripheral lymphocyte membrane were assayed with flow cytometry. The plasma concentrations of the cytokines IFN-gamma and IL-10 in CHC patients and normal controls were assayed by enzyme-linked-immunosorbent assay (ELISA). The samples were collected randomly from Xinjiang Autonomous Region, Zhejiang and the northern regions of Jiangsu Province in China. RESULTS: The levels of IFNGR alpha in CHC patients were significantly lower than that of normal controls (NC), especially among patients during the stable stage (P < 0.001), whereas there were no significant differences between CHC in active and stable stages. Among the patients of the three regions, there were no significant differences between patients from Xinjiang and Zhejiang provinces, but both had statistically significant difference compared with the patients from Jiangsu Province (P < 0.001). Plasma IFN-gamma and IL-10 concentrations in CHC patients decreased significantly, IFN-gamma in particular, but there were no significant differences in these levels between various stages of the disease. The IFN-gamma/IL-10 (Th1/Th2) ratio in patients was reversed. CONCLUSION: There may be defects in the IFN-gamma system in chronic HCV infected subjects and a low immune response, which may play an important role in the persistence of HCV infection.

[Population genetic analysis of Y-chromosomal single nucleotide polymorphism in six Chinese populations].

OBJECTIVE: To analyze the genetic polymorphism of 15 single nucleotide polymorphism (SNP) loci on the nonrecombining portion of the Y chromosome in 6 populations in China. METHODS: Allelic specific polymerase chain reaction and 2% agarose gel electrophoresis and 6% PAGE were used to analyze the genetic polymorphism of 343 unrelated males, representing 6 populations in China, including Fujian Hans, Sichuan Hans, Mongolian, Hezhen, Sibo and Hui from the South, Northeast and Northwest. RESULTS: Thirty haplogroups were observed, and 3 of them (H15, H16, H18) were seen in all of the six populations. Although the heterozygosity levels of the Hezhen, Mongolian, Sibo populations are similar and those of the other 3 populations (Fujian Hans, Sichuan Hans, Hui) are similar, the pairwise differences among haplogroups are significant. Analysis of molecular variance (AMOVA) and principal component (PC) analysis of the haplogroup distributions suggested highly different allele diversity between group I including Hezhen, Mongolian, Sibo and group II including Hui, Fujian Hans, Sichuan Hans. CONCLUSION: The above analyses show more significant variance components in Northeast/South populations and clearly reveal the geographic genetic relationship among the six populations in the Northeast/Northwest/South. These results confirm the complexity of the genetic structure of Chinese populations and make a significant contribution for constructing the contemporary human gene pool and tracing genetic dispersal trail from Chinese populations.

[Peri-uvulopalatopharyng oplasty otoacoustic emissions in patients with obstructive sleep apnea-hypopnea syndrome].

OBJECTIVE: To study the cochlear function of patients with obstructive sleep apnea-hypopnea syndrome (OSAHS). METHODS: Transiently evoked otoacoustic emissions (TEOAEs) and Distortion-product otoacoustic emissions(DPOAEs) of 28 patients with OSAHS were measured one week before uvulopalatopharyngoplasty (UPPP) and six months after UPPP. RESULTS: In these patients the prevalence of TEOAEs was 64.3%, its spectral energy was mainly distributed between 0.5-2 kHz, the amplitudes of DPOAEs between 0.5-8 kHz were reduced significantly, the prevalences of DPOAEs at 0.5-8 kHz were between 46.4%-89.3%. After UPPP, the prevalences of TEOAEs and DPOAEs were increased, the amplitudes of DPOAEs were in turn (22.50 +/- 16.70) dB SPL, (5.83 +/- 2.69) dB SPL, (0.69 +/- 3.83) dB SPL, (-2.50 +/- 2.45) dB SPL, (4.95 +/- 2.65) dB SPL, (4.48 +/- 3.07) dB SPL from 0.5 kHz to 8 kHz, the amplitudes of DPOAEs were significantly increased at 1 kHz, 2 kHz, 4 kHz, 6 kHz, 8 kHz. CONCLUSION: OSAHS can affect the cochlear function of the patients with OSAHS, UPPP can improve the cochlear function.

[Case-parental control study].

OBJECTIVE: To introduce the methodology on the design and statistical method as well as analysis of published data related to case-parental control study. METHODS: Data from a research on association between the human neural tube defects and the T allelic variant TIVS7-2 was analyzed by haplotype relative risk (HRR), genotype relative risk (GRR) and transmission/disequilibrium test (TDT) methods. RESULTS: Using the HRR method, HRR value was 1.33 (chi-square statistic is 1.4618 with P = 0.2266). The results of the GRR method showed that psi(1) value was 1.26 and psi(2) value was 0.98 (chi-square statistic is 3.1809 with 2 df, P = 0.2038). The transmission/disequilibrium test showed that TDT was 1.4516 with P = 0.2283. All results suggested that when a weak association of TIVS7-2 allele with neural tube defects was found, there was no evidence showing the statistical difference. CONCLUSION: The case-parental control design method avoids population stratification when parents used as controls. This method might be used to test the association between genes and disease, as well as to evaluate the gene environmental interaction.

[Preliminary study on the relationship between loads of human papillomavirus in cervical carcinoma and cervical intraepithelial neoplasia].

OBJECTIVE: To explore the relationship between loads of human papillomavirus in cervical carcinoma and cervical intraepithelial neoplasia. METHODS: From December 2001 to May 2002, 9075 married women aged from 35 to 50 years who lived in the areas with a high incidence of cervical carcinoma of Shanxi Province were screened high risk types of human papillomavirus (HPV) infection using hybrid capture II (HC-II). Of them, 2087 women with positive human papillomavirus further underwent colposcopy and multi-focal directed punch biopsies plus endocervical currettage. RESULTS: Two thousand and eighty-seven women were found with positive human papillomavirus infection, comprising 1402 (67.2%) women who were diagnosed as cervicitis, 663 (31.8%) women diagnosed as cervical intraepithelial neoplasia [including cervical intraepithelial neoplasia (CIN) I to III], and 22 (1.1%) women diagnosed as cervical squamous cell carcinoma (SCC). HPV-DNA loads of women with chronic and acute cervicitis were 150 +/- 11 and 108 +/- 13, respectively, with no significant difference between two groups statistically (P = 0.225). HPV-DNA loads for women with CIN I, CIN II and CIN III were 332 +/- 29, 358 +/- 35, and 370 +/- 31, respectively, all significantly higher than that of women with cervicitis (P = 0.000), but there were no significant differences among three groups (P > 0.05). HPV-DNA loads of women with cervical squamous cell carcinoma was 593 +/- 86, much higher than those of women with varied grades of cervical intraepithelial neoplasia and cervicitis (P < 0.05 and P < 0.01). CONCLUSIONS: The new technology using HC-II is an effective method to detect HPV infection in cervix. Viral loads of HPV-DNA increase with severity of cervical neoplasia. So it could be used for screening primary cervical carcinoma.

[DNA binding activity of nuclear factor-kappa B in human astrocytoma].

BACKGROUND & OBJECTIVE: Transcription factors of human cancer cells can regulate the expression of many malignancy-related gene products, relate to poor differentiation, and high metastasis of cancer cells, and also inhibit apoptosis of cancer cells. This study was designed to investigate the nuclear factor-kappa B (NF-kappaB)DNA binding activity in human astrocytoma tissue, and its significance with human astrocytoma development. METHODS: The NF-kappaB DNA binding activities of 12 cases of normal brain tissue, and 37 cases of human astrocytoma tissue were detected by electrophoretic mobility shift assay (EMSA), and photo densitometry. RESULTS: The absorbency of NF-kappaB DNA binding activity in human astrocytoma tissue was 134.2+/-24.1, higher than that in normal brain tissue (97.5+/-1.9)(P< 0.05). The NF-kappaB DNA binding activity in multiform glioblastoma tissue was highest (106.8+/-7.4), that in anaplasia Astrocytoma was 123.2+/-10.1, in astroblastoma was 139.3+/-16.8,and in astrocytoma was 160.2+/-18.6 (P< 0.05). CONCLUSION: The NF-kappaB DNA binding activity correlates with malignancy degree of human astrocytoma.

[Cochlear reimplantation].

OBJECTIVE: To analyze retrospectively the outcome of 6 cases of cochlear reimplantation to summarize its experience. METHODS: The operation method of cochlear reimplantation was similar to routine cochlear implantation. A "H" incision for the muscle-periosteum was used to shorten the operation time. RESULTS: Of all the cases, the electrodes are successfully and totally inserted into the cochlea. The causes for revision are different. The electrode was found to be not in the proper situ of the cochlea after the first operation in one patient. The out part was ruptured because of trauma in another patient. The causes of the other four patients were unknown. CONCLUSIONS: The incision should not be too small. The control electrode should be put beneath the periosteum. The site used to fix up the out part need be burnished. Unabsorbable suture should do not be used. It is important to insert the new electrode quickly after drawing the old one out. The time to accommodate the new electrode often needs 3-4 weeks.

[Study on the spectrum response characteristic based on the wide type bacteriorhodopsin film].

The photochromic retinal protein bacteriorhodopsin (BR) was found in the cell membrane of the Archaean Halobacterium salinarium. The excellent photochromic and photocycle properties of the BR provide the possibility of many applications in the filed of optical information processing. In this paper, the spectrum response characteristic of the wild type bacteriorhodopsin molecule film was studied by using pump-probe method. After the samples was excited by 532 nm YAG laser beam, the absorption spectra were probed by an optical fiber spectrum analysis (OSA). The absorption peaks at the ground state (B state) of the two samples are all at 562 nm wavelength. At 562 nm wavelength, the optical densities (OD) of the samples are about OD (WT1) 562 nm = 2.04 and OD (WT2) 562 nm = 1.37 respectively. The experiment results show that BR(WT) films have absorption that appears to strengthen with the probe time increasing in wavelength 550-650 nm, and this change phenomenon is described by spectra measured at different probe time. Appling the theoretical plot-fit of two exponentials to analyze the process of the absorption change it is found that this change includes two processes-fast process and slow process. Their corresponse time constants of BR(WT1) are about 11 ms and 60 s, and those of BR(WT2) about 24 and 30 s respectively.

[EGG and acoustic test for the diagnosis of laryngeal diseases].

OBJECTIVE: To explore the relationship between electroglottography/acoustic test parameter and disease diagnosis. METHOD: 3.0 software by Dr. Speech was used to test synchronically the electroglottography and acoustic test parameter of 503 cases so as to make a statistical analysis and comparison. RESULT: The shimmer was similar with the jitter on the inspection of polyp of vocal cord and vocal nodules, but the shimmer was more susceptive than the jitter. Compared with the healthy group, mean F0, jitter and shimmer of polyp of vocal cord increased, while the Harmonic-to-Noise decreased, so did the vocal nodules and carcinoma of larynx. Compared with the normal, the contact index of carcinoma of larynx increased, but the contact quotient remained on the same level. CONCLUSION: EGG, acoustic test number comparison and wave change provide an objective quantized index for the diagnosis of laryngeal diseases.

Islet cell engraftment and control of diabetes in rats after transplantation of pig pancreatic anlagen.

The insufficient supply of tissue, loss posttransplantation, and limited potential for expansion of beta-cells restrict the use of islet allotransplantation for diabetes. A way to overcome the supply and expansion problems is to xenotransplant embryonic tissue. We have shown that whole rat pancreatic anlagen isotransplanted into the omentum of rats, or xenotransplanted into costimulatory blocked mice, undergo growth and differentiate into islets surrounded by stoma without exocrine tissue. Isotransplants normalize glucose tolerance in diabetic hosts. Here, we show that embryonic day 29 porcine pancreas transplanted into the omentum of adult diabetic rats undergoes endocrine tissue differentiation over 20 wk and normalizes body weights and glucose tolerance. Unlike rat-to-rodent transplants, individual alpha- and beta-cells engraft without a stromal component, and no immunosuppression is required for pig-to-rat transplants. Herein is described a novel means to effect the xenotransplantation of individual islet cells across a highly disparate barrier.

Application of high-speed counter-current chromatography to the preparative separation and purification of baicalin from the Chinese medicinal plant Scutellaria baicalensis.

Baicalin was separated and purified for the first time from the traditional Chinese medicinal plant Scutellaria baicalensis Georgi by high-speed counter-current chromatography. Crude baicalin was obtained by extraction with methanol-water (70:30, v/v) from S. baicalensis Georgi. The separation was performed in two steps with a two-phase solvent system composed of n-butanol-water (1:1, v/v), in which the lower phase was used as the mobile phase at a flow-rate of 1.0 ml min(-1) in the head-to-tail elution mode. A total of 37.0 mg of baicalin at 96.5% purity was yielded from 200 mg of the crude baicalin (containing 21.6% baicalin) with 86.0% recovery as determined by HPLC analysis.

Caspase cleavage of tau: linking amyloid and neurofibrillary tangles in Alzheimer's disease.

The principal pathological features of Alzheimer's disease (AD) are extracellular amyloid plaques and intracellular neurofibrillary tangles, the latter composed of the microtubule-binding protein tau assembled into paired helical and straight filaments. Recent studies suggest that these pathological entities may be functionally linked, although the mechanisms by which amyloid deposition promotes pathological tau filament assembly are poorly understood. Here, we report that tau is proteolyzed by multiple caspases at a highly conserved aspartate residue (Asp421) in its C terminus in vitro and in neurons treated with amyloid-beta (Abeta) (1-42) peptide. Tau is rapidly cleaved at Asp421 in Abeta-treated neurons (within 2 h), and its proteolysis appears to precede the nuclear events of apoptosis. We also demonstrate that caspase cleavage of tau generates a truncated protein that lacks its C-terminal 20 amino acids and assembles more rapidly and more extensively into tau filaments in vitro than wild-type tau. Using a monoclonal antibody that specifically recognizes tau truncated at Asp421, we show that tau is proteolytically cleaved at this site in the fibrillar pathologies of AD brain. Taken together, our results suggest a novel mechanism linking amyloid deposition and neurofibrillary tangles in AD: Abeta peptides promote pathological tau filament assembly in neurons by triggering caspase cleavage of tau and generating a proteolytic product with enhanced polymerization kinetics.
