[Immediate effect of cardiac resynchronization therapy by tissue synchronization imaging and tissue tracking in patients with congestive heart failure].

OBJECTIVE: To evaluate the immediate effect of cardiac resynchronization therapy (CRT) by Doppler tissue imaging (DTI), tissue synchronization imaging (TSI) and tissue tracking imaging (TTI) in patients with congestive heart failure. METHODS: Ten patients with congestive heart failure who had cardiac resynchronization therapy were enrolled. The TSI and TTI imaging were performed by GE vivid 7 with M3s probe. The TTI image was obtained in diastole to determine delayed longitude contraction (DLC). The left ventricular ejection fraction (LVEF), the percentage of delayed longitude contraction segments (DLC), the standard deviation of the time to peak myocardial systolic contraction of 16 segments (Ts-SD), the standard deviation of the time to peak myocardial diastole of 16 segments (Td-SD), the systolic velocity of right ventricule (RV-Sm) and the average systolic velocity of mitral valve annulus (LV-Sm) were measured. The intraventricular dyssynchrony could be semi-quantified by TSI as 4 (red), 3 (orange), 2 (yellow), 1 (green), and the average value of 16 segments was defined as the TSI index. The immediate changes of these parameters were investigated when the pacemaker was turned on and off. The correlation of the Ts and TSI index was also analyzed. RESULTS: When the pacemaker was on, the LVEF improved significantly from (37 +/- 11.30)% to (46 +/- 10.10)% (P < 0.01), and LV-Sm increased significantly from (3.16 +/- 0.87) cm/s to (3.76 +/- 0.74) cm/s (P < 0.01), RV-Sm increased significantly from (6.79 +/- 1.78) cm/s to (7.75 +/- 1.92) cm/s (P < 0.01). DLCs decreased significantly from (35 +/- 6.04)% to (18.13 +/- 9.97)% (P < 0.01), Ts-SD decreased from (83.97 +/- 33.02) ms to (52.67 +/- 19.65) ms, P < 0.05, Td-SD decreased from (87.81 +/- 22.34) ms to (63.45 +/- 31.49) ms, P < 0.05 and TSI index reduced from 2.11 +/- 0.15 to 1.60 +/- 0.33 (P < 0.01) respectively. In addition, the reduction of TSI index correlated significantly with that of Ts-SD (r = 0.75, P < 0.05). CONCLUSIONS: CRT could immediately improve the systolic and diastolic synchrony of the left ventricle and ventricular function. TSI and TTI may be as the new effective modalities to assess the mechanical dyssynchrony. TSI index was direct and reliable in this study.

[Expressions of nuclear factor-kappaB and intercellular adhesion molecule-1 in endometriosis].

OBJECTIVE: To investigate the association of nuclear factor-kappaB (NF-kappaB) and intercellular adhesion molecule-1 (ICAM-1) with endometriosis. METHODS: Immunohistochemistry was used to examine the NF-kappaB and VEGF protein expression in 35 specimens of normal endometrium, 48 eutopic endometrium of endometriosis and 76 ectopic endometrium, and the results were analyzed statistically. RESULTS: The expressions of NF-kappaB and ICAM-1 were found mainly in the glandular epithelial cells of the three endometrium specimens, with the ectopic endometriotic tissues showing the highest and normal endometrium the lowest expressions (P<0.001). NF-kappaB and ICAM-1 expressions were positively correlated to normal, eutopic and ectopic endometrium (r=0.688, 0.773 and 0.777, respectively, P<0.01). CONCLUSION: NF-kappaB and ICAM-1 may participate in the process of the pathogenesis in endometriosis.

[Fragment length polymorphism of Wx-B1 gene digested by BamHI and its relation to amylose content of wheat].

There are two types of starch, amylose and amylopectin of flour. Amylose content (AC) is significantly negatively correlated with noodle quality. The waxy gene (Wx) encodes the granule-bound starch synthase (GBSS, EC 3.4.1.11), or Wx protein, is responsible for the synthesis of amylose in wheat. Common wheat is allohexaploid, it has three different Wx proteins: Wx-A1, Wx-B1, and Wx-D1. Consequently, amylose content and pasting properties of starch were influenced by the lack of the Wx-B1 protein, followed by that of Wx-D1, and Wx-A1. Using STS (sequence-tagged sites) primers, the sequences were amplified near the 4th intron region, and fragments amplified were digested by BamHI in 35 wheat cultivars. The results indicate that some fragments can be digested and the other cannot. The fragments digested showed two length polymorphism, and the length is negatively correlated with amylose content. The fragment in many cultivars with amylose content above 20% can be digested by BamHI, while those with amylose content below 20% cannot. All these indices demonstrate that there is variation exists in amplified region of Wx-B1 gene. This can be useful in breeding program to select wheat cultivars with different amylose content, and improve wheat quality.

[Molecular cloning and expression of an isotoxin gene, alpha-bungarotoxin, from Bungarus multicinctus].

Abstract: Snake venom contains a number of small proteins, enzymes and other components, which displays a broad spectrum of biological activities. With the ability of specifically binding on acetylcholine acceptor, alpha-bungarotoxins are not only useful molecular probes in investigating the mechanism of neural signal transmission, but also potential pharmic preparations for neural disease treatment. In current research,cDNAs of Bungarus multicinutus venom gland were synthesized using SMART cDNA amplification kit and then, alpha-bungarotoxin genes were cloned and sequenced. Total of 20 clones were sequenced representing 14 isotoxin mRNAs of alpha-bungarotoxins. Among those clones, a novel isotoxin gene was subcloned into two expression plasmids, alpha-BgTX/pQE30a and alpha-BgTX/pGEX-4T-1, and transformed into E. coli. After inducing with IPTG, fused protein of GST-alpha-BgTX was successfully expressed at level of 30% gross proteins of bacteria. More than 25% of fused protein was in the soluble fraction and the rest in inclusion body.

[Beneficial neurohormonal profiles of beta-blockades in chronic left heart failure].

OBJECTIVES: The aims of this study were to evaluate the effects of beta-blockers on neurohormonal factors in patients with chronic left heart failure (CHF). METHODS: 44 patients, 33 men and 11 women, with age of 60.1 +/- 10.6 years with chronic left heart failure (ejection fraction less or equal to 40% by UCG) were included in this study. All patients received conventional therapy and were randomly assigned either to a bisoprolol or carvedilol group. The dosage of beta-blockers were increased gradually to target or the tolerant dosages (bisoprolol 10 mg qd, carvedilol 25 mg bid) during 3 months in 36 patients. Maintenance dose was continued for 4 months. Plasma concentrations of renin activity (PRA), angiotensin II (Ang II), aldosterone (Ald) and the N-terminal portion of brain natriuretic (NT-proBNP) were assessed with RIA and ELISA at baseline and 3 and 7 months after staring beta-blocker therapy. Left ventricular ejection fraction (LVEF, Modify SIMPSON) was assessed at baseline and 7 months after starting therapy. RESULTS: (1) In patients with left heart failure, the baseline plasma level of PRA, Ang II and Ald were at normal range. N-terminal BNP concentration was much higher than 200 pg/ml as a result of impaired systolic function, as it elevated with increasing of NYHA grade. (2) The plasma level of NT-proBNP decreased significantly, as compared with that before therapy, but there is no significant change of plasma level of PRA, Ang II and Ald. (3) There were no significant differences between the event group and non-event group for the plasma level of renin-angiotensin and aldosterone during 7 months after starting beta-blocker. The Plasma levels of NT-proBNP were much higher in the event group than non-event group. (4) Multi regression analysis showed that the value of LVEF increased with the decreasing of NT-proBNP levels (beta = -0.389, P = 0.009) and increasing of Ang II level (beta = 0.341, P = 0.020) at baseline. After-therapy LVEF increased with the decreasing of NT-proBNP levels at titration-end (beta = -0.424, P = 0.020). CONCLUSIONS: The plasma level of NT-proBNP is more sensitive and accurate than the plasma level of PRA, Ang II and Ald in evaluation of severity and prognosis of CHF. beta-Blocker administration in patients with CHF decreases circulating levels of NT-proBNP and thus improves left ventricular function, but there is no significant effect on plasma level of PRA, Ang II and Ald.

[Effects of c-myc antisense oligodeoxynucleotide on the telomerase activity and the induction of apoptosis in HL-60 cells].

To investigate the effects of c-myc antisense oligodeoxynucleotide (ASODN) on the telomerase activity and the induction of apoptosis in HL-60 cells, and to explore the relationship between the telomerase activity and the expression of c-myc gene in HL-60 cells, after treatment by c-myc ASODN, the expression of c-myc was detected by RT-PCR, the apoptosis, cell cycle were detected with agarose gel electrophoresis and flow cytomety, and the telomerase activity was determined with TRAP-ELISA. The results showed that after blocking c-myc gene with ASODN for 72 hours, it is obvious that the expression of c-myc gene was inhibited. The percentage of S phase HL-60 cells decreased from 55.6% to 30%, the early apoptosis peak appeared (the percentage of apoptosis cells were 25.2%) and the DNA ladders were shown. OD(450 - 690) were 2.648 +/- 0.42, 2.324 +/- 0.36, 2.162 +/- 0.38, 1.952 +/- 0.14, 1.805 +/- 0.40, 1.616 +/- 0.41 and 2.466 +/- 0.29, respectively, as the cells were treated with 0, 1, 2, 3, 4, 5 micromol/L ASODN and 5 micromol/L SODN for 72 hours. The difference was significant when compared 3, 4, 5 micromol/L groups with 0 micromol/L ASODN group respectively (P < 0.05), but the difference was no significant when compared 1, 2 micromol/L ASODN and 5 micromol/L SODN groups with 0 micromol/L ASODN group (P > 0.05). It is concluded that the c-myc gene ASODN may induce the apoptosis of cells, inhibit cells from G(1) phase into S phase and regulate the telomerase activity down in HL-60 cells by blocking the expression of c-myc gene.

[Effects of dilution rates on the oscillatory behaviors of a very high gravity continuous ethanol fermentation system].

Continuous ethanol fermentation using very high gravity medium containing 280 g/L glucose, 5 g/L yeast extract and 3 g/L peptone was run at the dilute rates of 0.006 h(-1), 0.012 h(-1), 0.017 h(-1), 0.024 h(-1) and 0.032 h(-1) (based on the total working volume) in a combined bioreactor system composed of a stirred tank and three-stage tubular bioreactors in series. Oscillations marked by big fluctuations of residual glucose, ethanol and biomass were observed at the dilution rate of 0.012 h(-1). The Hopf Bifurcation theory was used to analyze and predict the occurring of these oscillations and the dilution rates that incited oscillations. Theoretical analysis revealed that oscillations can occur at designated specific growth rates and was validated by experimental results. The benefits of oscillations for the fermentation system were also discussed by comparing the fermentation results with those without oscillations.

[Continuous ethanol fermentation using self-flocculating yeast in multi-stage suspended bioreactors coupled with directly recycling of waste distillage].

A fermentation system composed of four airlift suspended-bed bioreactors in series and with a total working volume of 4800 mL was established. Continuous ethanol fermentation using self-flocculating yeast SPSC01, a fusant from Saccharomyces cerevisiae and Schizosaccharomyces pombe, and two-stage enzymatic hydrolyte of dry milling corn powder, was continuously run for 120 days. All of the backset distillage collected after distilling the final beer was used to mix the corn powder and no any other wastes except the solid residue of corn powder was discharged from the fermentation system, which guaranteed the distillage to be recycled at its maximum. The experimental results revealed that both ethanol and residual sugar in the final beer could be maintained relatively stable with their average levels of 93.6 and 7.9 g/L, respectively when the fermentation system was operated at the dilution rate of 0.05 h(-1). Parameter oscillations reported previously were also observed for the first and second bioreactors, but were effectively attenuated thereafter, which indicated that high yeast cell concentrations resulted from the self-immobilization of this special self-flocculating strain contributed to damp these oscillations. The monitoring of residual nitrogen and phosphor indicated that the accumulations of these nutritional elements occurred and the amount of these inorganic salts supplemented in the substrate should be decreased properly.

[Sequence analysis of the HA1 regions of hemagglutinin gene from influenza viruses (H3N2) isolated from children in Beijing in 2004 indicated antigenic variation].

OBJECTIVE: To identify variations in hemagglutinin genes from influenza viruses (H3N2) isolated from infants and young children with acute respiratory infection (ARI) between March, 2004 and April 2005. METHODS: RNAs from influenza A virus strains (subtype H3) isolated from specimens collected from ARI children were extracted followed by amplification for HA1 fragments from hemagglutinin (HA) genes by RT-PCR. The sequences of the fragments were defined by direct sequencing for the PCR products or the target inserts after the PCR fragments were cloned into the TA-cloning vector pBS-T and analyzed by bioinformatic software. RESULTS: Fragments of 987 bps of HA1 (encoding 329 amino acids) from a total of 32 strains of influenza A virus (subtype H3) isolated from the 2004 season and 1 from the 2003 season were amplified and the sequences were compared with vaccine reference strains recommended by WHO which were used in recent years. There were several consistent amino acid variations which involved in both antigenic epitopes A and B and receptor binding site (RBS) for isolated strains in the 2004 influenza season compared with the vaccine strains used during the recent years and the virus strains isolated in March 2004, indicated the antigenic drift of the viruses isolated in 2004 influenza season may lead to variant viruses. CONCLUSION: The variations of the HA genes from influenza virus (subtype H3) strains in the 2004-2005 influenza season were confirmed by sequence analysis for the HA1 regions of the hemagglutinin genes, which indicate that the antigenic drift would have been caused by the diversification of the genes and the efficacy of the recently used vaccines should be kept under close watch.

Cultivation of aerobic granules for simultaneous nitrification and denitrification by seeding different inoculated sludge.

Cultivation of aerobic granules for simultaneous nitrification and denitrification in two sequencing batch airlift bioreactors was studied. Conventional activated floc and anaerobic granules served as main two inoculated sludge in the systems. Morphological variations of sludge in the reactors were observed. It was found that the cultivation of aerobic granules was closely associated with the kind of inoculated sludge. Round and regular aerobic granules were prevailed in both reactors, and the physical characteristics of the aerobic granules in terms of settling ability, specific gravity, and ratio of water containing were distinct when the inoculate sludge was different. Aerobic granules formed by seeding activated floc are more excellent in simultaneous nitrification and denitrification than that by aerobic granules formed from anaerobic granules. It was concluded that inoculated sludge plays a crucial role in the cultivation of aerobic granules for simultaneous nitrification and denitrification.

[Studies on relics conservation by diffuse reflectance spectroscopy].

A diffuse reflectance spectroscopic method was developed to investigate relics conservation with a self-made fiber optics reflectance spectrophotometer. With this technique (1) identifying pigments on the color relics was done by comparing the shapes of reflectance spectra and the positions of reflectance peaks or first derivative peaks with those of related standard pigments; (2) changes of surface states of metals were monitored. From the investigation on the reflectance spectra of copper and its rust product, we can easily get some information about the metal surface, so diffuse reflectance spectroscopy shows special function in the research on metal surface; (3) the diffuse reflectance spectroscopic characteristics of the materials used in protection and restoration of cultural heritages were studied, which can help us choose proper protecting and restoring materials. From our studies it was verified that diffuse reflectance spectroscopy is a quick, simple and non-destructive analytical technique. The study makes diffuse reflectance spectroscopy a new means for relic protection, and opens a new application area for diffuse reflectance spectroscopy as well.

[Constructing recombinant plasmid with prostate-specific membrane antigen promoter and reverse enhancer and comparing the transcription activity of enhancers in different directions].

OBJECTIVE: To assess the specificity of prostate-specific membrane antigen(PSMA) promoter and enhancer in controlling gene expression and to compare the activity of enhancers in different directions for choosing the most suitable prostate-specific PSMA controlling elements. METHODS: PSMA enhancer gene was amplified with PCR, then the enhancer gene was subcloned into the expressing vector pEGFP-PSMA(Pro) reversely to construct the recombinant plasmid pEGFP-PSMA(E(r)-p), which was transfected into different cell lines such as LNCaP, PC-3,MCF-7,A549. Green fluorescent protein (GFP) expression was observed and compared to other recombinants constructed previously. RESULTS: The recombinant plasmid with reverse enhancer was successfully constructed, the PSMA promoter and enhancer showed modulating activity in PSMA-expressed cell line uniquely. PSMA enhancer could increase 30-fold transcriptional activity over the basal level achieved by PSMA promoter alone, and no impact of the direction on the activity of enhancer was noted. CONCLUSION: PSMA promoter/ enhancer is specific to PSMA-expressed cells. The transcriptional activity of reverse enhancer is similar to that of enhancer. PSMA promoter/enhancer has the potential for use in targeted gene therapy of prostate adenocarcinoma.

[Seroprevalence of antibody against human metapneumovirus in Beijing].

OBJECTIVE: To understand the seroprevalence of antibody against the newly identified human metapneumovirus (hMPV) in Beijing. METHODS: The antigenic specificity of hMPV N protein cloned into vector pET30a and then expressed in E coli was verified by using SDS-PAGE and Western blotting in 116 serum specimens. The plasmid pET30a without insert was used as control. Totally 710 serum specimens collected from non-respiratory infection patients visited the Outpatient Departments of Children's Hospital affiliated to Capital Institute of Pediatrics and Xuanwu Hospital, Beijing from April 1996 to March 1997 were tested for specific IgG antibody against hMPV N protein. RESULTS: The bands with expected molecular weight showed only on the membranes transferred by the expressed hMPV N protein and incubated with rabbit hyperimmune serum against hMPV N protein polypeptides as well as the collected human sera, indicating the specificity of the expressed hMPV N protein. Out of 710 specimens tested, 17.2% (122/710) were positive for antibody to N protein. Antibody positive rate was the lowest in 2 to 6 months old infants (3.1%); the rate declined from 13.2% in newborns to 6.1% in 1 to 2 months old infants, then to 3.1% in the 2 to 6 months group, and sustained at about 3.0% from 6 months group to 30 years of age, then increased to 28.1% in 30 to 39-year-old adults, 32.3% in 40 to 49-year-old adults and to 38.5% in the group over age of 50 years. CONCLUSION: The expressed hMPV N protein is reliable when it was used as antigen for testing specific IgG antibody against hMPV in human sera. The high seroprevalence of antibody against hMPV N protein and early age antibody acquisition suggest that hMPV has been circulating in Beijing and the importance of the virus as pathogen should be further analyzed.

[Hysteretic characteristics of atrazine desorption from fluvo-aquic soil].

Kinetic batch experiments are carried out in order to investigate the effects of a wide range of atrazine initial concentrations and consecutive desorption time on the desorption characteristics of atrazine from fluvo-aquic soil. The results reveal that, atrazine concentrations in soil solution gradually decreased with desorption time increasing, whose relationship can be expressed with empirically exponential functions. After consecutive five steps (or 5 days), when atrazine initial concentrations increased from 50 microg x L(-1) to 2000 microg x L(-1), the average desorption percentages were 23.1%, 30.4%, 33.0%, 36.4% and 38.5%, respectively. The relationships between the amount of atrazine adsorbed by soil colloids and corresponding atrazine concentrations in soil solutions may be described through the traditional and time-dependent desorption isotherms. The obvious discrepancies in two families of both traditional and time-dependent desorption isotherms from adsorption isotherm, which retention time last for 168 hours before commencement of desorption, were indicative of hysteresis. Two sets of Freundlich parameters derived from both desorption isotherms can quantify the hysteresis between desorption and adsorption isotherms. However, hysteresis coefficient, omega, was applicable only for the traditional desorption isotherms, H and lamda applicable for both.

[Role of mitochondria in Tubeimoside I-mediated apoptosis in human cervical carcinoma HeLa cell line].

OBJECTIVE: To investigate the role of mitochondria in TBMS1-medited apoptosis of human cervical carcinoma HeLa cell line. METHOD: Mitochondrial transmembrane potentia (deltapsim) was assayed by flow cytometry. Apoptotic induction by TBMS1 was determined by gel electrophoresis of fragmented DNA. Cytochrome c (Cyt c) was detected by Western blotting. RESULT: The results showed that TBMS1 induced apoptosis in HeLa cells, that TBMS1 decreased deltapsim and facilitated Cyt c release, and that TBMS1 induced apoptosis of HeLa cells dose-and time-dependently in accordance with increase of cytosolic Cyt c. TBMS1 had direct effect on isolated rat mitochondria which induced a time- and dose-dependent release of Cyt c from mitochondria. The results also showed that CsA protected partly HeLa cells from the effect of TBMS1. CONCLUSION: The mitochondrial apoptosis pathway has effects on TBMS1 induced human cervical carcinoma HeLa cell line apoptosis.

[Comparison of different pacing sites of the right ventricle on left ventricular mechanical synchrony and systolic performance using tissue Doppler imaging].

OBJECTIVE: Right ventricular apical pacing may induce cardiac desynchronize and deteriorate left ventricular systolic performance. We hypothesized that right ventricular outflow tract (RVOT) pacing could produce better mechanical synchrony and left ventricular contraction. METHODS: We enrolled nine patients without structural heart disease who underwent electrophysiological studies. The pacing sites (right apex, low septum, free wall and septum of RVOT of the right ventricle) were defined with fluoroscopy and ECG. The atrioventricular sequential pacing was applied every 5 minutes in a random order at a rate of 120 bpm. Tissue Doppler imaging was carried out with GE VIVID 7 for off-line analysis at each pacing site. The global systolic contraction amplitude (GSCA) was calculated as the average shortening amplitude of all 16 segments of left ventricle. RESULTS: The GSCA during pacing was 5.76 mm +/- 0.66 mm at free wall of RVOT and 5.66 mm +/- 1.00 mm at septum of RVOT, respectively. The GSCA at both sites was significantly higher than that at apical pacing 4.82 mm +/- 0.94 mm (P < 0.05) or low septum pacing 4.82 mm +/- 1.06 mm (P < 0.05). Moreover, segmental displacement analysis showed that the longitudinal displacement of lateral, posterior, and inferior walls significantly decreased at apical pacing compared with RVOT pacing, although no difference could be demonstrated in anterior and septum walls. Accordingly, the curve of the myocardial displacement at apical or low septum pacing was M-shaped, and had a negative wave at the end of the diastole in lateral, posterior, and inferior walls. The tissue velocity during isovolumic contraction period was also higher than systolic tissue velocity in these walls. The phenomenon could seldom be seen at RVOT pacing. CONCLUSION: RVOT pacing in patients without structural heart disease is associated with more favorable immediate myocardial contraction and mechanical synchrony compared with right apical pacing or low septum pacing.
