[Sequence analysis of the HA1 regions of hemagglutinin genes of influenza viruses (H3N2) isolated from children in Beijing from 1998 - 2004].

OBJECTIVE: To characterize the HA1 regions of hemagglutinin gene of influenza viruses (H3N2) isolated from children in Beijing from 1998 - 2004. METHODS: The HA1 regions of hemagglutinin gene were amplified by RT-PCR from the viruses isolated and identified as A3 (H3N2) from clinical samples collected from infants and children during the peak seasons of influenza between 1998 and 2004. PCR products were sequenced or cloned into T-A vector and were analyzed after being sequenced. RESULTS: The HA1 regions of hemagglutinin genes amplified from those isolates were 987 bp in length, encoding a protein of 329 amino acids in length. The identities of nucleotides and amino acids among these H3N2 isolates in Beijing and vaccines strains from 1998 - 2004 were 95.5% - 100.0% and 93.0% - 100.0%, respectively. The homology of the HA1 regions were related to the date of virus isolation, meaning the homology was higher among those strains isolated in nearer dates than others. Seven potential N-linked glycosylation sites in the HA1 regions located at amino acid positions 8, 22, 38, 63, 126, 165 and 285 were conserved in all the viruses analyzed. Two sites at 122 and 133 were inserted in those virus isolated after 1997, and another site at 144 appeared in those isolated after 1999. More amino acid substitutions located in the five putative antigenic sites or receptor binding sites were found more in the isolates than the isolates from previous year. Phylogenetic analysis showed new branches appeared continuously during 1998 - 2004. The strains isolated during winter in 2004 belonged to different branches, suggesting the appearance of new variants. CONCLUSION: Amino acid substitutions continuously occurred in the HA1 regions of hemagglutinin genes in influenza virus (H3N2) isolated from children in Beijing from 1998 - 2004, which might have resulted in antigenic drift and led to the appearance of new variants.

[Knockdown of Hsp701A induces K562 cells apoptosis by RNA interference].

AIM: To investigate the apoptosis of K562 cells induced by RNA interference(RNAi) targeting Hsp701A. METHODS: Small interference RNA (siRNA) targeting Hsp701A was generated and its eukaryotic expression vectors was constructed and transected into K562 cells. Proliferation, apoptosis and cell cycle of the transected cells was respectively detected by MTT colorimetry and FCM. RESULTS: The eukaryotic expression vector of siRNA against Hsp701A was successfully constructed and transfected into K562 cells, which markedly decreased the expression of Hsp701A on both mRNA and protein level. K562 cells transfected with the vector exhibited slower proliferation, increased apoptosis and increased G(0)/G(1) arrest. CONCLUSION: The inhibition of Hsp701A gene by RNAi may provide a foundation for the development of novel therapeutic strategy for chronic granulocyte leukemia (CML).

[Study on the CD and ORD of the crystal NaClO3].

Chiral isotropy material should be obtained in order to manufacture chiral optical waveguide. In the present paper, a chiral crystal of NaClO3 was grown from water solution by standard rocking techniques. Crystal incising and optical polishing were doneand a transparent 10.9 mm x 8.2 mm x 4.7 mm crystal NaClO3 was obtained. Along the six directions perpendicular to the surface of the crystal NaClO3, its circular dichroism and UV spectra were collected, and at the same time its optical rotatory dispersion was measured by a self-fabricated ORD apparatus. It was approved by our experiment that the CD, UV spectrum and the optical rotatory dispersion of the crystal NaClO were isotropic, which is not the same as those of quartz.

[Determination of carotenoids in flue-cured tobacco leaves during its growth by reversed-phase high performance liquid chromatography].

A reversed-phase high performance liquid chromatographic (RP-HPLC) method for the determination of carotenoids in flue-cured tobacco leaves was developed. Carotenoids were extracted from flue-cured tobacco leaves by acetone-water (90:10, v/v) solution containing 0.1% butylated hydroxytoluene (BHT). Plant proteins were eliminated by adding 0.1 g Pb(Ac)2 and by centrifugation (10000 r/min) for 5 min at 4 degrees C. Lutein, beta-carotene, neoxanthin, violaxanthin and other plant pigments were separated on a reversed-phase C18 column (3.9 mm i.d. x 150 mm, 5 microm), with a mobile phase of (A) methanol-isopropyl alcohol (1:1, v/v) and (B) water using a gradient elution at a flow rate of 0.5 mL/min. The optimum elution gradient was as follows: 0-10 min, 70% A + 30% B; 10-17 min, 100% A; 17-30 min, 90% A + 10% B. The recoveries of carotenoids in flue-cured tobacco leaves were 91.77%-97.42%, and relative standard deviations were 3. 46%-0.98%. This method was applied to determine carotenoids in flue-cured tobacco leaves during its growth with satisfactory results.

[Detection of respiratory syncytial virus in nasopharyngeal aspirates of children by using real-time polymerase chain reaction].

OBJECTIVE: Human respiratory syncytial virus (hRSV) is the leading cause of acute upper and lower respiratory tract infections in infants and young children worldwide. Pediatric RSV disease claims more than 1 million lives annually. With the rapid development of specific anti-RSV agents and the spread of respiratory infections, RSV detection techniques with higher sensitivity, specificity and quicker performance are badly needed. This study was designed to develop a real-time polymerase chain reaction (PCR) for detection of RSV in nasopharyngeal aspirates. METHODS: (1) The TaqMan probe and primers of real-time PCR for RSV subgroup A and subgroup B detection were designed from the conserved region in N protein encoding gene, respectively. The sensitivity of real-time PCR was evaluated by using the virus with known amount of PFU. The specificity of real-time PCR for RSV detection was assessed by cross testing 10 isolates of strains A, 10 isolates of strains B, and by testing a variety of other respiratory viruses positive samples. (2) Sixty-one stored RSV positive respiratory samples and 103 nasopharyngeal aspirates were detected by real-time PCR, virus isolation, immunofluorescence assay (IFA), and nested-PCR. RESULTS: (1) The sensitivity of the real-time PCR developed in this study for RSV subgroup A detection was 5.25 pfu, and for subgroup B was 3.75 pfu, the same as that of nested-PCR. (2) No positive results were found in cross testing of other viruses positive specimens. (3) Twenty-seven out of 30 (90%) of RSV A stored samples and 27 out of 31 (87.1%) of RSV B stored samples were positive by the real-time PCR. (4) Thirty-five (34.0%) out of the 103 specimens were found RSV positive by real-time PCR (7 of them were subgroup A and 28 subgroup B); 31 (30.1%) specimens were positive by nested-PCR (6 of them were subgroup A and 25 subgroup B); 22 (21.4%) were found positive for RSV with IFA (5 of them were subgroup A and 17 subgroup B); RSV was isolated from 9 (8.7%) specimens (6 of them were subgroup A and 3 subgroup B). All the specimens found to be negative by real-time PCR were negative by rest of the methods used in this study. CONCLUSION: The real time PCR method developed in this project with the TaqMan probe and primers is sensitive and specific for detecting RSV subgroup A and B in nasopharyngeal aspirates.

[Clinical and laboratory study of a complex translocation t (6; 21; 8) (p22; q22; q22) in two patients with acute myeloid leukemia].

OBJECTIVE: To investigate the clinical and laboratory characteristics of a complex translocation t (6; 21; 8) (p22; q22; q22) in two patients with acute myeloid leukemia. METHODS: Bone marrow (BM) samples were collected at presentation, prepared by short-term (24 hours) unstimulated culture and R-binding, for conventional cytogenetic assay (CCA). The complex translocation was assayed by fluorescence in situ hybridization (FISH) with a dual-color AML1/ETO-specific probe. AML1/ETO chimeric transcript was detected by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: In both cases CCA demonstrated a complex translocation, t (6; 8; 21) (p22; q22; q22), which was confirmed by interphase and metaphase FISH and AML1/ETO fusion transcript was detected by RT-PCR. Both the two patients were diagnosed as AML-M(2), but with different immunophenotype and therapeutic outcome. CONCLUSION: The t (6; 21; 8) (p22; q22; q22) is a rare variant of complex translocation of t (8; 21) (q22; q22). More such cases are needed for elucidating its clinical features and prognosis.

[SSR polymorphism on the waxy gene locus and their relationship to amylose content in wheat].

Variation of one simple sequence repeat (SSR)(AT (AT)n AT) in the 3 end of the waxy gene was analyzed in samples of 32 cultivars, including different wheat cultivars in Shandong region and representing a wide distribution range of amylose content (AC) in cultivated wheat. In this study, we found two fragments were present in all cultivars, one of 204bp, the expected size, and one of 225-346bp. 204bp fragment located on the chromosome 7DS, while 225-346bp fragment located on the chromosome 7AS. That is, Wx-Ala gene and Wx-Dla gene present in all wheat lines, but the larger fragment showed length polymorphism. The result showed that the amylose level is highly positive correlated with the length of the polymorphic SSR. Amplified fragment is long in high AC cultivars and electrophoresis migration rate is small, while the fragment is short in low AC cultivars and electrophoresis migration rate is large. AC is highly diverse among different lines with the various SSR genotypes. Although the SSR variation did not seem to have obvious function in the synthesis of the starch synthase encoded by the waxy gene, the correlation between the SSR and AC level could be used as molecular marker for quality improvement in wheat programs.

[Surveillance for influenza A virus infections in infants and young children in Beijing, China, 2001-2005].

OBJECTIVE: To characterize the prevalence and antigenic drift of influenza A viruses isolated during the period from 2001 to 2005 in infants and young children in Beijing. METHODS: MDCK cell culture, indirect immunofluorescence assay (IFA) and hemagglutination inhibition (HI) assay were used to isolate and identify type A influenza viruses (H1N1 and H3N2) from clinical samples collected from outpatients and inpatients who visited the Affiliated Children's Hospital because of acute respiratory infections from Oct. 2001 to Aug. 2005. The HA1 regions of hemagglutinin gene of H3N2 isolates were amplified by using RT-PCR followed by sequencing. RESULTS: Out of 7338 clinical samples collected during this surveillance period, 347 (4.7%) were positive for influenza A viruses, including 48 (13.8%) of H1N1, 273 (78.7%) of H3N2 and 26 (7.5%) of subtype-unidentified influenza A viruses. Although there was a prevalence season of influenza A from October each year to April of next year during the 2001-2004 period, it was worth noting that a consecutive influenza A activity was detected from Aug. 2004 to Aug. 2005, when some influenza A viruses were detected even in summer. The positive rate of H3N2 was 14.2% in August, 2005, which was equal to that of the peak season of 2003-2004. H3N2 were predominant in most of the influenza seasons during the surveillance period, and H1N1 was detected only in the influenza seasons of the 2001-2002 and 2004-2005 along with H3N2. The positive rates for both H3N2 and H1N1 were higher in specimens from outpatients than those from inpatients. A total of 46.6% (110/236) of the H3N2 were detected from children younger than 2 years of age, and 14.0% (33/236) were from children older than 5 years, whereas, more H1N1 was found in children older than 5 years (48.0%, 12/31) than in those younger than 2 years (6.5%, 2/31) during a period from Nov. 2003 to Aug. 2005. Sequence analysis of the HA1 regions of hemagglutinin of H3N2 isolated in a series of years revealed amino acid changes in the HA1 domain of H3N2 isolates in the antigenic sites (A-E) each year. CONCLUSION: H3N2 and H1N1 prevailed in each influenza season during the surveillance period in Beijing, and H3N2 strains were predominant. The data from all-year around surveillance of influenza in Beijing indicate that continuous surveillance throughout a year and use of both antigenic and molecular analysis will be more helpful for early identification of any antigenic variants as well as prevention and control of influenza by promoting development of vaccines.

[Role of Bcl-2 and Bax protein contents and their gene expression in Al-induced neurons apoptosis].

OBJECTIVE: To study the role of Bcl-2 and Bax protein contents and their gene expression in Al-induced neurons apoptosis. METHODS: Neurons from 0 - 3 day rats were cultured and treated with different concentrations of AlCl(3 x 6) H2O. The cell apoptosis was observed by the TUNEL method and under the scan electron microscope. Bcl-2 and Bax protein contents were detected by the immunochemistry method while their gene expressions were measured by the RT-PCR method. RESULTS: (1) DNA fractions in the TUNEL method increased with the rising aluminum concentration. Blebbings and apoptosis bodies on the surface of the neurons were clearly observed under the scan electron microscope. (2) Bcl-2 protein contents and their gene expression decreased with the rising aluminum concentration (P < 0.01, r = -0.695; P < 0.05, r = -0.647), while Bax increased at the same time (P < 0.01, r = 0.676; P < 0.01, r = 0.794), the value of Bcl-2/Bax was related with the aluminum concentration (P < 0.01, r = -0.655; P < 0.01, r = -0.777). CONCLUSION: The aluminum may induce neurons apoptosis. Bcl-2 and Bax protein contents and their gene expression may play an important role in Al-induced apoptosis.

[Transfection of dominant negative MyD88 decreases IL-8 production in bacteria-infected airway epithelial cells].

Interleukin-8 (IL-8) is an important activator and chemoattratant of neutrophils and has been implicated in airway inflammatory diseases. To explore the new gene therapeutic strategies for airway inflammation, plasmid expressing dominant negative myeloid differentiation protein (MyD88 DN) was constructed and transfected into human airway epithelial cell lines A549 and SPC-A-I. The cells were challenged with M. tuberculosis, P. aeruginosa or K. pneumoniae and the release of IL-8 was measured using ELISA. The results showed that the supernatants of M. tuberculosis and R. aeruginosa enhanced IL-8 release from the epithelial cells; and transfection of MyD88 DN diminished this effect. MyD88 DN also reduced IL-8 release from cells induced by live bacteria of P. aeruginosa or K. pneumoniae. These data suggest that MyD88 could be used as a target gene in the gene therapy of airway inflammation.

[Effects of chlorine ion and EDTA on cadmium availability to plants].

In a water culture experiment with rice and rape, different concentrations of chloride or EDTA were added to study the effects of chlorine ion and EDTA complexation on the uptake of cadmium by the plants. The results showed that the cadmium concentration in the shoots and roots of test plants decreased with increasing concentration of chloride or EDTA. With the increase of Cl- concentration, the cadmium content in rice shoot and root decreased from 212.2 mg x kg (-1) and 345. 1 mg x kg - to 34.1 mg x kg(-1) and 209.1 mg x kg(-1) , respectively. EDTA addition decreased the cadmium content in rice shoot and root from 212.2 mg x kg (-1) and 345.2 mg x kg(-1) to 50.0 mg x kg(-1) and 4.2 mg x kg(-1) , and that in rape shoot and root from 86.7 mg x kg(-1) and 149. 2 mg x kg(-1) to 22.2 mg x kg (-1) and 12. 3 mg x kg(-1 ), respectively. EDTA also affected the cadmium translocation from root to shoot. The complexation of Cl- or EDTA with cadmium reduced the uptake of Cd by plants, and the effect of EDTA was more obvious than that of Cl -.

[Histopathological changes post cryoablation in canine myocardium].

OBJECTIVE: The purpose of this study was to observe histopathological changes post cryoablation in canine myocardium, to characterize the specific ablation lesion post cryoablation. METHODS: Cryothermal ablation was applied on myocardium (both epicardium and endocardium) of 14 mongrel dogs with different ablation parameters (-25 degrees C x 4 min, -50 degrees C x 4 min, -75 degrees C x 4 min, -75 degrees C x 2 min, -75 degrees C x 6 min, -75 degrees C x 8 min). Lesion dimensions and histopathologic changes were observed. RESULTS: The discrete, sharply delimited lesions were detected in cryoablated myocardium. Histologically, cryoablation in all temperatures studied induced heterogeneous necrosis of the myocardium. Lesion dimensions are related to freezing time and temperature. CONCLUSION: Cryoablation is a feasible and preferably choice for clinical application due to its controllable myocardium lesions.

Detection of human betaV-tubulin expression in epithelial cancer cell lines by tubulin proteomics.

Tubulin, the constitutive protein of microtubules, is a heterodimeric protein with an alpha and beta subunit, encoded in vertebrates by six and seven different genes, respectively. Each tubulin isotype can be identified by its divergent C-terminal sequence. Nevertheless, two groups of beta-tubulin isotypes can be distinguished by sequence alignment; one includes betaI-, betaII-, betaIVa-, and betaIVb-tubulin, and the other includes betaIII-, betaV-, and betaVI-tubulin. betaIII-tubulin overexpression has been associated with microtubule destabilization and resistance to Taxol. Recent data indicate that mouse betaV-tubulin overexpression in CHO cells results in profound microtubule disorganization and dependence of cells on Taxol for growth. Mouse and human betaV-tubulin sequences display several differences, such as their respective extreme C-terminus, suggesting that they may have different effects on microtubule stability and different affinities for drugs. When high-resolution isoelectric focusing, in-gel CNBr cleavage, and mass spectrometry were combined, we detected for the first time the betaV-tubulin protein in human cell lines and found that it was highly expressed in Hey, an epithelial ovarian cancer cell line. Our data confirm that human and rodent betaV-tubulins are distinct and indicate that, regardless of species, betaIII- and betaV-tubulin may be expressed in a complementary pattern at the protein level. Therefore, both betaIII- and betaV-tubulin expression levels should be systematically determined to assess the role of differential tubulin isotype expression in the response of tumors to drugs targeting microtubules.

Enhanced immunogenicity of microencapsulated multiepitope DNA vaccine encoding T and B cell epitopes of foot-and-mouth disease virus in mice.

The role of poly(D,L-lactic-co-glycolic acid, PLGA) microparticles on enhancing immune responses of multiepitope DNA vaccines was investigated in vitro and in vivo. pcDNA-SG encoding T and B cell epitopes of foot-and-mouth disease virus (FMDV) was encapsulated into PLGA microparticles. PLGA microparticles could protect themselves from nuclease degradation in vitro. PLGA-pcDNA-SG microparticles could be uptaken by cells and expressed His-tagged SG immunogen in vitro and in vivo. A prolonged expression and presentation of SG immunogen were observed by confocal laser scanning microscopy in the lymphocytes from the mice incubated with PLGA-pcDNA-SG microparticles, compared with the mice immunized with naked pcDNA-SG. PLGA-pcDNA-SG microparticles displayed a significant stronger immunogenicity than naked DNA vaccines with a higher titer of virus-specific antibody, elevated IFN-gamma production and enhanced lymphocyte proliferation. PLGA-DNA microparticle could elicit augmented humoral and cellular responses with reduced amounts and times of immunization.

Quantitative structure-toxicity relationships of organophosphorous pesticides to fish (Cyprinus carpio).

This study was conducted to determine the relationships between 1381 chemical and structural parameters of 43 organophosphorus pesticides (OPs) and their toxicity to fish, Cyprinus carpio, using ChemOffice 8.03 and Dragon 2.1. By multivariate linear regression and intervariable regression analyses, various equations have been derived to calculate the lethal toxicity value, LC(50), for 43 OPs found in fish with different levels of toxicity. Results show that for all selected OPs, especially those of low toxic OPs (LC(50)< 2.5 mM), the equation, LC(50) = 56.259 - 13.071 lg K(ow)+17.510 MATS8P-17.455 Mor24u - 0.085 MW + 1.706 (lg K(ow))(2) + 2.306 (Mor14e)(2) + 6.849 Mor20 m (n = 43, F = 36.815, r = 0.942, r(adj)(2) = 0.862, SE = 2.899, p < 10(-6)), could account for 86.2% of the variability of the toxic effect. The steric and electronic characteristics and the hydrophobicity of OPs, in particular, are among the most important parameters determining the toxicity of OPs to fish. For the OPs with high toxicity, different structural parameters were introduced into the following two equations: LC(50)=3.795-1.195 (H1p)(2)-0.037 U-2.225 MATS3v-19.593 Tcon (n = 16, F = 56.820, r = 0.977, r(adj)(2) = 0.937, SE = 0.143, p < 10(-6)), where LC(50) is less than 2.5 mM, and LC(50) = 0.341-0.561 (HOMA)(2) + 0.231 HOMA (n = 3,r(adj)(2) = 1), where LC(50) is less than 0.3 mM. These results suggest that chemical and structural parameters could be useful in modeling chemical reactivity within homologous series of OP compounds and elucidating possible mechanisms associated with different levels of toxicity to fish.

[The changing etiology and therapeutic situation of atherosclerotic renal artery stenosis].

OBJECTIVE: To determine whether the etiology of renal artery stenosis changed. To investigate the clinical characteristics and current therapeutic situation of atherosclerotic renal artery stenosis (ARAS). METHODS: Retrospective analysis was performed in 144 cases of renal artery stenosis diagnosed by angiography (> or = 50% luminal narrowing) from 1979 to 2003. Eighty-seven of them were diagnosed as ARAS by special characteristics of angiography and clinical features. The clinical data of ARAS was compared with RAS caused by other diseases. RESULTS: The proportion of ARAS in all RAS was 60.4%. The proportion of ARAS was 28.9% during the period of 1980 to 1990 and 71.7% from the year of 1991 to 2003, respectively. The prevalence of hypertension was 90.8% among ARAS patients. Newly-progressive hypertension was more common among ARAS patients compared with RAS patients with other causes (37.9% vs 21.0%, P = 0.044). Renal dysfunction were more commonly seen among ARAS than non-ARAS patients in those with the same degree of stenosis (11.2% vs 6.9% for stenosis 50%-74%, P = 0.038; 35.8% vs 15.4% for stenosis > or = 75%, P = 0.046). Among ARAS cases, 60.9% were managed with antihypertensive agents only, 18.8% were managed by angioplasty with or without stents. After 1990, the improvement rate of blood pressure rose significantly but not for renal function. The improvement rate of blood pressure by angioplasty was similar with that by medication (84.1% vs 92.6% for stenosis 50%-74%, P = 0.39; 76.6% vs 80.1% for stenosis > or = 75%, P = 0.61). And the improvement or stable rate of renal function was not different (85.4% vs 85.7% for stenosis 50%-74%, P = 0.91; 77.8% vs 76.9% for stenosis > or = 75%, P = 0.83). CONCLUSION: ARAS is the predominant cause of RAS nowadays in stead of Takayasu arteritis. Medication is the mostly used management for ARAS, though more angioplasty with or without stents appear. The effect of medication for ARAS has improved since 1990. Most cases of ARAS could achieve ideal blood pressure control and similar renal function protection with either rational medication or angioplasty.
