[The effects of strontium in drinking water on growth and development of rat bone].

Effects of strontium at a high level in drinking water on growth and development of rat bone were studied. The results showed that Sr2+ concentration from 5 to 500 mg/L in drinking water could increase the contents of strontium in blood serum, urine, femur, mixilla and tooth in Wistar rats exposed to Sr2+ for 12 weeks with an obvious dose-response relationship. In addition, strontium at over 50 mg/L could decrease the contents of calcium in bone, increase the contents of calcium in tooth and bone density, and decrease the levels of calcium in blood serum except female rats at the 12th week. Effects of Sr2+ on body weight, body length, AKP activity of serum, calcium content of urine and breaking load of bended femur for rats were not found. However, there are differences in the effects of strontium on growth and development of bone between male and female rats. At the 12th week the content of calcium in blood serum decreased in male rats but increased in female rats in exposed groups. At the 4th and 8th weeks, urine Hop/Cr in male rats increased but it remained normal level in female rats. Sr2+ increased the bone density of mixilla in male rats but it did not increase that of femur in female rats. It is suggested that such changes may be a result of the differences in endocritic regulation and metabolic process between two sexes.

[Dynamic study on the mutagenic activity of organic concentration of water from one branch port of Huangpu River].

From 1991 to 1994, we studied the mutagenicity of drinking water in Shanghai by seasons by Ames test. The results indicated that the mean value of mutagenic activity of organic concentration on TA98 is 162 revertants per liter. Studies on the four branch ports of Huangpu river showed that they contributed 74% of the total mutagenicity, which accounted for the predominant fraction of the mutagenic activity of Shanghai drinking water. The mutagenicity of Hongkou branch was the highest, accounting for 1/3 of the total mutagenic activity of the 4 branches. Studies showed that the mutagenic activity of organic concentration of Hongkou branch water was declining year by year, and factors affecting the mutagenic activity were analysed.

[A gas chromatographic study on the characters of adsorbents in the enrichment of organic compounds from air samples].

A gas chromatographic method for the measurement of enriching characters of adsorbents used in preconcentration of organic compounds from air samples was studied. The character parameters suggested in this study include adsorption parameters (breakthrough volume or maximum sample volume) and desorption ones (minimum flashing volume and the lowest desorption temperature). The parameters of some low-molecular-mass compounds such as alcohols, saturated hydrocarbons, aromatics, esters and halohydrocarbons on GDX-301 were measured by using elution GC method. The maximum sample volumes of all the 20 compounds (with exception of methanol) got from these experiments are not less than 0.8 L at 35 degrees C for sampling tube (filled with 2 g GDX-301) and 0.3 L at 0 degrees C for focus tube (with 0.14 g GDX-301). The corresponding minimum flashing volumes of less than 177 mL and 13 mL respectively are needed to complete desorption at 180 degrees C, and the lowest desorption temperatures of not over 180 degrees C and 157 degrees C at flashing volume of 180 mL and 30 mL are lower than the maximum allowable operating temperature (250 degrees C) of GDX-301. The results showed that GDX-301 is a good adsorbent for enrichment of the organic compounds mentioned above. The following operating conditions are recommended: using the above sampling tube and focus tube, actual sample volume < or = 0.8 L, adsorption temperature < or = 35 degrees C and focus temperature 0 degrees C, desorption for 6 min from sampling tube and 1 min from focus tube at 180 degrees C with flashing rate 30 mL/min. Good correlations between the parameters and boiling point of the investigated compounds were found. It can be used to estimate the parameters of other organic compounds not measured in this study.

[Simplex optimization of the automatic gas chromatographic instrument].

A simplex optimization method developed for the automatic gas chromatography is described in this paper. The instrument was developed by ourself based on a gas chromatograph and an auto-sampler, which are controlled by a computer through several assisting circuit boards. A series of chromatographic function SCRF is used in this method for searching the best column temperature for gas chromatographic separation of a given sample. A real sample is used for probation of this method and the results are discussed.

[Influence of reperfusion following ischemia on microvessels and microcirculation of skeletal muscle].

In order to study the influence of reperfusion following ischemia on microvesseles and microcirculation of skeletal muscle, unilateral hindlimbs of 16 rabbits were subjected to normothermic ischemia for 2 and 5 hours by tourniquet. After release of the tourniquet, microcirculation of the peritenon on dorsum of the foot was observed for 1 hours by intravital microscope. At 1 hour and 72 hours following reperfusion, the anterior tibia muscle biopsiy were taken and the specimens were subjected to light and electron microscopic examinations. It was found that after release of the tourniquet, in the limbs undergone 2 hours ischemia, there was immediate and well distributed reflow in the microvesseles of peritenon though a few aggregates of red cells and increase in the number of adherent leukocytes occured in some venules, and the microvesseles of the skeletal muscle only showed signs of minimal injury, the muscle fibers could survive in the limbs undergone 5 hours of ischemia, however, there was serious disturbance of microcirculation in the peritenon, which was characterized by "no reflow" in most area and there was significant increase in the number of leukocytes adherent to venular endothelium, and the microvesseles of the skeletal muscle showed signs of severe injury, including remarkable swelling of the endothelial cell, disruption of the basement membrane and interstitial edema, and finally, most of the muscle fibers had necrosis occured. The results demonstrated that reperfusion following ischimia might result in microvascular injury and microcirculation disorder in the ischemic area. The degree of the injury and disorder depended on the duration of ischemic period, and was an important factor which determined the fate of the parenchymal cell.

Inhibition of melanoma-associated antigen expression and ecotropic retrovirus production in B16BL6 melanoma cells transfected with major histocompatibility complex class I genes.

We have previously reported that expression of the melanoma-associated antigen (MAA) recognized by MM2-9B6 monoclonal antibody in B16 melanoma was closely associated with C-type ecotropic retroviral particle production. Our present data show that this MAA is encoded by the env gene of an ecotropic retrovirus produced by B16 melanoma cells. Transfection of H-2Kb or H-2Kd genes into two individual clones isolated from B16BL6 melanoma, BL6-8 (H-2Kb-, H-2Db+) and BL6-2 (H-2Kb-, H-2Db-), resulted in a loss of MAA expression. Electron immunohistochemical analysis of melanoma cells and reverse transcriptase assay revealed that the loss of MAA expression in the H-2K gene-transfected cells paralleled the elimination of retroviral particles. In contrast, expression of the endogenous H-2Db gene or transfection with the H-2Dd or H-2Ld gene had no effect on MAA expression or retrovirus production. Northern blot analysis showed equivalent retroviral messages in retrovirus-producing and -nonproducing BL6 melanoma clones. Southern blot analysis revealed that H-2Kb-negative BL6 melanoma cells contain at least four different ecotropic retroviruses with different insertion sites that somatically emerged during malignant transformation or progression. Restriction enzyme analysis showed various changes in proviral DNAs from the H-2Kb- and H2Kd-transfected cells that failed to produce retroviral particles. The observed alterations in the patterns of PstI- and HindIII-digested proviral DNA were found to be due to the appearance of PstI and loss of HindIII restriction sites in the pol region as a result of several nucleotide substitutions. Thus, BL6 melanoma cells produce melanoma-specific ecotropic murine leukemia viruses that encode serologically detectable cell surface MAA. The transfection of BL6 melanoma cells with H-2Kb or H2Kd genes but not H-2Dd or H-2Ld genes resulted in a loss of MAA expression that was attributed to the changes in proviral DNA and loss of melanoma-specific ecotropic retrovirus particle production.

Oxidation of phenols, anilines, and benzenethiols by fungal laccases: correlation between activity and redox potentials as well as halide inhibition.

A comparative study has been performed with several fungal laccases for the oxidation of a series of phenols, anilines, and benzenethiols and for the inhibition by halides. The observed K(m) and kcat were correlated to the structure of substrate. The change in log (kcat/K(m)) was found to be proportional to the one-electron redox potential difference between laccase's type 1 copper site and substrate. This correlation indicated that the first electron transfer from substrate to laccase was governed by the "outersphere" mechanism. Compared to the electronic factor, the steric effect of small o-substituents (such as methyl and methoxy groups) was found to be unimportant. The depth of the laccase's type 1 copper site was estimated as approximately 10 A by comparing the steric effect among five 2-methoxyphenols whose 4-substituents ranged from 0.1 to 14 kDa in mass. The observed inhibition potency order of F- > Cl- > Br- was attributed to limited accessibility of laccase's type 2/type 3 trinuclear copper cluster site. Although the enzymes studied have homologous primary sequences and predicted similar backbone structures, the difference exhibited by each enzyme (in interacting with individual substrate or inhibitor) suggested the structural variation in their functional domains.

Conformational trapping in a membrane environment: a regulatory mechanism for protein activity?

Functional regulation of proteins is central to living organisms. Here it is shown that a nonfunctional conformational state of a polypeptide can be kinetically trapped in a lipid bilayer environment. This state is a metastable structure that is stable for weeks just above the phase transition temperature of the lipid. When the samples are incubated for several days at 68 degrees C, 50% of the trapped conformation converts to the minimum-energy functional state. This result suggests the possibility that another mechanism for functional regulation of protein activity may be available for membrane proteins: that cells may insert proteins into membranes in inactive states pending the biological demand for protein function.

A study of a series of recombinant fungal laccases and bilirubin oxidase that exhibit significant differences in redox potential, substrate specificity, and stability.

A series of fungal laccases (Polyporus pinsitus, Rhizoctonia solani, Myceliophthora thermophila, Scytalidium thermophilum) and one bilirubin oxidase (Myrothecium verrucaria) have been studied to determine their redox potential, specificity, and stability. Polyporus and Rhizoctonia laccases possess potentials near 0.7-0.8 V (vs. NHE), while other oxidases have potentials near 0.5 V. It is observed that higher redox potential correlates with higher activity. By EPR, no significant change in the geometry of type 1 copper (II) site is observed over this series. At the optimal pH, the two substrates studied, 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulfonic acid) and syringaldazine, show Km values ranging form 10 to 120 and from 1 to 45 microM; and kcat values ranging from 50 to 16 000 and 200 to 3000 per min, respectively. The enzymes are more stable in the neutral-alkaline pH range. The thermal stability is in the order of bilirubin oxidase equivalent to Myceliophthora laccase equivalent to Scytalidium laccase > Polyporus laccase > Rhizoctonia laccase. Based on these results and the sequence alignments made against Zucchini ascorbate oxidase it is speculated that structural differences in the substrate-activation site (a 'blue', type 1 copper center) control the redox potential range as well as substrate specificity, and the cystine content contributes to stability.

Infants' metaphysics: the case of numerical identity.

Adults conceptualize the world in terms of enduring physical objects. Sortal concepts provide conditions of individuation (establishing the boundaries of objects) and numerical identity (establishing whether an object is the same one as one encountered at some other time). In the adult conceptual system, there are two roughly hierarchical levels of object sortals. Most general is the sortal bounded physical object itself, for which spatiotemporal properties provide the criteria for individuation and identity. More specific sortals, such as dog or car, rely on additional types of properties to provide criteria for individuation and identity. We conjecture that young infants might represent only the general sortal, object, and construct more specific sortals later (the Object-first Hypothesis). This is closely related to Bower's (1974) conjecture that infants use spatiotemporal information to trace identity before they use property information. Five studies using the visual habituation paradigm were conducted to address the Object-first Hypothesis. In these studies, 10-month-old infants were able to use spatiotemporal information but failed to use property/kind information to set up representations of numerically distinct individuals, thus providing empirical evidence for the Object-first Hypothesis. Finally, infants succeed at object individuation in terms of more specific sortals by 12 months. The relation between success at our task and early noun comprehension is discussed.

The identification and characterization of four laccases from the plant pathogenic fungus Rhizoctonia solani.

Four distinct laccase genes, lcc1, lcc2, lcc3 and lcc4, have been identified in the fungus Rhizoctonia solani. Both cDNA and genomic copies of these genes were isolated and characterized. Hybridization analyses indicate that each of the four laccase genes is present in a single copy in the genome. The R. solani laccases can be divided into two groups based on their protein size, intron/exon organization, and transcriptional regulation. Three of these enzymes have been expressed in the fungus Aspergillus oryzae. Two of the recombinant laccases, r-lcc1 and r-lcc4, as well as the native lcc4 enzyme were purified and characterized. The purified proteins are homodimeric, comprised of two subunits of approximately 66kDa for lcc4 and 50-100kDa for the recombinant lcc1 protein. These laccases have spectral properties that are consistent with other blue copper proteins. With syringaldazine as a substrate, lcc4 has optimal activity at pH7, whereas lcc1 has optimal activity at pH6.

Preoperative evaluation of macrophage colony-stimulating factor levels in patients with endometrial cancer.

OBJECTIVE: Our purpose was to examine the relationship between preoperative serum levels of macrophage colony-stimulating factor, alone and in combination with CA 125, and the presence of prognostic clinicopathologic factors and subclinical metastases in women with endometrial cancer. STUDY DESIGN: Ninety-two women who underwent primary exploration for endometrial adenocarcinoma had preoperative serum samples evaluated for macrophage colony-stimulating factor and CA 125 levels. Multivariate analysis was used to determine the associations of surgicopathologic findings with macrophage colony-stimulating factor and CA 125 levels. Logistic regression analysis was used to identify factors associated with the risk of extrauterine disease. The association of macrophage colony-stimulating factor and CA 125 levels with stage, grade, and depth of myometrial invasion and histologic characteristics were analyzed with Fisher's two-tailed exact test. RESULTS: Elevated levels of macrophage colony-stimulating factor were not associated with depth of myometrial invasion, histologic grade, or histologic cell type; however, advanced stage (p = 0.02) and the presence of lymph node metastases (p = 0.04) were associated with elevated levels. Sensitivity and specificity of macrophage colony-stimulating factor for predicting extrauterine disease were 42% and 89%, respectively. If either an elevated macrophage colony-stimulating factor or an elevated CA 125 level was used to predict extrauterine disease, the sensitivity was increased to 67% but the specificity was decreased to 78%. Macrophage colony-stimulating factor elevations predicted lymph node metastases with a sensitivity of 50% and a specificity of 86%. A multivariate regression model showed CA 125 to be the most significant predictor of extrauterine disease; macrophage colony-stimulating factor also contributed prognostic information (p = 0.02). The sensitivity and specificity of the multivariate model for predicting extrauterine disease were 75% and 73%, respectively. CONCLUSION: Macrophage colony-stimulating factor and CA 125 are neither sensitive nor specific enough to be used as predictors of the presence or absence of extrauterine disease in patients with endometrial cancer.

Remembrance of odors past: enhancement by central facilitation of AMPA receptors.

Pharmacological facilitation of the alpha-amino-3-hydroxy-5-methyl-4-isoxazole (AMPA)-type glutamate receptor has recently been demonstrated to enhance synaptic responses, promote long-term potentiation (LTP) induction in freely moving rats, and facilitate learning and retention of information. The present study verifies and extends the behavioral action of allosteric AMPA receptor modulation by showing that the benzoyl-piperidine compound BDP-12 promotes retention of olfactory and transient spatial memory in a dose-dependent fashion; is only effective when given before but not after training, consistent with the hypothesis that glutamatergic facilitation enhances information encoding by means of action on the machinery involved in LTP induction; and, following suboptimal training in a paradigm of enduring memory, prolongs the ability of rats to retain odors by extending the decay of weak memory traces.

Serum levels of GM-CSF are elevated in patients with thrombocytopenia.

Serum levels of GM-CSF, IL-3 and IL-6 were measured in patients with immune thrombocytopenia (ITP), non-immune thrombocytopenia (NIT), autoimmune haemolytic anaemia (AIHA) and neutropenia. 8/10 children with ITP had elevated serum levels of GM-CSF (mean 18.4 pg/ml) while thrombocytopenic, but only two had detectable levels (mean 4.5 pg/ml) after normalization of the platelet count. In patients with NIT a significant inverse correlation between platelet count and serum levels of GM-CSF was observed. IL-3 and IL-6 levels were not significantly elevated in thrombocytopenic patients and only two of the nine patients with either AIHA or neutropenia had detectable levels of GM-CSF. Thus, GM-CSF may play a role in the response of severe thrombocytopenia.

Fine-structure mapping of meiosis-specific double-strand DNA breaks at a recombination hotspot associated with an insertion of telomeric sequences upstream of the HIS4 locus in yeast.

Meiotic recombination in Saccharomyces cerevisiae is initiated by double-strand DNA breaks (DSBs). Using two approaches, we mapped the position of DSBs associated with a recombination hotspot created by insertion of telomeric sequences into the region upstream of HIS4. We found that the breaks have no obvious sequence specificity and localize to a region of approximately 50 bp adjacent to the telomeric insertion. By mapping the breaks and by studies of the exonuclease III sensitivity of the broken ends, we conclude that most of the broken DNA molecules have blunt ends with 3'-hydroxyl groups.
