[Simulation of a decoupling generalized predictive control algorithm for temperature-humidity system].

A decoupling generalized predictive control algorithm was presented in this paper. This algorithm was proposed to control the temperature-humidity system of a practical air-conditioning environmental chamber. By introducing absolute humidity as an intermediate variable, temperature and humidity of the system can be decoupled. Then the two decoupled systems are controlled by generalized predictive control (GPC) algorithm at different sample rates. Many simulation experiments demonstrated the effectiveness of the algorithm. Besides, decoupling of temperature and humidity will be extremely helpful to the application of GPC on the real system. This is also the effective method to control the temperature and humidity system.

Antiganglioside antibodies in cerebrospinal fluid of children with neuropsychiatric lupus erythematosus.

OBJECTIVE: To determine antiganglioside antibodies (AGA) in cerebrospinal fluid (CSF) of children with neuropsychiatric lupus erythematosus (NPLE), and to investigate their association with clinical manifestations and other laboratory tests. METHODS: An enzyme linked immunoabsorbent assay was used to detect AGA in 22 CSF samples obtained from 18 children with systemic lupus erythematosus (SLE). These patients were further classified into clinical active NPLE (9 patients) and SLE (9 patients) groups. Indirect immunofluorescent assay was used for determination of CSF antinucleus antibodies (ANA). RESULTS: AGA could be found in CSF of all the 9 active NPLE children who had neuropsychiatric manifestations within 6 months before or after AGA was tested. A clear association was observed between AGA and brain CT scan. No detectable ANA was found in CSF samples even though high titer of ANA was present in corresponding serum samples of SLE patients. CONCLUSION: Detection of AGAs in CSF, more sensitive and specific than routine CSF examination and EEG, and positively associated with brain CT scan, is a simple and practical test for early judgment of childhood NPLE.

Epidemiological survey on clonorchiasis sinensis in Yangxin County of Hubei Province of PR China.

An epidemiological survey of clonorchiasis was conducted at Panqiao township of Yangxin County of Hubei Province from June to November, 1993. The positive rate of cercaria in the body of intermediate hosts, Parafossarulus stratulus and Alocinma longicornis was 12.25% and 3.84% respectively. Positive rates of metacercariae in the bodies of Pseudonaphona parva was 48.15%, Ctenopharyngodon idellus 17.24% and Hypophthalmichthys nobilis 18.18%. Positive rate of eggs in the feces of cats was 36.36% and pigs 16.67%. It has been confirmed that there is a natural focus of clonorchiasis sinensis at Yangxin County of Hubei Province. A total population of 6,865 in 20 sites of 10 production brigades of Panqiao township was surveyed for infection with Clonorchis sinensis. The average infection rate in the local residents was 5.80%. Male had a higher infection rate than female. The infected persons were mainly peasants and school girls and boys. Most of the infected persons had light infections (I0) without a serious clinical manifestations.

[Expression of nm23/NDPK, integrin, type IV collagenase and uPA in transplanted tumors with various metastatic potential in nude mice and surgical specimens].

OBJECTIVES: To observe the relationship between the metastasis of human lung carcinoma and the expressions of nm23/NDPK, integrins, type IV collagenase and uPA. METHODS: By ABC immunohistochemical staining, we observed the expressions of nm23/NDPK, integrins, type IV collagenase and uPA in four strains of subcutaneous transplanted tumors and 87 surgical specimens for human lung carcinoma. The metastatic potential of four xenografts varied from low to high among PLA-801C, Anip973, PLA-801D, PLA-801DL. The effects of beta 1 antibody and the RGD peptide on the PLA-801DL xenograft were studied in the nude mice. RESULTS: High expression of nm23/NDPK was observed in the four xenografts and specimens from the 87 patients. No association was found between expression of nm23/NDPK and lymph node metastasis. Low expressions of beta 1 subfamily integrins were observed in the four xenografts. beta 1 subunit antibody and RGD peptide had no effect on metastasis of PLA-801DL xenograft. The expressions of type IV collagenase were positively correlated with their metastatic capacity in the four xenografts and surgical specimens from the 87 patients. Anip973 and PLA-801D showed positive staining to uPA, while PLA-801DL and PLA-801C were negative. CONCLUSIONS: The expression of type IV collagenase is positively correlated with the metastatic capacity of lung carcinoma. Although high expression of nm23/NDPK, low expression of integrins, and various expression of uPA were observed, these were not correlated with metastatic capacity of lung carcinoma.

Liver injury model in mice induced by a cellular immunologic mechanism--delayed-type hypersensitivity-induced liver injury to picryl chloride and phenotype of effector cell.

Liver injury was induced in BALB/c mice by local delayed-type hypersensitivity (DTH) to picryl chloride (PC1). Distinct changes of biochemical parameters were observed including the elevation of serum alanine and aspartate aminotransferases, increase of liver lipid peroxides, as well as decrease of serum alkaline phosphatase. Damage was confirmed by histopathological findings such as hepatocellular necrosis, granulocyte infiltration, and fatty degeneration. The liver injury was passively transferred into naive syngeneic mice by infusing spleen cells from immune mice. The capacity of the splenocytes to induce liver injury in recipient mice was almost completely abolished by pretreatment of the cells with anti-Thy 1.2 or anti-CD4, but not anti-CD8 antibody. These findings suggest that the production of liver injury by a local DTH mechanism is possible and the subpopulation of T cells, Thy-1.2+, L3T4+, and Lyt-2- cells, is at least one of the effector cells that mediate the injury.

Preponderance of Fis-binding sites in the R6K gamma origin and the curious effect of the penicillin resistance marker on replication of this origin in the absence of Fis.

Fis protein is shown here to bind to 10 sites in the gamma origin of plasmid R6K. The Fis-binding sites overlap all the previously identified binding sites in the gamma origin for the plasmid-encoded pi initiator protein and three host-encoded proteins, DnaA, integration host factor, and RNA polymerase. However, the requirement of Fis for R6K replication depends on the use of copy-up pi-protein variants and, oddly, the antibiotic resistance marker on the plasmid. In Fis-deficient cells, copy-up pi variants cannot drive replication of R6K gamma-origin plasmids carrying the bla gene encoding resistance to penicillin (Penr) but can drive replication of plasmids with the same origin but carrying the chloramphenicol acetyltransferase gene encoding chloramphenicol resistance (Cmr). In contrast, R6K replication driven by wild-type pi is unaffected by the antibiotic resistance marker in the absence of Fis protein. Individually, none of these elements (copy-up pi, Fis deficiency, or drug markers) prevents R6K replication. The replication defect is not caused by penicillin in the medium or runaway replication and is unaffected by the orientation of the bla gene relative to the origin. Replication remains inhibited when part of the bla coding segment is deleted but the bla promoter is left intact. However, replication is restored by insertion of transcriptional terminators on either side of the gamma origin, suggesting that excess transcription from the bla gene may inactivate replication driven by pi copy-up mutants in the absence of Fis. This study suggests that vector sequences such as drug markers may not be inconsequential in replication studies, as is generally assumed.

Novel, ligation-dependent PCR assay for detection of hepatitis C in serum.

A simple, sensitive, and specific ligation-dependent PCR (LD-PCR) method for the detection of hepatitis C virus (HCV) RNA in serum is described. The assay uses two DNA capture probes for RNA isolation and two DNA hemiprobes for subsequent PCR. Each capture probe has a 3' sequence complementary to the conserved 5' untranslated region of HCV RNA and a biotin moiety at the 5' end capable of interacting with streptavidin-coated paramagnetic beads. Each hemiprobe contains a sequence complementary to the 5' untranslated region in juxtaposition to one another and a common sequence for PCR primer binding. In guanidinium thiocyanate solutions, the capture probes and the hemiprobes form a hybrid with their target, and the hybrid can be isolated from serum by the binding of the capture probes to the paramagnetic beads in the presence of a magnetic field. The hemiprobes can then be linked to each other by incubation with T4 DNA ligase to form a full probe that serves as a template for a PCR. When serial 10-fold dilutions of synthetic HCV RNA (10(7) to 10 molecules) were tested, there was a good correlation between the amount of PCR product and the initial number of RNA molecules, with a sensitivity of 100 HCV RNA molecules per reaction. Twenty-four specimens that had been tested by either a branched DNA probe (bDNA) assay (13 specimens) or a reverse transcription PCR (RT-PCR) assay (11 specimens) were also analyzed by LD-PCR. The results showed a good correlation among LD-PCR, RT-PCR, and the bDNA assay. However, both LD-PCR and RT-PCR were more sensitive than the bDNA assay when the HCV titer was low.

Cell cycle arrest in G0/G1 phase by contact inhibition and TGF-beta 1 in mink Mv1Lu lung epithelial cells.

We postulated that contact inhibition and transforming growth factor (TGF)-beta 1 may target the same molecules to negatively regulate the Mv1Lu cell cycle in G0/G1. Both contact inhibition and TGF-beta 1 suppressed the expression of a 45-kDa protein (p45); cyclins D2 and B1; cyclin-dependent protein kinase (Cdk)-4, Cdc-2, and Cdc-2-associated activity; and the phosphorylation of retinoblastoma tumor-suppressor protein (pRb) but did not affect the expression of cyclins D1, E, and A or the expression of Cdk-2 and Cdk-5. Expression of p45 reappeared 12 h after release from contact inhibition and 6-8 h after release from TGF-beta 1, while TGF-beta 1 prevented release from contact inhibition and maintained suppression of both p45 and cyclin D2. Additionally, cyclin D2 phosphorylation and its associated kinase activity were strongly inhibited by contact inhibition and TGF-beta 1. Thus suppression of p45, cyclin D2/Cdk-4, and cyclin B1/Cdc-2 expression and/or activities is targeted both by contact inhibition and by TGF-beta 1 and may define common mechanisms through which these negative growth signals are integrated.

Time reversal focusing applied to lithotripsy.

Time reversal of ultrasonic field allows a very efficient approach to focusing pulsed ultrasonic waves through lossless inhomogeneous media. Time reversal mirrors (TRM) are made of large transducer arrays, allowing the incident acoustic field to be sampled, time reversed and re-emitted. Time reversal processing permits a choice of any temporal window to be time reversed, allowing operation in an iterative mode. In multitarget media, this process converges on the most reflective target, i.e., the dominant scatterer. In this paper, the time reversal process is applied to track, in real time, a moving gall bladder or kidney stone embedded in its surrounding medium. We investigate the feasibility of a piezoelectric shock wave generator in which the focal zone is moved electronically to track the stone during a lithotripsy treatment. We show that TRM allows us to obtain sharp focusing on one bright point of the stone. The time of flight profile is then determined and used in a least-mean-square method to calculate the spatial coordinates of the stone.

Pharmacokinetics and efficacy of low-dose all-trans retinoic acid in the treatment of acute promyelocytic leukemia.

A clinical trial was conducted in order to evaluate the therapeutic effect and side-effects of low-dose ATRA in the treatment of acute promyelocytic leukemia (APL). We compared the pharmacokinetic features in normal individuals with single oral ATRA at 15 mg/m2 and 45 mg/m2, respectively. The results showed that maximal plasma concentration (Cpmax) with oral 15 mg/m2 ATRA was high enough (10(-6) M) to induce APL cell differentiation. Based on these results, 27 cases of de novo APL patients were treated with continuous oral ATRA at the dose of 15-20 mg/m2/day and 24/26 evaluable cases (92%) achieved clinical CR after 13 to 67 days of ATRA treatment. No patient experienced RAS and DIC. The Cpmax with a single dose of ATRA on day 1 of treatment and immediately at CR obtained with continuous ATRA in three cases demonstrated similar values in one patient and an approximately two-fold decrease in two patients. More importantly, compared with a relatively well-matched historic group of 20 APL patients treated with high-dose ATRA, our results suggest that low-dose ATRA is as effective as high-dose in treating APL patients and may provide advantages through decreased hyperleukocytosis and other side-effects.

Incorporation of nuclear matrix attachment regions into the herpes simplex virus type 1 genome does not induce long-term expression of a foreign gene during latency.

The nuclear matrix plays a critical role in DNA replication, gene transcription and RNA processing. Transcriptionally active genes are usually associated with the nuclear matrix through DNA sequences, matrix attachment regions or MARs, which tether looped DNA to the matrix. In stable transfection and in transgenic mice MAR elements placed at the flanks of genic constructs may enhance expression and insulate against position effect variability, suggesting that independent units of transcription are established insulated from the regulatory controls of their neighbors. Herpes simplex virus type 1 (HSV-1) establishes lifelong latency in the infected host. Latency repression of viral genes extends to foreign genes incorporated into the viral genome. We report here a test of the hypothesis that MAR elements, flanking a foreign gene in the HSV-1 genome, would act to insulate it from latency repression, achieving long-term expression. A recombinant virus was produced which has an expression construct inserted into the HSV-1 genome at the Us3 locus. The expression construct consists of the A MAR element on one flank, an HIV-LRT driving the lacZ gene and the B MAR element on the other flank. The A MAR element is a 3 kb pair fragment of the 5' portion of the chicken lysozyme gene and the B MAR element is a 2.6 kb pair fragment from the 5' end of the human beta-globin gene locus control region. The LTR is derived from a human immunodeficiency virus isolated from the brain of an AIDS patient. Virus was stereotactically injected in the hippocampus, olfactory bulb and striatum of rat brains. Intense blue reaction product indicating beta-galactosidase activity was found in cells in each injected area at 2 days after injection. At 14 days after injection beta-galactosidase activity was no longer detected at any of the injected sites. We conclude that the MAR element construct did not escape latency repression.

Suppression of haematopoiesis by sera from patients with aplastic anemia.

To describe the suppressive effect of sera from patients with aplastic anemia (AA) on heamatopoietic progenitor cells and explore further the features of serum inhibitor, we first examined the inhibiting activity of the serum on the colony formation of burst-forming units of erythrocyte (BFU-E) and colony-forming units of granulocyte/macrophage (CFU-GM). The potent inhibiting activity was demonstrated in six of 20 patients. Five of the six patients were severe AA (SAA). The suppressive effect did not change after elimination of monocyles and lymphocytes from target MNC. The haematopoietic inhibitory activity of sera from three of the six patients was almost totally neutralized by the MoAb against gamma-IFN. All these three patients were SAA. These data suggest that inhibitor of haematopoietic progenitor cells is present in most of SAA patients. In some patients, the inhibitor was gamma-IFN, which play an important role in the pathogenesis of SAA.

[Effects of electrical acupuncture on the cerebral blood flow and the pial microcirculatory blood flow in dogs].

Using electromagnetic flowmeter and Laser Coppler Microcirculatory flowmeter, we studied the effect of electrical acupuncture at Zusanli (ST 36) and Quchi (LI 11) on cerebral blood flow and pial microcirculatory blood flow in dogs. The results showed that the cerebral blood flow increased remarkably, while the cerebral vascular resistance (CUR) decreased 1 min. after the acupuncture start and 20, 30, 60, 90 min. after stopping of the stimulation, and the blood flow of the pial microvessels increased 1, 5, 10 min. after starting of acupuncture and 30 min. after stopping of the stimulation. It indicates that one of the mechanisms of treatment of cerebral vascular diseases with acupuncture is that acupuncture increases the cerebral blood flow and improve the pial microcirculation.

[The role of hypergastrinemia in the pathogenesis of intussusception in infants].

From 1988 to 1992 the serum gastrin levels of 124 patients with intussusception and 20 normal infants and 15 patients with intestinal obstruction under 3 years of age were measured. Serum cAMP levels of 10 normal infants and 10 patients with intussusception were determined. In animal experiment study, the pressures of ileum cavity and ileoceal valve were determined before and after injectting pentagastrin into the veins of 8 young dogs. The levels of gastrin (219.9 +/- 100.6 pg/ml) and cAMP (32.4 +/- 7.6 pmol/ml) were higher in patient group than in the control group. (89 +/- 3 pg/ml, P < 0.05 and 22.2 +/- 7.5 pmol/ml P < 0.05). The result of animal experiment showed that the effect of gastrin and etiology of intussusception are identical. The main physiological funcitons of gastrin is to increase the intestinal peristalsis and relax the ileocecal sphincter. Our study showed that hypergastrinemia may be the basis etiologic factor of intussusception. Main clinical feactures were explained by this new theory.

[Determination of five isoflavones in Belamcanda Chinensis by RP-HPLC].

A reversed phase high performance liquid chromatographic method for the quantitative determination of the five isoflavones (tectoridin, iridin, tectorigenin, irigenin and irisflorentin) in the rhizome of Belamcanda and Iris was developed. The column was packed with TSK gel ODS 80 TS (150 mm x 4.6 mm). Methanol-water (36%-60%) was used as mobile phase. The method was rapid, sensitive, precise and has good reproducibility. The five isoflavones in the crude drugs were completely separated within 40 min. Recoveries of the five constituents were 98.41%-101.93%. The crude drugs were analysed as follows: The powdered sample was refluxed with methanol for 6 hours. The extract was transferred into a 2 ml volumetric flask and diluted to mark with methanol. A definite amount of this sample solution was injected into RP-HPLC. The contents of the five constituents in the rhizomes of Belamcanda and Iris were calculated from the relevant peak heights or peak areas. The analytical results of determination for various species and habitats are shown in Tab 2-5.
