[The transfer and expression of human clotting factor IX in muscle mediated by electroporation].

OBJECTIVE: To probe into the feasibility of increasing hFIX cDNA transfer and expression in muscle. METHODS: The high-frequency electric field was used to promote both Lac-Z-encoding plasmid pCMV beta and hFIX-expressing plasmid G1NaMCIX to transfer and express in muscle. The effects of frequency and length of square pulse, as well as eletroporation time on hFIX expression were investigated. RESULTS: Electric stimulation could increase the transfer and expression of pCMV beta in muscle, the number of X-gal positive myofiber cells in electroporation-treated mice is 2.1 times larger than that of mice not treated by electroporation (P<0.01). The most optimal electric simulation condition for hFIX cDNA transfer and expression was obtained, under this condition, the highest level of hFIX antigen in plasma is (40+/- 5.4) ng/ml and 7 times higher than that of mice without electroporation P < 0.001). CONCLUSION: Electroporation is able to enhance hFIX cDNA transfer and expression in muscle efficiently.

Batroxobin against anoxic damage of rat hippocampal neurons in culture: morphological changes and Hsp70 expression.

Batroxobin, the thrombin-like enzyme, is used for therapeutic defibrination. We have found that batroxobin has good therapeutic effect in ischemic reperfusion rats and clinical practices in vivo. But we have not studied the neuroprotective effect of batroxobin on anoxic hippocampal neurons in vitro. The purpose of this study was to obtain further information on the mechanism of the batroxobin-induced neuroprotection and examine the neuroprotective effect on neurons exposed to anoxia. The effect of batroxobin on anoxic damages in cultured hippocampal neurons of neonatal rats was investigated by using morphological changes and heat shock protein 70 Kd (Hsp70) immunoreactive expression as indicators. The results indicate that batroxobin, besides its defibrination, may have a direct neuroprotective effect on anoxic damage of hippocampal neurons.

[Applied anatomical study of blood supply in human palate].

OBJECTIVE: To investigate the origins and distributions of palatal vasculature and the three dimensional architecture. METHODS: Materials consisted of 14 normal and 10 cleft palate head-and-neck specimens. After vascular infusion, specimens were seprately analysed by stereomicroscopy, radiography, histological study and scanning electron microscopy. RESULTS: The greater palatine artery was the main vessel to supply the hard palate and the ascending palatine artery provided the principal supply of soft palate. The branches of greater palatine artery formed abundant anastomoses with the neighbor vessels. The ascending palatine artery divided into two branches in the soft palate. The artery branches in hard palate took on tree-like appearance and were arranged parallel. The capillary network was loosely packed. Whereas, the arteries in soft palate were always found to be crooked. The angles of the branches from the main trunk were changeable. The capillary network was packed densely. The vascular origins and courses of the cleft palate specimens were the same as the normal person, but the distributions and anastomoses were different. CONCLUSIONS: The blood supplies of hard and soft palate come from different vascular vessels and their vascular architectures have their own character.

[Effect of toluene on urease activity of Lou soil].

Urease activity affected by toluene studying results showed as follows: Toluene had little action on Jackbean urease activity, but can remarkably increase soil urease activity in lower toluene concentrations and shorter time, the increasing variation was 1.48-3.96 times than without toluene treatment in seven soil samples tested. The variation was less than 3.98% when sterilized soil samples adsorbed the Jackbean urease. It may be because toluene can kill soil microorganism, release urease within cell and subsequently urease fixed into soil organic-inorganic colloidal. Different soil pH and microbiology can lead to vary differently for urease activity.

Modulation by divalent cations of GABA rho 1 receptor from human retina expressed in Xenopus oocytes.

OBJECTIVE: To investigate functional homooligomeric GABA rho 1 receptors expressed in Xenopus oocytes and the modulation of divalent cations. METHODS: GABA rho 1 cDNA from human retina was transcribed in vitro to obtain sense rho 1 mRNA, which was microinjected into Xenopus oocytes. Two-electrodes voltage clamp technique was performed to record GABA-induced currents. RESULTS: Expressed receptors were found to have similar properties to GABAc receptors characterized in the retina. Cl-currents induced by GABA were blocked by picrotoxin instead of bicuculline. GABA-induced currents reversed at -19 +/- 2.5 mV, and EC50 was 3.3 mumol/L. Zn++ modulated GABA-induced currents with an IC50 = 9.6 mumol/L. Ni++, Cu++ and Cd++ inhibited GABA rho 1 obviously, too. Their rank order of potency was Zn++ > Ni++ > Cu++ > Cd++. CONCLUSION: Zinc (10 mumol/L) inhibited GABA-induced currents in a competitive manner, and its action was sensitive to extracellular pH. Site-directed mutagenesis revealed that substitution of a single histidine residue (H44 and H48) failed to affect zinc sensitivity.

[Effect of high-vaccum-manual-chop technique in phacoemulsification].

OBJECTIVE: To study the effect of high-vaccum-manual-chop technique to reduce complications related to phacoemulsification. METHODS: The correlation between the relative energy complex parameter (RECP) and the postoperative corneal edema rate (CER) in manual-chop and phaco-divide groups was statistically studied. RESULTS: The intraoperative mean relative ultrasound power and the mean time of ultrasound phacoemulsification were increased with the increase of the lens nucleus hardness. A significant difference existed between the mean time of the ultrasound phacoemulsification of the two groups (t = 3.1467, P < 0.01). The comparison between the mean relative ultrasound power of the two groups has no significant difference (t = 0.1874, P > 0.05). Positive correlation was observed between RECP and CER (r = 0.97, P < 0.01). CONCLUSION: High-vaccum-manual-chop technique may reduce the postoperative complications related to phacoemulsification, and RECP can be taken as a reliable index to evaluate the safety of phacoemulsification.

[Preparation of green fluorescent protein retrovirus and its application in mediating gene transfer into retinal pigment epithelial cells].

OBJECTIVE: To determine whether human retinal pigment epithelial (RPE) cells can be infected by retrovirus and modified by retrovirus-mediated gene transfer. METHODS: Recombination retroviral vector pLNCX-GFP (green fluorescent protein, GFP) was generated by inserting 780 bp GFP cDNA fragment into the MCS site of pLNCX. pLNCX-GFP was transfected into ecotropic packaging cell line PhiX-Eco and amphotropic packaging cell line PhiX-Ampho and PA317. Retroviral titer was tested by counting GFP expression of NIH3T3 cells. Then RPE cells were infected by using GFP retrovirus-containing supernatant. RESULTS: GFP was expressed and retrovirus was produced upon pLNCX-GFP being transfected into packaging cell line. The GFP retrovirus was able to infect primary cultured human RPE cells and immortalized RPE cell line. CONCLUSION: The retrovirus can introduce a foreign gene into RPE cells efficiently, thereby it can be used as an important tool to deliver gene into RPE for therapy of fundus diseases.

[The relationship between postburn enterogenic hypermetabolism and decontamination of intestine].

OBJECTIVE: To explore whether postburn enterogenic hypermetabolism exists. METHODS: Eighty -- eight Wistar rats inflicted by 30% TBSA III degree burns were randomly divided into two groups, i,e. burn (B) and burn with gut decontamination (D) groups. The change in resting energy expenditure (REE) was observed during 0 to 10 postburn day (PBDs) in rats. The plasma contents of lipopolysaccharide (LPS), tumor necrosis factor (TNF) and interleukin-1 (IL-1) were determined on 1, 3, 5, 7 and 10 PBDs and their correlations were analyzed. RESULTS: The postburn REE and plasma contents of TNF, LPS and IL-1 were much higher in both groups than those before injury. When compared between the two groups, the above data in D group were lower in varying degrees than those in B group. It was shown by correlation analysis that REE was postively correlated to LPS, TNF and IL-1, respectively (gamma = 0.77 similar 0.9, P < 0.05 similar 0.01). CONCLUSION: Postburn hypermetabolism in rats could be ameliorated by gut decontamination, and the postburn plasma inflammatory mediators in rats could also be lowered by gut decontmination. This suggests that enterogenic hypermetabolism does exist after burn injury.

[In vitro study of the killing activities on H-2(d) murine normal and tumor cells by Ly49A gene transfected lymphocytes].

OBJECTIVE: To observe the killing activities of Ly49A gene transfected lymphocytes of C57BL/6 mice to normal and tumor cells of BALB/c mice. METHODS: pLXSN-Ly49A retrovirus vector was constructed and packaged with PA317 cell line. The lymphocytes of C57BL/6 mice were transfected by culture with virus producing PA317 cells. The Ly49A expression rate on the transfected lymphocytes was detected by flow cytometry and the killing activities of the transfected lymphocytes to normal and tumor cells of BALB/c mice were assayed by MTT method. RESULTS: The Ly49A expression rates of C57BL/6 mice lymphocytes transfected with pLXSN-Ly49A for 24 hours, of those transfected with pLXSN and nontransfected control were (46.67 +/- 0.35)%, (18.73 +/- 0.85)%, and (19.60 +/- 0.27)%, respectively. The killing activity of the transfected lymphocytes to 4T(1) tumor cells remained almost the same as that of the control (P > 0.05), but to normal fibroblasts decreased sharply (inhibiting rate 22% - 25%). CONCLUSION: The Ly49A transfected C57BL/6 mice lymphocytes could kill BALB/c mice tumor cells as effectively as the control did, but the activity decreased sharply to normal BALB/c mice cells, which would be instructive for resolving graft versus host disease after allogeneic bone marrow transplantation.

[HCV-PCR-Hybrid.ELISA for the detection of HCV RNA in serum samples from hepatitis patients].

BACKGROUND: To detect HCV RNA in serum samples from hepatitis patients by using the methods of HCV-Reverse transcription polymerase chain reaction and hybrid ELISA. METHODS: HCV RNA in serum samples was amplified by RT-PCR with one of the primer labeled with biotin. PCR amplified products were detected with microtiter plate hybridization with HCV specific probe, then determined with steptavidin-HRP assay. Anti-HCV in these samples was also detected with ELISA simultaneously. RESULTS: The positive rates of HCV RNA were 81.8% (72/88) in anti-HCV positive group and 57.9% (88/152) in the whole patients group. Serum HCV RNA and anti-HCV positive rates were compared and the consistent rate was 78.9% (120/152, P>0.05). The positive rate of HCV RNA was higher in H.ELISA (57.9%, 88/152) than that in electrophoresis (35.5%, 54/152)(P<0.01). CONCLUSIONS: The HCV-PCR-H.ELISA was a sensitive, specific, stable, semi-quantitative and easy to operate method. This HCV RNA detection was especially beneficial to early diagnosis during viramia period and was helpful for the evaluation of anti-HCV therapy.

[Orthodontic diagnosis and treatment of unilateral posterior cross-bite].

OBJECTIVE: The aim of this study is to investigate the clinical diagnosis and treatment of unilateral posterior cross-bite and to provide information for correct treatment of this disease. METHODS: The X-ray examination of patients' heads in the posterior-anterior position, analysis of pantomorphic films and model measurement were used for the diagnosis of unilateral cross-bite. According to these data, three types of unilateral posterior cross-bite would be classified, and different orthodontic appliances would be applied in the following treatment, including separate plate removable expansion appliances, Quad-helix appliances, narrowing dental arch and inter-arch elastics. RESULTS: A total of 78 cases of unilateral posterior cross-bite were treated by different techniques in a manner consistent with the classification. All of the clinical results of treatment were satisfactory and successful. CONCLUSION: If diagnosis is correct, and suitable treatment methods are chosen, unilateral cross-bite can be treated successful.

[Transforming growth factor-beta can induce differentiation of hypertrophic chondrocytes into osteoblast-like cells in epiphyseal plate].

OBJECTIVE: To investigate the effect of transforming growth factor-beta (TGF-beta) on the differentiation of hypertrophic chondrocytes in epiphyseal plate in order to understand its role in the mechanism of bone and cartilage formation. METHODS: We observed the effect of TGF-beta in varied time and dosage on the differentiation of hypertrophic chondrocytes in embryonic chick femora which were cultured in serum-free medium, using histochemical staining to detect the expression of alkaline phosphatase (ALP) and immunohistochemical staining to detect the expression of type I collagen and fibronectin. RESULTS: TGF-beta enabled the chondrocytes to express type I collagen and fibronectin, and it enhanced the expression of ALP in hypertrophic zone. Time-effect and dosage-effect relationships were observed. CONCLUSION: TGF-beta can induce the differentiation of hypertrophic chondrocytes into osteoblast-like cells in epiphyseal plate of embryonic chick femora. The inducing effect might be closely related to time and dosage of TGF-beta.

[Spectroscopic studies on the binding of effective component of Chinese herbs with human serum albumin].

Tanshione II A and Cryptotanshinone are the main effective components of the traditional Chinese medicine Salvia miltiorrhiza bung. They both have many kinds of physiologic activities. When a medicine enters the body, it always goes to the acceptor site and has pharmacodynamic action by way of storage and carriage of plasma. Serum albumin is the major serum protein, which can combine with many inner and outer source compounds. In this paper, we have studied the interaction of Tanshinone with the human serum albumin(HSA) in beta-CD buffer system by spectroscopic method. The results show that the absorption maximum of Tanshinone exhibit red shift upon binding to HSA and the absorbance enhance. Tanshinone have stronger fluorescence quenching effect on HSA and the dissociation constants of Tanshinone bound to HSA are calculated. The distance of protein Trp-214 to the first binding site is also calculated on the basis of energy transfer between HSA and bound Tanshinone.

[Research on the near-infrared (NIR) photon migration in multi-layered structures of biological tissues].

The phenomena of photon migration in multi-layered structures of biological tissue is very important in the field of non-invasive near-infrared optical diagnosis. In this paper, a two-layered structure consisting of the fat and the muscle of poke was used as the sample. Near-infrared light source and detector were located on the surface of the fat and a part of the photons reflected by the two-layered structure of the tissue were detected. The relationship between the optical current and the distance from source to detector and the relationship between the optical current and the thickness of the fat layer were given. In order to analyze the experiment results, Monte-Carlo simulation was used to calculate the photons migration traces in the uniform tissues and the Fresnel's law is used to determine the refraction and reflection components of a photon package at the boundary between the fat and the muscle. The results showed that the simulation data coincident well with the experimental results. The conclusion is that the experiment and the simulation method provided in this study may be useful to analyze the photon migration in a mult-layered tissue and to optimize the design of the transducer.

[The efficacy of the native flumazenil for acute poisoning with benzodiazepines].

OBJECTIVE: To evaluate the efficacy of the native flumazenil for acute self-poisoning with benzodiazepines. METHODS: One hundred and twenty-six patients with unconsciousness from benzodiazepines-induced self-poisoning were randomly divided into two groups: flumazenil group(Group II, 63 cases) were treated with flumazenil, and conventional-medicine group(Group I, 63 cases) with placebo(glucose, vitamin C, KCl). A modified Glasgow Coma Scale(MGCS) and Observer's Assessment of Alertness/Sedation Scale(OAA/S) were used in the assessment of consciousness. RESULTS: MGCS were increased by 5.3, 8.0, 9.4 and 7.3 at 15 min, 30 min, 60 min and 180 min after intravenous flumazenil(P < 0.01) and by 5.2, 7.7, 8.7 and 6.9 in comparison with conventional-medicine group(P < 0.01). OAA/S increased by 1.9 compared with conventional-medicine group(P < 0.01). No severe side-effects were found in the treatment. CONCLUSIONS: Flumazenil might improve benzodiazepines-induced unconsciousness markedly and may be the most effective antagonist of benzodiazepines.

Cytotoxicity of PM(2.5) and PM(2.5--10) ambient air pollutants assessed by the MTT and the Comet assays.

Ambient air particulate matters are classified into two distinct modes in size distribution, namely the coarse and fine particles. Correlation between high particulate concentration and adverse effects on human populations has long been recognized, however, the toxicology of these adverse effects has not been clarified. In the current report, the cytotoxic effects of the solvent-extractable organic compounds (SEOC) from fine particles smaller than 2.5 microm (PM(2.5)) and from coarse particles between 2.5-10 microm (PM(2.5-10)) were studied. Nine 24h consecutive monthly samples were tested to determine the correlation between cytotoxicity and total SEOC in two size fractions of particulate air pollution. Cytotoxicity of SEOC was measured by two micro-scale mammalian cells-based bioassays: the MTT cell proliferation assay, and the Comet assay for the detection of DNA damage. A well-defined mammalian cell line - Rat 6 rodent fibroblast was employed in the study. The SEOC extracts of air particulate matters were sub divided into two equal parts. One part was dissolved in DMSO, the other in KOH/hexane and then conjugated with bovine serum albumin to produce a lipid-soluble fraction for testing. The DMSO fraction would contain mainly the polycyclic aromatic hydrocarbons (PAH), alkanes and alkanols, while the lipid-soluble fraction would be enriched with fatty acids. The results from MTT assay showed that cytotoxicity of the PM(2.5) was much more severe than the PM(2.5-10), suggesting that toxic SEOC were confined to the fine particles. By and large, the DMSO solubles were much more toxic than the lipid solubles. The degree of cytotoxicity of the DMSO soluble samples is positively correlated to the amount of particulates present in the ambient air. For the PM(2.5), the winter samples were significantly more toxic than the summer samples in terms of cell killing, which seemed to be a direct reflection of the total loading of organic matter in the samples. Results from Comet assays showed that SEOC samples of PM(2.5) derived from winter months induced DNA damage at dosages resulting in no obvious cell killing in the MTT assay. Thus, long-term exposure to non-killing dosage of air pollutants may lead to the accumulation of DNA lesions, which may be one of the mechanisms responsible for the chronic adverse health effects of particulate air pollution.
