Geographical distribution of the Hbb haplotypes in the Mus musculus subspecies in eastern Asia.

We surveyed the geographical distribution of Hbb haplotypes of the house mouse, Mus musculus, in the former USSR and the northern part of China. Mice with the w1 haplotype were distributed from the coastal area of the Black Sea to the Maritime Province of Siberia in the former USSR and in the area north of the Yangtze River in China. Mice with the p haplotype were found in the areas surrounding those populated with the w1 haplotype mice. In the Maritime Province and the southern part of Siberia, the d haplotype was predominant. The correlation between the geographical variation of the Hbb haplotypes and the distribution of the Mus musculus subspecies in the eastern part of the Eurasian Continent, is discussed.

Comparative pharmacology of epibatidine: a potent agonist for neuronal nicotinic acetylcholine receptors.

Pharmacological properties of the (+)- and (-)-isomers of synthetic epibatidine, exo-2-(6-chloro-3-pyridyl)-7-azabicyclo-[2.2.1]heptane, were compared with nicotine and acetylcholine on several subtypes of chicken and human nicotinic acetylcholine receptors (AChRs). Both isomers of epibatidine behaved as extremely potent full agonists on chicken (alpha 3 beta 2, alpha 3 beta 4, alpha 4 beta 2, alpha 7, and alpha 8) and human (alpha 3 beta 2, alpha 3 beta 4, and alpha 7) neuronal AChRs expressed in Xenopus oocytes. Currents induced by epibatidine were effectively blocked by the nicotinic antagonists hexamethonium and mecamylamine. Apparent affinity was 100 to 1000-fold higher for epibatidine than for nicotine or acetylcholine. EC50 values ranged from 1 nM (for homomeric chicken alpha 8) to 2 microM (for homomeric chicken alpha 7). Epibatidine showed comparatively lower affinity for muscle-type AChRs from Torpedo and humans (EC50 values, 1.6 and 16 microM respectively). In binding assays, epibatidine was used on AChR subtypes immunoisolated from chicken brain and retina (alpha 4 beta 2, alpha 7, and alpha 8), the human neuronal cell line SH-SY5Y (alpha 3 and alpha 7), Torpedo electric organ (alpha 1 beta 1 gamma delta), or the human rhabdomyosarcoma cell line TE671 (alpha 1 beta 1 gamma delta). Both isomers of epibatidine exhibited extremely high affinity for all neuronal AChRs tested, with KI values ranging from 0.6 pM (human alpha 3 AChRs) to 0.6 microM (chicken alpha 7 AChRs). In contrast, epibatidine had lower affinity for Torpedo muscle-type AChRs (KI approximately 5 microM). Racemic [3H]epibatidine was an effective labeling reagent for human alpha 3 beta 2 AChRs, exhibiting a KD (0.14 nM) similar to the KI values observed for unlabeled (+)-epibatidine (0.23 nM) or (-)-epibatidine (0.16 nM).

[Clinical effects and immunological analysis of intraabdominal and intrapleural injection of lentinan for malignant ascites and pleural effusion of gastric carcinoma].

Twenty-one patients with malignant peritoneal or pleural effusions of gastric carcinomas were treated with intracavitary injection of lentinan (LNT). LNT was injected at a dosage of 4 mg/week for 4 weeks. In total, fifteen (71%) of twenty-one patients demonstrated clinical responses. Toxicity caused a high fever in only one case. LAK and ATK activities induced from peritoneal exudate cells (PEC) after culture with autologous tumor and interleukin-2 were examined before and after LNT injection. ATK activity was augmented, but LAK activity was reduced after LNT injection. These results indicate that intracavitary injection of LNT is a useful treatment for malignant effusions, and that LNT augments the induction of cytotoxic T-lymphocytes.

[Drug resistant mechanism of clinical isolates of Klebsiella pneumoniae].

Plasmid conjugation experiments of 34 gentamicin-resistant strains of K. Pneumoniae were done. The resistant genes of 31 strains (91%) were transferred partially or completely with a frequency of 10(-4)-10(-6). The results of plasmid analysis in 63 strains of K. Pneumoniae showed that 85kb and 58kb plasmids carrying resistant genes were responsible for bacterial resistance. 13kb plasmids or the smaller ones were not related to bacterial resistance. In spite of the aminoglycoside modifying enzyme, The profiles were different, but the plasmid patterns were almost the same.

[Detection of carcinogen in source drinking water in stomach cancer prevalent areas of Zanhuang county].

Benzo(a) pyrene, aflatoxin, sterigmatocystin and six kinds of nitrosamine and their precursors (secondary amine) in 36 specimens of source drinking water collected from stomach cancer prevalent areas of Zanhuang County during the autumn and spring periods were determined quantitatively with high performance liquid chromatography. Results showed drinking water for local residents was significantly contaminated with benzo(a) pyrene, with an average of 0.0148 microgram/L and the highest one of 0.0305 microgram/L, and especially in the autumn, simultaneous contamination with nitrosodimethylamine, nitrosodiethylamine, and nitrosodipropylamine could be found, with proportions of 38.5%, 76.9% and 76.9%, respectively. Aflatoxin B1, and G1 could also be found in drinking water in the autumn, but no sterigmatocystin could be detected.

Late-onset systemic lupus erythematosus: clinical and immunological characteristics.

Between January 1976 and December 1992, 17 patients on follow-up at Systemic Erythematosus (SLE) Clinic in the University Hospital, Kuala Lumpur had onset of the disease after the age of 50 years. This constituted about 4% of our total SLE patients. They formed a distinct subgroup of the lupus population with an insidious onset and have a benign course compared to the younger SLE patients. Arthritis and skin rashes were the commonest initial manifestations. Renal and central nervous system manifestations were uncommon but pulmonary involvement was frequent compared to young SLE patients. The prevalence of positive autoantibodies and hypocomplementaemia were lower. Disease activity showed no correlation with erythrocyte sendimentation rate, autoantibodies or complement levels. Overall prognosis in these late-onset patients was favourable with a good response to steroids and less frequent relapses.

[Bacterial resistance in clinical isolates of Klebsiella pneumoniae].

Ninety-four percent of 336 strains of Klebsiella pneumoniae were resistant to ampicillin, while less than 10% were resistant to cefotaxine and ceftazidine. Among all the aminoglycosides tested, amikacin had the strongest activity on K. pneumoniae. The resistant rate of nosocomial strains was higher than that of community strains. The results of susceptibility test showed that 42% (119/280) of K. pneumoniae strains were gentamicin-resistant. Based on the resistant pattern of K. pneumoniae, the aminoglycoside-modifying enzymes produced by the organisms were inferred. Among them 90% of 119 strains produced aminoglycosides-modifying enzymes AAC (3)-V, 8% (10/119) AAC (3)-V together with AAC (6')-Ib, AAC (6')-V or ANT (2"). The resistance of two strains was assumed to be permeability change. Besides, 58% (161/280) were sensitive strains without producing any aminoglycoside-modifying enzymes. Bacteria typing done by phage typing or Klebcin in 27 strains showed that most strains were from different origins with few exceptions. The results show the significance for controlling the nosocomial infections and reasonable use of antibiotics.

Radiofrequency catheter ablation of ventricular tachycardia in 26 patients.

Electrophysiology study and radiofrequency catheter ablation (RFCA) were performed in 26 patients with refractory sustained ventricular tachycardia (VT). After induction of VT, 12-lead electrocardiogram (ECG) was recorded and QRS morphology and axis of induced VT were studied to identify the origin of VT. The precise site of VT origin were localized by pace mapping and activation mapping carefully. RF energy was delivered through a big-tip deflectable electrode catheter when the earliest site of endocardial activation and a high-frequency and low-amplitude potential of Purkinje fiber, preceding surface QRS by more than 25 ms, were identified and/or a pace map was obtained showing identical QRS complexes in at least 11 of 12 ECG leads. VTs were ablated successfully in 24 of 26 patients (success rate was 92%). For successful ablation, it is essential that the pace map QRS morphology in 12 leads should be identical with that in spontaneous or induced VT as far as possible in performing pace mapping. Pace mapping is safe, simple and has no unfavourable effect hemodynamics although it takes longer time. Activation mapping takes shorter time and has a high success rate. QRS configuration in spontaneous VT can help to localize the site of VT origin. Deliberate mapping at the site suggested to bo the origin of VT by surface ECG can shorten the duration of mapping and increase the success rate of RFCA. RFCA of VT in patients without structural heart disease is effective, safe, and has a high success rate, so it may be considered as an early therapy for these patients.

[The level of serum gastrin and ulceration in cirrhotic patients with portal hypertension].

A higher incidence of gastric and duodenum ulcer was well recognized in patients with liver cirrhosis, but the mechanism has not been fully identified. In this study, serum gastrin, free portal pressure (FPP) were measured in 24 consecutively admitted cirrhotic portal hypertensive patients, and preoperative basic acid output (BAO) was measured in 13 randomized patients. Among the 24 patients, concomitant duodenal ulcers were found in 3 by both gastroduodenoscopy and barium series, and gastritis was found in all patients. It was found that most patients (71%) with liver cirrhosis have a elevated level in serum gastrin, whereas BAO is lower than normal in all patients, and the higher the FPP, the lower the BAO is. We believe that the congestive gastroduodenal mucosal lesion was underlying the ulceration most often seen in patients with portal hypertension.

[Localization of insulin-like growth factor I in the upper end of human fetal femur].

The localization of insulin-like growth factor I in the upper end of the human fetal femur was studied by immunohistochemistry on the undecalcified bone sections. Certain numbers of chondrocytes in germinal, proliferating and hypertrophic zones of the epiphysial growth plates, some osteoblasts and a few osteoclasts were immunoreactive to the antibody to IGF-I. The end-product of the DAB reaction was deposited in the osteoid and bone matrix of metaphysis. Also presented is a discussion on the local effects of IGF-I on the chondrocytes, osteoblasts and osteoclasts.

[Detections of hepatitis C virus RNA and NS3 antigen and their relation to liver histopathology].

To detect the distribution of hepatitis C virus and investigate the pathogenesis mechanisms of the viral infection in the liver tissues of the patients with acute or chronic hepatitis C, we examined HCV antigen expression by using the murine monoclonal antibody against HCV C33c peptide in the paraffin-embedded liver tissues from 28 patients with acute or chronic hepatitis C. The NS3 antigen was detected in 85.7% (24/28) of all the biopsy specimens. The distribution and staining density of the antigen immunoreactive signal varied according to different types of patients and the regions in the liver sections, but they obviously had a topographical relationship with the inflammatory-necrosis areas such as fatty and ballooning degeneration and focal necrosis in the liver tissues of nearly all the patients. In addition, the localization of HCV RNA investigated by in situ hybridization assay in 20 liver tissues the above 28 biopsy HD in the Chinese. They also provide valuable data for HD molecular diagnosis, genetic counselling and genetic health.

[Experimental study on targeting therapy of human hepatocellular carcinoma in nude mice using adriamycin-anti-transferrin receptor conjugate].

Conjugate between adriamycin and anti-transferrin receptor monoclonal antobody WuT9 was prepared with Dextran T10 as a bridge, and it was proved to retain both the drug cytotoxity and antibody activity. The conjugate or its individual components were administered to athymic mice bearing SMMC-7721 human hepato-cellular carcinoma (HCC) cells. These agents were given 2, 5, 8 days after inoculation of tumor cells (1 x 10(7) per mouse). Tumor size was measured daily (geometric mean diameter, GMD) and latent period recorded. All the animals were killed on day 35 to measure tumor weight and natural killer (NK) activity of splenic lymphocytes. The results showed that the latent period in the conjugate treated group was much longer than that in Adr treated group (33.0 days vs 25.0 days), and that GMD on day 35 in the conjugate treated group was much smaller than that in Adr treated group (3.80 +/- 1.9 mm vs 6.69 +/- 2.3 mm, P < 0.05). In addition, NK activity in the conjugate treated group was higher than that in Adr treated group (28.2 +/- 8.2% vs 15.1 +/- 3.6%, P < 0.05). The results suggest that adriamycin-anti-transferrin receptor conjugate might be useful in the treatment of human HCC.

[Analysis of beta-thalassemia mutations and prenated diagnosis in Chengdu population].

Ninety-five of non-differential diagnostic patients were detected by dot-blot analysis on enzymatically amplified DNA with a number of allele specific oligonucleotide probes complementary to the most common mutations in Chengdu population. Prenatal diagnosis was accomplished by the same procedure on enzymatically amplified amniocyte DNA. The result revealed fifty-eight cases of beta-thalassemia. Of the 73 chromosomes tested, twenty-eight (38.4%) had the codon 17(A-->T) mutation, twenty-one (28.8%) had the codon 41-42(-TTCT) mutation, fourteen (19.0%) had the IVS-I-654(C-->T) mutation; nt--28(A-->G) and nt--29(A-->G) mutations were six (8.2%) and four (5.5%) respectively.

[Secretory immunoglobulin A in human milk and infants' feces at 1-4 months after delivery].

OBJECTIVE: To determine the concentration of secretory immunoglobulin A (SIgA) in human milk at 1, 2, 3 and 4 months after delivery and in infants' feces in the same period and find out the effect of milk on the content of SIgA in infants' intestine. METHODS: 20 women and 12 breast feeding infants and 13 formula feeding infants were studied, and milk and infants' feces were collected at 1, 2, 3 and 4 months after delivery, respectively. The concentrations of SIgA in milk and feces were determined by radioimmunoassay. RESULTS: No apparent changes in the concentration of SIgA were found in human milk and feces from breast feeding and formula feeding infants at 1-4 months after delivery, but the concentration of fecal SIgA in breast feeding infants was significantly higher than that in the formula feeding infants, during the first 4 months after birth. CONCLUSIONS: Human milk can provide a large amount of SIgA for infants. We conclude from these data that human milk is beneficial in protecting the immunological function of infants digestive tract.
