The presence of tissue amyloid P component was determined by using a direct immunofluorescence technique on frozen sections of normal human placentae and umbilical cords from gestations of various durations. Amyloid P component was first detected at the 16th week of gestation and appeared to increase progressively in amount so as to be present in abundance in the term placenta. Placental amyloid P component was present in the perifetal capillary zone where basement membrane-like material and reticulin fibres are also found. Amyloid P component may be related to the maturation of the placenta.

Increased gastrocnemius/soleus muscle tone in children with cerebral palsy may cause an equinus of the ankle. Botulinum toxin type A (BTX), a neuromuscular blocking agent, reduces muscle tone in various neuromuscular disorders. The safety and short-term efficacy of BTX injections were evaluated in a prospective, 3-month, double-blind, randomized clinical trial involving 114 children with cerebral palsy and dynamic equinus foot deformity. Outcome was determined by observational gait analysis, ankle range-of-motion measurements, and quantification of muscle denervation by nerve conduction. Patients in the BTX group demonstrated improved gait function and partial denervation of the injected muscle. No serious adverse events were reported.
Using a panel of monoclonal antibodies for the immunophenotyping of haematological malignancies, we have made a direct comparison of the usefulness of indirect immunofluorescence using flow cytometry (IFC) alongside the alkaline phosphatase-anti-alkaline phosphatase (APAAP) immunoenzyme method. Analysis of 400 consecutive patient samples (blood and bone marrow) over a 2-year period, resulted in unequivocal phenotyping agreement by both methods in 98 per cent of cases. The positive results accounting for 2 per cent discordance were obtained by the APAAP method. The value of the technique in the clinical management of patients with leukaemia is documented together with guidelines for clinical laboratories.

A rapid and sensitive method was developed for simultaneous detection of nivalenol (NIV), deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-A-DON), and 15-O-acetyl-4-deoxynivalenol (15-A-DON) in wheat flour. Samples were extracted with acetonitrile-water (84 + 16), and the extract was filtered and purified by a column containing a combination of charcoal, celite, and other adsorbents. For screening analysis, the column eluate was only extracted with ethyl acetate. After evaporation of the solvent, the dried residue was redissolved in acetonitrile-water (2 + 8) and then analyzed by reversed-phase liquid chromatography (LC) with diode array detection. Recoveries of NIV, DON, 3-A-DON, and 15-A-DON from whole wheat flour spiked at 2 levels were 49-55, 92-97, 98-100, and 100-105%, respectively. To quantitate mycotoxin amounts lower than 1 ppm, purified column extracts were evaporated to dryness, derivatized with heptafluorobutyric anhydride, and analyzed by gas chromatography with electron capture detection (GC-ECD). Average recoveries of NIV, DON, 3-A-DON, and 15-A-DON from whole wheat flour spiked at 2 levels, were 45-52, 91-103, 81-85, and 84-92%, respectively. GC-ECD detection limits for all mycotoxins tested at a signal-to-noise ratio of 4:1 were < 30 ng/g. Results of GC-ECD analysis for whole wheat flour samples spiked with mycotoxins at 3 and 10 ppm compared well with results (2.8 and 9.9 ppm) for the same samples analyzed by LC.
