Evidence of androgen and estrogen receptors in the preen gland of male ducks.

Androgen receptors have been found in duck preen glands by using dextran-coated charcoal adsorption. They bound DHT with high affinity (KD = 0.2 nM), limited capacity (45 fmoles/mgP) and good specificity. They sedimented at 8 S in a sucrose gradient and were destroyed by pronase digestion and heating. An estrogen receptor having different binding specificity was also demonstrated. On the basis of a marked annual cycle of gonadal activity in ducks, this system appears appropriate for studying the regulation of sex steroid hormone receptors.

[Estrogen receptors and nuclear translocation in C3H independent mouse mammary tumors (author's transl)].

In C3H independent mouse mammary tumors, estrogen receptors were characterized and analyzed using dextrancoated charcoal, protamine sulfate and hydroxyapatite assays. The mean concentration of cytosol receptors was of 18.5 +/- 3.5 femtomoles per mg of protein stating that C3H tumor was a "receptor-positive" tumor. The cytosol estrogen-receptor complexes were translocated in vivo to the nucleus thus suggesting that the hormone-independency of the C3H tumor should be searched in later steps of the estradiol mechanism of action.

Regulation of estrogen receptors in ovarian-dependent rat mammary tumors. I. Effects of castration and prolactin.

The "ovarian-dependent" rat mammary tumors, induced by 9,10 dimethyl-1,2-benzanthracene, were assayed for their estrogen receptor content by dextran-coated charcoal adsorption and sucrose gradient ultra-centrifugation. The estradiol receptors bound estrogens with a high affinity (KD approximately 0.25 nM), limited capacity and high specificity, and sediment at 8 S in a sucrose gradient. The cytosol receptors were transferred to the nucleus after binding to estrogens either in vivo or in vitro. The tumor area regressed by 70% during the first 10 days of castration while the concentration of estradiol cytosol receptors decreased from 225 to 16 fmoles/mg of protein. Three to five days after in vivo administration of estradiol (2 mug daily) or prolactin (1 mg daily) the concentration of estrogen receptors was increased in spayed rats. In biopsy experiments, prolactin, but not estradiol, was shown to increase the estrogen receptor concentration when endogenous prolactin release was blocked by CB 154. Prolactin did not modify the intracellular distribution of the estrogen receptor or its binding affinity for estrogen. The uterine estrogen receptor sites were not modified by prolactin under the same conditions. We, therefore, suggest that in mammary tumors prolactin sensitizes the action of estrogens at the target level by increasing the concentration of their available receptor sites.

On the glandular origin of seminal plasma lipids in man.

The aim of this study is to examine the glandular origin of seminal fluid lipids in man. The triglycerides, the total cholesterol, the non-esterified fatty acid and the total phospholipids were measured in seminal plasma of vasectomized patients (n = 8) and control subjects (n = 15). The same parameters were measured in seminal plasma collected in three fractions from split ejaculates (n = 10). The total cholesterol and the non-esterified fatty acid are principally prostatic in origin. The phospholipids are secreted by the epididymis but also by the prostate. The origin of the triglycerides seems to be very varied.

Comparative study of semen in diabetic and healthy men.

This study examines the consequences of diabetes on male reproductive function. We have studied a group of 18 diabetics (15 type I, 3 type II) age 24 to 40 years (mean 34 +/- 5.3) with duration of diabetes from 2 to 30 years (mean 11.8 +/- 8.3). This group was compared to 20 semen donors. In diabetics higher sperm (p = 0.0063) and round cell (p = 0.031) concentrations/ml as well as a higher total spermatozoa count (p = 0.023) were observed. The percentage of abnormal sperms were also higher. None of the couples had consulted for sterility and they had a mean of 2 children. In this group of patients, diabetes was not a cause of subfertility. Specific spermiological alteration in diabetics might reflect a testicular damage secondary to their diabetes or disease.

[Oestrogen-induced proteins in breast cancer (author's transl)].

The hormone dependence and the prognosis of human breast cancers are currently based on assays of the estrogen receptor (RE) and progesterone receptor (RP) in the tumor. However, such prediction can not be fully accurate: RE may be non functional in some tumors and the estrogenic regulation of RP is not directly correlated with the effects of estrogens on cell growth. Here we described other estrogen induced intracellular, secreted or membrane proteins. Investigations of these proteins should allow a better understanding of the mechanisms by which estrogens stimulate the growth and spreading of breast cancer. Some secreted proteins could even be useful as circulatory markers of hormone dependence.
