Conformational change in rat liver phenylalanine hydroxylase associated with activation by phenylalanine or N-(1-anilinonaphth-4-yl)maleimide was investigated by measuring fluorescence spectra and fluorescence lifetimes of tryptophanyl residues as well as the probe fluorophore conjugated with SH groups of the hydroxylase. The fluorescence spectrum of tryptophan exhibited its maximum at 342 nm. It shifted by 8 nm toward longer wavelength accompanied by an increase in its intensity, by preincubation with 1 mM phenylalanine. The fluorescence intensity of tryptophan increased by 36% upon the activation. On the other hand, the binding of (6R)-L-erythro-tetrahydrobiopterin, a natural cofactor of the enzyme, induced a decrease in the fluorescence intensity by 79% without a shift of the maximum wavelength. The fluorescence lifetime of tryptophan of phenylalanine hydroxylase exhibited two components with lifetimes of 1.7 and 4.1 ns. The values of the lifetimes changed to 1.4 and 5.6 ns, respectively, upon the activation. It is considered that the change in the longer lifetime is correlated with the shift of the emission peak upon the activation. The values of both the lifetimes decreased to 0.64 and 3.6 ns upon the binding of (6R)-L-erythro-tetrahydrobiopterin, which is coincident with the decrease in the fluorescence intensity. Conjugation of N-(1-anilinonaphth-4-yl)maleimide with SH of phenylalanine hydroxylase brought about a decrease in both the fluorescence intensity and the value of the shorter lifetime of the tryptophanyl residues, while the longer lifetime remained unchanged.(ABSTRACT TRUNCATED AT 250 WORDS)

An IgE RIA kit (Sandwich method; Dainabott), is used to obtain the following results. (1) Standard curve: Since the range of reproduction rate show 3.16-7.07% (C.V.), the curve become steep. (2) Incubations under controlled situation: Both of the incubations are controlled at 15-30 degrees C for 2 h. (3) Reproducibility test: Coefficients of variation (C.V.) of intra-assay and inter-assay variation are 2.32-3.94% and 2.92-3.92% respectively. (4) Recovery test: A result of the recovery test range between 100.1-101.7%. (5) Dilution test: Multiple dilution effects are observed. (6) Average counts of the serum IgE for the controlled and diseased groups: The average counts of the serum IgE for the controlled group, atopic diseased group, allergic rhinitis group and allergic bronchial asthma are 144.9 +/- 183.2 IU/ml, 1,099.0 +/- 2,782.4 IU/ml, 1,150.9 +/- 2,063.3 IU/ml and 600.7 +/- 686.4 IU/ml respectively. The value of the diseased groups have tendency to show higher averages than the controlled group. Since the controlled and diseased groups show wide distributions of the serum IgE level, there is no significant difference of two variations. However the diseased groups have tendency to show higher ratio of the serum IgE level in blood than the controlled groups. These basic researches are quite meaningful, because they are able to apply for a supplemental diagnosis of the atopic and parasitic disease.
The homologous blood transfusion is often required for the replacement of intraoperative blood loss in the radical hysterectomy. Recently, we have tried elective autologous blood transfusion and hypotensive anesthesia to eliminate the complications caused by homologous blood transfusion. This study reports our experience at Kure National Hospital. Radical hysterectomy has been done in 27 cases with uterine cancer from June, 1985 to March, 1987. Homologous blood transfusion was required to one of 5 cases with autologous blood transfusion, none of 5 cases with autologous blood transfusion and hypotensive anesthesia, and 6 of 17 cases without either procedures. Autologous transfusion and hypotensive anesthesia seemed to be effective in reducing homologous blood transfusion.

Two thousand three hundred seventy gastric cancers from 2173 patients for which surgical resections were performed at the Tokyo University Hospital from 1967 to 1985, were analyzed. Cancers were classified into diffuse adenocarcinomas, intestinal adenocarcinomas, and an unclassified group. The proportional rate of the intestinal type was higher in the old than in the young, and higher in men than in women. The reverse was true of the diffuse type. The intestinal-diffuse ratio had increased recently, especially in patients with early cancers and in patients in their 50s and 60s, which suggested the efficacy of early screening methods for gastric cancers.
In many proteins fluorescence from single tryptophan exhibits a nonexponential decay function. To elucidate the origin of this nonexponential decay, we have examined the fluorescence decay function and time-resolved fluorescence anisotropy of a fluorophore covalently bound to a macromolecule by solving a rotational analogue of the Smoluchowski equation. An angular-dependent quenching constant and potential energy for the fluorophore undergoing internal rotation were introduced into the equation of motion for fluorophore. Results of numerical calculations using the equations thus obtained predict that both the fluorescence decay function and time-resolved anisotropy are dependent on rotational diffusion coefficients of fluorophore and potential energy for the internal rotation. The method was applied to the observed fluorescence decay curve of the single tryptophan in apocytochrome c from horse heart. The calculated decay curves fit the observed ones well.

This is the third report in a series on the inflammatory mediators and modulators released in organ culture from skin lesions of various ages, which were produced in vivo in rabbits by the military vesicant, sulfur mustard (SM). It describes the electrophoretic protein fractions and trypsin-inhibitory capacities of the various culture fluids and the amounts of alpha 1-proteinase inhibitor and alpha-macroglobulin proteinase inhibitors in these fluids. With one-dimensional electrophoresis, the albumin and beta-globulin fractions of protein in culture fluids varied little with the development and healing of the SM lesions. These fractions proportionally resembled the corresponding fractions found in serum. The alpha 1-globulin fraction was proportionally smaller than the corresponding fractions of serum as the lesions healed. The alpha 2-globulin fraction was proportionally smaller than the corresponding fractions of serum at all stages of lesion development and healing. The gamma-globulin fraction was proportionally larger as the lesions healed. With two-dimensional electrophoresis, about 68%, 46%, and 35% of the protein spots in culture fluids from representative 1-day and 6-day SM lesions and normal skin, respectively, matched those from serum. In each case, the large, diffuse, serum albumin spot represented about two-thirds of the protein present. Thus, gravimetrically, in normal skin and in both developing and healing lesions, the extracellular proteins were 80-90% of serum origin. The trypsin-inhibitory capacity (TIC) per milligram protein in the culture fluids of healing lesions was markedly less than the TIC per milligram protein in the fluids of peak lesions. This decrease correlates well with the decrease found in the alpha 1-globulin fraction, which contains alpha 1-antiproteinase (alpha 1-PI) (and alpha 1-macroglobulin [alpha 1M] in rabbits). The alpha 1PI and the alpha 1M-alpha 2M proteinase inhibitors were identified in the culture fluids by means of sodium dodecyl sulfate-polyacrylamide gel electrophoresis, Western blots, specific antibodies, and the immuno-peroxidase technique. The levels of both free and proteinase-complexed alpha 1PI and alpha M inhibitors in the culture fluids decreased as the lesions healed. In both developing and healing lesions, at least half of the alpha 1PI and alpha M inhibitors seemed to be complexed with proteinases. Thus, serum seems to be a major source of unbounded extracellular protein within acute inflammatory lesions, and serum proteinase inhibitors seem to be the host's major defense against local damage by proteinases from serum, infiltrating leukocytes, and activated fibroblasts.
Eight cases of uterine papillary serous carcinoma were studied clinicopathologically. Postoperatively, four were stage I and four were stage III. Operations combined with radiation and chemotherapy were performed on the stage I cases. Three of them died within one year, and the other one died 3 years and 7 months after treatment. The prognosis was extremely poor even in stage. I. UPSC is histologically similar to ovarian serous carcinoma and is characterized by a papillary architecture supported by a fibrovascular stalk and a lining epithelium of single or stratified atypical cuboidal epithelium. Psammoma bodies were found in 3 out of the 8 cases.
We investigated 24 patients with Prader-Willi syndrome by the high-resolution banding technique. Their history and clinical findings were also examined in some detail. Twelve had interstitial deletion of 15q; del(15) (q11.2q13) in 11 cases and del(15) (q11.2q12) in one case. Six revealed normal karyotypes at about 500-850 bands per haploid-set level. In an additional six cases, no deletion was detected. However, we took the results as tentative, as the observed karyotypes were at the 400-bands level. During the course of this study, it was realized that a small deletion in the proximal 15q could be easily overlooked when a mitotic spread around 400-bands or less per haploid-set level was used. There was no distinct difference in the clinical features of patients with interstitial deletion and those with a normal karyotype. Two cases in the latter group lacked some of the typical features of the former group, e.g. poor fetal vigor, neonatal feeding difficulty, hypotonia, and delayed motor development.
Five cases of poisoning by indigenous mushroom Hikageshibiretake (Psilocybe argentipes) are reported. As this mushroom contains psilocybin, in general, clinical features were similar to those seen by pure psilocybin. Acute toxic stuporous state with complete amnesia in the culminating period occurred in one case, psychedelic state with dreamy consciousness in one case and psychotic adverse reactions with vivid visual hallucinations with consciousness in three cases. There were accompanied with anxiety and panic reactions to subjective experiences. Though these toxic effects were usually short-lived, for management of such patients it is important to recognize that horrible emotional reactions and other harmful behavioral problems can also occur.
Fetuses of outbred mammalian species can survive despite disparities of participants' histocompatibility antigens. During pregnancy, maternal immune response is supposed to be suppressed by various immune suppressive mechanisms. In order to know whether fetal lymphocytes can suppress the maternal immune reaction, we investigated the suppressive effect of human newborns' T lymphocytes. Newborn T cells were isolated from the umbilical cord blood by means of the sheep red blood cell rosette forming method and treated with mitomycin C. The newborn T cells were added as regulator cells to one way mixed lymphocyte reaction (MLR) using maternal lymphocytes, paternal lymphocytes and unrelated donor's lymphocytes. In a control study, adult T cells were treated in the same manner. In any combination of the MLR, the response was substantially suppressed by the addition of cord T cells in comparison with the response by the addition of normal adult T cells. The suppression was dose-related with an increasing number of regulator cord T cells. The suppressive activity of cord T cells showed radiation resistance until 1,000 rads irradiation. In the kinetics study, cord T cells were found to suppress the recognition phase of MLR. These results suggest that cord T lymphocytes suppress the mother's immunological reaction to the fetal antigens.
This study of girls with Turner syndrome (16 cases) and boys with XXY (two cases) and XYY (eight cases) constitutions was designed to follow the longitudinal growth and to analyze the psychologic findings and the sleeping patterns in them. The growth curves of stature in Turner syndrome were below the 3rd percentile at all ages. The mean of SDS of height in cases less than 5 years old was -2.38, that in cases older than 6 years old -2.40. The shoulders in Turner syndrome have a tendency to broaden with time, in contrast to the development of the pelvis, which is not so good. The height of 47,XYY boys from infancy to 9 years of age was between the 10th and 90th percentiles, with one exception in which the height was above the 90th after 8 years of age. Relatively common psychologic findings in cases with Turner syndrome were as follows: low responsiveness to emotional stimuli, poor emotional expressions, passive and negative mental attitudes, low activity and low productivity, narrow and limited interests, passiveness and common sense adaptability, lower impulsiveness and aggressiveness, etc. The circadian oscillation was observed up to 3 years of age with shifting of the sleeping time to the night period. After that, day sleep gradually tends to decrease with age. Age variation in day and night sleep of an XXY boy was compared with that of a normal child. This showed the normal variation in circadian rhythms in early childhood, although the infant with this syndrome slept more, both day and night, than normal. In early childhood, no difference was noted. Similar studies were performed on a child with the chromosomal aberration XYY and revealed the sleep parameters to be quite normal in infancy and early childhood.
