Distribution of circulating mononuclear cells in short-term PUVA-treated psoriatic patients and healthy subjects.

Peripheral blood mononuclear cells (PBMC), as defined by monoclonal antibodies (OKT3, OKT4, OKT8, OK 1, Leu 7, Leu 11b) were determined in 10 psoriatic patients and in 10 healthy subjects before and after administration of short-term PUVA therapy. A comparison of the mean baseline percentages of the two groups showed a statistically significant increase in Leu 7+ cells (p less than 0.001) as well as a slight increase in OKM1 and OKT8 positive cells in the psoriatic subjects. After 21 exposures, these subsets showed a reduction towards control values, while a significant increase in OKT3 and OKT4 positive cells (p less than 0.01) could be observed only in the control group. These results indicate that short-term PUVA therapy is associated with changes in PBMC subpopulations. This modification, however, does not necessarily imply a disturbance of immune system function, including natural killer activity.

Amniotic fluid content of glucose, C-peptide and insulin in normal and diabetic pregnancies.

Glucose, C-peptide and insulin concentrations in amniotic fluid were measured in 63 women, 19 of whom had insulin-dependent diabetes. The amniotic fluid glucose concentration was higher in the diabetic (37.4 +/- 4.1) than in the normal women (19.4 +/- 5.1 mg/dl; P less than 0.01). The amniotic fluid insulin concentration was higher in diabetic than non diabetic women (15.9 +/- 3.1 versus 8.9 +/- 2.1 microU/ml; P less than 0.01). The C-peptide concentration was also higher in the amniotic fluid of diabetic women than normal (1.60 +/- 0.66 vs 0.26 +/- 0.15 nmol/l; P less than 0.001). The lecithin/sphingomyelin (L/S) ratio in the amniotic fluid was greater than 2 in 61 mothers; only two diabetic patients had an L/S ratio lower than 2. Five neonates of diabetic mothers were macrosomic and three of these were also hypoglycaemic. A negative correlation was observed in diabetic women between insulin and C-peptide concentrations in the amniotic fluid and Apgar score of the newborn infants both at 1 and 5 minutes; no correlation was found on the contrary with the amniotic glucose. In the prognostic evaluation of the fetus with a diabetic mother one must consider many parameters. Among these more consideration must be given to the levels of C-peptide and insulin in amniotic fluid than to glucose level.

[Electrophoretic and immunological characteristics of non-histone proteins of human skin tumors. I) Methods].

The object of the research is to classify the nuclear proteins of the cellular nucleus of some types of human skin tumours (melanotic and amelanotic melanomas, squamous epitheliomas). Nuclear proteins were extracted from the nuclei with a saline buffer. Nuclear proteins were extracted from a melanoma step by step. After prolonged centrifugation to sediment the DNA and the large ribonucleoprotein complexes, the nuclear proteins were chromatographed on columns of Biorex 70+ to remove proteins were chromatographed on columns of Biorex 70+ to remove the histones. Nuclear proteins from a squamous epithelioma were injected into a rabbit to induce specific antibodies.

[Electrophoretic and immunological characteristics of non-histone proteins of human skin tumors. II) Results].

In this communication we present the immunoelectrophoretic profiles of non-histone proteins of the cellular nucleus of some types of human skin tumours. The results show that the nuclear non-histone proteins of different types of tumours are quantitatively different from one another. The immunoelectrophoretic profiles of the samples show that an antigenic affinity between the skin tumours concerns most of the nuclear proteins of each sample.

[Chromatography of membrane proteins in sodium dodecyl sulfate. I) Methods].

The purpose of this study is to classify the membrane proteins from some cutaneous tumours (basal cell epitheliomas, squamous epitheliomas, melanomas) and possibly detect the ones which are characteristic of each type of tumour. The microsomal, mitochondrial and nuclear fractions were purified following the usual techniques. The membrane proteins were solubilized with sodium dodecyl sulfate, treated with iodoacetamide and chromatographed in SDS-acrylamide slab-gel.

Membrane proteins in human skin tumors.

The microsomal, mitochondrial, and nuclear fractions of cutaneous tumors have been investigated. Cutaneous tumors were homogenized and microsomal, mitochondrial, and nuclear fractions were purified. The membrane proteins were solubilized with sodium duodecyl sulfate (SDS). The membrane lipids were removed and membrane proteins were solubilized again in a small volume of SDS solution and chromatographed in SDS-acrylamide slab-gel. The plates were stained with Coomassie Brilliant Blue to show protein bands. The preliminary results show that the electrophoretic profiles of microsomal proteins are characteristic of some tumors.

Lipid synthesis from the liver in patients with psoriasis.

In psoriatic patients with hyperlipidemia we studied the hepatic lipid synthesis from (1-(14)C)-acetate in human liver biopsy specimens in vitro by a thin-layer radio-chromatography. In psoriatics type IV (according to Fredrickson) a significant increase in (1-(14)C)-acetate hepatic incorporation especially into phospholipids (25%) and triglycerides (52%) was observed; in type IIb increased lipogenesis was phospholipids (24.5%), free cholesterol (44.4%) and triglycerides (29%). Abnormal lipid metabolism often coexists with glucose intolerance in psoriasis; no correlation between hyperinsulinemia and augmented (1-(14)C)-acetate incorporation into hepatic triglycerides was found.

Fetal origin of amniotic fluid insulin in the human mother.

To investigate the origin of insulin in amniotic fluid amniocenteses were carried out in pregnancies with live, dead and anencephalic fetuses. Amniotic fluid insulin of pregnant women bearing live fetuses was 9.0 +/- 2.1 microU/ml; in six women with dead foetuses amniotic fluid insulin was not detected. A significant positive correlation was observed between gestational age and the amniotic fluid concentration of insulin. In the amniotic fluid of the four women bearing anencephalic fetuses, the amount of hormone was within normal limits (10.0 +/- 1.4 microU/ml). Intravenous glucose administration (0.33 g/kg body weight) to the mother doses not influence levels of insulin in amniotic fluid, but brought about changes in amniotic fluid glucose concentration. These findings support the conclusion that human amniotic fluid insulin is of fetal rather than maternal origin.

Urbach-Wiethe disease. Light and electron microscopic study.

A case of lipoid proteinosis was subjected to clinical, histologic and electron microscopic study. Palpebral biopsies showed a large dermal infiltration composed of closely interwoven granulo-filamentous material in the superficial and deep dermis. Collagen fibers were normal. Fibroblasts seemed to play an active role in the synthesis of this accumulating substance. Around the dermal capillaries there was a prominent concentric multilamination of the basal lamina which was considered a reaction of the endothelial cells to the irritation caused by compression of the accumulating substance.

Age-related mitochondrial lipoperoxidation in human skin.

Mitochondria were isolated from the epidermis of human subjects of different ages by a procedure that included pretreatment of the tissue with bacterial protease before homogenization. That epidermal mitochondria catalyze enzymatic lipid peroxidation in the presence of NADPH and chelated iron (ADP-Fe3+) is indicated by the production of malonaldehyde. The process does not appear to be influenced by age, and it is strongly enhanced in preparations containing a small amount of melanin.
