Functional restoration for recurrent laryngeal paralysis: an experimental study.

The purpose of this investigation was to exclude abductor disturbance and to maintain phonation during recurrent laryngeal nerve paralysis through a new surgical design. This new method consists of implantation of the ansa hypoglossi and sternothyroid muscle pedicle which has been shown to provide sufficient abductor function in a recent publication, and the selective severance of the abductor branch after neurorrhaphy of the recurrent laryngeal which is shown to be a sufficient adductor function in another study. The recovery of sufficient abductor function during inspiration and dyspnea was 67% and 89% of adductor function with phonation. However, after cutting an adductor nerve branch of the recurrent laryngeal nerve in cases of insufficient abduction, the vocal cord on the operated-on side regained abduction during inspiration. The effect of the ansa hypoglossi and sternothyroid muscle pedicle is also discussed.

Secretion of a potassium-rich fluid by the secretory coil of the rat paw eccrine sweat gland.

1. It is already known that the rat paw eccrine sweat contains high K(+) (greater than 150 mM) and low Na(+) concentrations (less than 70 mM). The present study was intended to clarify the site of K(+) secretion within the sweat gland, namely, the duct or the secretory coil. In vivo paw sweat was first induced by systemic pilocarpine injection or nerve stimulation. Both K(+) and Na(+) concentrations were studied in relation to the sweat rate to determine indirectly whether there is ductal secretion or reabsorption.2. Both Na(+) and K(+) concentrations in paw sweat agreed with the previous studies but did not show any saturation-type flow dependence at the high sweat rate range.3. A method has been developed to isolate a single segment of the secretory coil and induce sweat secretion directly from it in an in vitro condition.4. In the presence of fresh serum (30%, preincubated for 30 min at 56 degrees C) in the incubation medium, stable secretory activity due to 10(-6)M-Mecholyl could be maintained for 40 min or longer. The primary sweat thus induced contained low Na(+) (30 mM) and high K(+) (160 mM) concentrations.5. In the secretory coil sweat in vitro, K(+) concentration decreased and Na(+) concentration increased as the secretory rate fell either spontaneously or after addition of atropine or cyanide.6. It remains to be studied whether auxiliary ductal secretion or reabsorption is present at low rates of sweating in the rat sweat gland.7. It was concluded that the secretory coil of the rat paw sweat gland is the major, if not the sole, site of K(+) secretion.

Neurorrhaphy of the recurrent laryngeal nerve.

The authors have experimentally performed neurorrhaphy and its modifications and nerve grafting with end-to-end anastomosis of the recurrent laryngeal nerve in dogs. Recovery of a neurorrhaphy of the recurrent laryngeal nerve resulted in the bizarre and spasmodic movement of the vocal cords. These effects were recorded by means of electromyography and 16 mm cinematography to demonstrate restoration of nerve muscle function and vocal cord movements. From these results, it was concluded that these single modes were not adequate for restoration of laryngeal function, however, some specific attention is given to a few trophic changes of the affected muscles after a period of ten months postoperative.

Late effects of fetal mice x-irradiated at middle or late intrauterine stage.

Mice of F1 hybrids of (C57BL/6xWHT) strains were exposed in utero to 200 R of X-rays at 12 or 16 approximately 18 days post coitum. Mice of both sexes were allowed to liver through their life span, and the induction of tumors, growth retardation, and induction of degenerative diseases were examined. A significant enhancement of lung, pituitary gland, and ovary tumorigenesis was observed in mice irradiated at the late intrauterine stage. Also, incidence of liver and skin tumors was increased slightly in this group, whereas thymic lymphomas were not induced. Persistent growth retardation, several congenital malformation, and amyloid degeneration were found in mice irradiated at the middle intrauterine stage. However, no increases incidence of tumors was seen in this group. Moreover, incidnece of lymphoreticular tissue tumors, lung tumors, and leiomyosarcomas of uterus was significantly decreased by irradiation at the middle intrauterine stage from the unirradiated control and the late intrauterine irradiated group.

Prediction of radiosensitivity of individual mice by multidimensional analysis.

Ten physiological indices on each mouse were measured before an exposure to ionizing radiation. The exposure of mice to 700R of X-rays induced 45% mortality within 30 days. A prediction of death or life of each mouse was done by a multidimensional quantification. The highest success rate of prediction was obtained by using all ten indices and it resulted in 74.1%. Accordingly, it was possible to predict death of individual mice exposed to a dose around LD50 (30) with a high rate of success. When one selected five indices out of the ten indices by some criterion, the success rate of prediction decreased to 67.1%.

Polynucleotide kinase from rat-liver nuclei. Purification and properties.

A polynucleotide kinase, which catalyzes the phosphorylation of 5'-hydroxyl ends of deoxyribonucleic acid in the presence of adenosine triphosphate, has been purified 260-fold with a yield of 14% from 0.15 M NaCl extracts of rat liver nuclei. The purified enzyme has a pH optimum of 5.5. The enzyme is reversible inhibited by p-chloromercuribenzoate. The S0.5 value (ligand concentration required for a half-maximal activity) for ATP is 2.5 muM. A bivalent cation is essential for the reaction and S0.5 values for Mg2+, Ca2+ and Mn2+ are 3.3 mM, 4 mM and 0.05 mM respectively. Pyrophosphate remarkable inhibits the activity with I0.5 value (ligand concentration required for a half-maximal inhibition) of 0.2 mM, and sulfate, with I0.5 of 0.5 mM, whereas phosphate weakly inhibits the activity with I0.5 of about 20 mM. An apparent molecular weight of the purified enzyme is estimated to be 8 X 10(4) by gel filtration on a column of Sephadex G-150, and the Stokes radius of the enzyme molecule is shown to be about 0.36 nm. Sucrose density gradient centrifugation reveals that the enzyme has a sedimentation coefficient of about 4.4 S.
