Production of [14C]patulin by Penicillium patulum.

Factors affecting the production of [14C]patulin from [1-14C]acetate by replacement cultures of Penicillium patulum have been investigated. Incorporation of [1-14C]acetate into patulin reached a maximum with 6- to 8-day-old cultures incubated at 28 degrees C for 8 h in a replacement medium containing 0.1 M glucose, inorganic salts, and undiluted [1-14C]acetate. The specific activity of [14C]patulin obtained from this method was 34 mCi/mmol when 0.5 mCi of [1-14C]acetate was supplied to the replacement medium.

Production of antibody against aflatoxin B1.

Antibody against aflatoxin B1 was obtained after one multiple-site injection of bovine serum albumin-aflatoxin B1 conjugate into rabbits. The antibody has greatest binding efficiency for aflatoxin B1, less efficiency for B2, G1, and Q1, and least for aflatoxicol, G2, and M1. Sterigmatocystin, coumarin, and 4-hydroxycoumarin did not give a cross-reaction with the antibody. The sensitivity of the binding assay for detection of aflatoxin B1 is in the range of 0.2 to 2.0 ng per 0.5-ml sample. Detailed methods for the preparation of the conjugate, production of immune serum, and methods for antibody titer determination are described.

Preparation and characterization of aflatox-n B1-1-(O-carboxymethyl) oxime.

A method is described for the preparation and purification of aflatoxin B1-1(O-carboxymethyl) oxime from aflatoxin B1. The overall yield was about 73-83%. The new aflatoxin B1 derivative was characterized by mass, ultraviolet, infrared, and nuclear magnetic resonance spectral analyses, and was nontoxic to 8-day-old chicken embryos when tested at a concentration of 3.48 microgram/egg.

Production of antibody against ochratoxin A.

Antibody against ochratoxin A was obtained after repeated injection of different protein-ochratoxin A conjugates to rabbits. Among many protein-ochratoxin conjugates tested, bovine serum albumin-ochratoxin A was found to be the best antigen. The antibody is specific for ochratoxins A, C, and T, but is not specific for ochratoxins B, alpha, and other coumarin derivatives. The sensitivity for ochratoxin A detection using a binding assay is in the range of 0.5 to 10 ng/0.5-ml sample. Detailed methods for the preparation of protein-ochratoxin conjugates, preparation of immune serum, and methods for antibody titers are described.

Stability of aflatoxin B-1 and ochratoxin A in brewing.

The stability of aflatoxin B-1 and ochratoxin A in brewing was investigated by adding the purified toxins to the raw materials at 1 and 10 mug/g levels during mashing in a conventional micro-brewing process. The results indicate that both toxins are stable to heat and are insensitive to cooker mash treatment. Both mycotoxins were partially removed in the mashing and brewing processes. About 14 to 18% and 27 to 28% of the added toxins were found in the final beers brewed from starting materials containing 1 and 10 mug, respectively, of either toxin per g. The possible route of transmission of mycotoxins into beer is discussed.

Fate of ochratoxin A in brewing.

The fate of ochratoxin A in brewing was investigated by adding (3H)ochratoxin A to the raw materials at 1- and 10-mug/g levels during mashing in a conventional microbrewing process. The results indicated that large portions (28 to 39%) of the added toxin were recovered in spent grains, with less recovery in the yeast (8 to 20%) and beer (14 to 18%). About 38 and 12% of the added toxin at levels of 1 and 10 mug/g, respectively, were degraded during brewing.

Reduction of aflatoxins B1 and B2 with sodium borohydride.

Reduction of aflatoxin B1 and aflatoxin B2 with sodium borohydride quantitatively yielded new fluorescent derivatives, designated as aflatoxin RB1 and aflatoxin RB2. Mass spectrometric data showed that RB1 and RB2 were trihydroxy derivatives of B1 and B2, respectively. Nuclear magnetic resonance analysis revealed that new chemical shifts were present in aflatoxins RB1 and RB2 in addition to those of the parent aflatoxins. The new compounds had lower melting points and different ultraviolet and infrared spectra compared to aflatoxins B1 and B2 and the monohydroxy derivative aflatoxicol. They were lses toxic to chick embryos than the parent toxins. Since the reduction yields were quantitative and since the reduction products could be detected at low levels comparable to those for B1 and B2, the reduction reaction could be used as a confirmatory test for both aflatoxins B1 and B2. Preliminary results obtained from gas-liquid chromatography (GLC) analysis of the trimethylsilyl derivatives of aflatoxins RB1 and RB2 indicated that these compounds could furnish the basis for developing an analytical GLC method for aflatoxins B1 and B2.

Byssochlamyopeptidase A, a rennin-like enzyme produced by Byssochlamys fulva.

The production of a rennin-like enzyme by Byssochlamys fulva varied considerably with the isolates tested. Among the seven isolates tested, NRRL 2260, IMI 83277, and N.Y. 1 were good enzyme producers. The enzyme produced by isolate IMI 83277 was purified approximately 20-fold after (NH(4))(2)SO(4) precipitation, diethylaminoethyl-cellulose chromatography and Sephadex G-100 gel filtration. The partially purified enzyme has a pH optimum at 2.9 and a temperature optimum around 60 C. The enzyme appeared to be relatively stable at 40 C between pH 3.0 and pH 6.85. A name, byssochlamyopeptidase A, was proposed for this new enzyme. The milk-clotting activity of byssochlamyo-peptidase A is dependent on pH and appeared to be minimal at pH 6.2 or above. No extensive proteolysis has been observed during the milk-clotting process. The non-trichloroacetic acid-precipitable nitrogen titration curve on skim milk was comparable to that catalyzed by animal rennet.
