[The rupture of surgical gloves in a general hospital].

We studied the incidence of rupture of surgical gloves used at a general hospital: A) new gloves (n = 114); B) gloves used at a major general, gynecological or urological operation (n = 258), and C) recirculating surgical gloves (n = 86). A special techniques based on transmission of an electrical signal through the rupture was used to identify rupture. Rupture was present in 8.8, 28 and 26% of surgical gloves in groups A, B and C, respectively. Type of surgery or surgeon involved did not influence rupture of gloves in group A. However, rupture increased in relation to the duration of surgery (40% incidence in operations lasting more than 90 min). Our results suggest the need for better quality control of new surgical gloves and glove change after 90 min of operation.

Secretory character of a group of isoproterenol-induced polypeptides in mouse parotid glands.

The secretory nature of the isoproterenol-induced mouse parotid polypeptides C, D, E, F, and G (molecular weights 64,000, 61,000, 51,500, 38,000, and 37,000, respectively) is documented. Polypeptides C, D, E, F, and G, accumulated in response to successive daily stimulations with isoproterenol, were detected in a fraction enriched in hypertrophic parotid acinar cells. These cells, characterized by an increased content of cytoplasmic granules, maintain a secretory responsiveness to isoproterenol, which has been evidenced by light microscopy, enzymatic analysis, and unidimensional SDS-polyacrylamide gel electrophoresis. Thus, a parallelism in the loss and recovery of both secretory granules, alpha-amylase and polypeptides C, D, E, F, and G, was observed. Moreover, after secretion stimulation, polypeptides C, D, E, F, and G were detected in the fluid collected directly from parotid gland cannulation. Given the secretory character of polypeptides C, D, E, F, and G, mechanisms explaining both their progressive accumulation along the chronic administration of isoproterenol, as well as their progressive disappearance observed after suspending that treatment, are discussed.

Interaction between noradrenaline and diltiazem in rat isolated atria.

1. The present study describes the effects of diltiazem (DIL) on noradrenaline (NA) activity in the isolated right atria of the rat. 2. Interactions between DIL and NA showed that DIL significantly shifted the concentration-response curve of NA to the right in a concentration-dependent manner. 3. The antagonist between DIL and NA is not consistent with a competitive antagonist since the slope is significantly lower than the unity. 4. A high calcium solution (4.5 mM) significantly increased the chronotropic effects of noradrenaline. 5. These results suggest that influx of calcium through calcium antagonist sensitive channels may be involved in linking of chronotropic responses to activation of alpha 1-adrenoceptors.

The effect of diazepam on ventricular automaticity induced by a local injury. Evidence of involvement of "peripheral type" benzodiazepine receptors.

The effect of diazepam on ectopic cardiac automaticity has been examined in rats. We also investigated whether "central" or "peripheral type" benzodiazepine receptors are involved, as well as the role of calcium, on the possible effect of diazepam, by studying the interaction of this drug with either GABA, picrotoxin, RO 15-1788, PK 11195, diltiazem or Bay K 8644. A local injury of the rat isolated right ventricle produced a sustained abnormal rhythm which was completely abolished by diazepam (30-50 microM). This effect was not modified by the presence of either GABA (100 microM) picrotoxin (2 microM) or RO 15-1788 (5 microM) but it was reduced by the antagonist of "peripheral type" benzodiazepine receptors PK 11195 (0.1 microM). On the other hand the calcium channel blocker diltiazem (5 microM) and the calcium channel activator Bay K 8644 (3 nM), respectively, potentiated and reduced the effect of diazepam. It is concluded that diazepam effectively reduces ectopic cardiac automaticity in the rat. The "central type" benzodiazepine receptors are not involved in this effect, but it seems to be, at least, partially mediated by "peripheral type" receptors and is a calcium-dependent phenomenon.

Theophylline antagonizes the effect of diazepam on ventricular automaticity.

The effect of diazepam on ventricular automaticity induced by a local injury was investigated in the isolated right ventricle of the rat in the presence of the diazepam central antagonist, RO 15-1788, and of the adenosine antagonist, theophylline. Theophylline but not RO 15-1788 antagonized the inhibitory effect induced by diazepam on ventricular automaticity. The inhibitory effect of adenosine but not that of 2-chloroadenosine was potentiated in the presence of diazepam. The results suggest that the inhibitory effect of diazepam on ventricular automaticity results from the inhibition of adenosine uptake.

A mutation affecting basal body duplication and cell shape in Paramecium.

The thermosensitive mutant sm19 of Paramecium tetraurelia undergoes a progressive reduction in cell length and basal body number over successive divisions at the nonpermissive temperature of 35 degrees C. In spite of these defects, sm19 cells retain the same generation time as wild-type cells at 35 degrees C. Cytological observations at both electron and light microscopy levels reveal no other perturbation than the rarefaction of basal bodies and the rare (3%) absence of one or two microtubules in basal bodies or ciliary axonemes. The temperature-sensitive period, during the last 30 min of the cell cycle, corresponds to the phase of basal body duplication. Upon transfer back to the permissive temperature, all basal bodies are normally duplicated. The mutational defect is transiently restored by microinjection of wild-type cytoplasm or of a soluble proteic fraction from wild-type cell homogenates. Altogether, the cytological and physiological data support the conclusion that the sm19+ gene codes for a diffusible product required for the initiation of basal body duplication and would thus be the first identified gene involved in this process. Our data also indicate that in Paramecium basal body number is not coupled with control of the cell cycle, but helps determine the shape of the cell via the organization of the cytoskeleton.

The effects of propranolol stereoisomers on nomotopic and ectopic cardiac automaticity in the rat.

The effect of the two isomers of propranolol was tested on nomotopic and ectopic cardiac automaticity in the rat. The spontaneously beating isolated right atria of the rat was used as a model for studying the effect of these drugs on nomotopic automaticity. Ectopic automaticity was induced by local injury of the isolated right ventricle. Both dextro and levopropranolol reduce nomotopic and abolish ectopic automaticity (ID50 was 0.17 microM for d-propranolol and 0.96 microM for 1-propranolol) in the rat heart. We conclude that the membrane stabilizing activity, which is present functionally in both stereoisomers, plays a significant role in the effect of these drugs on cardiac automaticity. On the other hand, the property of competitive blocking of beta-adrenergic receptors, which mainly resides in the levoisomer of propranolol does not seem to be essential for this effect.

[Chemotherapy of advanced cervical carcinoma].

The authors describe a therapeutic trial of advanced cervical carcinoma using adriamycin and Methotrexate (84 cases), cis-platinum (9 cases), bleomycin and mitomycin C (18 cases), and a combination of cis-platinum, bleomycin and mitomycin C. Good results were never obtained.

[Tumor markers in cervical carcinoma].

The value of seven tumor markers to differentiate preinvasive and invasive gynecologic tumors is studied. CEA beta -hCH. alpha -FP and alpha-glycoprotein/prealbumin quotient allow such differentiation, while TPA, LDH-V and PRL have no value as markers of the gynecologic tumors and to not distinguish between preinvasive and invasive tumors.

[Our experience in the surgical treatment of cervical carcinoma].

The authors study 60 cases of invasive carcinoma of the cervix uteri belonging to several tumor stages. They analyze their past history, histology, lymph node invasion and symptomatology. The authors indicate the employed therapy and the five year survival rate, that was 82.3% for stage Ib tumors, 0% for stage IIA and 78.7% for stage IIB. The data for stage IIA are not significant statistically because the small number of the studied cases.

Relation of simple clotting tests to clotting factor levels in liver disease.

The prothrombin time with Manchester and ox thromboplastins, the 'P & P' test of Owren and Aas, the partial thromboplastin time, the thrombin time and assays for factors II, V, VII, VIII:C, IX and X were performed by one observer in 18 patients with liver disease and 27 normal subjects; the prothrombin time and partial thromboplastin time were also carried out on another 28 similar patients by various observers. Routine liver function tests were measured in all patients. The prothrombin time with Manchester thromboplastin was well correlated with other clotting tests, and performing the other tests did not add to the information. Discriminant function analysis confirmed that clotting tests did not distinguish between different types of liver disease. Correlations between clotting tests and liver function tests reflected liver cell damage but were also influenced by acute phase reactions.
