Influence of some mononucleotides and their corresponding nucleosides on the metabolism of carbohydrates in the isolated rat diaphragm muscle.

1. The influence of ATP on glucose metabolism was studied in the isolated rat diaphragm; it was shown that ATP increases the oxidation of glucose and the aerobic conversion of glucose into lactate, whereas it decreases glycogen synthesis. There was no influence of ATP on the anaerobic formation of lactate from glucose. 2. A maximum effect of ATP on the oxidation of glucose (about 160% increase) was obtained in the presence of 10mm-ATP; in the presence of 2mm-ATP the effect was about 65%, and was approximately constant from 10 to 90min. incubation period. 3. In a phosphate-free tris-buffered medium the oxidation of glucose was considerably decreased, but the percentage stimulation by ATP was about the same as in a phosphate-buffered medium. 4. ATP was shown to increase the oxidation of fructose, glucose 6-phosphate, glucose 1-phosphate, fructose 1,6-diphosphate and, to a much smaller extent, pyruvate. 5. ADP stimulated the oxidation of glucose to the same extent as ATP at a concentration of 2mm and the effect with AMP was only slightly less; IMP and adenosine had only a small stimulatory effect at this concentration, whereas inosine had no effect.

The role of dopaminergic agonists in congestive heart failure.

Congestive Heart Failure is a clinical syndrome characterised by myocardial dysfunction and sympathetic activation. Plasma norepinephrine (NE) levels have been related to the poorest survival. Large-scale clinical trials have proved the clinical benefits of Angiotensin Converting Enzyme inhibitors in reducing the risk of death and hospitalisation. However, mortality remains high in the treated group underlining the need to explore new therapeutic approaches. Specific activation of peripheral dopamine receptors exerts profound hemodynamic effects and neurohormonal control such as peripheral and renal vasodilation, diuresis and natriuresis and inhibition of NE release. Z1046, a mixed D1/D2-like agonist, reduces peripheral and renal vascular resistance increasing renal blood flow. In anaesthetised dogs the compound strongly reduces plasma NE without increasing plasma renin activity and plasma aldosterone. The inhibition of NE could be the basis of Z1046 potent cardioprotective effect observed in a dog model of myocardial ischemia and reperfusion, in which the severity of ventricular arrhythmias was markedly reduced, resulting in higher survival. These findings suggest that chronic oral treatment with specific dopaminergic agonists is able to alleviate the hemodynamic burden on the myocardium, and to suppress the sympatho-adrenal activity.

Prevalence of opiate use among young men in Italy, 1980 and 1982.

An epidemiological study on opiate use among young men undergoing examination for army draft was carried out in 1980 and 1982. The following six cities and surrounding provinces were selected for the study: Bologna, Florence, Genoa, Milan, Rome and Turin. There were 15,266 respondents in 1980 and 15,874 in 1982. For each respondent, a urine sample was tested for the presence of opiates and a questionnaire on demographic and socio-economic variables was completed. The study revealed 247 opiate cases with a prevalence rate of 1,618 per 100,000 in 1980, and 339 opiate cases with a prevalence rate of 2,136 per 100,000 in 1982. The number of opiate cases and demographic and socio-economic characteristics of respondents varied greatly from province to province. The group of unemployed respondents had significantly higher prevalence rates than the other occupational groups. A methodology was used that made it possible to estimate the number of opiate abuse cases that did not come to the attention of the authorities. On the basis of the study it was estimated that there were 68,000 opiate users in 1980 and 92,000 in 1982 in the entire country.

Further studies on rat, mouse and guinea pig muscle esterase activity.

Esterase activity was investigated in several skeletal muscles of different rodent species employing as substrates a series of alpha and beta naphthyl esters of different aliphatic chain lengths. Results with these chemically homogenous substrates show there is marked heterogeneity in the levels of esterase activity in several skeletal muscles of the same animal and among the same muscles from different species depending also on the sex, and the type and stereospecificity of the substrate.
