Response of human gingival fibroblasts to prostaglandins.

The effects of various prostaglandins (PGs) on the functions of human gingival fibroblasts (Gin-1 cells; ATCC CRL 1292) were examined by phase-contrast microscopy, cell-counting and radioautographic experiments. Tested PGs were PGA1, PGA2, PGB1, PGB2, PGD2, PGE1, PGE2, PGF1 alpha, PGF2 alpha, PGI2, 6-keto-PGF1 alpha, 9 alpha-11 alpha-methanoepoxy-PGF2 alpha, and thromboxane (TX) B2. PGA1 and PGD2 at 30 microM caused morphological deformation of Gin-1 cells. All the PGs tested at 30 microM suppressed the proliferation of Gin-1 cells in the logarithmic growth phase. Furthermore, all the PGs tested at 10 microM suppressed DNA synthesis, collagen synthesis, and noncollagenous protein synthesis in confluent Gin-1 cells, while exerting no effect on GAG synthesis. The concentrations of PGs used are beyond those found in healthy gingiva. However, in periodontitis the local concentrations of some PGs within the gingiva are expected to be extremely elevated beyond the physiological level. These results suggest that PGs may play an important role as a negative regulator in metabolism and some pathologic gingival conditions by suppressing the functions of gingival fibroblasts.

Isolation, purification, and partial characterization of an autocrine periodontal ligament cell chemotactic factor.

Periodontal ligament (PDL) cells are believed to play a critically important role in the regeneration of the periodontium. We have suggested that polypeptide growth factors can enhance periodontal regeneration by stimulating PDL cell chemotaxis and mitogenesis. This manuscript describes the identification of a novel chemotactic factor isolated from human PDL cells which we named PDL-CTX. PDL-CTX induces the directed migration of human PDL cells in vitro and was found to be a more potent chemotactic agent than other known growth factors. Additionally, PDL-CTX has no chemotactic effect on gingival fibroblasts or gingival epithelial cells. Both tryptic digestion and boiling abolished PDL-CTX's biological activity. The designed purification method included Mono-S cation exchange, heparin-sepharose affinity, and microbore reverse-phase HPLC. The purified factor has a relative molecular weight of approximately 7000 daltons based on sodium dodecyl sulfate (SDS) gel analysis. The amino acid composition and partial amino acid sequence were determined from HPLC-purified material. These were determined to be unique. Further investigation of the biological functions of PDL-CTX on PDL cells and other ligament cells should help improve our understanding of ligament repair.

Clinical and laboratory studies on a patient with early onset periodontitis and her family members. A case report.

Extensive clinical, microbiological, hematological, and immunological studies were performed on a patient with early onset periodontitis (EOP) and two other members of the family. The proband, a 27-year-old female, had early onset periodontitis and a high level of serum rheumatoid factors (RF) with no diagnosable medical disease. Her mother had lost all her teeth at the age of 50 because of advanced periodontitis, while her elder sister was unaffected by periodontitis. Neither the proband's periodontally-affected mother nor her unaffected sister exhibited a detectable level of RF. In this study, we examined: 1) serum immunoglobulin G (IgG) antibody titers against putative periodontal pathogenic bacteria; 2) peripheral neutrophil functions; 3) phenotypic analyses of peripheral lymphocyte subpopulations; and 4) peripheral lymphocyte functions (T cell proliferative activity, ability of cytokine [interleukin (IL)-2, tumor necrosis factor-alpha, interferon-gamma, IL-6 and IL-8] and IgG and IgM productivity). High antibody titers to Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, and Campylobacter rectus were detected in the sera of the proband, as were high serum antibody titers to P. gingivalis in the mother and to C. rectus in the unaffected sister compared to the non-periodontitis affected subjects. The proband also showed enhanced neutrophil chemotaxis; a high percentage of pan-B cells; and high productivity of IL-6, IgG, and IgM compared to individuals who were not periodontally affected. The mother showed slightly low helper/induced T cells (Th/i) suppressor/cytotoxic T cells (Ts/c) ratios due to the elevated count of Ts/c, and high IFN-gamma productivity compared to control subjects.(ABSTRACT TRUNCATED AT 250 WORDS)

[Effect of an additional cathode on the electrolytic-polishing of titanium specimens with non-aqueous electrolyte].

The purpose of this study was to polish electrolytically a large maxillary denture base-shaped titanium specimen with non-aqueous electrolyte. The test specimen was prepared by bending a sheet of titanium plate. An additional cathode was made by cutting off part of a cylindrical cathode. The effects of the shape and position of the additional cathode were investigated. Selected factors of the additional cathode were as follows: the connecting point between the additional cathode and the cylindrical cathode, the end position of the additional cathode, and the distance between the additional cathode and the titanium specimen. An acceptable polished titanium surface was obtained when the connecting point was below the electrolyte surface, the end position of the additional cathode was at a similar level to the bottom of the specimen, and the distance between the additional cathode and the specimen was 10 mm.

Mitogenic and chemotactic responses of human periodontal ligament cells to the different isoforms of platelet-derived growth factor.

A major focus of studies that center on regeneration of the periodontium is to determine the efficacy of the use of polypeptide growth factors. Platelet-derived growth factor has been reported to be a possible agent for clinical use. PDGF has various isoforms. Therefore, we decided to study the mitogenic and chemotactic responses of human periodontal ligament (PDL) cells to recombinant human PDGF-AB, AA, and BB. Addition of each isoform of PDGF to in vitro mitogenesis assays induced PDL cell proliferation in a dose-dependent manner. The maximum mitogenic effect was evident at the concentration of 100 ng/mL. In these assays, PDGF-BB was found to be the most potent mitogen. PDGF-AB elicited an intermediate response, and PDGF-AA was the least effective. The results of chemotaxis assays closely parallel those of the mitogenesis assays. PDGF-BB exhibited the most potent chemotactic effect. The maximal effect was observed at 10 ng/mL. The findings of these experiments indicate that PDGF-BB is more effective than the other isoforms in promoting mitogenesis and chemotaxis of PDL cells in vitro, and may therefore be a suitable ethical pharmaceutical for use in periodontal regeneration procedures.

Unique intronic variations of HLA-DQ beta gene in early-onset periodontitis.

Human leukocyte antigen (HLA) class II beta chain plays an important role in the recognition of foreign antigens in immune reactions. Different forms of immune reaction may be concerned with initiation and progression of infectious diseases such as periodontitis. In this study we examined the frequency of HLA class II serotype and the variation of HLA class II beta gene in periodontitis patients. HLA serotypic frequencies in 70 Japanese patients with periodontitis and 26 individuals with periodontal health were examined. No HLA serotype specific to any type of periodontitis was observed. In order to detect differences among some HLA serotypes, restriction fragment length polymorphism (RFLP) analysis was undertaken with cDNA probes for HLA-DR beta and HLA-DQ beta genes in 20 subjects (15 patients and 5 healthy individuals). Atypical BamHI and EcoRI restriction sites were found in the HLA-DQ beta gene from 3 patients with early-onset periodontitis. In addition to these 20 subjects, an additional 80 subjects (40 patients and 40 healthy individuals) were screened for the atypical BamHI restriction site using the polymerase chain reaction method. It was detected in 7 patients with early-onset periodontitis, 1 patient with adult periodontitis, and 3 healthy subjects. No clinical differences except age were found between patients with this gene variation and other patients. Interestingly, all 3 healthy subjects with this gene variation were from subjects whose family members developed early-onset periodontitis with the gene variation. Atypical BamHI and EcoRI restriction sites and 41-nt repeated sequence were found in the intron before the third exon of HLA-DQB gene. These results suggest that these intronic gene variations may be useful as gene markers for a subpopulation of early-onset periodontitis and might affect immune reactions such as antigen recognition.

[Variables effecting casting accuracy of quick heating casting investments].

Recently, several new products of investments for "quick heating" have been put on the Japanese market. The total casting procedure time for this quick heating method involves only one hour; 30-minutes waiting after the start of mixing before placing the mold directly into the 700 degrees C furnace and 30-minutes heating in the furnace. The purpose of this study was to evaluate two variables effecting casting accuracy using these new investments. The effect of thickness of the casting liner inside the casting ring and the effect of waiting time before placing the mold into the 700 degrees C furnace were evaluated. A stainless-steel die with a convergence angle of 8 degrees was employed. Marginal discrepancies of the crown between the wax patterns and castings were measured. The size of the cast crown became larger when the thickness of the ring liner was thick and when the waiting time before placing the mold into the furnace was long. These results suggest that these new investments have the advantage of providing sound castings using short-time casting procedures. However, it is necessary to pay careful attention to the casting conditions for obtaining reproducible castings.

Oblique cine MRI for the evaluation of aortic regurgitation: comparison with cineangiography.

The results of oblique cine magnetic resonance imaging (MRI) through a long axis slice of the left ventricle in 40 patients was compared with cineangiography in order to determine the usefulness of cine MR imaging for the evaluation of aortic regurgitation (AR). A diastolic dark low-intensity flow signal extending from the aortic valve into the left ventricle was seen in all 30 patients with AR, and in none of the 10 patients without AR. The length and area of the low-intensity signal due to the regurgitant flow were planimetered, and the ratio to the length and area of the left ventricle was calculated. The best correlation with angiography was with the ratio of the area of the low-intensity signal (ALIS) to the area of the left ventricle (ALV); it correctly classified the degree of regurgitation in 28 of the 30 patients (93.3%). The ALIS/ALV ratio was under 25% in all 11 patients with angiographic mild AR, 26-50% in 10 of the 11 patients with moderate AR, and over 51% in 7 of the 8 patients with severe AR. Thus the results point to a high degree of correlation between long axis oblique cine MRI and cineangiography in the identification of AR and in providing a semiquantitative estimation of its severity.

Interleukin-8 is a major neutrophil chemotactic factor derived from cultured human gingival fibroblasts stimulated with interleukin-1 beta or tumor necrosis factor alpha.

Inflammatory mediators produced by cells in the gingiva have been implicated in the initiation and progression of periodontal disease, a common infectious disease. In this study, we examined the biological activity of neutrophil chemotactic factors and the kinetics of expression of interleukin-8 (IL-8) mRNA derived from normal gingival fibroblasts in response to inflammatory mediators in an in vitro model. Gingival fibroblasts stimulated by either recombinant human interleukin-1 beta or recombinant human tumor necrosis factor alpha produced neutrophil chemotactic factors after 4 h, whereas expression of cell-derived IL-8 mRNA was detected within 1 h after stimulation. Furthermore, in a neutralization assay, rabbit anti-recombinant human IL-8 antiserum inhibited neutrophil chemotactic activity to basal levels. These results provide evidence that gingival fibroblasts synthesize potent chemotactic factors such as IL-8 in the presence of the inflammatory mediators interleukin-1 beta and tumor necrosis factor alpha. The activity of these factors may contribute to neutrophil-mediated processes in the pathogenesis of periodontal disease.

Primary angiosarcoma of the right atrium detected by magnetic resonance imaging.

A 20-year-old woman with a primary angiosarcoma of the right atrium is reported. The patient had a cardiorespiratory arrest due to cardiac tamponade with bloody pericardial effusion. Magnetic resonance imaging (MRI) revealed a tumor, which was corroborated by selective coronary angiography. Open-heart surgery was performed. The tumor relapsed however, and she died four months after operation. The tumor was undetectable by echocardiography, but MRI demonstrated a heterogeneous mass with focal areas of high- and low-signal intensity in the right atrium, suggesting that MRI may allow characterization of cardiac tumors.

A new procedure of the tensile fatigue test for dental materials.

A new tensile fatigue test using a stainless steel screw was introduced. A transparent PMMA rod was selected as the test material. A hole was drilled and tapped into the rod; a screw was put into the rod with dental cement or without cementation. Three types of cyclic loads were applied to the test piece; the number of times the load was applied when the initial crack was observed and when the final fracture occurred were recorded. Surface observation showed that actual fatigue fractures occurred in the test piece. The preferred applied load waveform was the cyclic haver-triangles load because of the small coefficient of variance. Fatigue strengths of test pieces with and without cementation were different. Cementation improved the fatigue properties, and the type of cement had an effect on the fatigue properties. Although the number of final fractures minus the number of initial cracks was the constant regardless of the type of cement used.

Development of a new investment for high-frequency induction soldering.

This study developed a new investment for induction soldering using high frequency induction heating. Ninety-five mass% magnesia clinker and 5 mass% dental stone were selected for the main constituents. The magnesia investment itself was scarcely affected by induction heating, so the addition of metal powders such as Fe, Ni, Co were investigated. Among these three powders, the addition of 10 mass% cobalt powder was most effective. This investment needed only 40 seconds of high frequency induction heating to go from room temperature to 900 degrees C without preheating. Thermal expansion of this investment in a vacuum atmosphere was 1.25% at 1000 degrees C. Dimensional changes during induction soldering were measured using German-silver and silver solder. When the new magnesia investment containing 10 mass% Co powder was used, the dimensional change was -0.2%. This contraction was less than when a magnesia investment without metal additives was used.

[A fundamental study of 99mTc-ethyl cysteinate dimer (99mTc-ECD)].

In vitro fundamental studies of 99mTc-ethyl cysteinate dimer (99mTc-ECD), a newly developed radiopharmaceutical for cerebral perfusion scintigraphy, were performed, and were compared with those of 99mTc-HM-PAO, which has been widely applied to clinical use. 99mTc-HM-PAO should not only be injected soon after reconstitution, but also be avoided mixture with saline or drugs such as Diamox, because of its radiochemical purity decreases with the passage of time and the addition of saline and Diamox. On the other hand, the radiochemical purity of 99mTc-ECD reached a constant level of 94-96% at 30 to 180 min after reconstitution, and it remained stable against the addition of saline and Diamox. Therefore, it was suggested that 99mTc-ECD could be injected from an already saline-infused venous-line or mixed with Diamox. However, early injection after reconstitution should be avoided, because 99mTc-ECD did not become stable enough to maintain sufficient radiochemical purity until 30 min after reconstitution. The present studies are suggested that the red blood cell component is largely related to the metabolism of 99mTc-ECD in blood. Furthermore, from study of the metabolic rate using rabbit blood, it was suggested that a species difference existed in esterase activity in blood.

Monoclonal antibody to a specific antigen from Wolinella recta ATCC 33238.

Four monoclonal antibodies (m-Abs) to a specific antigen (150-kDa protein antigen) isolated from Wolinella recta ATCC 33238 by acid extraction were obtained. The four antibodies were all of the IgG1 subclass and exhibited equally high specificity for the antigen. A battery of 14 strains or oral bacteria were screened for cross-reactivity with each mAb by ELISA and Western blot analysis. Weak cross-reactivity to some strains was observed which differed depending upon the mAb. Immuno-electromicroscopic studies were performed with a mAb which revealed that the 150-kDa antigen was localized to the cell surface of W. recta ATCC 33238.
