A statistically standardized muscle cell culture bioassay measuring the effect of swine serum on muscle cell proliferation.

We have developed a statistically standardized bioassay for quantifying the effect of swine serum on the proliferation rate of cultured L6 myogenic cells. The intra-assay coefficient of variation for this assay is 2.5%. Over 29 experiments, the relationship between the assays response to 2.5% control swine serum and its response to 2.5% serum plus 10(-7) M insulin is linear (r2 = .9). This relationship can be used to adjust experimental data obtained in different experiment to a common control value so that valid inter-assay comparisons can be made. The inter-assay coefficient of variation is reduced four- to five fold by this adjustment procedure. We believe this standardized assay provides a useful system for identifying and isolating unknown growth factors that affect muscle growth rate in an economically important species.

[Pericardial manifestations of Lyme disease].

Among cardiac manifestations of Lyme disease, pericardic involvement, as the main symptom, seems to be exceptional. We report two cases of pericarditis with typical clinical features. In both cases, pericarditis was the only clinical sign of the disease. Diagnosis was based on significant high serological antibodies levels with no evidence of any other etiological factor. Penicillin was administered and lead in one case to negativation of the serology. As cardiac involvement can occur without any preexisting cutaneous lesion, the frequency of pericardic involvement in Lyme disease is certainly underestimated.

Gut-associated lymphoid tissue and 1,2-dimethylhydrazine intestinal tumors in the rat: an histological and immunoenzymatic study.

The association between chemically-induced intestinal carcinoma and gut lymphoid patches was studied in 20 male Sprague-Dawley rats 7 months after the first of 16 weekly injections of 1,2 dimethylhydrazine (DMH). The lymphoid patches of DMH-treated rats and of 14 untreated control animals were systematically studied histologically on sections of "swiss-rolled" whole intestine. It was found that 78% of the small-intestine carcinomas and 73% of the colorectal carcinomas were associated with intestinal lymphoid patches. Furthermore, misplaced and often atypical glandular crypts were often found in the parafollicular or interfollicular areas of lymphoid patches, in treated as well as in control animals. These glands could be the origin of the lymphoid-patch-associated carcinoma. Immunohistological staining with monoclonal antibodies (MAbs) against T-lymphocyte antigens or anti-IgM serum labelling B lymphocytes clearly localized early carcinoma and atypical glands in the T-dependent, interfollicular and parafollicular area of lymphoid follicles. An MAb directed against la-antigen stained some well-differentiated carcinomas and some atypical glands found in control rats. On the other hand, lymphoid patches, when not invaded by a carcinoma, were not modified in their number, size, morphology or cellular composition in DMH-treated rats as compared to control animals.

[Portal circulation following Warren's operation. Long-term control using computerized tomography].

Computed tomography with contrast injection was carried out in 18 patients who had undergone a Warren procedure for portal hypertension due to cirrhosis of the liver more than five years previously. The results show that it is not possible to drain only a part of the venous portal territory. The portal circulation does not consist of two portions, with different pressure relationships. Pressure difference across the splenorenal anastomosis is greater than that into the mediastinal veins. Postoperative development of a hepatofugal circulation continues for a long period and is not confined to the early phase only. This phenomenon is, however, not uniform. In particular, there are variations in the extent of the collateral circulation and in the maintenance of liver blood flow.

Ocular manifestations of adult Niemann-Pick disease: a case report.

Niemann-Pick Disease Type B in a 33-year-old man is described. No family, racial or hereditary factors were noted in this case as in other reports in the literature. The ocular abnormalities in our patient were mild myopia, a mild generalized colour abnormality, and fundi showing a lipid ring-form opacity about the foveolas resulting in minimal visual impairment. Sphingomyelinase deficiency was found with the resulting deposition of phospholipid occurring most probably in the Müller cell. No neurological features were present and the patient was otherwise well.

Congenital lower nasolacrimal duct anomaly: a case report.

A congenital anomaly of the nasolacrimal system not previously described is reported. The apposition of the distal end of the nasolacrimal duct was not with the nasal cavity as normally occurs prior to birth but to the external skin. As a result the child presented with difficulty with tear drainage and consequent infection--"neonatal conjunctivitis" followed by a facial abscess at the left lower border of the nose. Recent embryological studies suggest that this anomaly is due to failure of complete separation and migration of the epithelial rod most distal from the surface ectoderm to the inferior meatus of the nasal cavity. This distal remnant of the nasolacrimal duct thus remains juxtaposed to the surface ectoderm of the face on the lateral aspect of the alar nares. The modern concept of the embryological development of the nasolacrimal system is reviewed.

Orbital cellulitis and preseptal cellulitis in childhood.

A study was made of 112 children with preseptal cellulitis, orbital cellulitis, subperiosteal abscess, and orbital abscess. These were consecutive admissions to the Royal Alexandra Hospital for Children, Sydney, during the period June 1976 to August 1985. The average age was 3 years 6 months. The clinical and radiological signs for each group were defined. Of the 112 children, 43.8% (with an average age of 3 years 3 months) had a clinical history of an upper respiratory tract infection. Trauma was a factor in 13.4% of patients. Haemophilus influenzae was recovered from blood culture in two patients and from conjunctival culture in seven patients. The average age of this group was 3 years 7 months. Plain radiographs taken of 53 patients revealed evidence of paranasal sinus disease in 29 (54.7%). Computerized tomography was performed in 14 patients. Of seven patients requiring surgical drainage of orbital or subperiosteal abscess (average age 7 years 2 months), five had clinical upper respiratory tract infection.

The effect of oestriol and tamoxifen on oestradiol induced prolactin secretion in anaesthetised rats.

In ovariectomized rats oestradiol and oestriol administered sc for 5 days increased basal plasma prolactin in a dose related manner to similar maximal values. The data fitted log dose-response curves. Oestradiol was about 12 times more potent than oestriol. These dose response relationships were retained on thyrotrophin releasing hormone (TRH) administration (50 ng iv, blood sampled 5 min post TRH). In addition, significantly higher maximal response values for both oestradiol and oestriol were now seen. The relative potency also increased in oestradiols favour. We subsequently examined the effect of co-administering oestriol with oestradiol on plasma secretory responses to a single dose of oestradiol in young male rats: Oestriol (30 micrograms) potentiated increases in basal plasma prolactin, TRH stimulated prolactin secretion and radioimmunoassayable pituitary prolactin in rats in response to a single sc injection of oestradiol (10 micrograms). Tamoxifen, administered once also potentiated these responses to a single sc injection of oestradiol. However, if the tamoxifen was administered sequentially i.e. 48 and 24 h before or 48 and 24 h before and simultaneous with the oestradiol the potentiation was completely reversed.

Peripheral neuropathy and myopathy in chronic alcoholism.

Nineteen chronic alcoholics referred for investigation of their medical complications and for alcohol re-education were subjected to quadriceps muscle biopsy and detailed peripheral nerve electrophysiological studies. Thirteen patients showed type II muscle fibre atrophy by histomorphometric analysis. The presence of atrophy did not correlate with peripheral neuropathy as determined by clinical examination or with objective indices of nutritional status. There was no association of muscle atrophy with neurophysiological evidence for neuropathy and both occurred independently of each other. It was concluded that chronic alcoholic myopathy was due to a toxic effect of ethanol on the muscle fibres and was not merely secondary to a peripheral neuropathy.

[Surgical treatment of snoring by correction of nasal and oropharyngeal obstruction].

32 patients with habitual snoring and 2 patients with obstructive sleep apnea syndrome underwent nasal and/or palatopharyngeal surgery. 12 patients with combined nasal septoplasty, submucous resection of the inferior turbinate bones and palatopharyngoplasty gained total relief from snoring. The same result was achieved in 10 children with removal of extremes of tonsillar and adenoid enlargement. From 7 patients with nasal surgery alone 4 experienced total improvement in the snoring, 3 perceived no long-term relief. 3 adults with PPP alone gained some change in their snoring. Uvulectomy in 2 adults had no influence on the snoring situation.

[Side effects of drugs on the larynx].

The ENT doctor is often confronted with pathological changes or functional disturbances of the larynx of uncertain aetiology. With regard to differential diagnosis various drugs should always be taken into consideration that have been prescribed by other specialists. This is pertinent particularly in three groups of pathological changes: Paralysis of the recurrent nerve, dyskinesia and laryngospasm. Exsiccation of the mucous lining, loss of kinaesthesia, development of local oedema and hyperplasia. Voice changes due to androgenic and anabolic steroids. The causative drugs and their mode of action are discussed.

[The 3-dimensional voice field].

Extension of the voice field (voice profile) by a third variable which characterizes the phonation has various advantages as well as drawbacks. Since the voice field evaluates the laryngeal function, the use of aerodynamic variables improve the representation of the laryngeal efficiency in the voice field. On the other hand, the measurement of these variables complicates the method. When voice quality measures (presence of singing formant, sharpness) are used as the third dimension, the limits of the voice field are unique references for the measures. However, the voice quality at the physiologic limits is not representative of the voice as produced in singing or speaking. Measures which characterize the "hoarseness" of the voice yield important results with respect to the perturbation of the voice production (jitter, shimmer, additive noise). The perturbation measures evaluate the laryngeal function at defined points in the frequency--intensity area. However, in forte phonation, the natural perturbation is reduced, in piano phonation, the perturbation is increased, and the results cannot be interpreted in relation to the perturbation magnitude within the voice field. Besides, the measurement of perturbation requires digital signal processing. If EMG data are used for the voice field, as it is shown for the M. vocalis, the voice field reveals interesting relationships between acoustic and physiologic features. For instance, in the piano phonation, the EMG signal power is constant on a low level. In the forte phonation, in the chest register, the EMG signal power shows a medium level and a high level in the head register. In the falsetto, however, the EMG signal power is low.(ABSTRACT TRUNCATED AT 250 WORDS)

Amiodarone-induced enhancement of doxorubicin and 4'-deoxydoxorubicin cytotoxicity to rat colon cancer cells in vitro and in vivo.

The mechanisms of the resistance of intestinal cancer to anthracyclines were studied on an experimental model of rat colon cancer cells. The accumulation of anthracyclines in the nucleus of living cancer cells was observed by fluorescence microscopy. This accumulation depended on both the capacity of anthracyclines to penetrate into the cell and the activity of an efflux mechanism extruding the drug from the cell. We found that 4'-deoxydoxorubicin was superior to 4 other anthracyclines in its capacity to penetrate into confluent colon cancer cells. Amiodarone, an antiarrhythmic agent used in cardiology, inhibited the efflux mechanism efficiently and increased the toxicity of anthracyclines to the colon cancer cells. Association of amiodarone and 4'-deoxydoxorubicin was able to cure 5 of 13 rats that had been inoculated i.p. previously with syngeneic colon cancer cells. This association could be of interest in the treatment of human colon cancer.
