[Study on mRNA differential display in Mycobacterium tuberculosis H37Rv and H37Ra].

OBJECTIVE: To study genetic differences between H37Rv and H37Ra by mRNA differential display, paving the way for obtaining virulence-related gene of Mycobacterium tuberculosis (MTB). METHODS: Total RNA was extracted from MTB H37Rv and H37Ra, mRNA differential display was conducted following reverse transcription, polymerase chain reaction, polyacrilamide gel electrophoresis and radiography. The differential bands were recovered, reamplified, sequenced, and then homologous query was done. RESULTS: 10 differential bands between H37Rv and H37Ra were recovered, 2 of which were homologous to H37Rv DNA after query from NCBI database of GenBank. CONCLUSIONS: mRNA differential display is an effective, sensitive method, can display differences between different strains intra- and extra-species and be used in searching Mycobacterium tuberculosis virulence-related gene. Obtained differential bands in this study may cover MTB virulence-related gene.

[Expression of bcl-2 gene in relation to histologic type and grade, and prognosis of breast cancer].

OBJECTIVE: To ascertain whether bcl-2 gene expression is related to histologic type and grade, and prognosis of breast cancer and its prognosis. METHODS: Expression of bcl-2 protein was examined immunohistochemically in 85 cases of breast cancer patients with > or = 5 years of follow-up. RESULTS: Expression of bcl-2 protein was detectable in 68.2% of breast cancer specimens. It was not related to histologic type, but to histologic grade of the cancer. In grade I, bcl-2 was positive in 95.2% of the specimens which was significantly higher than that in grade II (64.6%) and grade III (25.0%)(P < 0.01). The positive rate of bcl-2 expression was 75.9% in patients who survived > or = 5 years as compared to that (51.6%) in patients survived < 5 years (P < 0.05). CONCLUSION: Expression of bcl-2 occurs more frequently in well differentiated breast cancer. It is associated with better prognosis of the patients.

Study of total quantity control of air pollutants in planning industrial district.

The total quantity control of pollutant emissions for an industrial district is determined by coefficient (A) method (Qian, 1990). It is suggested that average daily concentrations of air pollutants should be estimated in relation with metrological parameters, such as wind directions, wind speed and atmospheric stability in the period of monitoring by Gaussian model (SEPB, 1991), and that the sources of pollution should be redistributed on the basis of the result of monitoring with a view improving local atmosphere environment.

[Recombinant human IGF-1 prevents type 1 diabetes in female non-obese diabetic mice].

OBJECTIVES: To study whether recombinant human (rhIGF-1) can prevent the onset of type 1 diabetes in female non-obese diabetic (NOD) mice, and its influence on their immune system. METHODS: Ten mice were injected intraperitonally (i.p.) rhIGF-1 37.5 micrograms.kg-1.d-1 at 4 weeks old for 2 weeks at treated group; and the same volume of normal solution (NS) was used in 10 mice as control group. All mice were killed at 40 weeks old or 2 weeks after the diagnosis of diabetes. Blood glucose of tail vein blood was measured weekly using a glucometer. At the end of the experiment, serum C-peptide levels and antibodies to glutamic acid decarboxylase (GAD) were detected with RIA and ELISA methods, respectively. T cell subsets of spleen were undertaken two-color FACS analysis using Cy-chrome anti-CD4 and FITC-anti-CD8 monoclonal antibodies; pancreatic histopathology and immunohistochemistry of T cell were conducted. RESULTS: Treatment with rhIGF-1 at 4 weeks old reduced the total incidence of diabetes (40% vs 90%, P < 0.05) and delay its onset [(26.25 +/- 3.68) weeks vs (21.6 +/- 6.7) weeks] compared with control group (P < 0.05). RhIGF-1 showed the growth effect on NOD mice, because the wet weight of spleen and pancreas of groups treated with rhIGF-1 were higher than that of control mice (P < 0.01, respectively). Only 3.24 (0.92 islets/mouse) was observed in the control group at death, but they increased to 13.2 (3.14 islets/mouse) in the rhIGF-1 group. The insulitis score decreased from 0.738 +/- 0.194 to 0.098 +/- 0.035 in the later group. A marked reduction of insulitis severity of pancreatic glands was also observed. A strong proliferative response of CD8+ T subsets in splenocytes occurred in rhIGF-1 treated mice, and the imbalance of CD4+/CD8+ subgroup in control mice (2.03 +/- 0.72) was corrected in the later group (0.53 +/- 0.22, respectively, P < 0.01). The lymphocytes infiltrating islets in the rhIGF-1 group showed a reduction of CD4+ cells, and they existed only outside of islets. CONCLUSIONS: These data demonstrated the preventive effects of rhtIGF-1 on the onset of type 1 diabetes and the severity of insulitis in female NOD mice. The mechanisms of these effects may be related to the induction of a CD8+ Ts cell response that inhibits the spontaneous development of autoreactive CD4+ T subsets response, or inhibiting the homing of CD4+ T cells to islets.

[Establishment of cisplatin-induced multidrug resistant human epithelial ovarian cancer cell line 3AO/cDDP and its expressions of multidrug resistance proteins].

OBJECTIVE: To establish multidrug resistant ovarian cancer cell line 3AO/cDDP by exposing parent cell line intervally and repeatedly to high-level concentration of cisplatin, and to detect expressions of lung resistance protein (LRP), multidrug resistance-associated protein(MRP) and P-glycoprotein (P-gp). METHODS: Using the corresponding dose calculated from clinical chemotherapy, we established 3AO/cDDP, with 3AO exposed intervally and repeatedly to high-level concentration of cisplatin at 10 micrograms/ml for 24 hours each time; LRP, MRP and P-gp expressions were detected with flow cytometry(FCM). RESULTS: 3AO/cDDP was established after 4.5 months with stable resistance and the drug resistant index was 1.62 which was similar to clinical resistance degree. Low expression levels of MRP and P-gp were found in both 3AO and 3AO/cDDP, and no statistical difference between the two lines (P > 0.05). However, LRP expression level in 3AO/cDDP was significantly higher than that in 3AO with FCM detection (P < 0.01). CONCLUSIONS: 3AO/cDDP, induced by exposing 3AO to high-level concentration of cisplatin intervally and repeatedly, is an ideal model for investigation of cisplatin resistance. Cisplatin resistance in 3AO/cDDP could be related to high LRP expression and be not associated with MRP or P-gp.

Immunosuppressive effects of rubidatum in mice.

AIM: To study the effect of rubidatum (Rub) on immune function in normal mice. METHODS: Serum lysozyme concentration (SLC) was measured using micrococcus lysodiekticus as a substrate. Delayed type hypersensitivity (DTH) was determined by measuring the thickness of the right hind footpad 24 h after the injection of 1 x 10(8) washed sRBC (50 microL 10% sRBC). Serum hemolysin concentration was determined by OD measuring at A540 after the serum was treated with 2-mercaptoethanol. Phagocytic function of peripheral leukocyte (Leu) were determined by the incorporated radioactivity of [3H]TdR. The hemolytic activity of plaque forming cell (PFC) was determined by measuring the lymphocytemediated hemolysis of sheep red blood cell in vitro. T- and B-lymphocyte transformation (TLT and BLT) were induced by phytohemagglutinin (PHA) and lipopolysacharide (LPS) respectively and measured by the incorporation of [3H]TdR. RESULTS: Rub 125, 500, 2000 mg.kg-1.d-1 p.o. to BALB/c (or NIH) mice decreased the SLC; inhibited the phagocytosing functions of peripheral leukocytes; diminished the hemolytic activity of PFC; decreased the HC50; inhibited the DTH reaction; and showed inhibitory effects on TLT and BLT. CONCLUSION: Rub has immunosuppressive effects on immune system in mice by affecting M phi, T, and B lymphocyte, which suggests that Rub has inhibitory effects on both nonspecific and specific immune function.

[Study on relationship between levels of leptin in maternal blood, amniotic fluid, and umbilical blood and newborn's weight].

OBJECTIVE: To explore the relationship between leptin's quantity in maternal blood, amniotic fluid, umbilical blood and maternal and newborn's weights. METHODS: Levels of leptin in maternal blood, amniotic fluid, and umbilical blood of 34 pregnant women were detected by radioimmunoassay (RIA) method. Results were analyzed by the correlative analysis. RESULTS: (1) Leptin can be detected either from maternal blood or amniotic fluid or umbilical blood. (2) There was an obvious correction between newborn's weight and leptin's quantity of maternal blood (relative coefficient r = 0.53, P < 0.01), amniotic fluid (r = 0.73, P < 0.01) and umbilical blood (r = 0.67, P < 0.01). (3) There was an obvious correction between maternal weight and leptin's quantity of maternal blood (r = 0.61, P < 0.01). (4) There was an obvious correction between leptin's quantity of amniotic fluid and umbilical blood (r = 0.72, P < 0.001). (5) There was not obvious correction between maternal weight and leptin's quantity of amniotic fluid and umbilical blood (r = 0.22, r = 0.18, respectively. P > 0.05). CONCLUSION: It may be of important significance in detection of leptin's quantity in amniotic fluid and maternal blood in pregnant women for predicting the weight and growth of newborns.

[The use of interposed colon for endothoracic anastomosis in esophageal surgery].

OBJECTIVE: To study the use of interposed colon for endothoracic anastomosis in esophageal surgery. METHODS: From May 1991 to December 1993, 28 cases were selected with carcinoma of the middle and/or lower segment of esophagus. After tumor resection, a segment of transverse colon with peduncle was anastomosed in its peristaltic direction to the two ends of the severed esophagus. The vagus nerve innervating esophagus and stomach was intact, the anastomical position of the esophagus-cardia hiatus and the stomach were preserved. RESULTS: No anastomosis leakage, reflux esophagitis or the symptoms caused by the endothoracic stomach occured in this group. The digestive function of the patients remained good. The 1-, 3-, 5-year survical rate was 96.6%, 92.8% and 71.4%, respectively. CONCLUSION: The interposed colon for endothoracic anastomosis is a comparatively ideal method for the reconstruction of esophagus following resection of cancer of mid and lower thind of the esophagus.

[Relationship between the expression of cell cycle regulators and microsatellite instability in colorectal carcinomas].

OBJECTIVE: To explore the relationship between the expression of cell cycle regulators and microsatellite instability (MSI) in colorectal carcinomas(CRC). METHODS: The expression of cell cycle regulators cyclin D1, P16, P21WAF1/CIP1, P53, Rb and PCNA were detected by immunohistochemical staining, and MSI at six microsatellite loci (D18S34, D17S799, D5S409, TCF-2, Rb and P53) were examined by PCR-SSLP in 56 cases of colorectal carcinomas. RESULTS: The overall positive rate of MSI of CRC was 44.6% (25/56). MSI was positive at 1 locus in 7 cases, at 2 and more than 2 loci in 18 cases. In CRC with MSI, the positive rate of cyclin D1 and P53 protein was significantly lower, while that of P21 protein was significantly higher than those without MSI (P < 0.05). The positive rate of P16 in CRC with MSI was higher than those without MSI. The expression of Rb proteins was similar in frequency in CRC with or without MSI. CONCLUSION: Overexpression of P21 may play an important role in MSI genesis in CRC. It may also be related to decreased expression of cyclin D1 and P53 and overexpression of P16.

[Effect of lost bite force on the expression of bFGF in rat molar periodontal ligament].

OBJECTIVE: To observe the dynamic change of the expression of basic fibroblast growth factor (bFGF) in rat molar periodontal ligament(PDL), combined with other experiments to probe into the relation of the expression between bFGF and type I collagen mRNA. METHODS: Animal model was established by extracting the left maxillary molars. Immunohistochemistry was applied. RESULTS: The expression of bFGF obviously increased from the first day to the second week (P < 0.05), Came back to that of normal bite force group at the third week. There were strong negative relative relationship between the expression of bFGF and that of type I collagen mRNA. CONCLUSION: The expression of bFGF increased under lost bite force in rat molar PDL. The effect of bite force on the expression of PDL type I collagen mRNA is closely related to the expression of bFGF.

[Myocardial protection and immunoregulation of minor bupleurum decoction and its decomposed preparations on coxsackie B3m viral myocarditis in mice].

OBJECTIVE: To study the effect of Minor Bupleurum Decoction (MBD) and its decomposed preparations, decomposition-1 (DC-1) and decomposition-2 (DC-2) in myocardial protection and immunoregulation on coxsackie virus B3m induced myocarditis in mice. METHODS: Murine model of experimental myocarditis caused by coxsackie virus B3m was developed in Balb/c mice, and were treated by MBD, DC-1 and DC-2. Natural killer (NK) cell activity, T-lymphocyte subsets and histopathological change of myocardium were examined at various times after MBD, DC-1 and DC-2 were taken. RESULTS: (1) MBD and DC-2 could increase the NK cell activity significantly, and also regulate the T-lymphocyte subsets, while DC-1 did not have this kind of effect; (2) All MBD, DC-1 and DC-2 could protect myocardium obviously, the effect of MBD is better than those of its decomposed preparations. CONCLUSION: MBD and DC-2 had bi-directional regulation on NK cell activity and T-cell subsets, all the 3 groups could obviously protect myocardium.

[Changes of blood vessels in glucocorticoid-induced avascular necrosis of femoral head in rabbits].

OBJECTIVE: To evaluate the effects of fatty tamponade in medullary cavity (serious intramedullary fatty infiltration) on the changes of blood vessels in femoral heads during the pathological process of glucocorticoid (GC)-induced avascular necrosis (AVM) of the femoral heads. METHODS: The animal model of AVN was established with injection of dexamethasone (DEX) at a high dose of 2.5 mg x kg(-1) x d(-1) in rabbits. Histopathological and morphological changes of blood vessels in the femoral heads were investigated during GC-treatment and after the cessation scanning electron microscope, light microscope, and image analysis. RESULTS: At the 2nd week of DEX-treatment, the fatty tamponade in the medullary cavity appeared in the femoral heads. Intramedullary vascular sinusoids were pressed by an excess of lipocytes and became narrow. The impressions of lipocytes on the surface of vascular sinusoids were definitely displayed on the vascular casts and ink-perfused slides of the femoral heads. These changes were pronounced with the prolongation of the treatment and the vascular sinusoids gradually lost their characteristics. Image analysis showed that the vascular area in the femoral heads continuously decreased. At the 8th week, the effects of fatty tamponade were the most marked. Intramedullary vessels became very sparse and the vascular area decreased to 1/4 of the controls. At the 6th week after the GC-treatment cessation, intramedullary fatty infiltration still existed. The blood vessels were fine and sparse, and the structural features of vascular sinusoids disappeared in the load-bearing regions of the femoral heads. At the 4th week, typical osteonecrosis focuses appeared in the femoral heads. The focuses became larger and the degree of osteonecrosis was increasing with the time of experiment. CONCLUSION: The fatty tamponade in the medullary cavity is one of the important pathological factors causing ischemic damage to the femoral heads, and plays an important role in the early stage of GC-induced AVN.

[Effects of selective preoperative intra-arterial infusion chemotherapy on apoptosis and proliferation of rectal cancer cells].

OBJECTIVES: To study the effects of anti-cancer drugs on apoptosis and proliferation of rectal cancer cells in vivo. METHODS: 114 patients with rectal cancer were treated by selective preoperative intra-arterial infusion chemotherapy. The rectal tumor tissues were derived before chemotherapy and 7 - 12 days after this treatment. In the tissues, apoptotic cells were examined by TUNEL method, and the expression of PCNA was examined by S-P staining. RESULTS: Before chemotherapy, the mean AI of tumor cells was 5.81 per thousand before chemotherapy and 10.99 per thousand 7 - 12 days after chemotherapy (P < 0.01). The mean PI of tumor cells before chemotherapy and 7 - 12 days after chemotherapy was 49.27% and 57.38% (P < 0.01). The mean AI of the high PI group increased more evidently than that of the low PI group. CONCLUSIONS: This treatment induced apoptosis of rectal cancer cells and inhibit tumor cell proliferation. The mean AI of the high PI group was higher than that of the low PI group. It is suggested that this operation should be performed shortly after chemotherapy.

[Prognostic factors of clinical curative effect for malignant obstructive jaundice].

OBJECTIVE: To explore prognostic factors for clinical treatment of patients with malignant obstructive jaundice. METHODS: 17 variables from 216 consecutive patients with malignant obstructive jaundice admitted from 1990 to 1997 were included for statistical analysis. RESULTS: The overall mortality was 19.9% (43/216), and the morbidity 37.5% (81/216). The morbidity of radical operation was higher than that of palliative operation (P < 0.01). There was a highly significant correlation between mortality, morbidity, ASA grade and duration of jaundice (P < 0.01). No correlation was seen between the mortality, type of operation and cause of jaundice. There was a highly significant correlation between the morbidity and the type of operation. CONCLUSIONS: The choice of operation in patients with malignant obstructive jaundice is radical operation. Early diagnosis and choice of treatment are essential to improving carative effect.

[A study on the expression of p16 protein and bcl-2 protein in cutaneous eyelid tumors].

OBJECTIVE: To investigate the relationship between p16 (tumor suppressor) and bcl-2 (apoptosis inhibitor) gene expression and the incidence and development of eyelid malignant tumors. METHODS: Streptavidin-biotin-peroxidase complex immunohistochemical method was used to study the expression of p16 gene and bcl-2 gene in 96 cases with eyelid malignant tumors. RESULTS: In the 96 cases, including 40 cases of basal cell carcinoma (BCC), 33 squamous and 23 sebaceous carcinomas, their p16 protein positive (nuclear staining) rates were 70.0%, 54.6% and 56.5% respectively. The p16 positive rate was negatively correlated to the degree of tumor histological differentiation, and the rate difference between the high differentiated carcinoma and the low differentiated one was significant (P < 0.05). bcl-2 protein positive expression was detected in the cytoplasm. All of the 40 BCCs were bcl-2 positive and nearly all the tumor cells showed positive cytoplasmic expression, while in the 33 specimens of squamous cell carcinoma, only did one show positive focal reaction and the staining in other 32 cases was relatively faint. None of the 23 sebaceous carcinoma expressed bcl-2. CONCLUSIONS: The expression of p16 protein is related to the occurrence and degree of differentiation of eyelid malignant tumors. The over-expression of bcl-2 protein suggests that suppression of apoptosis might play a role in the tumorigenesis of BCC.

[Analysis on Helicobacter pylori infection and its related diseases in 1,442 children in Tianjin city].

OBJECTIVE: The aim of this study was to investigate the prevalence of Helicobacter pylori (Hp) infection in children in Tianjin and to study the relationship between the Hp infection and its related diseases. METHODS: (1) Serologic examination for Hp - IgG; (2) Rapid urease test for Hp (Hp - Rut); (3) Giemsa staining for Hp; (4) Gastric fluid examination for Hp by PCR. Hp infection were diagnosed by at least two criteria. RESULTS: (1) Of the 1 442 children, 651 were Hp - IgG positive. The total positive rate was 45.15%, Positive rates of Hp - IgG among children under the age of 1 year, 3 years, 6 years, 10 years and to 14 years were 11.19%, 19.23%, 44.21%, 54.20% and 61.46%, respectively. (2) Positive rates of Hp - IgG were 31.18% (164/526) and 53.17% (48/916) in asymptomatic children and in patients with gastrointestinal symptoms. The rate was significantly higher in the later group (P < 0.005). (3) Hp infection rate in patients with duodenal ulcer (DU) was 90.76%, higher than that in other gastrointestinal diseases. (4) Histological examination showed that Hp infection rates were 33.20% and 83.97% in mild and moderate or severe gastritis, respectively. CONCLUSIONS: (1) The prevalence of Hp infection in children in Tianjin was high. (2) Rate of Hp infection increased with age and Hp infection rate was higher in children with gastrointestinal symptoms than with asymptomatie. (3) There was a high correlation in children with gastroduodenal diseases and Hp infection especially with DU. (4) Hp positive rate correlated with the pathohistologic severity of gastritis.

[Effects of hypoxia alone or exercise combined on capillarization of rat gastrocnemius muscle and its mechanism].

OBJECTIVE: To study the effects of hypoxia and hypoxia-combined-exercise on capillary density and vascular endothelial growth factor (VEGF) and its receptor KDR of skeletal muscle in rats. METHODS: Myosin-ATPase histochemistry was used to assay the size and capillary density of skeletal muscle. VEGF and its receptor KDR were studied by immunohistochemistry. RESULTS: Five-week hypoxia (simulated 5,000 m altitude) resulted in a decrease in cross-sectional area of skeletal muscle fiber and an increase in capillary density (CD), but the capillary/fiber ratio (C/F) remained unchanged. After 5-week-exercise at high altitude (1 h/d, 6 d/w), the muscle fibers did not undergo atrophy. At the same time, CD and C/F were increased. VEGF protein was found primarily in the matrix between muscle fibers; VEGF receptor-KDR was shown mainly in endothelial cells of capillary. VEGF was more strongly stained in the skeletal muscle of hypoxia-combined-exercise rats than the other two groups. CONCLUSION: Hypoxia itself can not induce neovascularization, while hypoxia-combined-exercise rats show capillary proliferation in skeletal muscle. VEGF and its receptor might play roles in this process.

[Factor XIIa-inhibited diluted thromboplastin time can reflect coagulation process].

OBJECTIVE: To establish a screening test that can reflect two stages of coagulation process. METHODS: With FXII a being blocked, the effects of various dilution of thromboplastin on clotting time were observed. FX a activity was determined by chromogenic assay. RESULTS: (1) At high concentration of thromboplastin, FXII a-inhibited diluted thromboplastin time (FXII ai DTT) of pooled normal plasma and FXI deficient plasma was very similar, but at low concentration, FXII ai DTT was in order of FVIII and FIX deficient plasma > FXI deficient plasma > pooled normal plasma. (2) FXI consumption by immunologic method induced FXII ai DTT of pooled normal plasma prolonged, and addition of FXI to FXI deficient plasma shortened FXII ai DTT. (3) Alpha-thrombin blocked by hirudin at different time (10 - 80 seconds) resulted in decreased FXa generation, and the earlier the block, the more the decrease. Under the same condition, the amount of FXa generation was in order of platelet-rich plasma > platelet-poor plasma > FXI deficient plasma. CONCLUSION: (1) Our data support the two-stage hypothesis and confirm the important role of FXI in the amplification stage. (2) FXII ai DTT as a screening test for coagulation process may be practicable.
