[Anti-virus effect of aralosides].

The result showed that the anti-virus effect of aralosides on infections with poliovirus II, ECHO delta virus, adenovirus II, herpes simplex virus I, coxsackie B3 virus and coxsackie A16 virus was remarkable. Aralosides could inhibit the development of cytopathic effect (CPE) and protect cultural cells from being infected with the above viruses.

[Electrophysiological investigation on decussate communication of bulbospinal inspiratory neurons with phrenic nucleus at level of spinal cord].

The experiments were carried out on anesthetized, vagotomized, immobilized and artificially ventilated cats, rabbits and rats. The rhythmic respiratory activities on both sides of the phrenic nerves existed after a hemisection between the 2nd and 3rd cervical cord, while the activities of the phrenic nerve ipsilateral to the hemisection disappeared following a subsequent mid-sagittal section from the 3rd to 5th cervical cord. The amplitude of the phrenic activities on either side was lowered, more remarkably on the ipsilateral side, by the hemisection. These results indicate that there exists a decussate communication of the bulbospinal inspiratory neurons with the phrenic nucleus at the level of spinal cord. In other words, the phrenic nucleus receives inspiratory drives via descending pathways on both sides of the cervical cord, mainly on the ipsilateral side.

[Clinicopathologic and immunohistochemical studies of midline malignant reticulosis in palate].

Fourteen patients (10 males and 4 females) with midline malignant reticulosis (MMR) in palate were studied. The symptoms were most commonly swelling and ulceration. In some cases, the lesion involved nasal, maxillary sinus and facial skin, but no regional lymph nodes were involved. A necrotizing infiltration of atypical lymphoid cells (ALCs) mixed with inflammatory cells was confirmed as the histopathologic feature of MMR. In 2 of 14 cases, ALCs formed a continuous sheet in several high-power fields with a few inflammatory cells. This overlapping feature may be a transitional form between MMR and malignant lymphoma. Immunohistochemical staining results indicated that in 10 of 14 cases, ALCs were CD3 positive; in 7 of 14 cases, ALCs were UCHL1 positive; and in all 14 cases, ALCs were L26 negative. We deem MMR a T-cell lymphoma in substance, but it should be treated as a histopathologic entity distinct from malignant lymphoma.

[A cohort study on the relationship between the mortality of cerebro-vascular diseases and farmer smokers].

The relationship between the mortality of cerebro-vascular diseases and farmer smokers in Shifang County was studied. The results indicated that the mortality of cerebro-vascular diseases in the male groups of cigarette-smokers older than 65 and cigar-smokers older than 55 was significantly higher than that of the nonsmokers (P < 0.05), the RR being 1.68-3.22. Also, the mortality in the female groups of cigarette-smokers older than 55 and cigar-smokers older than 65 was significantly higher than that of the non-smokers (P < 0.05), the RR being 1.99-3.19. The sex-specific mortalities of the other age groups revealed no significant differences in spite of some inequalities in smoking (P < 0.05). Age should be one of the risk factors for the death of cerebro-vascular diseases regardless of the sex. The mortality rose with the increasing accumulated amount of smoking. The relationship was not significant between the mortality of cerebro-vascular diseases and short-term smokers with small dose (P > 0.05). However, when the accumulated amount reached certain degree, i.e. the smoker consumed cigar more than 270 kg or consumed cigarette more than 10,000 packs, the relationship between the behavior of cigar-smoking and cigarette-smoking and the mortality became apparent, the RR being 2.53-3.91 (P < 0.01).

[Diagnosis and treatment of hydronephrosis: report of 100 cases].

One hundred patients with hydronephrosis were treated for 10 years. In 88 patients operations confirmed the causes of obstruction including pyelo-ureteral junction obstruction (pujo, 46 cases), anomalous (15), retroperitoneal fibrosis (15), ureteral polyps and valves and others (12). In accordance with etiology and renal function pyeloplasty was carried out in 31 cases, nephrectomy 37 and nephrostomy 12; other operation was done in 20 cases. The successful rate of pyeloplasty was 91.3%. Early diagnosis and treatment of hydronephrosis is key for reserving renal function. Renal r-scan, percutaneous nephrostomy are useful in judging renal function. Dual-Flex stent internal drainage is better than external drainage when pyeloplasty is done.

[Optimization of the processing technology of Aconitum kusnezoffii Reichb. by orthogonal test].

A study on the processing technology of Aconitum kusnezoffii by orthogonal test was carried out by assaying as criteria the changes in contents of the total alkaloids and ester type alkaloids before and after processing. The optimum experimental conditions were obtained after treating the experimental data with a formula for the multiple criteria, and the optimal processing results were achieved by having the material macerated with water, cut into thick pieces and then steamed under high pressure (127 degrees C, 0.15MPa) for 3 hours.

A behavioural training programme for chronic schizophrenic patients. A three-month randomised controlled trial in Beijing.

This paper discusses the effectiveness of an in-patient rehabilitation programme administered by nurses that combines life skills training, active encouragement, and a token economy. Fifty-two chronic schizophrenic patients with prominent negative symptoms who had been continuously in hospital for at least a year were randomly assigned to the experimental or control group. The training and associated reinforcement schedule were administered daily to experimental subjects by two specially trained rehabilitation nurses. Control subjects did not receive training or reinforcement but were individually asked to perform the same daily tasks and participate in the same activities as the experimental-group subjects. Patients in both groups received their previous dosage of medication throughout the trial. After three months the severity of negative symptoms, as assessed by blind evaluators, decreased in both groups of subjects, but the improvement in the experimental group was much greater than that in the control group. These findings demonstrate the efficacy of behavioural interventions for chronic schizophrenic in-patients in China and highlight the importance of changing the role of Chinese psychiatric nurses from that of custodians who control patients' behaviour to that of therapists who provide psychological and behavioural treatment.

Identification and determination of the enantiomers of moprolol and their metabolites in human urine by high-performance liquid chromatography and gas chromatography-mass spectrometry.

A simple and sensitive high-performance liquid chromatographic (HPLC) method using chiral derivatization was developed to screen and determine the enantiomers of moprolol and their metabolites in human urine. The recovery of (+)- and (-)-moprolol from urine was 70.8-81.1% at different concentrations. The coefficients of variation (C.V.) were less than 3.2 and 6.5% for intra- and inter-assays, respectively. Moprolol could be detected in urine up to 24 h after oral administration of a 50-mg dose of moprolol. Unconjugated and conjugated enantiomers of moprolol and their metabolites were analyzed by gas chromatography (GC). A gas chromatographic-mass spectrometric (GC-MS) confirmatory method was established to identify the metabolites of moprolol. The double derivation procedure for moprolol and their metabolites with S-(-)-methyl chloroformate [(-)-MCF] and N-methyl(trimethylsilyl)trifluoroacetamide (MSTFA) gave very good GC-MS properties of the derivatized compounds and provided reliable structural information for their confirmation analysis. This is the first published report on the use of a GC-MS method for the detection of the enantiomers of moprolol and their metabolites in human urine.

Biochemical markers of bone turnover in women with surgically treated carcinoma of the breast.

Biochemical markers of bone turnover were measured in fasting urine and blood samples obtained from 38 postmenopausal women with previous surgical treatment of breast cancer combined with adjuvant chemotherapy, tamoxifen, or placebo. Significantly elevated urinary pyridinoline as nmol mmol-1 creatinine (47.5 and 42.5 in tamoxifen and placebo treated patients compared with 26.3 in normal controls, both P < 0.001) and deoxypyridinoline (11.9 and 10.5 compared with 6.3, P < 0.001 and P = 0.002 respectively) were found with unchanged urinary hydroxyproline, serum alkaline phosphatase and procollagen I carboxyterminal peptide (PICP). These findings suggest enhanced bone resorption resulting from the humoral osteoclast activating effect of the previous breast cancer or underlying carcinoma recurrence. Alternatively the raised pyridinium excretion might indicate an altered crosslinking composition of bone collagen. No specific effect on bone metabolism was found with tamoxifen treatment as all measured parameters were similar in both tamoxifen ex-users and non-users. This confirmed the safety of tamoxifen therapy with respect to bone.

Defective major histocompatibility complex class I expression on lymphoid cells in autoimmunity.

Lymphocytes from patients with insulin-dependent diabetes mellitus (IDDM), a chronic autoimmune disease, have recently been shown to have decreased surface expression of MHC class I antigens. Since IDDM and other autoimmune diseases share a strong genetic association with MHC class II genes, which may in turn be linked to genes that affect MHC class I expression, we studied other autoimmune diseases to determine whether MHC class I expression is abnormal. Fresh PBLs were isolated from patients with IDDM, Hashimoto's thyroiditis, Graves' disease, systemic lupus erythematosis, rheumatoid arthritis, and Sjogren's syndrome. Nondiabetic and non-insulin-dependent diabetes mellitus patients served as controls. MHC class I expression was measured with a conformationally dependent monoclonal antibody, W6/32. Freshly prepared PBLs from the autoimmune diseases studied and the corresponding fresh EBV-transformed B cell lines had decreased MHC class I expression compared with PBLs from normal volunteers and non-insulin-dependent (nonautoimmune) diabetic patients. Only 3 of more than 180 donors without IDDM or other clinically recognized autoimmune disease had persistently decreased MHC class I expression; one patient was treated with immunosuppressive drugs, and subsequent screening of the other two patients revealed high titers of autoantibodies, revealing clinically occult autoimmunity. Patients with nonautoimmune inflammation (osteomyelitis or tuberculosis) had normal MHC class I expression. Autoimmune diseases are characterized by decreased expression of MHC class I on lymphocytes. MHC class I expression may be necessary for self-tolerance, and abnormalities in such expression may lead to autoimmunity.

Decreased arteriolar endothelium-derived relaxing factor production during the development of genetic hypertension.

In the present study, basal and stimulated production of endothelium-derived relaxing factor (EDRF) were examined in young (4-5 week-old) and mature (10-12 week-old) spontaneously hypertensive rats (SHR) and were compared with those of age-matched Wistar Kyoto (WKY) normotensive rats. Nw-nitro-L-arginine methyl ester (L-NAME) was used as a probe for basal EDRF production in vivo. We also determined the dose-response effects of acetylcholine (Ach, 10(-9)-10(-5) M), sodium nitroprusside (NP, 10(-9)-10(-5) M), and norepinephrine (NE, 3 x 10(-10)-10(-6) M), on third-order arterioles (3A) in the two age groups. Relaxation of 3A vessels in response to Ach was significantly lower (p < 0.01) in mature SHR than in WKY rats. No significant differences were observed in responses to Ach in young animals or in responses to NP in either age groups. Preincubation with L-NAME (2 x 10(-4) M) shifted the dose-response curves of NE to the left in mature WKY rats (n = 6), but not in mature SHR (n = 6). These data indicate that both basal and stimulated production of EDRF are decreased in small arterioles in the striated muscle microcirculation at an established stage of genetic hypertension. These data suggest that a diminished EDRF response by arterioles may be in part responsible for the elevated microvascular reactivity observed in genetic hypertension.

[Prevalence of HBV and HCV infections in plasma donors with recently increased ALT level].

HCV and HBV markers in sera were examined in a group of donors (71 cases) who had a history of elevated ALT, and were compared with another group of blood donors (217 cases) who had no such a history. The results showed that the positive rates in both two groups of donors were 14.08% and 6.45% for anti-HCV, 3/8 and 2/13 for anti-HCV-IgM in anti-HCV positive cases, 2.82% and 0.46% for HBsAg, 32.39% and 25.35% for anti-HBs, 45.07% and 33.18% for anti-HBc, respectively (P < 0.05-0.01). It showed a good correlation between the positive rates of HCV and HBV markers and the times of ALT elevation.

Modulation of the activity of a human granulocyte-macrophage colony-stimulating factor/interleukin-3 fusion protein (pIXY 321) by the macrocyclic lactone protein kinase C activator bryostatin 1.

We have examined the effect of the macrocyclic lactone protein kinase C (PK-C) activator bryostatin 1 on the proliferative capacity and lineage commitment of CD34+ human bone marrow cells exposed to the granulocyte-macrophage colony-stimulating factor/interleukin-3 (GM-CSF/IL-3) fusion protein pIXY 321. pIXY 321 administered at a dose of 10 ng/mL was as effective as the combination of plateau concentrations of recombinant (r) IL-3 and rGM-CSF (e.g., 50 ng/mL) in stimulating the growth of day-14 committed myeloid progenitors (colony-forming units granulocyte/macrophage [CFU-GM]). In the large majority of samples tested, coadministration of 0.5 to 100 nM bryostatin 1 with either pIXY 321 or the combination of rIL-3 plus rGM-CSF led to modest but significant increases (e.g., 30 to 75%) in the number of CFU-GM, compared to administration of growth factors alone. The degree of bryostatin 1-induced potentiation, however, was considerably less than that previously observed in the case of cells exposed to either rIL-3 or rGM-CSF, where increases of 100 to 150% were regularly noted. While at least 50% of day-14 CFU-GM stimulated by either pIXY 321 or the combination of rIL-3 plus rGM-CSF were of the pure or mixed eosinophilic variety, coadministration of bryostatin 1 resulted in a dramatic inhibition of eosinophilic colonies and a corresponding increase in pure and mixed neutrophil and macrophage colonies. Although coadministration of recombinant granulocyte colony-stimulating factor (rG-CSF) or recombinant colony-stimulating factor-1 (rCSF-1) mimicked the capacity of bryostatin 1 to increase the total number of pIXY 321-induced day-14 CFU-GM, these growth factors, unlike bryostatin 1, were not capable of inhibiting eosinophilic colony formation. Furthermore, whereas addition of neutralizing antibodies to G-CSF or CSF-1 blocked the capacity of these growth factors to potentiate colony formation in the presence of pIXY 321, it did not abrogate the effect of bryostatin 1 on progenitor cell growth or lineage commitment. Finally, in contrast to its effects on committed myeloid progenitors, bryostatin 1 did not increase the growth of erythroid (burst-forming units-erythroid [BFU-E]) and multipotent (multipotent colony-forming units [CFU-GEMM]) progenitors stimulated by pIXY 321, but instead inhibited colony formation at higher concentrations (e.g., 10 to 100 nM).(ABSTRACT TRUNCATED AT 400 WORDS)

Effects of ilexonin A on cAMP metabolism in platelets.

Ilexonin A (Ile A) is an effective compound isolated from Ilex pubescens Hook et Arn which has been found to inhibit platelet aggregation and Ca2+ influx in platelets. The present experiments were designed to observe the effects of Ile A on platelet metabolism. Verapamil (Ver) was used as control. The results indicated that thrombin significantly reduced the level of platelet cAMP, while Ile A and Ver obviously antagonized this effect. The inhibition of the activity of platelet phosphodiesterase by Ile A was much stronger than that by Ver. The results suggest that Ile A may be beneficial in the treatment of cardiovascular disease.

High level expression of human prourokinase cDNA in Chinese hamster ovary cells.

We have used Chinese hamster ovary (CHO) cells to express high levels of human prourokinase gene cDNA with recourse to construction of good expression vector, the improvement of transfection technique and gene coamplification. First, we constructed expression plasmid pMG10102 by placing pro-UK cDNA under the control of SR alpha promoter/SV40 polyadenylation signals and expressed it transiently in COS-7 cells. Expression level was about 5 times higher than with SV40 early promoter. Linear plasmids pMG10102 and pSV2-dhfr were then cotransfected into CHO-dhfr cells by calcium phosphorate coprecipitation and cells were cultured in selective medium. Twenty transformants expressing pro-UK were picked, the range of expression levels was 12.5-100IU/10(6) cells/day. When subjected to stepwise selection of methotrexate (MTX), the stable cell lines were obtained that secreted up to 400-500IU/10(6) cells/day. Western blot analysis showed that molecular weight of secreted recombinant pro-UK was the same as that of natural pro-UK, which is 52 kDa, and more than 60% of expression production was single chain urokinase (rscUK) without protease inhibitor in medium.

[Spiral shaped bacteria in the human gastric biopsy].

Biopsy specimens from the gastric mucosa of 149 patients who underwent gastroduodenal endoscopy for upper gastrointestinal complaints were studied by light microscopy and culture. Spiral shaped bacteria were detected in four of the specimens by smears with Gram stain. The positive rate was 2.68%, but these bacteria and HP did not grow in culture. The characteristic helical morphology of the bacteria appears to be similar to that of the bacteria found in the stomach of cats and dogs. And what of significance in these cases is the presence of spiral shaped bacteria in association with chronic gastritis.
