The Task and Ego Orientation in Sport Questionnaire: construct equivalence and mean differences across gender.

Research on achievement goal orientation in sport has relied primarily on traditional statistical methodology to examine group mean differences. Unfortunately, examination of the measurement model is generally ignored prior to means testing. This study reports an application of structural equation modeling (SEM) in testing measurement invariance and latent mean structure of the Task and Ego Orientation in Sport Questionnaire (TEOSQ; Duda & Nicholls, 1989) using male and female college students. A confirmatory factor analysis for testing invariance revealed invariant measurement properties and factor structures across gender, indicating that task and ego orientation are similarly conceptualized by male and female students. Subsequent testing of latent mean structures, however, showed significant gender differences with respect to ego orientation, but no difference in task orientation. The SEM procedures used in the present study demonstrate additional construct validity and internal consistency reliability for the TEOSQ and, by confirming its factor structure, provide a sound psychometric basis for its continued use in substantive studies focusing on the comparison of achievement goal orientation across gender.

Testing the simplex assumption underlying the Sport Motivation Scale: a structural equation modeling analysis.

Self-determination theory (Deci & Ryan, 1985) suggests that motivational orientation or regulatory styles with respect to various behaviors can be conceptualized along a continuum ranging from low (a motivation) to high (intrinsic motivation) levels of self-determination. This pattern is manifested in the rank order of correlations among these regulatory styles (i.e., adjacent correlations are expected to be higher than those more distant) and is known as a simplex structure. Using responses from the Sport Motivation Scale (Pelletier et al., 1995) obtained from a sample of 857 college students (442 men, 415 women), the present study tested the simplex structure underlying SMS subscales via structural equation modeling. Results confirmed the simplex model structure, indicating that the various motivational constructs are empirically organized from low to high self-determination. The simplex pattern was further found to be invariant across gender. Findings from this study support the construct validity of the SMS and have important implications for studies focusing on the influence of motivational orientation in sport.

Translation initiation region plays an important role in the expression of human thrombopoietin in Escherichia coli.

A mutant human thrombopoietin (TPO) gene with a modified translation initiation region (TIR) sequence was created by site-specific mutagenesis based on the PCR technique. This mutant TPO gene encoded the same amino acid sequence as wild-type TPO gene. The wild-type TPO gene was expressed in E. coli with very low efficiency. The mutant TPO gene could reach an expression level of up to 10% of total cellular proteins in E. coli, which was much higher than the wild-type gene. The recombinant protein was mainly in the form of inclusion body which could acquire in vitro activities of human thrombopoietin after refolding.

Amount of care given and caregiving satisfaction: a latent growth curve analysis.

We examined the wear-and-tear hypothesis using data from 4 annual interviews with 130 (128 White) middle-aged daughters caring for their physically impaired, elderly mothers. We formulated a latent growth curve model hypothesizing that increases in the amount of care given by daughters caused a decrease in caregiving satisfaction, independent of caregiving duration. We found considerable individual variability and change in both caregiving satisfaction and the amount of care given in univariate latent growth curve analyses. Contrary to the wear-and-tear hypothesis, a multivariate latent growth curve analysis revealed duration of caregiving had no effect on either initial caregiving satisfaction or change in satisfaction. An elaborated wear-and-tear model was supported, however. The mechanism for decline in satisfaction is an increase in the amount of care given.

Magnetic resonance imaging of neurogenic tumors of the thoracic inlet: determination of the parent nerve.

To investigate the value of magnetic resonance imaging (MRI) in determining the parent nerves of neurogenic tumors in the thoracic inlet, analysis of MR images was performed in nine patients with surgically resected neurogenic tumors in the thoracic inlet (two neurofibromas and one schwannoma of the vagus nerve, three schwannomas of the brachial plexus, and two schwannomas and one ganglioneuroma of the sympathetic nerves). These MR images were compared with surgical and pathologic findings. The multidirectional capability and excellent tissue contrast of MRI facilitated recognition of the location, shape, and extent of the tumors. MRI, which permitted an easy understanding of the spatial relation between the tumors and the subclavian vessels, scalenus muscles, and brachial plexus, was useful in determining the nerves of origin. Two neurofibromas, four of six schwannomas, and one ganglioneuroma were recognized to extend along the axes of the parent nerves on MR images. MRI is useful in determining the parent nerve of neurogenic tumors in the thoracic inlet and is helpful in planning surgical treatment of these tumors.

Synergistic interaction of the Neu proto-oncogene product and transforming growth factor alpha in the mammary epithelium of transgenic mice.

Transgenic mice expressing either the neu proto-oncogene or transforming growth factor (TGF-alpha) in the mammary epithelium develop spontaneous focal mammary tumors that occur after a long latency. Since the epidermal growth factor receptor (EGFR) and Neu are capable of forming heterodimers that are responsive to EGFR ligands such as TGF-alpha, we examined whether coexpression of TGF-alpha and Neu in mammary epithelium could cooperate to accelerate the onset of mammary tumors. To test this hypothesis, we interbred separate transgenic strains harboring either a mouse mammary tumor virus/TGF-alpha or a mouse mammary tumor virus/neu transgene to generate bitransgenic mice that coexpress TGF-alpha and neu in the mammary epithelium. Female mice coexpressing TGF-alpha and neu developed multifocal mammary tumors which arose after a significantly shorter latency period than either parental strain alone. The development of these mammary tumors was correlated with the tyrosine phosphorylation of Neu and the recruitment of c-Src to the Neu complex. Immunoprecipitation and immunoblot analyses with EGFR- and Neu-specific antisera, however, failed to detect physical complexes of these two receptors. Taken together, these observations suggest that Neu and TGF-alpha cooperate in mammary tumorigenesis through a mechanism involving Neu and EGFR transactivation.

Angiotensin II activation of Ca(2+)-permeant nonselective cation channels in rat adrenal glomerulosa cells.

A Ca(2+)-permeant, nonselective cation channel was observed in cell-attached and inside-out membrane patches from rat adrenal glomerulosa cells maintained in primary cell culture. In cell-attached patches under near physiological ionic conditions, single-channel currents exhibited a reversal potential near -10 mV, inward rectification, a nearly linear slope conductance between 0 and -80 mV of 17.4 pS, and voltage-dependent block at potentials more negative than -80 mV. Channels exhibiting identical conductance and gating properties were observed in inside-out patches; however, channel gating ran down within minutes in this configuation. In the inside-out configuration, channel gating did not require cytosolic Ca2+ (Ca2+ < 10(-9) M), and inward rectification was relieved by removal of intracellular Mg2+. Relative ionic permeability was calculated using reversal potential measurements from inside-out patches under bi-ionic conditions. The channel discriminated poorly among monovalent cations (PLi > PK > PCs > PNa) and was not significantly permeable to anions. The channel was permeable to Ca2+, exhibiting a relative permeability ratio of 0.29 PCa/PNa) when measured with 110 mM Ca2+ on the intracellular face and a permeability ratio of 4.38 (PCa/PNa) with 110 mM Ca2+ on the extracellular face. Channel gating behavior was episodic with open times ranging from milliseconds to tens of seconds and closed times lasting up to several minutes or longer. Channel gating appeared to be relatively voltage independent except that mean channel open time and open probability were reduced by membrane hyperpolarization. In cell-attached patches, bath application of 1 nM angiotensin II (ANG II) increased the channel open probability, primarily affecting channels exhibiting a low open probability, primarily affecting channels exhibiting a low open probability before stimulation. With the use of nystatin perforated-patch current clamp to measure membrane potential, ANG II was observed to induce large transient membrane depolarizations, consistent with activation of an inward current. We hypothesize that this channel is an important component of ANG II-induced membrane depolarization and Ca2+ influx during stimulation of aldosterone secretion.

Measurement of regional myocardial blood flow in rats by unlabeled microspheres and Coulter channelyzer.

A relatively inexpensive, expeditious, new nonradioactive microsphere method for measuring regional myocardial blood flow (RMBF) was developed with unlabeled microspheres and a Coulter Channelyzer. To validate the efficiency of this method, hearts from rats were perfused ex vivo by retrograde aortic cannulation. Unlabeled microspheres of varying size were injected into a side arm in the aortic cannula or added to blood samples collected from the rats. Microspheres were then recovered from the cardiac tissue and blood samples. It was found that > 97% of perfused microspheres (diam > 9.4 microns) were retained in the myocardium and that 94.8 +/- 2.2% of the trapped microspheres were recovered and counted successfully using a Counter Channelyzer. The percent recovery of microspheres from 2- and 0.5-ml blood samples were 95.4 +/- 2.3 and 95.3 +/- 3.1%, respectively. Blood flow to the anterior and posterior halves of the ventricular free walls and septum were measured in six rats; excellent agreements were found between the results yielded by 10-, 15-, and 20-microns unlabeled microspheres injected simultaneously. The transmural flow gradients in the left ventricular free wall estimated by 10- and 15-microns spheres did not significantly differ from each other. Thus the method developed here provides a new alternative for measurement of RMBF, which currently allows at least three measurements for nontransmural gradient RMBF and at least two measurements for transmural gradient RMBF.

Microcirculatory changes in rat skeletal muscle in sepsis.

The aim of this study was to confirm that microvascular perfusion was abnormal during the early phases of normotensive sepsis and to determine whether these changes were due to the development of tissue edema. Skeletal muscle red blood cell (RBC) flow was studied in rats made septic by cecal ligation and perforation (CLP). After anesthesia with halothane, arterial and venous cannulae were inserted and, in the treatment group, a CLP performed. At 6, 24, and 48 h after entry into the study, the incidence of microcirculatory absence of flow in the extensor digitorum longus muscle (EDL) was examined with intravital microscopy. The number of capillaries containing RBCs were counted over a 60-s interval, and the flow status of each capillary was recorded. A significant increase in the number of stopped-flow capillaries was observed in the CLP group (p < 0.01) as compared with time-matched controls. In both groups the number of capillaries with stopped flow was greater than in naive animals. The severity of absence of flow was negatively correlated with the systemic hemoglobin concentration. These changes were not associated with an increase in tissue wet/dry weight ratio or albumin flux. This study shows that sepsis was associated with increased RBC flow heterogeneity. These changes, which occur within 24 h of the septic insult, are a persistent feature of the evolving septic process in the absence of tissue edema. These observations support the view that extrinsic compression of the microcirculation by tissue edema is not the primary cause of alterations in microcirculatory flow in sepsis.

[Effects of frostbite on some factors of blood coagulation system in rats under hypoxia].

The changes of some factors of blood coagulation system in rats following frost-bite of both hind feet under hypoxia were investigated. Male Wistar rats weighed 200 +/- 20g were divided into four groups: normal control (C); frostbite at normoxia (FN); frostbite during acute hypoxia (FAH) and frostbite during hypoxia after altitude acclimation (FHAC). Bleeding time and clotting time, rate of clot-retraction, plasma content of 6-keto-PGF1 alpha and TXB2 were determined following exposure to cold. The results showed that bleeding time and clotting time were shortened, and rate of clot-retraction was decreased, plasma content of 6-keto-PGF1 alpha and TXB2, T/P ratio were increased significantly after exposure to cold in all frostbite groups, but these changes were more prominent in FHAC than those in FN and FAH. The results demonstrated that there were changes in blood coagulation system following cold injury, blood coagulability was increased. These changes were closely related to the degree of frostbite. In addition, the degree of cold injury was aggravated by altitude acclimation and this may play an important role in the pathological process of dysfunction leading to necrosis of local frostbite tissue.

[Effects of freezing and hypoxia on serum creatine kinase activity in rats].

Serum creatine kinase (CK) activity after cold exposure in rats acclimated to cold, hypoxia, or combined factors (both cold and hypoxia) were observed under normoxic [correction of normoxie] and hypoxic condition in order to study the degrees of damage to the tissues. The results showed that resistance to frostbite increased in rats acclimated to cold; resistance to frostbite decreased markedly in rats acclimated to hypoxia, i.e. negative cross-acclimation exists between hypoxia and cold. The changes in serum CK activity in rats exposed to both cold and hypoxia for 4 weeks were essentially the same as those in hypoxia-acclimated rats, and were different significantly from those in cold-acclimated rats. It indicates that the effects of hypoxic exposure were stronger than those of cold exposure, its mechanism remains an area for further study.

Characterization of eosinophils generated in vitro from CD34+ peripheral blood progenitor cells.

Methods for isolation and cultivation of CD34+ peripheral blood progenitor cells (PBPCs) have facilitated their use in autologous transplantation and as potential targets for gene therapy. In this work, we present the possibility of using these isolated cells to study lineage-specific hematopoietic differentiation. We have shown that differentiating PBPCs faithfully replicate transcriptional events that occur during maturation of the eosinophil lineage; messenger RNAs encoding the five eosinophil granule proteins were detected by reverse-transcriptase polymerase chain reaction (RT-PCR) after 2-3 days of cytokine-stimulated growth. Only three of the five proteins were detected by immunofluorescence staining after 14 days of cytokine-stimulated growth; the percentage of Charcot-Leyden crystal protein (CLC)-containing cells (16-18%) exceeded that of eosinophil peroxidase (EPO)-containing cells (7-8%), which in turn exceeded that of eosinophil-derived neurotoxin (EDN)-containing cells (2-4%). While the electrophoretic mobilities of both CLC and EPO synthesized by differentiating PBPCs were similar to those of their normal counterparts, immunoreactive EDN was found to be heterogeneous and of higher molecular weight that EDN found in mature eosinophils. It is not clear whether our results, which show progressive, but incomplete, differentiation of PBPCs into eosinophils, reflect a lack of knowledge as to what factors are essential for complete differentiation in vitro or relate to the inherent capacity of PBPCs to differentiate along this lineage.

[High-resolution CT of epidermoid carcinoma in peripheral lung fields: radiologic-pathologic correlation].

We correlated high-resolution CT (HR-CT) images of 30 surgically resected epidermoid carcinomas in peripheral lung fields with pathologic findings. We classified peripheral epidermoid carcinomas into three types based on HR-CT findings. All tumors showed a notch along their margin. Type 1 shows scanty speculations without the convergence of surrounding lung structures. Type 2, which has two subtypes, shows a spiculated border with the convergence of peripheral lung structures. Type 2a has fine irregularities in its border, while type 2b has few fine irregularities. The convergence of peripheral lung structures seen in type 2 carcinoma is caused by the presence of scar tissue within tumors. Speculations corresponded to tumor extension or lymphocytic infiltration, sometimes along pulmonary vessels or interlobular septa, and fine irregularities seen in type 2a corresponded to summation of fibrous thickening of alveolar septa. Nine tumors showed varying-sized cavities, which were caused by the circulatory disturbance and/or central necrosis seen in tumor nests. In conclusion, HR-CT images were well correlated with pathologic findings of the resected specimens; however, some type 2a tumors may mimic adenocarcinoma on HR-CT findings.

[Detectability of simulated nodules on chest phantom by Fuji computed radiography: comparison of different nodule sizes, configurations and tube voltages].

The detectability of simulated nodules placed on a chest phantom and shown on FCR images has been studied. Different tube voltages (130 kVp, 100 kVp), presence or absence of compensation filter, various sites and different sizes (5 mm/10 mm) and configurations (spherical or broad based) of the nodule were examined. 112 FCR chest phantom images were evaluated by ten radiology specialists utilizing ROC analysis. The detection rate for nodules was influenced in general by the site. The detection rate was as follows in decreasing order: intercostal, overlaid with one rib, two ribs, diaphragm and heart. Detection of nodules of 10 mm in height was mainly influenced by the density of the lung where the nodule was located, whereas that of nodules of 5 mm in height was more dependent on the site of the nodule, probably greatly influenced by the amount of structure mottle. Spherical nodules of 5 mm in height were more frequently recognized than the broad-based ones. High kVp (130 kVp) resulted in a higher detection rate for both sizes of nodules than low kVp (100 kVp). Detection of spherical nodules overlying the heart or diaphragm was improved with the use of a compensation filter.

Studies on the reaction model for the immobilized Streptomyces aureofaciens's protoplasts used to convert lincomycin.

A reaction kinetic model is proposed for immobilized S. aureofacien's protoplasts which can facilitate the conversion of lincomycin into clindamycin. The simulation result of the model has been given. Effects of various kinetic parameters of the reaction on the conversion rate are given in the formula. The formulas which can be used to calculate the maximum conversion rate and the optimal reaction time have been given. The results of model simulation are in good agreement with the experimental results.

[Early changes in cardiac function and beta-ARs in heart after severe scalds in rat].

The changes in left ventricular function and ventricle beta-adrenoceptors (beta-ARs) were measured in rat undergoing a full thickness scalded burn of skin of 30% total body surface area. The results showed that there were a significant decrease in left ventricular systolic pressure (LVSP), maximum rising and falling rate of left intraventricular pressure (LV +/- dp/dtmax) and density of ventricular beta-ARs. Correlation analysis showed that the change in beta-ARs density correlated with the change in LVSP, LV +/- dp/dtmax positively.
