Lymphangiogenesis in tumor tissues and 5'-Nase, Flt-4, Flk-1 expressions.

OBJECTIVE: To investigate lymphangiogenesis in tumor tissues and 5'-Nase, Flt-4, Flk-1 expressions. METHODS: 5'-Nase and Aplase expressions and the morphology of lymphatics in gastric carcinoma tissues were observed with enzyme histochemistry. Flt-4 and Flk-1 expression in gastric carcinoma and sarcoma tissues was studied by immunohistochemistry. CNE-2z cells were implanted into the subdermal layer of nude mice and hyaluronic acid was injected at the same sites. After 4 or 5 weeks, nasopharyngeal carcinoma (NPC) xenograft tissues were cut for immunohistochemistry and enzyme histochemistry, and lymphangiogenesis, 5'-Nase, Alpase, Flt-4 and Flk-1 expression in NPC tissues was studied. Hyaluronic acid contents in sera of the patients with NPC and mice with NPC xenografts were measured by radioimmunoassay. RESULTS: There were a lot of lymphatics and solid strip-like tissues in gastric carcinoma and NPC xenograft treated with hyaluronic acid. The total number of lymphatics (26.9 +/- 14.2/HP) increased significantly in gastric carcinoma tissues. Flt-4 and Flk-1 positive expression was found in gastric carcinoma, sarcoma tissues and NPC xenograft. Most of the vessels with positive Flt-4 were lymphatics. Flk-1 expression was only found in the walls of blood vessels. Hyaluronic acid content (363.52 +/- 98.15 ng/ml) in sera of patients with NPC was higher than that of the patients with chronic nasopharyngeal inflammation (306.29 +/- 27.15 ng/ml), with a significant difference (P = 0.039). The hyaluronic acid content in sera of nude mice with NPC (2120.59 +/- 490.22 ng/ml) was markedly higher than that in normal mice (1588.99 +/- 502.12 ng/ml), with a significant difference between them (P = 0.007). CONCLUSION: There are a lot of lymph capillaries with positive 5'-Nase in gastric carcinoma and NPC xenograft treated with hyaluronic acid. Flt-4 positive expression is shown in gastric carcinoma, sarcoma tissues and NPC xenografts. The results suggest there is lymphangiogenesis in these malignant tumor tissues. Hyaluronic acid contents in sera of patients with NPC and nude mice with NPC increase, which may promote the formation of lymphatics.

[Preliminary clinical observation on treatment of chronic rhinitis with rhinitis spray].

OBJECTIVE: To investigate the clinical effect of Rhinitis Spray (RS) in treating chronic rhinitis. METHODS: Eighty patients with chronic rhinitis were divided into three groups, and treated with RS, ephedrine and normal saline respectively. RESULTS: The short-term effective rate in the three groups was 89.3%, 66.7% and 8.0% respectively. RS was effective in alleviating symptoms, increasing IgG level in nasal discharge, improving ventilation of nasal cavity and transfer function of cilia of nasal mucosa obviously. CONCLUSION: The therapeutic effect of RS in treating chronic rhinitis is good.

[Primary study on characteristics of insulin secretion rate, metabolic clearance rate and sensitivity in non-insulin-dependent diabetic subjects from multiplex diabetic pedigrees].

OBJECTIVE: To investigate the characteristics of insulin secretion rate (ISR), metabolic clearance rate (MCR-I) and sensitivity and to explore their relationship with obesity in non-insulin-dependent diabetic subjects from multiplex diabetic pedigrees (MDP). METHODS: Fifteen subjects with normal glucose tolerance and 11 non-insulin-dependent diabetic patients from MDP were included in the study. Frequently sampled intravenous glucose tolerance test (FSIVGTT) was performed. Glucose, insulin (INS) and connecting-peptide (C-P) concentrations were measured. A computer procedure devised by our laboratory was used to calculate the value of ISR at each time point, then MCR-I was acquired. Insulin sensitivity index (SI) was calculated according to minimal model technique about glucose in FSIVGTT. RESULTS: The ISR curve in control group was biphasic, while in non-insulin. In non-insulin-dependent diabetic group, areas under the curves of C-P (AUCC) and ISR level (AUCS) measured during 0 approximately 16 min were 7.9 nmol.min(-1).L(-1) +/- 2.8 nmol.min(-1).L(-1), and 6.1 nmol +/- 2.2 nmol, respectively, which were significantly lower than those in control group 17.7 nmol.min(-1).L(-1) +/- 4.92 nmol.min(-1).L(-1) and 12.3 nmol +/- 3.9 nmol (P < 0.01). The two parameters were slightly higher than those in control group 155 nmol.min(-1).L(-1) +/- 44 nmol.min(-1).L(-1) vs 101 nmol.min(-1).L(-1) +/- 30 nmol.min(-1).L(-1) and 76 nmol +/- 26 nmol vs 54 nmol +/- 20.0 nmol (P < 0.05)measured during 16 approximately 180 min. There was no significant difference, between the two groups about the amount of insulin secretion during 3 hours (82 nmol +/- 28nmol vs 68 nmol +/- 21 nmol, P = 0.2). In control group, there were significant positive correlation, between AUCS, waist-hip ratio (WHR), and body surface area, (BSA) and significant negative correlation between MCR-I, SI and WHR, BSA (P < 0.01), and also between MCR-I and SI. In non-insulin-dependent diabetic group, AUCS were significantly correlated with body mass index (BMI) (P < 0.01), and MCR-I had significant negative correlation with AUCC (P < 0.01). CONCLUSIONS: There are obvious impaired first phase insulin secretion after glucose challenge in non-insulin-dependent diabetic subjects from MDP. Decrease in endogenous MCR-I might be an important factor to hyperinsulinemia and insulin resistance. Increased insulin secretion, decreased MCR-I and insulin sensitivity can be observed in abdominal obese subjects of control group.

[Association of HLA-DRB1 allele and the susceptibility to alveolar echinococcosis in the west of China].

OBJECTIVE: To investigat the association of HLA-DRB1 alleles and the susceptibility to alveolar echinococcosis (AE). METHODS: Thirty-five patients with AE in the high prevalence areas in the west of China were investigated for HLA-DRB1 gene by PCR/SSP technique. The results were compared with 104 normal healthy peoples. RESULTS: Frequency of HLA-DRB1 * 040x was 26% in patient group, which was significantly higher than in control group (9.62%), with a relative risk (RR) of 4.45 (chi(2) = 13.67, P < 0.01), and with an etiological fraction of 0.20. Frequency of HLA-DRB1 * 0701 allele was decreased in patient group (patient 2.86%, control 13.94%; chi(2) = 6.67, P < 0.05), with preventable fraction of 0.30. While the frequencies of other DRB1 alleles were not significantly different. CONCLUSION: Susceptibility to AE is strongly associated with the HLA-DRB1 * 040x allele. The high prevalence of human AE in this region was associated with genetic factor. HLA-DRB1 * 0701 geners resistant to Echinococcus multilocularis metacestode infection.

[Detection of remnants after removal of medullary thyroid carcinoma].

OBJECTIVE: To assess whether calcium stimulation test or somatostain-receptor (SS-R) imaging could early detect remnants after removal of medullary thyroid carcinoma (MTC). METHODS: Calcitonin stimulation (calcium element 3 mg/kg weight, i.v. 10 min), carcino-embryonic antigen (CEA), gastrin (GST) and vasoactive intestinal peptide (VIP) were tested in 14 patients with postoperative MTC. SS-R imaging was used to localize the remnants after removal of MTC in patients with elevated calcitonin. RESULTS: Calcitonin stimulation test showed that the peak value of serum calcitonin was elevated in all patients, of whom 7 had the elevated vatue of basic calcitonin and peak calcitonin. In the 7 patients, SS-R imaging showed normal CEA, GST and VIP. CONCLUSIONS: It is necessary for patients after removal of MTC to perform calcitonin stimulation test for detecting remnants early. SS-R imaging may be useful for localizing remnants and metastatic foci. CEA, GST and VIP are not significant in monitoring MTC after operation.

[Anisodamine in prevention and treatment of sepsis of severely burned patients].

OBJECTIVE: To observe the preventive effect of anisodamine on possible sepsis of patients with major burns and the effect of anisodamine on patients with sepsis. METHODS: Forty-two patients with extensive burn admitted to our burn institute from April 1998 to November 1999 were divided randomly into two groups: treatment group (T group) and control group (C group). In the T group, all 20 patients received fluid resuscitation regimen with anisodamine, and in the C group, 22 patients received the regimen with no anisodamine. A tonometry catheter was positioned in the stomach, connecting with the automatic gas analysis machine (Datex-Engstrom Corporation, Dutch) for determining gastric intramucosal pH (pHi). The plasma concentrations of diamine oxidase (DAO) and endotoxin were measured. Correlation analysis between pHi, DAO and endotoxin were made respectively during early stage of postburn. All the parameters in 7 patients with sepsis before and after administration of anisodamine were compared with those in 6 patients with sepsis without use of anisodamine. RESULTS: The incidence of sepsis in the T group was lower (20.0%) than that in C group (40.9%). The gastric pHi value in the early period of postburn was significantly higher in the T group than in the C group (P < 0.05). Concurrently, the plasma concentrations of DAO and endotoxin were significantly lower in the T group than in the C group (P < 0.05 or 0.01). A significant negative correlation was seen between the gastric pHi and respective values of DAO, endotoxin (P < 0.05 or 0.01). There were a decrease in gastric pHi, and an increase in plasma DAO and endotoxin level in patients with septic episode; however all the parameters after administration of anisodamine were improved compared with those in septic patients without use of anisodamine. CONCLUSIONS: Intestinal ischemic injury plays an important role in provoking sepsis during early postburn period. Anisodamine is effective in restoring intestinal circulation both in the shock phase and after the development of sepsis.

[Protective effect of nitric oxide on pancreas and its relation to sulfhydryl compounds and oxygen free radicals].

OBJECTIVE: To investigate the effect of endogenous nitric oxide (NO) on acute necrosis pancreatitis in rats and its relation to sulfhydryl compounds and lipid peroxidation. METHODS: Acute necrosis pancreatitis in rats was induced by retrograde sodium taurocholate (5%) infusion into the pancreatobiliary duct (1 ml/kg body weight), and N(G)-nitro-L-arginine (L-NNA) was used as the inhibitor of endogenous NO. The effect of endogenous NO on pancreatic injury, serum amylase level, the pancreatic tissue levels of sulfhydryl compounds, and malonaldehyde (MDA, the end product of lipid peroxidation) was evaluated, respectively. RESULTS: Sodium taurocholate administration induced evident pancreatic tissue edema and acinar necrosis, and intrapancreatic hemorrhage occurred in 2/7 rats. Both serum amylase and tissue MDA [(1.25 +/- 0.28) nmol/mg x pr vs. (0.5 +/- 0.03) nmol/mg x pr, P < 0.05] were significantly increased, but tissue sulfhydryl compounds were decreased markedly. Pretreatment with the NO inhibitor, L-NNA (12.5 mg/kg body weight), significantly intensified acinar necrosis and increased the intrapancreatic hemorrhage (10/12). L-NNA also resulted in a further increase of serum amylase and tissue MDA [(3.0 +/- 0.40) vs. (1.25 +/- 0.28) nmol/mg x pr, P < 0.05], but it had no effect on the tissue sulfhydryl compounds. CONCLUSION: Endogenous NO has the effect of pancreatic protection, and its antioxidation may be responsible, at least in part, for the protective mechanisms. Sulfhydryl compounds may not be involved in NO's pancreatic protection mechanisms.

[The expression and clinical significance of p80 protein in anaplastic large cell lymphoma].

OBJECTIVE: To investigate the relationship of expression of NPM-ALK cohesive gene protein with the subtypes and prognosis of anaplastic large cell lymphoma (ALCL). METHODS: Nineteen cases of ALCL with CD30 positivity were studied. Immunohistochemical staining was performed in paraffin sections by using polyclonal antibody anti-p80 and other monoclonal antibodies. The clinical data of these patients were analyzed and all patients were followed-up. RESULTS: The neoplastic cells in 9 out of 19 ALCL expressed p80 protein. Histopathologically, all 9 cases with p80 expression were common type and small cell type. None of these 9 cases were Hodgkin like type (P < 0.05). Immunologically, all the 9 positive cases were T and null cell phenotype and none of them was B cell or other phenotype (P < 0.05). Clinically, the patients with p80 expression were younger than those without p80 expression (P < 0.05). The prognosis of the former was worse than in the later (P < 0.05). CONCLUSIONS: p80 positive ALCL could be a new clinicopathological entity. The polyclonal antibody anti-p80 might be an useful marker for the differential diagnosis and prognosis estimation of ALCL.

[Subcutaneous panniculitis-like T-cell lymphoma: expression of cytotoxic-granule-associated protein TIA-1 and its relation with Epstein-Barr virus infection].

OBJECTIVE: To study the clinicopathologic features, the expression of cytotoxic-granule-associated protein TIA-1 and Epstein-Barr virus (EBV) infection in subcutaneous panniculitis-like T-cell lymphoma (SPTCL). METHODS: Totally 17 cases of SPTCL were studied. Immunohistochemical staining was done for TIA-1, CD45RO, CD3, CD20 and CD68, and in situ hybridization for EBV encoded small nuclear RNA (EBER1/2). RESULTS: In the 17 cases of SPTCL, the ratio of male-to-female was 1:1.1 and the median age was 24 years old. The main clinical manifestations were asymptomatic plaques, nodules and masses. Histologically, the pattern of lymphoid cells infiltration was panniculitis. The infiltrated lymphoid cells were pleomorphic and their mitotic figures were few (0 - 2/hpt). In the tumor tissue, there might be epithilioid granulomas, multinuclear cells, beanbag cells, small pieces of necrosis and small vessels with lymphoid cells infiltration in the vascular wall. Immunohistochemically, all the cases showed a T-cell phenotype and the expression rate of TIA-1 was 100% (16/16). By in situ hybridization, the positive rate of EBV-EBER1/2 was 29.4% (5/17). CONCLUSIONS: SPTCL has a specific histopathologic picture. It may originate from cytotoxic T lymphocytes or NK cells. Some of them may be accompanying with EBV latent infection.

[The effects of zinc supplementation on postburn nutritional status of zinc-deficient scalded rats].

OBJECTIVE: To investigate the effects of zinc supplementation on postburn nutritional status of zinc-deficient scalded rats. METHODS: Weanling Wistar rats were fed with zinc-deficient diets for one week to induce zinc-deficient status. The rats were inflicted with 20% TBSA deep partial thickness burn and thereafter fed with diets of low, normal and high zinc contents. The scalded rats fed with normal zinc-containing diet before and after injury were taken as normal control group. The changes of body weight, cumulative nitrogen balance and the contents of DNA and protein in jejunum mucous, liver, kidneys, spleen and testis were employed to represent the nutritional status of the rats. RESULTS: The rats fed with diets with normal and high contents of zinc were superior to those with low content of zinc in terms of body weight, food conversion rate, cumulative nitrogen balance and organ protein contents. The serum zinc level, alkaline phosphatase (ALP) activity and the DNA contents in jejunum mucous, kidneys, spleen and testis increased significantly after zinc supplementation. CONCLUSION: Zinc supplementation was helpful to improve postburn nutritional status in scalded rats. And relative higher amount of zinc supplementation might be beneficial to raise the DNA content in testis and kidneys when compared to that with normal zinc supplement.

[The study of beta-fibrinogen gene - 455 G/A, - 148 C/T, 448 G/A polymorphisms and their association with plasma fibrinogen levels].

OBJECTIVE: To analyze the frequency of beta-fibrinogen (beta-Fg) gene - 455 G/A, - 148 C/T, 448 G/A polymorphisms and their association with plasma fibrinogen levels in Han nationality in Guangdong Chinese. METHODS: The beta-Fg gene - 455 G/A, - 148 C/T, 448 G/A polymorphisms of 156 individuals were analyzed by restriction fragment length polymorphism (RFLP). Plasma fibrinogen levels were determined by turbidimetry. RESULTS: The frequencies of A(- 455), T(- 148) and A(448) allele were 0.276, 0.285 and 0.272, respectively. There were strong linkages of G(- 455), C(- 148) and G(448), and of A(- 455), T(- 148) and A(448), the correspondence was more than 97%. The plasma fibrinogen levels in the presence and absence of A(- 455) allele were (3.13 +/- 0.74) g/L and (2.89 +/- 0.57) g/L (P < 0.05); of T(- 148) allele were (3.12 +/- 0.73) g/L and (2.89 +/- 0.58) g/L (P < 0.05); and of A(448) were (3.13 +/- 0.74) g/L and (2.89 +/- 0.57) g/L (P < 0.05), respectively. The plasma fibrinogen levels of the three polymorphisms with the mutant gene are significantly higher than that in the wild type. CONCLUSION: The three polymorphisms loci are strong linkage disequilibrium. It suggests that beta-Fg gene - 455 G/A, - 148 C/T, 448 G/A polymorphisms are associated with plasma fibrinogen levels.

[The change and distribution of endothelin-1 in lung of hypoxic pulmonary hypertension rat].

This study was aimed to determine the distribution and change of ET-1 in the lung of rat with chronic hypoxic pulmonary hypertension. Immunohistochemical analysis (Avidin-Biotion Complex) was used to localize the ET-1 immunoreactivity in the rat lung. The ET-1 concentration in plasma and in lung homogenate was measured by radioimmunoassay. In normal rats, the mean ET-1 concentration in venous plasma was 2.25 +/- 0.68 ng/L, in arterial plasma 1.52 +/- 0.63 ng/L and in lung 1.75 +/- 0.46 ng/L. There was no significant increase of ET-1 level in rats exposed to hypoxia for 2 hours, but there was significant increase of ET-1 after 24 hours hypoxia, and the high levels of ET-1 were maintained in the sustained hypoxia. There was significant correlation between the ET-1 level in arterial plasma(or in lung homogenate) and the maen pulmonary artery pressure. Similaryly, there was significant negative correlation between ET-1 in arterial plasma(or in lung homogenate) and PaO2. Immunohistochemical analysis revealed that the ET-1 immunoreactivity was seen in the endothelium of pulmonary arteries, particularly more positive staining was seen in the band between the endothelium and the smooth muscle cells. Chronic hypoxia elevated the mean pulmonary arterial pressure, caused vessel remodelling and the right ventricular hypertrophy. These changes were accompained by an increase of ET-1 in plasma and lung homogenate. The expression and production of ET-1 were localized to endothelium and airway epithelium in the lungs.

[The lymphocyte subset and blood interleukin-2 level in children with Henoch-Schönlein purpura before and after cimetidine treatment].

The objective of this study was to evaluate the effect of cimetidine treatment on cellular immunity in children with Henoch-Schönlein purpura. T cell subset was detected by double marker flow cytometry and the level of IL-2 in peripheral blood was measured by MTT method, before and after cimetidine treatment. The results showed that CD3+, CD4+ T cell subset, CD4+/CD8+ ratio, and blood IL-2 level increased after the treatment (P < 0.05). There was no significant change in CD8+ T subset after the treatment. These findings indicate that cimetidine can promote T lymphocyte proliferation and differentiation, especially TH1 lymphocyte activity.

[Expressions of apoptosis-associated proteins in prostate carcinoma and their clinicopathological significance].

This study was made to explore the expressions of P53, P16, Bc1-2 and Bax protein in prostate carcinoma and their relations to histopathological grading, clinical staging and prognosis of this tumor. Immunohistochemical staining method was used to assess the expression of P53, P16, Bc1-2 and Bax protein in 51 cases of prostate carcinoma. The positive rates of P53, P16, Bc1-2 and Bax proteins were 31.37%, 54.90%, 21.57% and 74.51% respectively. The expression of P53 and Bc1-2 showed positive correlation with the histopathological grading and clinical staging of this tumor (P < 0.05). However, the expression of P16 and Bax showed negative correlation with the grading and staging of the tumor (P < 0.05). The expression of P53 protein correlated with the survival rate of these patients. There was a relation between P53 and Bc1-2, and a relation between Bc1-2 and Bax also (P < 0.05). These findings suggest that expressions of P53, P16, Bc1-2 and Bax may be helpful to the grading, staging and prognostic evaluation of prostate carcinoma.

[Study on IGF-I regulation the proliferation of rat condylar chondrocytes in vitro].

This study was aimed at the mechanism for the changes in the proliferation of cultured rat mandibular condylar chondrocytes. Flow cytometry was used to explore the changes of cellular DNA content and cell cycles of passaged condylar chondrocytes. The results showed that when the mandibular condylar chondrocytes were cultured with IGF-I for 24 hours and 96 hours, the cellular proliferation in the experimental group enhanced higher than that in the control group. It was implied that IGF-I may be a mitogen on rat mandibular condylar chondrocytes. The mitogenic effect of IGF-I had a lag period.

[Tn5 mutagenesis and the characteristics of indole-3-acetic acid biosynthesis in Alcaligenes faecalis A1501].

The IAA production of Alcaligenes faecalis wild type strain A1501 depended on addition of L-tryptopham(trp). A1501 could grow well in minimal medium without Trp, but could not produce IAA. The results indicated that there was Trp-dependent IAA biosynthesis pathway in A. faecalis A1501. A1501 shows a characteristic of cell density-dependent IAA biosynthesis. The Tn5-induced mutant bank of A1501 was constructed using Tn5 mutagenesis and one Trp auxotrophic mutant AT63 was selected and identified. The Tn5-induced mutant AT63 could synthesis 2.24 micrograms/mL IAA in minimal medium without L-Trp. It indicated there were at least two IAA biosynthesis pathways in A. faecalis, one needed Trp, the other needed IGP as precursor. The results of southern hybridization also suggested that the Tn5-insertion might locate in gene locus encoded IAA synthetase.

[Screening of acrystalliferous mutants from Bacillus thuringiensis and their transformation properties].

After elevating growth temperature of YBT-1463 and other 8 Bacillus thuringiensis parental strains to 42 degrees C, 9 acrystalliferous(Cry-) mutants were obtained, and then a series of partial plasmid-cured mutants were further obtained from 3 Cry- mutants BMB151, BMB154 and BMB160 by elevating temperature to 44 degrees C. One of which, BMB170, was further treated by elevating temperature to 46 degrees C and adding 0.05% SDS as the plasmid-curing agent. The resulting mutant, which was designated BMB171 and had no resident plasmid, was finally obtained. The studies on the transformation efficiency and stability of Cry- mutants showed that the transformation frequencies, as well as stability of several exogenous plasmids(pHT3101, pBMB1736, etc.) introduced into Cry- mutants by electroporation, were correlated with the numbers of resident plasmids bearing by the relevant recipients, the patterns of plasmid replicons and the sizes of those exogenous plasmids. Meanwhile, a much higher electro-transformation frequencies of Cry- mutants than the parental strain YBT-1463 was found, and the highest of the transformation frequency, in BMB 171, could reach up to 10(7) transformants/microgram DNA, and the stability of those exogenous plasmids in BMB171 were also higher than in other Cry- mutants and YBT-1463 strain.

[Boot colonization of wheat by lux-AB genes marked Pseudomonas fluorescens Xl6L2].

Colonization density of Pseudomonas fluorescens Xl6L2 marked with luxAB genes in wheat rhizosphere in asepsis rhizobox-Calcareous chao soil microcosms reached the maximum(4.60 log cfu.g-1 root) 3 days after seeds coated with Pf.Xl6L2 sown, then declined to a relatively stable lower level(2.45 log cfu.g-1 root) in 0-2 cm root segment. Dynamics of colonization of Pf.Xl6L2 in wheat rhizosphere under field conditions was the same as in microcosms, Pf.Xl6L2 could move to the place of 10 cm of root from seeds under field conditions, distance of horizontal movement of Pf.Xl6L2 in field soil was not over 40 cm during 125 days of plant growth.
