[Structural polymorphism of the provirus integrated in the genome of mammalian cells transformed by Rous sarcoma virus].

The structural organization of integrated Rous sarcoma virus (RSV) genome in 8 different mammalian tumour cell lines has been studied. The different types of provirus rearrangements were found, e.g.: breakpoint mutations, deletions of RSV structural genes, elimination of the entire viral genome. The structural changes of proviruses appeared during long-term cultivation of tumour cell lines in vitro. The ability of transformed cells to produce complete infectious RSV correlate with the structural peculiarities of proviruses, although the tumorigenic properties of these cells are retained in all cases.

[Human T-lymphotrophic retroviruses].

Investigations of retroviruses, possible etiological agents of T-cell human leukemias and acquired immunodeficiency syndrome are reviewed. Main attention is paid to the genome structure and function.

[Isolation and characteristics of antibodies against synthetic fragments of oncoproteins. II. Antiserum against the transforming protein pp60src of Rous sarcoma virus].

To generate the antibodies to the transforming protein of Rous sarcoma virus (RSV) pp60src, rabbits were immunized with the peptide, corresponding to 415-421 sequence of pp60src. These antibodies immunoprecipitate pp60src in RSV-transformed chicken and mammalian cells, and also some proteins (45, 85 and 120 kDa), which could be autophosphorylated in vitro. It was shown that 415-421 sequence of pp60src is not recognized by the antibodies to pp60src from RSV-induced tumour bearing rabbits (TBR serum). In contrast to TBR serum, antibodies, generated against synthetic peptide, corresponding 415-421 sequence of pp60src couldn't be phosphorylated in vitro, when [gamma-32P]ATP is added to the immune complex. The antipeptide antibodies, bound to pp60src did not block phosphorylation of TBR immunoglobulins, added to this immune complex. Hence, 415-421 sequence of pp60src RSV containing the major tyrosine phosphorylation site does not take part in the kinase reaction in vitro.

[Transcription of cellular oncogenes in human tumors].

The analysis of 11 various oncogenes expression in different human tumors showed that each tumor is characterised by a specific functioning program of these genes. In 40-50% of tumors the oncogenes ras, fos and myc are expressed. All other oncogenes are either considered to be "silent" or are expressed only in few cases. The increased expression of sis and myb oncogenes is observed in metastases.

[Biochemical and physiocochemical characteristics of a type C virus isolated from spontaneous lymphosarcoma of CC57Br strain mice].

The properties of murine oncornavirus produced by cells of spontaneous lymphosarcroma of CC57Br mice are described. In addition to the properties common for C-type RNA tumor viruses such as 60-70 S high molecular weight RNA, the presence of RNA-directed DNA polymerase and murine gs I (intraspecies) antigen and typical morphology in electron microscope, etc., the virus under study is characterized by instability of virions, unusual features of the DNA polymerase system and by the absence of demonstrable oncogenicity either in laboratory animals or in tissue cultures.

[Molecular biology criteria for functioning of the genome of Rous sarcoma virus in transformed rat XC cells].

Integration, transcription and translation of Rous sarcoma virus (RSV) genome was analyzed in transformed rat fibrosarcoma XC cells (non-virus producing). About ten copies of RSV genome were integrated into the host cell genome. Different types of viral RNA were identified in the cytoplasmic fraction whose sedimentation values were nearly the same as for RSV-specific RNA in virus-producing chicken cells. These RNA were translated in polysomes and directed synthesis of gag, env and src proteins. In comparison to chicken cells in transformed XC cells the cleavage of gag precursor Pr 76gag could not be observed.

[Nuclear oncoprotein expression in lung precancer and cancer at various stages of tumor progression studies at the level of light- and electron-immunohistochemistry].

68 cases of lung carcinoma, 3 carcinoids and 15 fibrosing alveolitis with foci of adenomatosis and bronchiolo-alveolar carcinoma were studied. Oncoproteins c-fos, c-jun, c-ets-1, c-myc L and L-myc were identified in the tumour and surrounding tissue. Expression of c-fos was revealed in 79 of 138(59.4%) of proliferative and dysplastic changes of lung epithelium; c-jun in 40 of 61 (65.6%), c-ets-1 in 22 of 41 (53.7%), c-myc in 41 of 96(42.7%) and L-myc in 15 of 61 (24.6%), mainly in altered bronchial epithelium with a positive reaction to the antibodies against neuron specific enolase and S100 protein. More pronounced expression of nuclear oncoproteins, heterogeneity of their location in tissues, frequent cytoplasmic location in tumour cells were typical for lung carcinoma.

[Cell oncogene expression in normal, metaplastic, dysplastic epithelium and squamous cell carcinoma of the uterine cervix].

Immunohistochemical analysis of the protein expression c-myc, ets 1, ets 2, TPR-met, c-fos, c-jun, c-ras-pan, p53, yes, src in 79 samples of normal, metaplastic squamous epithelium, intraepithelial and invasive squamous cell carcinoma of uterine cervix was performed using polyclonal rabbit antibodies to the synthetic peptides homologous active areas of corresponding oncoproteins. Higher content of myc, fos, ets2, p53, ras is noted in metaplasia, dysplasia and in tumours as compared to the normal tissues. Protein myc is revealed in the cytoplasm at a grave dysplasia and in the nucleus in the intraepithelial carcinoma: this may serve as a criterion at a differential diagnosis of these conditions. Expression of the oncoproteins fos, ets2, p53, src in the metaplastic squamous cell carcinoma was higher than in the true squamous cell (ectocervical) carcinoma. When compared to the advanced carcinomas, increase of ets2, p53, and at some degree that of myc, the increase is noted in the latter. Invasive carcinoma with a high level of oncoproteins showed a tendency to the synchronization of myc and ras expression. Poor prognosis was associated with a low level (before treatment) of the expression of the majority of the oncoproteins studied.

[In vivo identification of YB-1 protein, interacting with the enhancer of human papillomavirus (HPV) type 18, using antibodies to a synthetic peptide].

Enhancer sequences of human papilloma virus (HPV) type 18 were used for screening of HeLa cells cDNA library in lambda gt11 using the protein binding method. Clones with YB I gene homology sequences were isolated. This gene is coding the protein which binds the regulatory region of Y gene of main histocompatibility complex (HLA 11). The YB I transcripts were revealed in all tested samples of cervical carcinomas. To analyze the protein the rabbit antibodies were produced to synthetic peptide, which corresponds to the most hydrophilic region of the protein. This antipeptide serum allowed to identify the nuclear 42K protein in HeLa cells as well as in normal fibroblasts.
