Alteration of the aurophilic interactions in trimeric gold(I) compounds through charge transfer. Behavior of solvoluminescent Au(3)(MeN=COMe)(3) in the presence of electron acceptors.

The ability of the triangular gold(I) complex, Au(3)(MeN=COMe)(3), which as a solid displays the novel property of solvoluminescence (see: Vickery, J. C.; Olmstead, M. M.; Fung, E. Y.; Balch, A. L. Angew.Chem., Int. Ed. Engl. 1997, 36, 1179) to function as an electron donor has been demonstrated through spectroscopic studies and isolation of crystalline adducts with organic acceptor molecules. Four such adducts with nitro-9-fluorenones have been isolated and subject to single-crystal X-ray diffraction study. These are deep yellow [Au(3)(MeN=COMe)(3)].[2,4,7-trinitro-9-fluorenone], red [Au(3)(MeN=COMe)(3)].[2,4,5,7-tetranitro-9-fluorenone], red [Au(3)(MeN=COEt)(3)](2).[2,7-dinitro-9-fluorenone], and red [Au(3)(MeN=COEt)(3)](2).[2,4,7-trinitro-9-fluorenone]. The solid-state structures of [Au(3)(MeN=COMe)(3)].[2,4,7-trinitro-9-fluorenone] and [Au(3)(MeN=COMe)(3)].[2,4,5,7-tetranitro-9-fluorenone] consist of columns in which the planar gold(I) trimers and the nearly planar nitro-9-fluorenones are interleaved with the gold trimers making face-to-face contact with the nitroaromatic portion of the electron acceptor. Thus the organic acceptors disrupt the aurophilic interactions present in crystalline [Au(3)(MeN=COMe)(3)] itself. However, in [Au(3)(MeN=COEt)(3)](2).[2,7-dinitro-9-fluorenone] and [Au(3)(MeN=COEt)(3)](2).[2,4,7-trinitro-9-fluorenone], aurophilic interactions are found which produce dimers, [Au(3)(MeN=COEt)(3)](2), with nearly trigonal prismatic Au(6) cores. These dimers are interleaved with the nitro-9-fluorenone molecules to again form extended columns in which the components make face-to-face contact. Despite the fact that the gold atoms in [Au(3)(MeN=COMe)(3)] and [Au(3)(MeN=COEt)(3)] are in exposed sites and only two-coordinate, there is no evidence of additional coordination of the nitro-9-fluorenones with gold centers in the crystalline adducts.

A study of toxicity of 5-fluorouracil on bovine trabecular meshwork cells in vitro.

In order to explore whether the conventional use of 5-fluorouracil (5-Fu) had any toxic effects on trabecular meshwork cells, bovine trabecular meshwork cells were cultured in vitro and exposed to 5-Fu at different concentrations. The cellular morphology, ultrastructure, mortality and phagocytosis were studied under light microscopy, transmission electron microscopy and methods of Wright's stain. It was found that the toxic effects of 5-Fu on the cells were in a dose-dependent mode. 1 x 10(-1) mg/ml of 5-Fu caused a large part of cells rounded up, while 1 x 10(-2) mg/ml of the drug only a rough appearance of the cell surface. Exposure to 1 x 10(-2) mg/ml of 5-Fu made mitochrone swollen and rough endoplasmic reticulum enlarged, with the cell mortality being 50.5%. The latex microspheres engulfed in cytoplasm in cells receiving 1 x 10(-1) and 1 x 10(-2) mg/ml of 5-Fu were significantly decreased as compared with those in the control group (P < 0.01). It was concluded that the safe concentration of 5-Fu on bovine trabecular meshwork cells was 1 x 10(-2) mg/ml and the conventional dosage of 5-Fu in clinical practice would not cause injury to trabecular meshwork cells.

Creating discrete joint densities from continuous ones: the moment matching-maximum entropy approach.

An approach for converting continuous densities into discrete ones with a pre-assigned set of support points is developed. The algorithm performs this conversion in a most skeptical, least-informative way, by finding the discrete distribution with the maximum entropy that satisfies a set of moment constraints derived from the stated continuous distribution, such as means and variances. Individualized drug therapies based on multiple model control rely on the availability of discrete distributions to generate the underlying model set. The methods developed herein are especially compatible with individualized drug therapies based on multiple model control that rely on the availability of discrete densities to generate the model set.

Farnesyl diphosphate synthase is abundantly expressed and regulated by androgen in rat prostatic epithelial cells.

Farnesyl diphosphate synthase (FPPS) has been identified as an androgen-response gene in the rat ventral prostate using a highly sensitive PCR-based cDNA subtraction technique. FPPS is an essential enzyme that catalyzes the synthesis of farnesyl diphosphate (FPP), which is required for cholesterol biosynthesis as well as protein prenylation. We have characterized the expression of FPPS in the rat prostate in response to androgen manipulation. Northern blot analysis showed that castration induced a 10-fold down-regulation of FPPS mRNA within 24 h in the ventral prostate and androgen replacement up-regulated FPPS mRNA rapidly in the regressed ventral prostate of a castrated rat. The expression of FPPS was also regulated by androgen in the lateral and dorsal prostate, indicating that FPPS is important to androgen action in all three lobes of the prostate. Western blot analysis showed that FPPS protein level was also regulated by androgen in the prostate. Northern blot analysis of tissue specificity indicated that FPPS was most abundantly expressed in the ventral prostate of a mature rat and was responsive to androgen manipulation in the prostate and seminal vesicles, but not in other tissues. In situ hybridization study showed that FPPS mRNA was localized to the prostatic epithelium. Interestingly, the expression of FPPS was elevated in Dunning rat prostate tumor cell lines. The above findings suggest that FPPS has the potential to play an important role in androgen action and prostate cancer progression.

Peptide-triggered conformational switch in HIV-1 RRE RNA complexes.

We have used NMR spectroscopy to determine the solution structure of a complex between an oligonucleotide derived from stem IIB of the Rev responsive element (RRE-IIB) of HIV-1 mRNA and an in vivo selected, high affinity binding Arg-rich peptide. The peptide binds in a partially alpha-helical conformation into a pocket within the RNA deep groove. Comparison with the structure of a complex between an alpha-helical Rev peptide and RRE-IIB reveals that the sequence of the bound peptide determines the local conformation of the RRE peptide binding site. A conformational switch of an unpaired uridine base was revealed; this points out into the solvent in the Rev peptide complex, but it is stabilized inside the RNA deep groove by stacking with an Arg side chain in the selected peptide complex. The conformational switch has been visualized by NMR chemical shift mapping of the uridine H5/H6 atoms during a competition experiment in which Rev peptide was displaced from RRE-IIB by the higher affinity binding selected peptide.

Endothelium-dependent vasorelaxation independent of nitric oxide and K(+) release in isolated renal arteries of rats.

1. We investigated whether K(+) can act as an endothelium-derived hyperpolarizing factor (EDHF) in isolated small renal arteries of Wistar-Kyoto rats. 2. Acetylcholine (0.001 - 3 microM) caused relaxations that were abolished by removal of the endothelium. However, acetylcholine-induced relaxations were not affected by the nitric oxide (NO) synthase inhibitor N:(omega)-nitro-L-arginine methyl ester (L-NAME, 100 microM), by L-NAME plus the soluble guanylate cyclase inhibitor 1H-[1,2,4]oxadiazolo[4,3,-a]quinoxalin-1-one (ODQ, 1 microM) or by L-NAME plus the cyclo-oxygenase inhibitor indomethacin (10 microM). In rings precontracted with high-K(+)(60 mM) physiological salt solution in the presence of L-NAME, acetylcholine-induced relaxations were abolished. 3. L-NAME-resistant relaxations were abolished by the large-conductance Ca(2+)-activated K(+) channel inhibitor charybdotoxin plus the small-conductance Ca(2+)-activated K(+) channel inhibitor apamin, while the inward rectifier K(+) channel inhibitor Ba(2+) or the gap junction inhibitor 18alpha-glycyrrhetinic acid had no effect. Acetylcholine-induced relaxation was unchanged by ouabain (10 microM) but was partially inhibited by a higher concentration (100 microM). 4. In half of the tissues tested, K(+)(10 mM) itself produced L-NAME-resistant relaxations that were blocked by ouabain (10 microM) and partially reduced by charybdotoxin plus apamin, but not affected by 18alpha-glycyrrhetinic acid or Ba(2+). However, K(+) did not induce relaxations in endothelium-denuded tissues. 5. In conclusion, acetylcholine-induced relaxations in this tissue are largely dependent upon hyperpolarization mechanisms that are initiated in the endothelium but do not depend upon NO release. K(+) release cannot account for endothelium-dependent relaxation and cannot be an EDHF in this artery. However, K(+) itself can initiate endothelium-dependent relaxations via a different pathway from acetylcholine, but the mechanisms of K(+)-induced relaxations remain to be clarified.

Effect of top excision and replacement by 1-naphthylacetic acid on partition and flow of potassium in tobacco plants.

The effect of removal of the shoot apex of 92-d-old tobacco plants and its replacement by 1-naphthylacetic acid (NAA) on sink-source relationships and on the flows and partitioning of potassium and water has been studied over a short-term period of 7 d (intact control plants) or 8 d (decapitated and NAA-treated plants). For determining flows an upper, middle and lower stratum of three leaves each were analysed. Within the study period three new leaves were formed in control plants and 57.7% of the total dry matter increment during the experimental period was allocated to the apex and these newly formed leaves. An even higher proportion of the K+ taken up (93.8%) was deposited in these organs and this was imported via xylem (72%) and phloem (28%). Only 18.7% and 9.8% of the total dry matter increment were found in the previously present upper leaves and the roots, respectively, and substantial net K+ export occurred from middle and lower leaves and roots. Decapitation removed the dominant phloem sink and caused marked changes in sink-source relationships. After decapitation the net increase in root dry matter was twice that of control plants. 56.2% of the total net increments in dry matter and 70% of the absorbed K+ were deposited in upper leaves (below the excised apex). There was only slight net K+ export from the middle leaves. Application of NAA on the cut surface of the stem stump did not change the growth of plants that much, apart from a substantial increase in stem growth, correspondingly it stimulated the partitioning of K+ into the upper leaves and most dramatically into the stem, which deposited 64.5% or 27% of the K+ uptake, respectively. In these plants K+ uptake was increased and the K+ concentrations in upper, middle and lower leaves were increased from 4.7, 5.4 and 5.6 to 5.1, 6.1 and 6.1% of dry matter, respectively. Possible mechanisms of this effect of NAA on the improvement of K+ concentration in tobacco leaves are discussed in detail.

Effects of alcohol on blood pressure and production of vascular aldosterone and corticosterone.

AIMS: The aims of this study were to search for the role of alcohol in blood pressure regulation in rats and to investigate the effects of alcohol on the production of vascular aldosterone and corticosterone. METHODS: Male Wistar rats received alcohol 0.7 g x kg(-1) x day(-1) (alcohol-treated group 1) or 1.4 g x kg(-1) x day(-1) (alcohol-treated group 2) or 2.1 g x kg(-1) x day(-1) (alcohol-treated group 3), orally, for 3 months, and blood pressure was monitored by a pressure transducer. Systolic blood pressure increased in Wistar rats treated with alcohol compared to control rats. Mesenteric artery perfusion ex vivo was performed and pressor responses to norepinephrine were determined. The pressor responses to norepinephrine in mesenteric arteries treated with alcohol were significantly increased. The perfusate from the mesenteric arteries was collected and applied to a Sep-Pak C 18 cartridge column for reverse-phase high-performance liquid chromatography and aldosterone and corticosterone levels were determined by radioimmunoassay, aldosterone was decreased but corticosterone was increased in the perfusate from arteries treated with alcohol. RESULTS: Reverse transcriptase polymerase chain reaction showed that alcohol inhibited the expression of 11beta-HSD2 and CYP11B2 mRNA in mesenteric arteries. CONCLUSIONS: These results reveal that alcohol is able to induce hypertension and provide evidence that alcohol inhibits the transcriptions of both 11beta-HSD2 and CYP11B2 in the vasculature, leading to lower aldosterone and higher corticosterone production in vessels, and increased vasoconstrictor responses to norepinephrine.

[Correlation between electroretinographic findings, clinical phenotypic and genotypic analysis in Duchenne and Becker muscular dystrophy].

OBJECTIVE: To explore the relationship between electrophysiological changes, clinical phenotype and genotype in Duchenne and Becker muscular dystrophy(DMD/BMD), to address the expression and roles of dystrophin and its isoforms on the retina, and to inquire into the molecular mechanism of the abnormal electroretinogram(ERG) on DMD/BMD patients with different genotype. METHODS: Gene deletions were screened by multiplex DNA amplification with eleven primers on twenty-two consecutive patients with DMD and BMD, and then, the ERG was tested according to international ERG standard. RESULTS: ERG phenotype was associated with the site of DMD gene defects rather than the severity of the phenotype. Patients with deletion in the central region of the gene had more severe changes in the scotopic ERG as compared to those with gene non-deletion. CONCLUSION: The ERG genotype-phenotype correlation suggests that DP260 may play the most important role in the retinal neurotransmission.

[Combustion characteristics of municipal solid waste in fluidized bed].

For the experimental investigation on the combustion characteristics of Municipal Solid Waste(MSW) in Fluidized Bed, a lab scale Fluidized Bed facility was constructed. Single and mixed municipal solid waste burning in Fluidized Bed showed that dry waste can be burnt quickly at the bed temperature of 500 degrees C, and furnace temperature increased about 30-50 degrees C. Many kinds of combustion runs were conducted in this Fluidized Bed combustion facility. The parameters examined were air flow rate(from 5.5 m3/h to 7.5 m3/h), form of fuels(scrap or whole), moisture content and so on. Concentration of CO2, CO, SO2 and NOx in the flue gas were monitored and recorded every 5 seconds. The temperatures along the reactor were recorded every 10 seconds.

[Relationship of vitamin D receptor polymorphism with breast cancer].

OBJECTIVE: To detect the association between vitamin D receptor polymorphism and breast cancer. METHODS: Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method was used. Two restriction fragment length polymorphisms in the 3'region of vitamin D receptor (VDR) gene, Taq I and Apa I, were tested for association with breast cancer risk in 86 females with breast cancer and 134 healthy female controls. RESULTS: Allele frequencies of the 3'Taq I polymorphism showed a significant association (P=0.0004, OR:5.39,CI:1.81-17.20). The further study of genotype found the association of Tt, tt with breast cancer. The haplotype analysis of Apa I and Taq I showed a linkage disequilibrium between t-allele and A-allele. The frequency of tA haplotype was higher in breast cancer patients than in controls (P=0.001), indicating that tA haplotype is associated with breast cancer. The alleles and haplotype of the two loci had not any difference among the clinical subgroups. CONCLUSION: There is association between vitamin D receptor polymorphism and breast cancer.

[Complex segregation analysis of systemic lupus erythematosus].

OBJECTIVE: To explore the genetic model of systemic lupus erythematosus(SLE). METHODS: Complex segregation analysis was performed by using statistical analysis for genetic epidemiology-REGTL(SAGE-REGTL). The genetic model and gene frequency were estimated. 300 pedigrees with SLE were collected from 300 patients as probands. RESULTS: The complex segregation analysis found the genetic model of SLE to be additive,the gene frequency 0.336. The authors believe the possibility of some different modes coexisting under different circumstances. The genetic effect on young man is stronger than that on old one. CONCLUSION: This study suggests that the genetic mode of SLE could be the major gene trait, and additive mode is the best fitted one.

[Quantitative measurement on degradation degree of ecosystem in hilly region of western Liaoning Province].

Exactly measuring the degradation degree of ecosystems is the basis and precondition of restoring ecosystems. At present, degradation degree is usually analyzed qualitatively. In this paper, quantitative classification method was used to study the degradation degree of ecosystem in hilly region of western Liaoning, which was measured by the degradation degree of habitat. The result shows that the habitat in the northern slope of mountains has been degraded to certain stage between shrubbery and pioneer tree forest, which provides theoretical basis for vegetation restoration practice. Moreover, the result is beneficial for studying the degradation degree of ecosystems in other regions or of other types.

[Selection of optimal ultrasonic extraction process of Elaeagnus angustifolia L. by uniform design].

The influences of ultrasonic frequency, ultrasonic time and solvent content on the extraction rate of fat oil in Elaeagnus angustifolia L. were studied. The optimum extraction condition was obtained. The results showed that the ultrasonic extraction method of fat oil in Elaeagnus angustifolia L. could save time, improve extraction rate and need not be hot compared with traditional extraction method.

[The measurement of multifocal electroretinography].

PURPOSE: Discussing the testing procedure of multifocal electroretinography(ERG). METHODS: The testing procedure of 131 multifocal ERG records was reviewed. RESULTS: Some variable factors existed in the measurement of multifocal ERG. These factors included the subject's co-operation, the mounting of electrode, the fixation of tested eye and adjustment of focus. The good records of multifocal ERG depend upon the skill of the operator. CONCLUSION: The testing procedure of multifocal ERG must be standardized so as to ensure the reliability and repeatability of the results.

The electroretinogram in patients with retinal vascular occlusion.

PURPOSE: To measure and analyze the electroretinogram (ERG) in patients with retinal vascular occlusion. METHOD: Fifty-eight cases (59 eyes) of retinal vascular occlusion and the fellow eyes of 47 cases were tested with Ganzfeld ERG. The scotopic and photopic ERG, and oscillatory potentials were tested according to the ERG standard of ISCEV. RESULTS: The abnormal rates of OPs and b wave were higher in retinal vascular occlusion. The abnormal ERG appeared mostly in CRVO among four types of retinal vascular occlusion. The comparisons of some amplitudes and latencies between the fellow eyes and the affected eyes showed statistically significant difference. CONCLUSION: The oscillatory potentials and b waves have important roles in evaluating the retinal function of patients with retinal vascular occlusions. There are some clinical significance for comparing ERG between the affected eye and the fellow eye.

[The application of wavelet analysis of remote detection of pollution clouds].

The discrete wavelet transform (DWT) is used to analyse the spectra of pollution clouds in complicated environment and extract the small-features. The DWT is a time-frequency analysis technology, which detects the subtle small changes in the target spectrum. The results show that the DWT is a quite effective method to extract features of target-cloud and improve the reliability of monitoring alarm system.

Construction of evolutionary tree models for renal cell carcinoma from comparative genomic hybridization data.

Renal cell carcinoma is characterized by an accumulation of complex chromosomal alterations during tumor progression. Chromosome 3p deletions are known to occur early in the carcinogenesis, but the nature of subsequent events, their interrelationships, and their sequence is poorly understood, as one usually only obtains a single "view" of the dynamic process of tumor development in a particular cancer patient. To address this limitation, we used comparative genomic hybridization analysis in combination with a distance-based and a branching-tree method to search for tree models of the oncogenesis process of 116 conventional (clear cell) renal carcinomas. This provides a means to analyze and model cancer development processes based on a more dynamic model, including the presence of multiple pathways, as compared with the fixed linear model first proposed by Vogelstein et al. (N. Engl. J. Med., 319: 525-532, 1988) for colorectal cancer. The most common DNA losses involved 3p (61%), 4q (50%), 6q (40%), 9p (35%), 13q (37%), and Xq (21%). The most common gains were seen at chromosome 17p and 17q (20%). The tree model derived from the distance-based method is consistent with the established theory that -3p is an important early event in conventional (clear cell) renal cancer and supports the prediction made from the branching tree that -4q is another important early event. Both tree models suggest that there may be two groups of clear cell renal cancers: one characterized by -6q, +17q, and + 17p, and another by -9p, -13q, and -18q. Putative prognostic parameters were -9p and -13q. The distance-based tree clarifies that -8p (present in 12% of tumors) is a late event, largely independent of other events. In summary, tree modeling of comparative genomic hybridization data provided new information on the interrelationships of genetic changes in renal cancer and their possible order, as well as a clustering of these events. Using tree analysis, one can derive a more in-depth understanding of the renal cancer development process than is possible by simply focusing on the frequencies of genetic events in a given cancer type.
