Immunofluorescent study of the skin of nine patients with mesenchymal, inflammatory scleroderma (mixed connective tissue disease) revealed immunoglobulin and complement deposition at the basement membrane or within blood vessel walls. The skin specimens of ten patients with systemic scleroderma were negative for immunofluorescence. It is proposed that basement membrane or vascular (or both) immunofluorescence is an excellent means of identifying the infrequent patient who has scleroderma and myositis or lupus erythematosus in whom a corticosteroid response may occur.
The clinical and histopathological picture of a case of Juvenile Xanthogranuloma present in a new-born, in which all the investigations and analytic results were normal, is commented upon. The patient presents a case of exclusive skin affectation.
A male patient is presented is clinical and histopathological characteristics are typical of Ehlers-Danlos Syndrome (cutaneous and articular hyperelasticity and cutaneous fragility with the presence of atrophic scars). Looking into the family history we think that other relatives had the same disease and they had died during their childhood due to internal hemorrhages. The patient we present did not have significant hemorrhages in any moment. The elastic fibers in the dermal papilla are reduced giving rise to the appearance of being increased in the middle and deep dermis.
A case of Lichen Nitidus exclusively localized on the arms and palms of an 8 year old girl with ten years of evolution and a good response to treatment, is described.
A case of Multiple Trichoepithelioma with extensive skin involvement without familial history is reported. Some of the lesions showed spontaneous remission over the years, and in two, the microscopcal examination, demonstrated malignant changes (Basal cell Epithelioma).
A unique, superficial, lobulated, benign vascular tumor of the skin, found in the acral areas of adult males, is described. The combination of three different elements--arterial, venous, and transitional vascular channels--make up the predominant histologic features of this tumor. The transition of fibromuscular channels indicates a venous lesion, but the acral location of the lesions and the structure and staining characteristics suggest that the lesion is a hamartomatous proliferation of the Sucquet-Hoyer canal of the true glomus.
The pattern of distribution of T and B lymphocytes was investigated in a group of patients with contact, atopic, and mixed dermatitis by means of immunofluorescence and T-cell rosette tests. A significant increase (P less than .001) in B lymphocytes with surface IgD but without changes in the lymphocytes bearing other immunoglobulins or C3 and T cells was noted in patients with contact dermatitis when compared with a control group. A significant increase (P less than .001) in B lymphocytes carrying IgE and a significant decrease (P less than .001) in the total numbers of T cells were noted in patients with atopic dermatitis, and a significant increase in B lymphocytes positive for IgD (P less than .001) and IgE (P less than .003) with a decrease in the number of lymphocytes bearing IgG were the most important changes observed in patients with mixed contact and atopic dermatitis.
Determination in peripheral blood T and B lymphocytes performed in progressive systemic scleroderma (8 patients); dermatomyositis (5 patients); and inflammatory or mesenchymal scleroderma ('mixed' connective tissue disease) (5 patients) demonstrated a marked increase in peripheral B cells with T cell depression in patients with mesenchymal scleroderma. Patients with progressive systemic scleroderma and dermatomyositis demonstrated also a peripheral T depression, but no consistent changes in B cell population were found. The clear relationship between a B lymphocyte increase and elevated immunoglobulins, antinuclear and antiribonucleoprotein antibody and positive direct immunofluorescence of skin (basal membrane and blood vessels) defines inflammatory or mesenchymal scleroderma in which a disturbance in humoral immunity is involved. B lymphocyte determination can help to measure these changes in immunoreactivity.
Two pretibial epidermolysis bullosa families, with a total of four cases directly examined and another eight cases known from references, were studied. Both families showed an autosomal dominant inheritance. In one family there was a case of "bullous epidermolysis albo-papular of Pasini". All cases were compared with those of the literature.

Flow cytometry was used to investigate the participation of reactive oxygen species, other than singlet oxygen, in the cytotoxic effect of photodynamic therapy (PDT) with 5-aminolevulinic acid (ALA)-induced protoporphyrin IX (PpIX) in vitro in A-431 squamous cell carcinoma (SCC) cells and human skin fibroblasts (HSF). We used propidium iodide to determine cellular cytotoxicity, hydroethidine to measure intracellular superoxide anion (O2-) and dihydrorhodamine 123 to assess intracellular hydrogen peroxide (H2O2) content. Our data support the importance of the incubation time with ALA in the selectivity of PDT with ALA against SCC cells, inducing minimum damage on normal HSF. Photoradiation mortality curves of the response of these cell lines to ALA-induced PpIX photosensitization correlated with the extent of photosensitizer accumulation. Intracellular O2- production correlated with cell death, increasing both in a light dose-dependent fashion in ALA treated cells. This correlation was not observed with H2O2-intracellular production. These results suggest the effectiveness of PDT with ALA in vitro in SCC, the significant participation of O2- in its phototoxic mechanism, and the usefulness of flow cytometry in the study of the cytotoxic effect of ALA-induced PpIX PDT.
