Survival, mutagenesis, and host cell reactivation in a Chinese hamster ovary cell ERCC1 knock-out mutant.

Positive selection-negative selection gene targeting was used to disrupt the nucleotide excision repair gene ERCC1 in a Chinese hamster ovary cell line, CHO-K1. Southern and Northern analysis showed that a cell clone isolated by this targeting approach, CHO-7-27, had an ERCC1 gene structure consistent with targeted disruption of ERCC1 exon V, and did not express ERCC1 mRNA. CHO-7-27 was further characterized with respect to UV and mitomycin C sensitivities, and was shown to exhibit severe mutagen sensitivity phenotypes consistent with those of other CHO cell ERCC1 mutants. Mutation frequency experiments showed that CHO-7-27 was UV-hypermutable at the hypoxanthine-guanine phosphoribosyltransferase locus. Experiments assessing host cell reactivation of viral DNA synthesis for UV-irradiated adenovirus showed that CHO7-27 exhibited a severely deficient HCR phenotype similar to that of UV20 cells. Our results demonstrate that CHOK1 cells are hemizygous for the ERCC1 gene, and show that the comparatively mild mutagen sensitivities and lack of severely deficient HCR phenotypes of conventionally derived CHO-K1 ERCC1 mutants, in contrast to the severe phenotypes of CHO-AA8-derived mutants, are not due to any intrinsic genetic differences between CHO-K1 and CHO-AA8 parental cell lines.

[Expression of the bcl-2 and bax oncoprotein in TCC and its clinical significances].

To investigate the role of bcl-2 (an apoptosis suppressing oncogene) and bax (an apoptosis accelerating oncogene) in the development of TCC. We investigated bcl-2 and bax expression by means of immunohistochemical technique in 34 cases of TCC and in 9 cases of normal bladder tissue. Bcl-2 was positive in 44.44% of normal bladder tissue and in 82.35% of TCC. Bcl-2 staining intensity was significantly stronger in TCC than that in normal bladder tissue. Intensity and positivity of bcl-2 also increased with increasing grades of TCC. Bax was positive in 88.89% of normal bladder tissue and in 52.94% of TCC. Bax staining intensity was significantly weaker in TCC than that in the normal bladder tissue. These results suggested that increased expression of bcl-2 and decreased expression of bax in TCC play an important role in the development of TCC.

[Effects of nimodipine on calcium currents in isolated outer hair cells of guinea pig cochlea].

The effects of nimodipine on calcium currents in outer hair cells isolated from guinea pig cochlea were investigated using a whole-cell recording patch-clamp technique. External application of nimodipine (10 mumol/L) caused a decrease of voltage-dependent calcium currents and an elevation of current-voltage curve. The half-maximal inhibition concentration of nimodipine inhibiting the voltage-dependent calcium currents was 60.14 nmol/L, and the maximal inhibition was 34.16%. The present results demonstrate that nimodipine reversiblly and partially blocks the voltage-dependent calcium currents in isolated outer hair cells of guinea pig cochlea in a concentration-dependent manner. It is suggested that nimodipine may be used in the treatment of Meniere disease through decreasing the influx of Ca2+ into the outer hair cells.

[Experimental study on increasing arterial oxygen saturation by healthy exerciser under hypoxia].

The study was carried out among 41 healthy subjects born in Shanghai (Sea level). They were divided into two groups. Group one (10 subjects) was studied at sea level in a hypobaric chamber (3000m). Group two (31 subjects) was studied in high altitude plateau (Xigatse at 3836m). Arterial oxygen saturation (SaO2), Heart Rate (HR), blood pressure (BP) and myocardial oxygen uptake (MVO2) were measured noninvasively with Nelleor N-200 pulse Oxymeter and HEM-812F-Digital Automatic Blood Pressure Monitor. SaO2 increased significantly (from 87.7 +/- 1.7% to 95.3 +/- 1.0% P<0.01 and from 86.4 +/-1.0% to 91.8 +/- 0.6%, P < 0.05, respectively) by healthy exerciser through 15 min in both groups. After 3 min exercise SaO2 was still significantly higher than before exercise. Then HR, BP and MVO2 have not change significantly. The study results suggested that this kind of exerciser may help increasing the SaO2 and thus could be used to prevent and treat acute hypoxic response to some extent.

Effects of thrombolytic therapy on recanalization in different starting time of treatment after acute myocardial infarction.

OBJECTIVE: To investigate the effects of thrombolytic therapy on recanalization in different starting time of treatment after acute myocardial infarction. METHODS: 172 cases with intravenous thrombolytic agents were divided into 4 groups: < or = 2 hours, > 2-4 hours, > 4-6 hours and > 6-12 hours, according to the different starting time of therapy after onset of symptoms. RESULTS: The recanalization rates were 78.6%, 74%, 39. 6% and 14.3% in < or = 2 hours, > 2-4 hours, > 4-6 hours and > 6-12 hours, respectively. The recanalization rate within 4 hours was significantly higher than those in the groups of > 4-6 hours AND > 6-12 hours. The time interval from the initiation of thrombolytic therapy to reperfusion was increased as the starting time of thrombolysis after onset of symptoms was delayed. CONCLUSIONS: Starting time of thrombolysis within 4 hours after onset of acute myocardial infarction is of the best effect on recanalization.

Changes in the prevalence of benign prostatic hyperplasia in China.

OBJECTIVE: To understand the rapid increase of benign prostatic hyperplasia (BPH) in China by a comparative study of the nutrition status and BPH morbidity between urban and rural areas. METHODS: A study was conducted in 1993-1995 in Beijing on the influence of diet on the prevalence of BPH in 413 rural and 419 urban male inhabitants over 40 years of age. RESULTS: The results indicated distinctive differences in dietary patterns and the prevalence of BPH between rural and urban inhabitants. CONCLUSION: The changing prevalence of BPH in China can be attributed to the increased daily intake of total calories, fat and animal protein, and the decreased daily intake of vegetables and whole grain, the sources of phytoestrogens.

[Apoptosis in the early stage of compensatory renal growth following uninephrectomy in the young and old rats].

OBJECTIVE: To investigate the role of apoptosis, one of the major factor to maintain homeostasis of cell number of organs, in the early process of compensatory renal growth, which have been considered to including cellular hyperplasia and hypertrophy, following uninephrectomy. METHOD: DNA fragment gel electrophoresis, tissue sections photolebeling technique and in situ nick translation assay were used to analyze apoptotic cells in the compensatory and normal kidney in the young and old Wistar rats. RESULTS: Apoptosis affected normal kidney of the old group, but not the young. In the old rat's normal kidney, apoptosis index was higher in cortical tubules than that in the medullar tubules. In the compensatory remnant kidney, apoptosis was diminished in old rats, especially in the cortex, but increased in the young rats, especially in the medulla. CONCLUSIONS: Compensatory renal growth includes cellular hypertrophy, hyperplasia and apoptosis. In the old, compensatory renal growth is mainly cellular hypertrophy, and reduction of cell death. In the young, compensatory renal growth is mainly cellular hyperplasia and hypertrophy, increased apoptosis may be responsible for the elimination of excess hyperplastic renal cell.

[Effects of streptomycin on ionic currents in isolated outer hair cells of guinea pig cochlea].

The effects of streptomycin on ionic currents in outer hair cells isolated from the cochlea of guinea pig were investigated using a whole-cell recording patch-clamp technique. External application of streptomycin (1 mmol/L) caused a decrease of the potassium currents and calcium currents, respectively. The half-maximal inhibition concentration of streptomycin checking the voltage-dependent calcium currents was 56.08 mumol/L, and the ratio of maximal inhibition 21.71%. The present results demonstrate that streptomycin partially irreversibly blocks the voltage-dependent calcium currents in dissociated outer hair cells of guinea pig cochlea in a concentration-dependent manner. The effects of streptomycin of the potassium currents may be, at least in part, the result of the inhibition of Ca2+ influx. It suggests that the inhibition of Ga2+ influx may be one of the ototoxic mechanisms of streptomycin on outer hair cells.

A panel of VNTR markers in pigs.

By cloning tandemly repeated sequences from the pig genome by use of non-porcine minisatellite probes for library screening, five novel polymorphic VNTR loci were isolated: three minisatellites and two satellite-like loci. Four of them could be mapped onto chromosomes by linkage analysis and/or in situ hybridization. They were assigned to Chromosomes (Chrs) 5, 6, 14, and 16. Physical mapping on both presumed satellites and on one of the minisatellites revealed that the former resided near or at the centromere and the latter towards the chromosome ends. The location of the minisatellite is of particular interest since, together with data on three other minisatellites previously isolated, it supports the idea that, as in humans, minisatellites may preferentially be subtelomeric also in pigs.

An endosomal beta COP is involved in the pH-dependent formation of transport vesicles destined for late endosomes.

In this paper, we show that beta COP is present on endosomes and is required for the formation of vesicles which mediate transport from early to late endosomes. Both the association of beta COP to endosomal membranes as well as transport vesicle formation depend on the lumenal pH. We find that epsilon COP, but not gamma COP, is also associated to endosomes, and that this association is also lumenal pH dependent. Our data, thus, indicate that a subset of COPs is part of the mechanism regulating endosomal membrane transport, and that membrane association of these COPs is controlled by the acidic properties of early endosomes, presumably via a trans-membrane pH sensor.

Demonstration of a dynamic, transcription-dependent organization of pre-mRNA splicing factors in polytene nuclei.

We describe the dynamic organization of pre-mRNA splicing factors in the intact polytene nuclei of the dipteran Chironomus tentans. The snRNPs and an SR non-snRNP splicing factor are present in excess, mainly distributed throughout the interchromatin. Approximately 10% of the U2 snRNP and an SR non-snRNP splicing factor are associated with the chromosomes, highly enriched in active gene loci where they are bound to RNA. We demonstrate that the splicing factors are specifically recruited to a defined gene upon induction of transcription during physiological conditions. Concomitantly, the splicing factors leave gene loci in which transcription is turned off. We also demonstrated that upon general transcription inhibition, the splicing factors redistribute from active gene loci to the interchromatin. Our findings demonstrate the dynamic intranuclear organization of splicing factors and a tight linkage between transcription and the intranuclear organization of the splicing machinery.

[A prospective study on causal factors of female infertility with laparoscopy and transvaginal B-ultrasound].

OBJECTIVE: To study the casual factors of female infertility with laparoscopy and B-ultrasound. METHODS: Eighty-five infertile women were evaluated by serial transvaginal B-ultrasound scans for follicular development and ovulation and by laparoscopic examinations 2 to 6 days after ovulation. RESULTS: The patterns of follicular development and ovulation detected by B-ultrasound were: (1) ovulation from normal follicle (ONF) (56.47%); (2) ovulation from two follicles at the same time (O2F) (5.88%); (3) ovulation from small follicle (OSF) (16.47%); (4) ovulation from polycystic ovaries (PCO) (3.53%); (5) luteinized unruptured follicle syndrome (LUFS) (7.06%); (6) anovulation (10.90%). Laparoscopic examinations showed that the incidence of partial and complete tubal obstruction was 32.94% and pelvic adhesions 35.29%. Pelvic tuberculosis was found in 20.00%, which was the main disease leading to pelvic adhesions (46.67%), and tubal obstruction (60.71%). Endometriosis (EMS) was discovered in 36 cases (42.35%), among them, 30 (83.33%) had patent tubes. The stigmas or/and corpus hemorrhagica were seen in 60 out of 70 patients in whom ovulation had been diagnosed by B-ultrasound. 6 cases were defined as "mechanically unruptured follicle syndrome". CONCLUSIONS: Abnormal follicular development may result in female infertility. Pelvic tuberculosis is another main causal factor of infertility. Endometriosis related infertility may rather result from other factors than tubal obstruction. Laparoscopic examinations combined with transvaginal B-ultrasound scans are very useful approaches in the etiological study of female infertility.

[Sequencing of right terminal fragment of canine adenovirus type 1 vaccine strain DNA].

Current studies show that adenoviruses are more advantageous as expression vectors than other viruses. Canine adenovirus type 1 (CAV-1) vaccine strain, CLL (cannaught laboratory limited), as vectors will play more important role for the prophylaxis of zoonosis than human adenoviruses. Studies on molecular biology of CAV-1 or CLL are necessary for construction of vectors of CLL. In this paper, the right terminal of CLL DNA cleaved by Hind II was sequenced. Authors found that it contained a 238 bp inverted terminal repeats (ITR). There are three copies of 40 bp short repeat sequences in the ITR. According to the DNA structural model of human adenovirus type 2, the 40 bp short repeats are the sites that are combined with nuclear factor 1 (NF-1) that is an initiation factor of DNA replication of virus, so they are essential structure of CLL DNA.

[Histologic examination and immunohistochemical analysis of estrogen and progesterone receptors of eutopic and ectopic endometrium in patients with endometriosis].

OBJECTIVE: To study the differences of histologic dating and steroid receptors between ectopic and eutopic endometrium in patients with endometriosis. METHODS: Histologic examinations were done on eutopic and ectopic endometrium in 28 patients with pelvic endometriosis, and estrogen and progestrone receptors concentrations were determined by immunochemical ABC method in 15 patients. All the ectopic tissues were biopsied from peritoneal lesions. RESULTS: In 67.9% patients, the ectopic endometrium development was retardated and 14.3% accelerated as compared with eutopic endometrium, and only 17.9% identical. The estrogen receptors (ER) and progesterone receptors (PR) contents of ectopic endometrium were significantly lower than those of eutopic endometrium in glandular cells (P < 0.01, P < 0.05 respectively), while differences were not significant in stroma cells. CONCLUSIONS: The advantage of the immunochemical technique is that it can definitely locate the site of receptors in the histologic section. The difference in histologic dating and ER and PR concentrations in the eutopic and ectopic endometrium may be accounted for by the different steroid environments and pathogenic theory of peritoneum metaplasia.
