Technical considerations for assessing alterations in skeletal muscle sarcoplasmic reticulum Ca(++)-sequestration function in vitro.

A multiple measurement system for assessing sarcoplasmic reticulum (SR) Ca(++)-ATPase activity and Ca(++)-uptake was used to examine the effects of SR fractionation and quick freezing on rat white (WG) and red (RG) gastrocnemius muscle. In vitro measurements were performed on whole muscle homogenates (HOM) and crude microsomal fractions (CM) enriched in SR vesicles before and after quick freezing in liquid nitrogen. Isolation of the CM fraction resulted in protein yields of 0.96 +/- 0.1 and 0.99 +/- 0.1 mg/g in WG and RG, respectively. The percent Ca(++)-ATPase recovery for CM compared to HOM was 14.5% (WG) and 10.1% (RG). SR Ca(++)-activated Ca(++)-ATPase activity was not affected by quick freezing of HOM or CM, but basal ATPase was reduced (P < 0.05) in frozen HOM (5.12 +/- 0.18-3.98 +/- 0.20 mole/g tissue/min in WG and from 5.39 +/- 0.20-4.48 +/- 0.24 mumole/g tissue/min in RG). Ca(++)-uptake was measured at a range of physiological free [Ca++] using the Ca++ fluorescent dye Indo-1. Maximum Ca(++)-uptake rates when corrected for initial [Ca++]f were not altered in HOM or CM by quick freezing but uptake between 300 and 400nM free Ca++ was reduced (P < 0.05) in quick frozen HOM (1.30 +/- 0.1-0.66 +/- 0.1 mumole/g tissue/min in WG and 1.04 +/- 0.2-0.60 +/- 0.1 mumole/g tissue/min in RG). Linear correlations between Ca(++)-uptake and Ca(++)-ATPase activity measured in the presence of the Ca++ ionophore A23187 were r = +0.25, (P < 0.05) and r = +0.74 (P < 0.05) in HOM and CM preparations, respectively, and were not altered by freezing. The linear relationships between HOM and CM maximum Ca(++)-uptake (r = +0.44, P < 0.05) and between HOM and CM Ca(++)-ATPase activity (r = +0.34, P < 0.05) were also not altered by tissue freezing. These data suggest that alterations in maximal SR Ca(++)-uptake function and maximal Ca(++)-ATPase activity may be measured in both HOM and CM fractions following freezing and short term storage.

Coexistent cutaneous T-cell lymphoma and B-cell malignancy. French Study Group on Cutaneous Lymphomas.

BACKGROUND: The coexistence of cutaneous T-cell lymphoma (CTCL) and a B-cell malignancy (BCM) is rare. OBJECTIVE: Our aim was to assess the clinical and pathologic aspects of coexistent CTCL and BCM and to examine potential explanations for this association. METHODS: We report six cases of concurrent CTCL and BCM in which B- and T-cell lineages were demonstrated by immunologic studies. The literature includes 13 additional cases. All 19 CTCL-BCM cases are reviewed. RESULTS: CTCL either preceded or followed the BCM, which was a low-grade malignancy in most cases (16 of 19). Possible explanations for the association include a genetic predisposition, underlying viral infection, chemotherapy-induced carcinogenesis, stimulation of a B-cell clone by malignant helper T cells, and alterations in progenitor cells before determination of B- and T-cell lineage. CONCLUSION: An alteration in progenitor cells, with subsequent oncogenic activation of variable origin, might account for most cases of coexistent CTCL and BCM.

Preservation of sarcoplasmic reticulum Ca2(+)-sequestering function in homogenates of different type composition following sprint activity.

To examine the effect of exercise on sarcoplasmic reticulum function in muscle tissue of different fibre composition, adult male Wistar rats weighing 388 +/- 23 g (x +/- SE) ran intermittently on a treadmill until fatigue. Fatigue was induced by 15-20 min of running performed at 52 m/min on an 8 degrees incline in periods of 2.5 min of exercise separated by 2 min of recovery. Analysis of sarcoplasmic reticulum Ca2+ ATPase activity determined in homogenates indicated no difference (p > 0.05) between age-matched control and exercised tissue for the soleus (SOL; 0.121 +/- 0.012 vs. 0.156 +/- 0.018 mumol.mg-1 protein.min-1), red gastrocnemius (RG; 0.381 +/- 0.022 vs. 0.354 +/- 0.022), or white gastrocnemius (WG; 0.526 +/- 0.05 vs. 0.471 +/- 0.031). Similarly, both total ATPase and Mg2+ ATPase activities were unaffected by the exercise in any of the tissues examined. Exercise also failed to alter sarcoplasmic reticulum Ca2+ uptake in homogenates of the SOL (1.43 +/- 0.15 vs. 1.38 +/- 0.19 nmol.mg-1 protein.min-1), RG (3.74 +/- 0.29 vs. 3.59 +/- 0.24), and WG (5.98 +/- 0.48 vs. 5.41 +/- 0.50). At fatigue, glycogen depletion was similar in all tissue types and amounted to 65.1% in the SOL (172 +/- 9 vs. 60 +/- 16 mmol.glucosyl units-1.kg-1 dry weight), 74.4% in RG (164 +/- 8 vs. 42 +/- 6), and 79% in the WG (167 +/- 9 vs. 35 +/- 9). It is concluded that exercise by itself does not alter sarcoplasmic reticulum Ca2(+)-sequestering function in tissues of primarily different fibre composition when determined in homogenates in vitro.(ABSTRACT TRUNCATED AT 250 WORDS)

[Salazosulfapyridine-induced eruption. Pustular erythroderma and pseudo-lymphomatous syndrome: 2 cases].

We report two observations of drug eruption due to salazosulfapyridine which occurred 15 to 21 days after introducing the molecule. The two patients were of Carabian origin and presented with fever, erythrodermia which became pustular and a pseudolymphomatous syndrome. Favourable outcome was observed 4 to 6 weeks later. The diagnosis of toxidermia was made based on the strong imputability of salazosulfapyridine in both cases. Severe immunoallergic reactions to this molecule have been described, including fatal cytolytic hepatitis and Lyell syndrome.

[Cutaneous metastasis disclosing adrenal cortical carcinoma].

Metastases to the skin from internal malignant neoplasms are uncommon and often preterminal event. Cutaneous metastases from an adrenal cortical carcinoma have rarely been reported even in the advanced stages of the disease. A patient was initially seen with a small cutaneous lesion on the cheek and was found to have a large adrenal cortical tumor. Histologic comparison proved that the cutaneous lesion was a metastase from an adrenal cortical carcinoma.

[Extra-abdominal desmoid tumor. Diagnostic, prognostic and therapeutic aspects].

We report the case of a 19 year-old female patient who consulted for a recurrent desmoid tumor of the back which had initially developed during childhood. Its clinical aspect was reminiscent of a localized subcutaneous tumor, but surgical exploration revealed multiple ramified extensions spreading over a wide area deep into the underlying muscular tissue. In spite of two attempts at wide surgery, complete resection was not achieved. EAD tumors are a distinct category between fibromatosis and fibrosarcoma. There is a direct contrast between the benign histological structure and the lack of a metastatic potential, and the fact that they are highly aggressive locally with a great proportion of relapses occurring after resection. Prognosis, which is good in terms of survival, is determinant in the choice of treatment which must remain conservative in the majority of cases. Wide, non mutilating surgery offers the best chances of cure without sequelae. When surgery may be mutilating, 50-60 Gy of radiotherapy (RT) is a good alternative and affords a high rate of complete regression over a long period of time. The value of adjuvant RT after surgical resection has yet to be determined. Hormonotherapy and other drugs are currently being evaluated.

[Development of occupational asthma. Follow-up R.F.E. of 23 cases].

Occupational asthma can either improve or develop into chronic respiratory insufficiency. It is often difficult to predict which course will be followed. Here we report the follow-up of respiratory function of 23 patients with occupational asthma of at least one year's standing, due to various substances; several prognostic factors are discussed.

Heat-induced changes in respiratory pathways and mitochondrial structure during microcycle conidiation of Neurospora crassa.

Changes in both respiratory pathways and mitochondrial structure of Neurospora crassa occurred under conditions of microcycle conidiation. Upon heat-treatment at 46 degrees C, conidia developed a highly cyanide-insensitive, hydroxamate-sensitive respiration associated with morphological alterations in mitochondrial membranes; such changes were time-dependent. When heat-treated conidia were shifted down to 25 degrees C, the alternate, hydroxamate-sensitive respiration decreased significantly, paralleling the recovery of well-cristated mitochondria with an electron-dense matrix in the germ tubes. The decrease in hydroxamate-sensitivity was associated with two periods of increase in cyanide sensitivity corresponding to the events of germination and precocious proconidial budding.

Differential deoxyribonucleic acid synthesis during microcycle conidiation in Neurospora crassa.

In conidia of Neurospora crassa germinating at 25 degrees C, DNA synthesis measured by incorporation of tritiated adenosine reaches a maximum soon after the outgrowth of the germ tube (6--7 h after inoculation). In conidia heat-treated at 46 degrees C (for 15 h), a maximum of incorporation of the DNA precursor occurs already 1 h after inoculation, then the incorporation progressively declines until the end of the heat-shock. When such conidia are shifted to 25 degrees C, a maximum of DNA synthesis occurs during the development of the presumptive conidiophore as at the outgrowth of normal germ tubes. This wave of DNA synthesis is followed by a second maximum of DNA synthesis, occurring only in the microcyclized cultures, when the premature differentiation of proconidia takes place. Prevention of this second wave of DNA synthesis with hydroxyurea or 5-fluorodeoxyuridine respectively reduces or fully inhibits such induced conidial differentiation.

[Anticonidiogenic effect of hydroxyurea on Neurospora crassa].

Conidiation induced in a low sugar-ammonium medium (or by other means such as full starvation or heat shock) is almost fully prevented by hydroxyurea. In a parallel manner the DNA/RNA and DNA/protein ratios are prevented from increasing.

[Knowledge and prevention of tick-bite borreliosis: survey of the population in Alsace, an endemic area].

INTRODUCTION: European borreliosis, known to the general public as 'Lyme disease' in analogy with the American form, which it differs from in many points, is endemic in the area of Alsace and is a public health problem. The level of knowledge and prevention of the population with regard to the disease has never been assessed in France. MATERIAL AND METHODS: A survey was conducted in all the national health examination centers in Alsace using a self-applied questionnaire. The data collected concerned the socio-demographical characteristics, knowledge on the disease, history of tick bites, fear the disease creates, frequency of visits to the forest, prevention habits when visiting the forest and the attitude adopted in the case of a tick bite. RESULTS: Out of the 600 persons included, 99 (16.5 p.cent) had been bitten by ticks once or more over the past year. The existence of Lyme's disease was known to 74 p.cent of the consultants, 63 p.cent claimed they were worried by the disease and 43 p.cent knew that the first manifestation is redness spreading over the skin. Twenty-seven percent wore trousers and long sleeves when visiting the forest and 19 p.cent inspected their body carefully on their return. The persons least well informed were also those who did little to protect themselves against tick bites. They often were under 30 years old, lived in urban settings and had few diplomas. Those who had frequent spare time in forest and those who had a history of tick bites were the best informed and protected themselves better. The fear of the disease was associated with better knowledge and more appropriate behaviour. DISCUSSION: This study shows that a large percent of the population in Alsace is exposed to tick bites. Tick bite borreliosis is relatively well known, but protection remains insufficient. Better knowledge of the disease is related to better prevention. Information and teaching campaigns for the general public could specifically target the young people, urban dwellers and those with few diplomas. Specific messages of prevention could be delivered to the most exposed at-risk subjects (i.e. those bitten by ticks or having leisure in forest) at the places of their leisure or medical consultations.
