Purification and characterization of EpiD, a flavoprotein involved in the biosynthesis of the lantibiotic epidermin.

The plasmid-encoded epidermin biosynthesis gene, epiD, of Staphylococcus epidermidis Tü3298 was expressed in Escherichia coli by using both the malE fusion system and the T7 RNA polymerase-promoter system. EpiD was identified by Western blotting (immunoblotting) with anti-maltose-binding protein (MBP)-EpiD antiserum. EpiD and the MBP-EpiD fusion protein, which were mainly present in the soluble protein fraction, were purified from the respective E. coli clones. Purified EpiD showed the typical absorption spectrum of an oxidized flavoprotein with maxima at 274, 382, and 453 nm. The coenzyme released from EpiD by heat treatment was identified as flavin mononucleotide. S. epidermidis Tü3298/EMS11, containing a mutation within epiD, was unable to synthesize active epidermin. This mutated gene, epiD*, was cloned in E. coli and expressed as an MBP-EpiD* fusion protein. DNA sequencing of epiD* identified a point mutation that led to replacement of Gly-93 with Asp. Unlike MBP-EpiD, the fusion protein MBP-EpiD* could not bind flavin mononucleotide. We propose that EpiD catalyzes the removal of two reducing equivalents from the cysteine residue of the C-terminal meso-lanthionine to form a --C==C-- double bond and is therefore involved in formation of the unusual S-[(Z)-2-aminovinyl[-D-cysteine structure in epidermin.

Regulation of Staphylococcus xylosus xylose utilization genes at the molecular level.

We have investigated the regulation of the operon encoding xylose utilization in Staphylococcus xylosus C2a and Staphylococcus carnosus TM300. For in vivo studies, transcriptional fusions of the xylAB regulatory region to the lipase gene from Staphylococcus hyicus were constructed. Repression of lipase activity depended on a functional xylR gene and an xyl operator palindrome downstream of the promoter, while induction was obtained in the presence of xylose. Inactivation of either xylR or the xyl operator led to constitutive expression in the absence of xylose. Crude protein extracts from xylR+ staphylococci led to gel mobility shifts of the xyl regulatory DNA in the absence but not in the presence of xylose. A copper-phenanthroline footprint of the shifted band revealed protection of 28 phosphodiesters from cleavage in each strand of the xyl operator. Thus, the Xyl repressor covers the DNA over more than 2.5 helical turns. Glucose repression of the xyl operon occurs at the level of transcription and is independent of a functional xylR gene. A potential cis-active sequence element for glucose repression is discussed on the basis of sequence similarities to respective elements from bacilli.

Cloning and expression of various staphylococcal genes encoding urease in Staphylococcus carnosus.

The urease genes from Staphylococcus xylosus C2a, Staphylococcus aureus U500, and S. aureus Newman were cloned in Staphylococcus carnosus using the plasmid vectors pCA43 and pCA44. The resulting respective recombinant plasmids pUra 402, pUraUH66, and pUra17 contained chromosomal DNA fragments with sizes of 5.6, 5.8, and 6.8 kb, respectively. Investigations on urease expression of the donor and recombinant strains in media with various nitrogen sources revealed that S. xylosus C2a produced urease constitutively at the highest specific activity. All of the recombinant strains had significantly lower urease activities than their DNA-donor strains. The nickel-dependence of urease was demonstrated in S. aureus U500 by a plate diffusion assay.

[Management of drug-induced priapism].

The authors demonstrate the management of erectile impotence by using Papaverin or the combination of Papaverin-Regitin and its overdosing. The overdosing should be put in mind when using these drugs. The authors also demonstrate three types of priapisms. In according to the management of 72 cases priapism, the authors are advising the irrigation of the corpus cavernosum with Dopamin after punction.

Organization, promoter analysis and transcriptional regulation of the Staphylococcus xylosus xylose utilization operon.

The Staphylococcus xylosus xyl genes were cloned in Staphylococcus carnosus by complementation to xylose utilization. Xylose isomerase assays under inducing (xylose present) and non-inducing (xylose absent) conditions indicated the presence of a regulated xylA gene on the recombinant plasmid. The nucleotide sequence (4520 bases) revealed three open reading frames with the same polarity. They were identified by sequence homologies as xylR, encoding the Xyl repressor, xylA, encoding xylose isomerase and xylB, encoding xylulokinase. Primer extension analyses indicated constitutive transcription of xylR and xylose-inducible transcription of xylA. Promoter consensus sequences were found upstream of both transcriptional start sites. A transcriptional terminator between xylR and xylA separates the different transcriptional units. Potential regulatory elements were identified by sequence analysis and suggest a repressor-operator mechanism for the regulation of xylAB expression.

Short- and long-term effects of a dopamine agonist (lisuride) on sex-specific behavioural patterns in rats.

Lisuride induces in juvenile male and especially in female rats a significant increase of the male-typical initiating activity in tests on social play-fighting behaviour. In neonatally castrated as well as in prenatally stress exposed males, castrated in adulthood, which exhibit under androgen substitution alone bisexual or even predominantly heterotypical sexual behaviour, additional treatment with lisuride resulted in a temporary normalization of sexual orientation in the homotypical male direction, limited to the duration of treatment. In castrated androgen-treated females, lisuride induces a partial conversion of sexual orientation to the heterotypical male direction. When intact newborn females or neonatally castrated males were treated with lisuride during the early postnatal differentiation period of the brain (day 2-12) or, on the other hand, during the peripuberal maturation period (day 26-40), it was found that early postnatal as well as peripuberal activation of the dopaminergic system in females resulted in permanent partial masculinization of sexual orientation. Comparable trends were observed after peripuberal androgen administration in females. In neonatally castrated males, the demasculinized play-fighting as well as the sexual orientation could be normalized--at least in part--by early postnatal or peripuberal lisuride administration. These findings confirm once more our previous reports that neurotransmitters can act directly as organizers of the brain. This holds true not only for the differentiation period but also for the maturation period of the brain.

The influence of fetal adrenals on the androgen levels during brain differentiation in human subjects and rats.

Measurements of plasma total and free testosterone (T) levels in human subjects from fetal to postpubertal life showed about twofold higher total T and 15-fold higher free T levels in female fetuses than in female adults. The ratios between the sexes were only moderate in fetal life. Between the 17th and 31st week of pregnancy the ratios (male:female) of total T were found to be 6.6 in week 17, 1.5 in week 22, 2.3 in week 28 and 1.2 in week 31 of pregnancy compared to 16.2 in adulthood. The corresponding ratios of free T were calculated to be 5.6 in week 17, 1.4 in week 22, 0.9 in week 28 and 0.7 in week 31 of pregnancy compared to 34 in adulthood. In amniotic fluids, we measured even an overlapping of T values between the two sexes. The reason for the observed striking difference of T levels between the sexes in fetal and postpubertal life may be the high adrenal activity and secretion rate in fetal life during brain differentiation. In rats, the contribution of adrenals to plasma T levels is only moderate and much smaller than in human beings. As measured in adult female rats, the portion was found to be about 20% only, contrary to about 60% in women. The main sources of T in female rats appear to be the gonads. The mainly gonadal secretion may be the reason that exposure of pregnant rats to stress diminished the T levels in male fetuses, but did not significantly elevate the T levels in females.(ABSTRACT TRUNCATED AT 250 WORDS)

Laparoscopic appendectomy in 625 cases: from innovation to routine.

Invented in 1982 by Semm, a gynecologist, laparoscopic appendectomy began to be used in operative surgery in 1987. Since then, we have modified and performed this promising procedure in > 700 patients. The data of the first 625 cases indicate that laparoscopic appendectomy is safe and practicable. With regard to patient age as well as the acuteness of the inflammation, the patients represent a cross section of the normal appendicitis population. The major benefits and possible problems of the method are presented and discussed.

[Laser-assisted laparoscopic appendectomy].

Our constantly good experience with approximately 300 laparoscopic appendectomies (almost 520 at present (Feb. 90)) encouraged us in 1988 to introduce Laser technology into laparoscopic abdominal surgery. 22 patients underwent Laser-aided laparoscopic appendectomy. We obtained good results by means of using both an CO2-Laser and a Nd-YAG-Laser.

[Laparoscopic placement of T-drainage in choledocholithiasis].

This case report illustrates the feasibility of bile duct revision and T-tube insertion by laparoscopic approach. The procedure largely employs the traditional action. Laparoscopy demands more skill, particularly regarding the suture of the common bile duct, but does not interfere its realisation.

Transcription inhibition of SV40 by in vitro DNA methylation.

SV40 DNA was methylated in vitro with prokaryotic or eukaryotic DNA cytosine-5-methyltransferases and the inhibition of transcription by methylation was studied in Xenopus oocytes. Methylation with the prokaryotic HhaI or HpaII methyltransferases did essentially not inhibit transcription of SV40. Methylation with a rat liver methyltransferase led only to minor inhibition of the SV40 early genes, but to a complete shut shut off of the SV40 late genes. Partial methylation showed that methylation of both, the regulatory region and the 5' end of the SV40 late genes, was necessary for the effect on transcription. The TK gene could be inactivated by eukaryotic methylation of either the promoter and the first 50 nucleotides of the gene or the 3' rest of the gene. Insertion of the SV40 enhancer, not containing methylatable CpGs, into the TK upstream region, had no influence on the inhibition of TK gene transcription by methylation.

Cloning, expression and characterization of the Sau3AI restriction and modification genes in Staphylococcus carnosus TM300.

The genes encoding the restriction enzyme (ENase) and modification enzyme (MTase) of Staphylococcus aureus 3A (recognition sequence 5'-GATC-3') have been cloned in Staphylococcus carnosus TM300 using the vector pCA44. Clones carrying both genes were isolated from DNA libraries prepared with MboI + BamHI. The DNA region encoding M.Sau3AI was subcloned on a 3.66-kb EcoRV fragment in vector pT181mcs. Plasmids purified from the clones were resistant to digestion with Sau3AI, indicating that the sau3AIM gene was expressed and the product was functional in S. carnosus. Cell lysates of clones with both activities encoded on plasmid pSEM7, cut DNA with the same pattern as Sau3AI, showing that the sau3AIR gene was also expressed and the ENase was functional in S. carnosus. Sequence analysis shows that both genes are transcribed in the same direction and encode polypeptides with calculated Mrs of 56,477 for R.Sau3AI and 47,300 for M.Sau3AI. Efforts to clone one or both genes in Escherichia coli have so far failed.

DNA methylation inhibits transcription by RNA polymerase III of a tRNA gene, but not of a 5S rRNA gene.

Methylation of cytosine in the DNA inhibits the transcription by RNA polymerase II in higher eukaryotes, but has no influence on RNA polymerase I transcription. The effect on RNA polymerase III was unknown, so far. Two polymerase III genes: a type 1 5S rRNA gene and a type 2 tRNA gene were methylated in vitro with a purified eukaryotic DNA methyltransferase (EC2.1.1.37) and their transcription was analyzed in Xenopus oocytes. The 5S rRNA gene, an oocyte 5S rRNA gene from X. laevis which is subject to developmental inactivation, was not affected by methylation. Conversely, transcription of the tRNA gene was 80% inhibited by methylation with the eukaryotic methyltransferase. HhaI and HpaII methylation left its transcription unaffected.

Escherichia coli 30S mutants lacking protein S20 are defective in translation initiation.

The 30S ribosomal subunits derived from Escherichia coli TA114, a a temperature-sensitive mutant lacking ribosomal protein S20, were shown to be defective in two ways: (a) they have a reduced capacity for association with the 50S ribosomal subunit which results in the impairment of most of the functions requiring a coordinated interaction between the two subunits; (b) they are defective in functions which do not require their interaction with the large subunit (i.e., the formation of ternary complexes with aminocyl-tRNAs and templates, including the formation of 30S initiation complexes with fMet-tRNA and mRNA). The 30S (-S20) subunits seem to interact normally with both template and aminoacyl-tRNA individually, but appear to be impaired in the rate-limiting isomerization step leading to the formation of a codon-anticodon interaction in the P site.
