Crossreactivity between Qa-1 region and H-2K antigens.

Antiserum (A.TL X A.SW)F1 anti-A.TH is specific for the products of the Qa-1a region. It reacts only weakly or, under certain circumstances, not at all with the immunizing cells, but it reacts strongly with 100% of H-2f and H-2p lymphocytes. The molecules that are the target of this strong heteroclitic reactivity are controlled by a gene in the K region, most likely the K locus itself. The results are interpreted in terms of crossreactivity of Qa-1-specific antibodies with Class I major histocompatibility complex molecules. This crossreactivity underscores the relatedness of Qa and Class I loci and strengthens the argument that Qa loci are members of the Class I family.

H-2 haplotypes carrying identical D but different L alleles.

The B10.SAA48 congenic line was derived by transferring the H-2 haplotype of a wild mouse onto the background of the inbred strain C57BL/10Sn (abbreviated as B10). The line carries the Dw3 allele in combination with an L allele different from that present in other Dw3 strains. One possible explanation of this finding is that crossing over occurred between the D and L loci. The determinants H-2.m64 and H-2.m65 represent an inclusion doublet in which the latter never occurs without the former. Typing of B10.W lines with Ld-specific CTLs reveals the absence of this allele in all the lines, including those carrying an allele serologically similar to Ld.

Polymorphism of Qa and Tla loci of the mouse.

Congenic lines carrying H-2 haplotypes derived from wild mice were typed serologically with polyclonal and monoclonal antibodies specific for Tla, Qa-1, and Qa-2 antigens. The typing revealed the presence of a minimum of five Tla, four Qa-1, and three Qa-2 alleles in the 32 lines. Two new Tla, two new Qa-1, and one new Qa-2 alleles could be described. This polymorphism of Tla and Qa loci is lower than that detected at the K and D loci in the same lines. The serological typing for Qa-2 antigens correlates remarkably well with previously published results of typing with cytolytic T lymphocytes. This correlation supports the identify of loci detected by the two methods.

The minimal length of the differential segment in H-2 congenic lines.

One hundred and four H-2 congenic lines were typed for alleles at seven loci, Qa-1, Qa-2, Tla, C3, Ce-2, Pgk-2, and Upg-1, residing distal to the H-2 complex. The results of the typing were used to estimate the length of the segment of chromosome 17 derived from the donor strain of each line--that is, the minimal length of the differential segment. The results indicate that only lines derived by intra-H-2 crossing-over in such a way that they inherited the right-hand portion of H-2 from the inbred partner have the telomeric half of chromosome 17 identical with that of the inbred-partner strain. In other lines the differential segment is at least 3 to 10 cM long. It is argued that in some lines the entire telomeric half of chromosome 17 might be of donor-strain origin.

Evidence for placing the Neu-1 locus within the mouse H-2 complex.

Congenic lines B10.KPA42, B10.KPA132, B10.SNA57, B10.DRB62, and B10.WOA105 carry H-2 haplotypes of wild mice on the genetic background of the strain C57BL/10Sn. Two of the lines (B10.DRB62 and B10.WOA105) have H-2 haplotypes indistinguishable from H-2v of B10.SM. The H-2 haplotype of one line (B10.SNA57) seems to have arisen from H-2v by recombination between the D and Qa-2 loci. The H-2 haplotypes of the remaining two lines probably arose from H-2v by recombination between the C4 and D loci. Since all six and no other lines carry the rare Neu-1a allele, the neuraminidase-1 locus is probably located proximal to the H-2D locus. Typing of H-2s recombinants for the enzyme acid phosphatase liver, the processing of which is controlled by the Neu-1 locus, suggests that the locus resides between the E alpha and D loci, that is in the S region.

H-2 antigen frequencies among wild mice from Chile.

Thirty-five wild mice (Mus musculus L.) were captured at four different localities in Chile. The mice were typed for the presence of 15 K-, 11 D-, 14 A-, and two E-region H-2 antigens, using the complement-antibody microcytotoxicity assay. The mice from the sample representing the largest locality had a characteristic antigenic profile distinguishing them from other mouse populations thus far studied. With respect to class I H-2 antigens, the profile was characterized by the presence of antigens H-2K.16, 31, 103, 19, 108, 21, 33, 26, 115, and H-2D.4, 106, 30, 32, 111, 114, and 18 (in order of decreasing antigen frequency), and by the absence of antigens H-2K.15, 17, 20, 113, 116 and H-2D.2, 9, 110, and 112. The profile was, furthermore, characterized by the relatively high frequency of antigens H-2K.16, 31, and H-2D.4 and 106. The profile of class II antigens was also unique to the Chilean population but less conspicuously so than that of the class I antigens. Analysis of antigenic associations suggested that among the 34% blank H-2K alleles there were at least two coding for relatively frequent but as yet unidentified K antigens. Similarly, among the 57% blanks at the H-2D locus there were at least two frequent alleles encoding unidentified D antigens. Analysis of genetic distances suggested similarity between South American mice and mice from coastal regions of Europe.
