Partial purification and characterization of a yeast extracellular acid protease.

During screening of 143 yeasts for proteolytic milk coagulating activity, a strain belonging to the species Cryptococcus albidus var. aerius was found which produced extracellular protease in shake culture. An enzyme preparation was obtained from the cell-free broth by ammonium sulphate precipitation. It possessed an optimum pH for milk-clotting at 5.5 to 5.7 at 35 degrees C. Maximum stability occurred between pH 3.5 and 5.5. The optimum temperature for the enzyme was 45 degrees C. Activity of the enzyme was inhibited by copper+2, iron+2, and mercury+2 ions.

[Tissue expression of carbohydrate antigen 19-9 (CA 19-9) in adenomatous lesions of the colorectum].

Serum levels of tumor markers almost not detectable in precancerous states or early cancer, the behavior of the tumor associated antigen CA 19-9 in colorectal carcinogenesis was analysed. In order to investigate the adenoma-carcinoma sequence, tissue antigenic expression was measured in adenomas, characterized by different size and histology, and compared with that found in normal colonic mucosa. Tissue content of CA 19-9 was determined by an immunoperoxidase technique (ABC-POD) in 88 colonic polyps, and correlated to the degree of histological dysplasia, and dimension of adenoma. Tissue content of CA 19-9 was also evaluated in non-adenomatous mucosa obtained by endoscopic biopsy. Among the 88 polyps, 50 showed the tubular histological type, while 31 resulted tubulovillous and 7 villous. High degree of dysplasia was present in 7 adenomas (7.9%) and focal carcinoma was observed in 6 (6.8%). Positivity for CA 19-9 was registered in 60.2% of adenomas. No correlation was found between tissue-CA 19-9 and degree of dysplasia, size of adenoma and villous component. However, a statistically significant correlation was observed between expression and cellular distribution of the antigen (chi 2 = 98.07, p < 0.00001). Our data confirmed CA 19-9 expression in adenomas, but it is unlikely this tissue antigen proves to be a reliable marker of adenoma-carcinoma sequence.

[Facial hemispasm and anomalies of the terminal portion of the vertebral artery (author's transl)].

A revision is suggested of the concept of "essential hemifacial spasm": 23 cases have been studied in whom the facial spasm was the only symptom present, while in 8 other cases in addition to the spasm of facial muscles other symptoms were noticed suggesting a pathology of the ponto-cerebellar angle. In many cases the symptom "spasm" and other concomitant symptoms are a direct consequence of an irritation due to compression on the nerve root (and/or on other nervous structures) owing to stretch of the distal tract of the vertebral artery or of the initial segment of the basilar artery. In five cases the AA. studied the vertebro-basilar circulation and in four patients they have actually found pathological elongations of the great vessels. In 18 other cases it was only possible to assume the presence of analogous alterations. The AA. suggest a study of the vertebro-basilar circulation in all the patients with "essential hemifacial spasm".

Intravenous administration of lisuride in the treatment of neuroleptic malignant syndrome.

The neuroleptic malignant syndrome (NMS) is a very rare but life-threatening complication of neuroleptic treatment. The mortality of NMS has been estimated at 8-30% and the most common cause of death is respiratory failure. Signs and symptoms of NMS are attributed to impairment of dopaminergic neurotransmission in the central nervous system. We describe two cases of NMS successfully treated with intravenous lisuride in combination with oral L-Dopa.
