Experimental pathogenicity of a presumed monoxenous trypanosomatid isolated from humans in a murine model.

Two strains of a presumed lower trypanosomatid isolated from immunocompetent and HIV-infected humans in French West Indies were investigated in vitro and in vivo in a murine experimental model. The ability of parasites to grow in vitro in bone marrow-derived macrophages and their virulence in vivo were assessed. For in vivo infection, two groups of BALB/c mice were inoculated either by the subcutaneous or intravenous route with 10(7) promastigotes at day 0. Infection was monitored by measuring parasite load in liver, spleen, foot pad, popliteal, and mesenteric lymph nodes and brain from day 7 to day 150 post-infection using a microtitration technique. Parasites multiplied in mouse macrophages in vitro. In vivo, both strains proved infective to mice and capable of visceralization and dissemination in the popliteal and mesenteric lymph nodes, liver, spleen, and even brain. Both strains elicited a strong humoral response against trypanosomatid antigen in mice, which cross-reacted with Leishmania antigen. Contrasting with the straightforward dissemination of parasites, the infection was strikingly well tolerated by the murine host with no clinical signs and minimal tissue changes around parasitized macrophage infiltrates.

[Experimental Schistosomiases. I. Study of S. mansoni fecondity as regards to its adaptation to different Biomphalaria glabrata strains (author's transl)].

We compared the infestation of different strains of B. glabrata from Brasil (Recife). Guadeloupe, Martinique and Porto-Rico with 6 to 8 miracidia of S. mansoni (from Recife. We noted the four following points: 1. The planorbid snails from Martinique and Guadeloupe had a low resistance to infestation. 2. The guadeloupean snails showed the lesser rate of positivity and the lower medium amount of emitted cercaries but, in the four strains of snails, the level of the issued cercaries is quite the same. 3. many planorbid snails in the groups studied during more than 2 months, showed significant periodic variations in the emission of cercaries, We thought that those variations might be caused by the alternate maturations of sporocysts born from the same miracidium or from different miracidia. 4. Infestation by S. mansoni had a strong effect on fecondity of the snails but the laid eggs had a normal development.

[Ecology of a focus of cutaneous leishmaniasis in the Thies region (Senegal, West Africa). 2. Epidemiological and clinical peculiarities of the human disease].

During their epidemiological survey of a cutaneous leishmaniasis focus, the authors study the epidemiological and clinical aspects of 60 proven human cases observed in the studied area (Thies region). The disease can occur at any age but is more frequently seen in the pre-adolescent (10-15 years), adolescent (15-20 years) and adult (20-40 years) age classes. During the year, 76.5% of the lesions appear between july and the end of november. The spontaneous evolution of the disease seems to have a one year duration. Less than 10 lesions are observed in the majority of the cases (91.5%). The lesions are mainly located on upper and/or lower limbs and are generally of humid ulcerated type, often covered by a deep scar; a lymphatic nodular dissemination from a lesion occurs in 25% of the cases.

[Ecology of a focus of cutaneous leishmaniasis in the area of Thies, Senegal, West Africa. 4. Spontaneous infestation and biology of Phlebotomus duboscqi Neveu-Lemaire 1906].

Following their epidemiological investigation on the cutaneous leishmaniasis focus of Keur Moussa, the authors study the sandfly fauna. The species Phlebotomus duboscqi is abundant (1532 samples out of 8411 sandflies collected between December 1976 and June 1979) and located inside the rodent burrows which have been found to be their larval breeding sites. Five P. duboscqi females out of 356 dissected were infected by Leishmania major promastigotes.

[Ecology of a cutaneous leishmaniasis focus in the Thies region (Senegal, West Africa). 5. Spontaneous infection and disease reservoir role of various wild rodent species].

The authors describe spontaneous infection of Mastomys erythroleucus and Tatera gambiana by Leishmania strains typed as L. major. They argue the reservoir role to these two species and the one of Arvicanthis niloticus previously found infected. The possible relations between the fluctuations of rodent populations and the incidence of the human disease are then discussed.

[False reactions in hydatic serology in order to improve its value (author's transl)].

We have studied false reactions in hydatic serology in order to improve its value. We have found false positive reactions in two cases of tumor (hepatic sclerocholangiomatosis and pleural mesothelioma) and among children showing cor's milk protein intolerance when serum is tested with an antigen extracted from ox's lung. As for false negative reactions, they are scarcely due to circulating antigens. We have found, for 34 falsely negative serums, IgG, IgM and above all IgE levels lower than the levels of patients showing a positive serology.

[Experimental schistosomiasis. II. Infection of Biophalaria glabrata by 2 miracidia of Schistosoma mansoni].

The authors have infected 127 B. glabrata by 2 miracidia of S. mansoni, either on the Same day, or introducing a second miracidium after 3, 7 and 16 days. 1 All the groups of planorbid snails had a low percentage of positivity (31 to 41 %). 2 The first cercarial emissions, in the 4 groups were scattered in the time, the delay being in correlation with the second infection. 3 In the snails reinfected after 3 and 7 days occured the highest emissions, those with the simultaneous double infection or second infection delayed to 16 days, had the lesser emissions. 4 An interval cycle of about 3 weeks was discovered for the highest emissions. All those phenomena are probably due to competition between the sporocysts born from both miracidia. Moreover, evident reduction of the fecundity in the positive snails were shown only after the beginning of the cercarial emissions, while a normal, or even increased fecundity was established in the prepatent period of the infected snails.
