Interferon production in male homosexuals with the acquired immune deficiency syndrome (AIDS) or generalized lymphadenopathy.

We measured serum interferon (IFN) levels and leukocyte IFN production in vitro in homosexual men with AIDS or generalized lymphadenopathy and in asymptomatic homosexual or heterosexual men. Acid-labile interferon alpha was detected in the serum of two patients with AIDS and in three out of nine patients with lymphadenopathy, but not in the sera of healthy homosexual or heterosexual controls. The presence of serum interferon in patients with AIDS or lymphadenopathy was correlated with a marked deficiency of their leukocytes to synthesize interferon alpha in response to viruses or tumor cells, but did not significantly interfere with the production of interferon gamma in response to lectin or bacterial enterotoxin. Our results suggest an inverse correlation between serum interferon alpha and the amount of interferon alpha produced in leukocyte cultures in male homosexuals with AIDS or generalized lymphadenopathy.

Four-year experience with a recombinant hepatitis B vaccine.

Three hundred and forty-three healthy adults were vaccinated with five different lots of recombinant hepatitis B vaccine. Three hundred and forty (99.1%) individuals produced antibodies against hepatitis B surface antigen (anti-HBs). Peak anti-HBs concentrations were significantly higher in females and younger individuals. All anti-HBs positive individuals developed antibodies to the common determinant "a" of HBsAg. The vaccine was well tolerated, without severe side reactions. Persistence of anti-HBs was followed in 130 individuals for 3, and in 15 for 4 years after the first vaccination of these two groups. 21.7% and 32.3%, respectively, no longer had protective levels of anti-HBs after this time. The persistence of anti-HBs was dependent on peak anti-HBs levels, with consistent kinetics of anti-HBs decline. Revaccination of individuals whose specific antibody levels had fallen below 10 IU/l led to a prompt anti-HBs response. Comparison with individuals vaccinated with plasma-derived hepatitis B vaccine revealed no substantial differences between the two vaccines.

HIV antigen, HIV antibody and serum interferon in a patient with encephalopathy.

Acute encephalopathy was associated with the presence of human immunodeficiency virus type I (HIV I) antigen and seroconversion to anti-HIV I in a 27-year-old homosexual man. Examination of consecutive sera from the patient revealed circulating interferon (IFN) alpha which became detectable with the appearance of HIV I antigen but before development of anti-HIV I. Serum IFN was present for only a limited time and was not demonstrable after neurological symptoms resolved. It may be speculated that circulating antiviral activity contributed to the clinical manifestations of acute HIV I infection.

Hepatitis in marmosets.

Some species of marmosets are susceptible, not only by parenteral inoculation but also by oral exposure, to human hepatitis A virus present in sera or feces. The stools of animals inoculated parenterally or orally contained fecal antigen during certain times of the incubation period and the early, acute phase of the disease; viruslike particles were present in feces of orally infected animals and such feces were infectious when inoculated into marmosets. The fecal antigen crossreacted both with the fecal virus particles and the immune-adherence antigen (see also papers by Purcell et al and Hilleman et al). The MS-1 and CR-326 strains of hepatitis A appeared antigenically similar or identical whereas the GB strain was antigenically different and may be associated with the recently defined type of hepatitis termed hepatitis C or hepatitis non-A/non-B. On repeated challenge hyperegic responses with diffuse liver cell necrosis occurred in some immune animals and this phenomenon must be taken into account in any future vaccination studies.

[Lymphocyte heterogeneity of the T system: characterization of an allotypic antiserum recognizing a sub-species antigenic determinant in non-human primates].

To determine the membrane properties in T-lymphocyte population from Saguinus primates or lymphoblastoid cell lines of the same origin, an allotypic antiserum was prepared inoculating two Saguinus illigeri with 3.5 x 10(10) viable 70N2 cells obtained in-vitro, from a lymphoma induced by inoculation of Herpesvirus Saimiri in a Saguinus oedipus. The 70N2 cells carried the virus genome in an episomal way; it did not produce infectious virus and it formed spontaneous rosettes (99%) with sheep erythrocytes. The resulting allotypic antiserum inhibited the formation of E rosettes and specifically reacted with lymphoblastic T-cell lines and peripheral lymphocytes of Saguinus oedipus while it did not cross react with these cells or peripheral lymphocytes of S. illigeri or nigrifons, S. jacchus, Saimiri sciureus , Papio Sp. and human primates. These results show that two antigenic populations exist within Saguinus T-lymphocytes; one is common to all members of the species and the other is specific for subspecies related to the spontaneous rosette receptors and it is present in lymphoblastic cell lines-T of the same origin.

[A new variant of the simian T-lymphotropic retrovirus type I (STLV-IF) in the Sukhumi colony of hamadryas baboons].

Polymerase chain reaction (PCR) was developed for the detection of simian T-lymphotropic virus type 1 (STLV-1) infection of P. hamadryas and direct sequencing using oligo-nucleotide primer pairs specific for the tax and env regions of the related human T-lymphotropic virus type 1 (HTLV-1). Excellent specificity was shown in the detection of STLV-1 provirus in infected baboons by PCR using HTLV-1-derived primers. The nucleotide sequences of env 467bp and tax 159bp of the proviral genome (env position 5700-6137, tax position 7373-7498 HTLV-1, according to Seiki et al., 1983) derived from STLV-1-infected P. hamadryas were analysed using PCR and direct sequencing techniques. Two STLV-1 isolates from different sources (Sukhumi main-SuTLV-1 and forest stocks-STLV-1F) were compared. Two variants of STLV-1 among P. hamadryas with different level of homology to HTLV-1 were wound (83.8% and 95.2%, respectively). A possible role of nucleotide changes in env and tax sequenced fragments and oncogenicity of STLV-1 variants is discussed.
