14-3-3 proteins bind to histone and affect both histone phosphorylation and dephosphorylation.

14-3-3 proteins appear to play a critical role in Ca(2+)-stimulated secretion in permeabilized chromaffin cells. 14-3-3 proteins have been reported to be both stimulators and inhibitors of protein kinase C (PKC). We have found that 14-3-3 proteins, isolated on the basis of their ability to enhance secretory activity, stimulated histone phosphorylation by PKC, but they had no effect on myosin light chain phosphorylation by PKC. 14-3-3 proteins were also found to inhibit the rate of [32P]histone dephosphorylation but not the rate of [32P]myosin light chain dephosphorylation. Cross-linking experiments and affinity chromatography demonstrated that 14-3-3 proteins bind to histones. These results suggest that at least some of the reported effects of 14-3-3 proteins on PKC activity may result from 14-3-3 proteins binding to histone.

Differences in stage at presentation of breast and gynecologic cancers among whites, blacks, and Hispanics.

BACKGROUND: One of the possible assumptions for the higher mortality from cancer of blacks versus whites is that blacks tend to be diagnosed relatively more often with later stage disease. This study examined the stages at diagnosis for female breast cancer and other gynecologic cancers among blacks, non-Hispanic whites, and Hispanic whites. METHODS: Data for Dade County were analyzed in this study, which was based on the tumor registry data from the Florida Cancer Data System from 1981-1989. The stages at diagnosis were classified as in situ, local, regional, and distant according to the International Classification of Diseases for Oncology coding rules. RESULTS: The percentage of early stage diagnoses (in situ and local stage) was significantly lower among blacks and Hispanic whites than among non-Hispanic whites, but only for breast and cervical cancer. There was a significant increase in the percentage of early stage diagnoses of breast cancer among all three groups during 1981-1989, whereas this increase was found only in Hispanic whites for cervical cancer. The percentage of in situ cervical cancers was much lower in women older than age 45, especially among women older than age 65, in all three groups. CONCLUSIONS: These data suggest that screening programs need to be targeted differently for specific age and ethnic groups.

Effect of beta-lymphocyte- and NPC-derived EBV-LMP1 gene expression on in vitro growth and differentiation of human epithelial cells.

The effect of expression of the Epstein-Barr-virus (EBV) latent membrane protein (LMP1) derived from B-lymphocytes (B) and nasopharyngeal carcinoma (NPC) (C) on the in vitro growth and differentiation of a human keratinocyte line, Rhek-1, was analyzed in clonal growth and in in vitro differentiation assays. In contrast to the polygonal parental cells, the B-LMP1-expressing sublines were spindle-shaped while the C-LMP1-expressing cells were pleomorphic. Both B- and C-LMP1-expressing sublines showed increased proliferation as evidenced by: (1) higher colony-forming efficiency (CFE) and larger colony size at reduced serum levels; (2) an increased number of epithelial cell layers formed in the air-liquid-interface culture system and (3) increased expression of proliferative cell nuclear antigen (PCNA). At low serum concentration, the C-LMP1-expressing sublines formed larger colonies than those expressing B-LMP1. In the air-liquid-interface culture system, both B- and C-LMP1-expressing lines showed reduced epithelial differentiation resulting in reduced stratification and reduced involucrin expression similar to those of the cancer cell line, Siha. The results of the present study indicate that the expression of LMP1 in human keratinocytes is associated with morphological transformation and predisposes these cells to a more neoplastic phenotype. The structural difference between the 2 genes responsible for the functional differences and transforming ability will be pinpointed in further experiments.

Cancer in south Florida Hispanic women. A 9-year assessment.

BACKGROUND: Cancer incidence varies among race and ethnic subgroups. Relatively little information exists on cancer among Hispanics, and none of it it stratified by race. METHODS: To explore and compare cancer incidence rates of each site of cancer among white Hispanic, white non-Hispanic, black Hispanic, and black non-Hispanic women in Dade County, South Florida, we accessed 9 years of cancer incidence data. Dade County's Hispanic population is the second largest of any US county. For each site, incidence rates, rate ratios, and 95% confidence intervals are presented. RESULTS: Among white Hispanics, there were statistically significant lower rates of cancers of the oral cavity, esophagus, colon, pancreas, lung, breast, ovary, bladder, kidney, melanoma, Kaposi's sarcoma, and non-Hodgkin's lymphoma than among white non-Hispanics. Black Hispanics had significantly lower rates of cancers of the oral cavity, stomach, rectum, lung, cervix, and bladder than black non-Hispanics. However, white Hispanic women had significantly higher rates of cancers of the liver, gallbladder, and uterine cervix than white non-Hispanic women. No in situ breast cancer cases among blacks of either ethnic subgroup was found. CONCLUSION: Both white Hispanic and black Hispanic women generally had lower rates of cancer than their race-specific non-Hispanic counterparts. However, incidence rates among Hispanics differ substantially by race; failure to distinguish them may lead to erroneous conclusions.

Cloning and expression of a novel, highly truncated phosphoinositide-specific phospholipase C cDNA from embryos of the brine shrimp, Artemia.

A novel, highly truncated form of a cDNA encoding Artemia phosphoinositide-specific phospholipase C (PLC), designated PLC-beta x, was isolated from a brine shrimp cDNA library. The full-length cDNA is of the beta-type, it is 2855 base pairs long, and it contains an open reading frame encoding 489 amino acids. The deduced amino acid sequence of PLC-beta c cDNA shows novel features. It lacks several hundred amino acids at the 5' end, as compared to PLC-beta s in the higher species. It contains conserved domains X and Y, but domain X is highly truncated at the 5' end (only 14-25 conserved amino acids as compared to about 150 amino acids in the higher eukaryotic organisms). Northern blot hybridization showed that the PLC-beta x cDNA corresponds to a 4.4-kilobase mRNA. Northern blot hybridization with a cDNA probe from the 5' end and PCR performed upstream from domain Y showed that PLC-beta x is not a cloning artifact due to fusion of an unrelated clone into the coding region of the PLC-beta homologue. A functional PLC and new protein bands on SDS-PAGE were observed after subcloning full-length PLC-beta x cDNA, as well as a fragment containing the conserved regions, into expression plasmid vectors and transfecting into Escherichia coli. 1 mM lithium markedly stimulated expression in E. coli.

Mapping of amino acid residues in the C mu 3 domain of mouse IgM important in macromolecular assembly and complement-dependent cytolysis.

By analyzing the effects of single site mutations of a TNP-binding mouse IgM we have identified amino acid residues clustered in two regions in the C mu 3 domain that are important in the complement-dependent cytolytic activity of polymeric IgM. Some of the mutations also impaired IgM polymerization. For one of these clusters, D432G, P434A, and P436S, which lies on the fy2 and fy3 strands and their connecting loop, polymerization was little affected and the effect on the cytolytic activity of the polymer fraction was taken to imply direct involvement of the residue in C1 binding. The other cluster, involving residues D356A K361A and D417G, is situated at the other end of the C mu 3 domain closer to the center of the Fc mu disc. The D356A K361A and D417G mutations significantly impaired polymer formation, suggesting that these residues are necessary for proper folding or packing of the C mu 3 domains and may affect cytolysis only indirectly. Some other mutations had little or no effect on polymerization or cytolytic activity (E423A, E527G), whereas some mutations impaired only IgM polymerization without affecting cytolytic activity (D344A, K361A, K443A P544G). In others the defect in polymerization was so profound that only the monomer formed (H430A/N/Q and K438G). Our results also suggest that the C1 binding site of IgM is not strictly homologous to the C1 binding site of IgG. Although mutation of E318 of IgG has been shown to reduce its cytolytic activity, mutation of the homologous residue in IgM, E423, was without effect as were mutations of other flanking-charged residues. Proline at 436 in IgM and 331 in IgG may, however, be a common element.

Cancer among Hispanic males in south Florida. Nine years of incidence data.

BACKGROUND: Dade County (Florida) is the largest county in the United States in which a majority of the population is Hispanic. To describe and compare cancer incidence among white and black Hispanic and non-Hispanic males in South Florida and to identify those sites showing important racial-ethnic differences between sub-groups, incidence rates were determined from 9 years of prospective population-based cancer surveillance data. METHODS: All incident cancer cases diagnosed among male residents of Dade County for the period January 1, 1981, through December 31, 1989, were obtained from the statewide tumor registry for Florida, the Florida Cancer Data System. Race- and ethnic-specific age-standardized incidence rates and standardized rate ratios to compare rates among racial-ethnic subgroups for each site are presented. RESULTS: Compared with white non-Hispanics, white Hispanic males had significantly higher rates of cancers of the liver, gallbladder, nasal cavity, penis, and thyroid; acute lymphocytic leukemia; and Kaposi's sarcoma. Significantly lower rates were found for cancers of the esophagus, stomach, colon, rectum, pancreas, lung, testes, bladder, and kidney, and for mesothelioma, melanoma, non-Hodgkin's lymphoma, and chronic lymphocytic leukemia. Black Hispanics had significantly lower rates only of cancers of the esophagus, lung, and prostate than did black non-Hispanics. CONCLUSION: Differences were found in incidence rates by race-ethnicity; while incidence rates for most sites among white Hispanic males were lower than among white non-Hispanics, higher rates for several sites are of particular concern and merit further investigation to determine differences in risk factors and exposures.

Finger cooling by contact with cold aluminium surfaces--effects of pressure, mass and whole body thermal balance.

Finger skin temperature change during contact with a cold aluminium surface was studied in 20 subjects (10 men and 10 women). Contact pressure (0.1 N, 5.9 N and 9.8 N), contact material mass (large one, mass 3559 g, small one, mass 108 g), surface temperatures (-7 degrees C, 0 degree C, +7 degrees C) and whole body thermal balance were controlled as independent factors. The contact experiments were performed in a small chamber and only the first section of the index finger of the left hand was in contact with the aluminium surface. The results indicated that all the factors studied had significant effects on the contact skin temperature change with time. The study confirmed that a modified Newtonian model with two components can accurately describe the contact skin temperature change with time. The study resulted in three predictive models for critical skin temperature when in contact with cold aluminium. The results indicated that metal surfaces in contact with bare hands should not be below 4 degrees C surface temperature. Lower temperatures require insulating material or the wearing of protective gloves.

Intraperitoneal 5-fluoro-2'-deoxyuridine with escalating doses of leucovorin: pharmacology and clinical tolerance.

In a preceding study, we established the tolerance and pharmacokinetic behavior of 5-fluoro-2'-deoxyuridine (FdUrd) given by the intraperitoneal (IP) route. A dose of 3 g daily x 3 days was found satisfactory for Phase II study and exploration of biochemical modulation. Therefore, the current study was conducted to study the tolerance and pharmacokinetics of such a dose-schedule and route of FdUrd combined with escalating doses of leucovorin (LV). Fourteen patients were entered and 13 were evaluable for tolerance determination. Pharmacologic determinations of IP FdUrd and 5-Fluorouracil (FUra) derived from it and LV were obtained by HPLC methods on 11 occasions. Findings were compared with the preceding study of FdUrd alone. LV did not appear to alter the tolerance of IP FdUrd even in the four patients receiving the highest dose of LV (640 mg). Toxicities included nausea, vomiting, and rarely neutropenia and diarrhea. Pharmacokinetic parameters indicate a parallel rate of egress of FdUrd and LV from the peritoneal cavity. The pharmacologic advantage for FdUrd is at least 3 logs as previously reported and one log for LV. Evidence of antitumor effect was noted particularly among untreated patients with gastrointestinal primaries. We conclude that IP FdUrd 3 g and LV in doses of up to 640 mg x 3 days are well tolerated. Since FdUrd is more potent, has an even greater hepatic clearance and shows greater potential for modulation with LV than FUra, it may be the preferred fluoropyrimidine for subsequent studies via the IP route in the treatment of carcinomas with prominent peritoneal spread. The pharmacologic advantage for leucovorin is limited but it is a good marker for peritoneal clearance since it parallels FdUrd clearance.

Colonoscopic follow-up of colorectal carcinoma.

PURPOSE: The value of routine colonoscopy in the prevention or early detection of metachronous carcinoma of the colon and rectum remains unproven. This study attempts to clarify this issue. METHODS: An analysis of a personal series of 460 patients with primary colorectal carcinoma treated by the second author between 1972 and 1990 was reviewed. After various exclusions, there remained 231 patients who had been prospectively followed by colonoscopy with a mean follow-up period of 5.6 years. RESULTS: In 48 (21 percent) patients, there were synchronous adenomas present at the time of the initial resection for carcinoma and 35 (73 percent) of these patients subsequently developed metachronous adenomas, being recurrent in 22. Ninety-five (52 percent) of the 183 patients without synchronous adenomas eventually developed metachronous adenomas, so that overall 130 (56 percent) patients developed a metachronous adenomas. Four patients developed a metachronous carcinoma that was either Dukes A or B, and all remain well at the time of final follow-up. These metachronous carcinomas were found after a mean interval of seven and three-quarter years. All four patients had synchronous adenomas and all developed metachronous adenomas on multiple occasions before the metachronous carcinoma was detected. Thus, a subset consisting of only 22 patients produced all four metachronous malignancies--a rate of 18 percent. CONCLUSION: It would appear that the presence of synchronous adenomas with the subsequent development of recurring metachronous adenomas is significant and warrants a more intensive follow-up program to ensure the early diagnosis and more likely cure of any metachronous carcinoma.

Inhibitory effects of chlorpheniramine and astemizolum on contraction of isolated rat and rabbit uteri induced by oxytocin and PGF2 alpha.

The contraction of isolated rat and rabbit uteri induced by oxytocin and PGF2 alpha was markedly inhibited by chlorpheniramine (Chl) and astemizolum (Ast), both of which also decreased the resting tension of uteri, and their spontaneous contraction. The inhibitory effects of both drugs were dose-dependent. At high concentrations, Chl 7.4 x 10(-4) mol/L and Ast 10(-4) mol/L could counteract the contraction of the uteri induced by Oxy and PGF2 alpha, and their spontaneous contraction as well. They decreased the resting tension to the lower level. The mechanism of their non-special relaxed action on uteri could not be completely explained only by their H1-receptor blocking action. Whether they act by blocking calcium channel or by inhibiting calmodulin (CaM) remains to be further explored.

The measurement of DNA content in Wilms' tumor and its clinical significance.

DNA content analysis with paraffin-embedded tissue was performed by flow cytometry on extracted nuclei from 42 Wilms' tumors. DNA histograms showed diploid in 13, tetraploid in 12, and aneuploid in 17. The frequency of DNA aneuploid was 40.48%. The 2- and 5-year survival rates were 84% and 75.16%, respectively, for patients with the DNA diploid pattern; the 2- and 5-year DNA aneuploid survival rates were 47.06% and 41.18% (P < .03 and P < .05), respectively. The 2- and 5-year survival rates for DNA tetraploid were in between, both 55.90%. For patients with stage I and II, the difference in the 2- and 5-year survival rates between DNA diploid plus tetraploid and DNA aneuploid was even more significant (P < .004 and P < .002, respectively). Thus, DNA aneuploid is an important indicator of poor prognosis in Wilms' tumor, especially for stages I and II.

Management of ipsilateral supracondylar femur fractures following total knee arthroplasty.

A complete review of the literature revealed 195 fractures in 12 published studies of ipsilateral supracondylar femur fractures occurring about a total knee arthroplasty. This review included general surveys, multicenter studies, and case reports. Based on these studies, the authors divided the fractures into nonoperative and operative treatment groups. The fracture types were also subclassified into nondisplaced (Neer type 1) and displaced (Neer types 2 and 3). Statistical analysis of each type of fracture in terms of the various treatments and clinical outcome was performed. This study revealed satisfactory results in 83% of the patients with nondisplaced fractures that were treated without surgery. In contrast, 64% of all patients with displaced fractures that were treated with or without surgery had satisfactory outcomes. This difference is statistically significant (P < .05). Of the patients with displaced fractures, nonoperative versus operative results were not statistically significant (P < .9) (67% satisfactory vs 61%). A treatment algorithm is proposed based on the above data.

Study of natural diamonds by dynamic nuclear polarization-enhanced 13C nuclear magnetic resonance spectroscopy.

The results of a study of two types of natural-diamond crystals by dynamic nuclear polarization (DNP)-enhanced high-resolution solid-state 13C nuclear magnetic resonance (NMR) are reported. The home-built DNP magic-angle spinning (MAS) 13C NMR spectrometer operates at 54 GHz for electrons and 20.2 MHz for carbons. The power of the microwave source was about 30 W and the highest DNP enhancement factor came near to 10(3). It was shown that in the MAS spectra the 13C NMR linewidths of the Ib-type diamond were broader than those of IaB3-type diamond. From the hyperfine structure of the DNP enhancement as a function of frequency, four kinds of nitrogen-centred and one kind of carbon-centred free radicals could be identified in the Ib-type diamond. The hyperfine structures of the DNP enhancement curve that originated from the anisotropic hyperfine interaction between electron and nuclei could be partially averaged out by MAS. The 13C polarization time of DNP was rather long, i.e. 1500 s, and the spin-lattice relaxation time (without microwave irradiation) was about 300 s, which was somewhat shorter than anticipated. Discussions on these experimental results have been made in this report.

Expression of Ki67 antigen, epidermal growth factor receptor and Epstein-Barr virus-encoded latent membrane protein (LMP1) in nasopharyngeal carcinoma.

The expression of Ki67 antigen, epidermal growth factor receptor (EGFR) and the Epstein-Barr virus (EBV)-encoded latent membrane protein (LMP1) in nasopharyngeal carcinoma (NPC) was immunohistochemically determined. In cases with sufficient material western blot analysis was applied to analyse the LMP1 expression. Biopsies from 20 Chinese and 3 Swedish patients with NPC were included in the study. Our results demonstrated a nuclear Ki67 staining, a membrane EGFR staining, and a dot-like cytoplasmic and/or membrane LMP1 staining pattern in tumour cells of NPC. The proportion of Ki67-positive cells correlated with tumour stage. A strong expression of EGFR was frequently seen in patients with tumour stages III and IV and was paralleled by a higher proportion of Ki67-positive cells. The majority of the LMP1-positive cases strongly expressed EGFR and had a higher proportion of Ki67-positive cells, indicating a possible effect of EBV LMP1 on the proliferation of tumour cells in NPC. The increased expression of EGFR and Ki67 in NPC at late tumour stage indicates their possible use in malignancy grading of NPC.

Epstein-Barr virus (EBV)-encoded membrane protein LMP1 from a nasopharyngeal carcinoma is non-immunogenic in a murine model system, in contrast to a B cell-derived homologue.

Epstein-Barr virus (EBV)-encoded LMP1 gene derived from a nude mouse passaged nasopharyngeal carcinoma (NPC) of Chinese origin (C-LMP1) and its B cell (B95-8 prototype)-derived counterpart (B-LMP1) were compared for their ability to induce tumour rejection in a mouse mammary adenocarcinoma system. Each of the two LMP1 genes was introduced individually by retroviral vectors into a non-immunogenic mammary carcinoma line, S6C, that originated in an ACA (H-2f) mouse. Syngeneic ACA mice were immunised for 3 consecutive weeks with irradiated B- or C-LMP1 expressors or control cells. The immunised and control mice were then challenged with graded numbers of viable cells from the corresponding cell line. Only the B-LMP1 expressing cells were highly immunogenic. Up to 10(5) cells were rejected in pre-immunised mice, whereas at least 10(2) cells grew in non-immunised controls. No rejection response was detected against the C-LMP1 expressing cells which grew equally well in control and immunised mice, with a minimum inoculum of 10(2) cells in the majority of the clones. In a previous study, we found numerous sequence differences between B- and C-LMP1. The question of whether any of these differences is related to the non-immunogenicity of C-LMP1 needs further investigation. Meanwhile, our findings raise the possibility that the NPC cells may escape host rejection by the development of a non-immunogenic LMP1 variant under the impact of immunoselection.

Mutations of the VHL tumour suppressor gene in renal carcinoma.

Multiple, bilateral renal carcinomas are a frequent occurrence in von Hippel-Lindau (VHL) disease. To elucidate the aetiological role of the VHL gene in human kidney tumorigenesis, localized and advanced tumours from 110 patients with sporadic renal carcinoma were analysed for VHL mutations and loss of heterozygosity (LOH). VHL mutations were identified in 57% of clear cell renal carcinomas analysed and LOH was observed in 98% of those samples. Moreover, VHL was mutated and lost in a renal tumour from a patient with familial renal carcinoma carrying the constitutional translocation, t(3;8)(p14;q24). The identification of VHL mutations in a majority of localized and advanced sporadic renal carcinomas and in a second form of hereditary renal carcinoma indicates that the VHL gene plays a critical part in the origin of this malignancy.
