Experimental cerebral tumors have been induced by transplacental ENU. The morphologic study of the brains of treated rats revealed that cellular hyperplasias appear at the 30th day of extrauterine life in the paraventricular white matter, i.e., before the already known "early neoplastic proliferations". Cytofluorimetric investigations failed to demonstrate differences between treated and control rats during the 1st month. On the contrary, adenylate cyclase activity is very high in that period. The duration of the latency period is discussed.

Tumours of the nervous system have been induced by transplacental ENU. Until the fourth month of life the tumoural lesions appear as mixed glial proliferations or oligodendroglial foci. From the fourth month on they develop as glial micro- and macrotumours or as isomorphic and polymorphic oligodendrogliomas. The adenylate cyclase activity studied during these two distinct phases of tumour development was markedly reduced in brain tumours, independent of their cellular origin, compared with the level in the normal brain. On the other hand, the activity of the enzyme responsible for the synthesis of cyclic AMP is significantly increased during the first period of tumour development when early neoplastic proliferations are present.

Analgesia and locomotor activity are genetically differentiated in C 57 and DBA mice. In fact, DBA strain, unlike C 57, is very sensitive to the analgesic effects of morphine. On the contrary, morphine elicits an increase of locomotor activity only in C 57 mice. We have used this genetic approach to study the in vitro response of vas deferens contractions to morphine and methionine-enkephalin. The results obtained are the following: 1. The percentage of morphine inhibition of the electrically evoked contractions of the longitudinal muscle of the vas deferens is greater in DBA strain, which is sensitive to the analgesic effects of morphine, than in C 57 mice in which morphine exerts a stimulating effect of the locomotor activity; 2. Met-enkephalin has been found to be more active than morphine on the same preparation; 3. The inhibitory effects of Met-enkephalin appear to be greater in C 57 than in DBA mice; 4. Different doses of Naltrexone are required to reverse the effects of morphine and Met-enkephalin; 5. Cumulative doses of Met-enkephalin and morphine induce different responses in the vas deferens of C 57 and DBA mice. The results emphasize the usefulness to study analgesic activity in these strains of mice.

Between July 1968 and December 1976, 9364 consecutive patients with coronary insufficiency underwent direct myocardial revascularisation using aortocoronary bypass (ACB). Among these patients 8017 had ACB alone, and the remaining 1347 had ACB in addition to correction of other cardiac and vascular lesions. In the series of patients having ACB alone the hospital (early) mortality was 3.9%, but for 2383 surgical patients in 1976 this figure was 2.2%. Operative mortality was higher for women (6.85%) than for men (3.5%), but late survival was about the same for both sexes. Among surviving patients 88% were improved or symptom-free after undergoing ACB. According to our data, at the end of eight years 80% of the patients will be survivors. These results, when compared with other series of patients treated medically, indicate the superiority of surgical treatment both in terms of symptomatic relief and also in increased life expectancy.

A simple procedure for the preparation of beta-N-acetylhexosaminidase (EC 3.2.1.52), free from other glycosidases, from commercial bovine serum albumin is described. It has been fractionated into three peaks on a DEAE-cellulose column. The Km and V values for 4-methylumbelliferyl-beta-D-N-acetylglucosaminide (4MU-gluNAc), 4-methvlumbelliferyl-beta-D-N-acetylgalactosaminide (4MU-galNAc), p-nitrophenyl-beta-D-N-acetylglucosaminide, and p-nitrophenyl-beta-D-N-acetylgalactosaminide for each was established. The inhibition by several structural analogues on 4MU-gluNAc and 4MU-galNAc hydrolysis by the enzyme present in peak 1 was investigated.
