Cell-mediated immunity to varicella zoster virus in children being treated for cancer.

Stimulation of lymphocytes by varicella-zoster virus (VZV) antigen was measured in vitro for 37 healthy adults and children and for 35 children who developed herpes zoster or varicella while receiving anticancer therapy. For 35 of the control group the history of varicella infection correlated with the in vitro response of lymphocytes to VZV antigen. A specific lymphocyte response was observed following 32 of the 37 episodes of VZV infection in the 35 children with malignancy. Re-evaluation 7--12 months following the acute infection revealed that the in vitro response to VZV antigen was lost in six of 19 patients who remained in remission and in all patients who relapsed and received reinduction therapy. Although there was some suggestion that development of a cell mediated response early in the infection decreased the incidence of skin dissemination of the virus in herpes zoster, no consistent correlation could be made between lymphocyte responsiveness in vitro and dissemination of disease or the duration of the appearance of new lesions.

Inosiplex for localized herpes zoster in childhood cancer patients: preliminary controlled study.

By multiple criteria, inosiplex-a reputed stimulator of virus-sensitized lymphocytes-had no demonstrable therapeutic effects in a preliminary controlled study of patients with localized herpes zoster and cancer. Lymphocytes from the six drug-treated patients were no more responsive to varicella-zoster antigen and phytohemagglutinin than were lymphocytes from seven patients who received a placebo.

Fascioloides magna infection in white-tailed deer of southeastern United States.

Livers of 4,023 white-tailed deer (Odocoileus virginianus) from 13 southeastern states were examined for flukes; 514 (12.8%) harbored Fascioloides magna. Other species of liver flukes were not found. Forty-two nidi of infection were found, encompassing 56 counties in 9 states. As many as 125 flukes were recovered from a single deer. Although F magna was widely distributed throughout southeastern United States, most enzootic areas were confined to the coastal plain physiographic province. Since deer serve as reservoirs for F magna infections in domestic livestock, it was concluded that every precaution should be exercised to prevent introduction of liver flukes into areas now free of this helminth.

Studies of endoparasites of the mourning dove (Zenaida macroura) in the southeast United States.

From September, 1973, through November, 1974, 255 mourning doves (Zenaida macroura) were collected in the southeastern United States and examined for endoparasites. Thirteen species of endoparasites were found and included six species of protozoans, one trematode, two cestodes, and four nematodes. New host records included Sarcocystis sp., Echinostoma revolutum, Hymenolepis sp., Aproctella stoddardi, Ascaridia columbae, and Dispharynx nasuta.

Correlation of cell kinetic and clinical response to chemotherapy in disseminated neuroblastoma.

Mitotic and labeling indices of bone marrow tumor cells were determined from patients with disseminated neuroblastoma. Although pretreatment values were variable, the median indices indicated that only a small proportion of tumor cells were proliferating. The pretreatment indices could not be correlated with presenting clinical features or with the response to chemotherapy. Studies were repeated after 7 daily doses of cyclophosphamide (150 mg/sq m) and serially after Adriamycin (35 mg/sq m) given on Day 8. Changes in the mitotic and labeling indices during the first course of therapy could be directly correlated with the clinical response as evaluated after 4 months of induction chemotherapy. In all patients who attained a complete or partial remission, an increase in these indices was observed after 7 days of cyclophosphamide. If this increase was associated with an observed kinetic effect of Adriamycin in the G1, S, and G2 phases of the cell cycle, a complete remission was attained. If, following Adriamycin, kinetic evidence of an effect was not present in all three phases of the cell cycle, only partial remission was attained. No clinical response to therapy was observed in those patients in whom the mitotic index and labeling index did not increase after 7 days of cyclophosphamide.

Intestinal nematodes of white-tailed deer in southeastern United States.

The small intestine, large intestine, and cecum of 975 white-tailed deer (Odocoileus virginianus) from 83 localities in 13 southeastern states were examined for nematodes. In order of prevalence, the following parasites were found: Capillaria bovis, Eucyathostomum webbi, Monodontus louisianensis, Nematodirus odocoilei, Oesophagostomum venulosum, Trichuris sp, Cooperia punctata, Trichostrongylus longispicularis, Strongyloides sp, Cooperia oncophora, Cooperia spatulata, Cooperia sp, and Trichostrongylus calcaratus. Over one-half of the deer examined were free of intestinal nematodes, and most infected deer harbored few parasites. Of the most prevalent species, C bovis, N odocoilei, O venulosum, and Trichuris sp were widely distributed, whereas M louisianensis and E webbi were restricted in distribution. Severe pathogenicity was associated with only heavy Strongyloides sp infections in 2 penned deer. The findings of this study suggested that white-tailed deer of the Southeast are insignificant in the epizootiology of important intestinal nematodes of domestic livestock.

Cell kinetics and chemotherapy in neuroblastoma.

The proliferative behavior of neuroblastoma cells in bone marrow was evaluated before and after perturbation by chemotherapy. The data presented indicated that the proliferating fraction of the tumor cells before therapy was small. In most of the patients, initial therapy with a non-cell-cycle specific agent increased the mitotic and labeling indices. If initial therapy was followed by a cell cycle-specific agent, variable kinetic responses were observed. Although the number of patients was small and the follow-up was short, there was a correlation between the clinical response and the kinetic changes induced by the chemotherapy. Evaluation of cell kinetic changes during the initial course of chemotherapy might identify early those patients who are unlikely to have a good clinical response. Similar studies in the evaluation of new drugs to aid in identifying agents that favorably change cell kinetics and thus therapy for children with neuroblastoma was urged.

Parasitism among white-tailed deer and domestic sheep on common range.

Parasitism was studied in white-tailed deer (Odocoileus virginianus) and domestic sheep (Ovis aries) which shared a common range in eastern West Virginia. Of 30 species of internal parasites, 11 were found in deer and 22 in sheep. Five parasites, Sarcocystis sp., Cysticercus tenuicollis, Oesophagostomum venulosum, Cooperia punctata, and Gongylonema pulchrum, occurred in both deer and sheep. An index of similarity of 17.2 suggests that the parasite faunas of these hosts are distinct, and that it is unlikely that white-tailed deer are reservoirs of common parasites of domestic sheep in the southern Appalachian region.

Evidence for recruitment and synchronization in leukemia and solid tumors.

In malignant cell populations which are sensitive to drug therapy it does not seem necessary to potentiate treatment with perturbation of the proliferative characteristics. However, in some relatively drug-resistant populations significant advantages can be obtained by recruiting resting cells into the drug-sensitive cell cycle. Further studies of growth-regulatory mechanisms for malignant cell populations may help a great deal in designing better methods of recruiting the resting cells back into active cell division. Synchronization of the dividing cells in a specificially drug-sensitive phase of the cell cycle is another method of potentiating chemotherapeutic effectiveness in relatively drug-resistant cell populations. The biochemical changes induced by sequential drug administration may be equally important in sensitizing the malignant cell to subsequent drug administration.

Helminth parasitisms among intermingling insular populations of white-tailed deer, feral cattle, and feral swine.

Helminth infections among free-ranging, intermingling populations of white-tailed deer (Odocoileus virginianus), cattle (Bos taurus), and swine (Sus scrofa) on an island off the Georgia coast were studied. Of 39 species of helminths collected, 19 were found in deer, 17 in cattle, and 13 in swine. Of 28 species of helminths recovered from ruminants, 8, viz, Capillaria bovis, Cooperia punctata, Dictyocaulus viviparus, Gongylonema pulchrum, G verrucosum, Haemonchus contortus, Moniezia benedeni, and Trichostrongylus axei, occurred in both deer and cattle. Common liver flukes (Fasciola hepatica) infected cattle and swine but not deer. Only 1 helminth, G pulchrum, infected deer, cattle, and swine. The findings suggested that helminths harbored by the host species are distinct, with little exchange occurring.
