[The intraindividual variation of fibrinolytic activity in volunteers (author's transl)].

In five volunteers fibrinolytic activity has been measured five times over a period of six weeks during increasing values of circadian rhythm. Euglobulinlysis-time, paper fibrinolysis, fibrinogen, plasminogen, alpha1-antitrypsin, alpha2-makroglobulin, thrombino-coagulase-time and fibrin-split-products were used in one study. An increase of fibrinolytic activity in the mooning was observed in all volunteers. Intraindividual variation of values is considerable but less than variation of values compared interindividually. Moreover variation was smaller at noon than in the time before noon. In clinical trial of the short-time effect of drugs the period of the plateau of the fibrinolytic activity should be used because of its smaller variation of values. Furthermore, in this period the intraindividual variation of the fibrinolytic activity is smaller than during increasing slope of the circadian rhythm of fibrinolysis. Intraindividual control should be granted.

Evidence for lysosomal enzyme recognition by human fibroblasts via a phosphorylated carbohydrate moiety.

Adsorptive endocytosis of five different lysosomal enzymes from various human and non-human sources was susceptible to inhibition by mannose and l-fucose, methyl alpha-d-mannoside, alpha-anomeric p-nitrophenyl glycosides of mannose and l-fucose, mannose 6-phosphate and fructose 1-phosphate. A few exceptions from this general scheme were observed for particular enzymes, particularly for beta-glucuronidase from human urine. The inhibition of alpha-N-acetylglucosaminidase endocytosis by mannose, p-nitrophenyl alpha-d-mannoside and mannose 6-phosphate was shown to be competitive. The loss of endocytosis after alkaline phosphatase treatment of lysosomal enzymes supports the hypothesis that the phosphorylated sugars compete with a phosphorylated carbohydrate on the enzymes for binding to the cell-surface receptors [Kaplan, Achord & Sly (1977) Proc. Natl. Acad. Sci. U.S.A.74, 2026-2030]. Endocytosis of ;low-uptake' forms of alpha-N-acetylglucosaminidase and alpha-mannosidase was likewise susceptible to inhibition by sugar phosphates and by alkaline phosphatase treatment, suggesting that ;low-uptake' forms are either contaminated with ;high-uptake' forms or are internalized via the same route as ;high-uptake' forms. The existence of an alternative route for adsorptive endocytosis of lysosomal enzymes is indicated by the unaffected adsorptive endocytosis of rat liver beta-glucuronidase in the presence of phosphorylated sugars and after treatment with alkaline phosphatase.

Pituitary somatotrophs contain [Met]enkephalin-like immunoreactivity.

Serial sections from epoxy resin-embedded rat anterior pituitaries were sequentially immunostained for endorphin, [Met]enkephalin, and growth hormone, respectively. We found that [Met]enkephalin immunoreactivity was confined to the growth hormone producing cells. Corticotropin/endorphin cells in the anterior pituitary from both normal and adrenalectomized rats did not contain any [Met]enkephalin immunoreactivity. When anterior pituitary cells were maintained in monolayer culture for 10 days, [Met]enkephalin immunoreactivity was still located in the growth hormone-producing cells although the staining was weaker than in the somatotrophs in pituitary tissue fixed immediately after death of the animals. This suggested that somatotrophs synthesize [Met]enkephalin. However, this cannot be proved conclusively until biosynthesis experiments have been performed. The following conclusions were drawn from these findings. (i) Anterior pituitary [Met]enkephalin is not an extraction artifact derived from beta-endorphin with which it shares the NH2-terminal pentapeptide sequence. (ii) In the anterior pituitary, beta-endorphin is not the precursor to [Met]enkephalin. [Met]Enkephalin in somatotrophs may be of brain origin and in the somatotrophs may be bound to intracellllar receptors as has been shown for luteotropin releasing hormone in gonadotropic cells.

Biochemical effects of gum arabic, gum tragacanth, methylcellulose and carboxymethylcellulose-Na in rat heart and liver.

Repeated oral administration of commonly used suspending media, gum arabic, gum tragacanth, methylcellulose, and carboxymethylcellulose-Na to rats caused uncoupling of oxidative phosphorylation in liver and heart mitochondria and partial inhibition of mixed function oxidases of liver endoplasmic reticulum, as measured by 2-biphenylhydroxylation and 4-biphenylhydroxylation. There were considerable differences between the compounds with regard to potency and reversibility of these effects. Only methylcellulose at a concentration of 0.5% did not alter mitochondrial function and mixed function oxidases. It is recommended as suspending medium for the use in pharmacological and toxicological experiments.

Granules and Golgi vesicles with differential reactivity to ACTH antiserum in the corticotroph of the rat anterior pituitary.

Slices of unembedded rat anterior pituitaries, fixed with a periodate-lysine-paraformaldehyde (PLP) fixative, were incubated with guinea pig antiserum to ACTH and stained with a peroxidase-conjugated IgG fraction of anti-guinea pig gamma-globulin serum from rabbits. The fine structure of the stained cells was identical to that of the ACTH-secreting cell, as described by Siperstein and coworkers. Immunoreactive granules were mainly located at the periphery of the cell. Numerous granules of the inner cytoplasm and also the Golgi complex were nonreactive to the antiserum. [Evaluation of blocking and activation response of focal EEG abnormalities by opening and closing the eyes (author's transl)].

Blocking and activation of constant focal EEG abnormalities by opening and closing the eyes were assessed by visual analysis in 173 EEG recordings of 173 patients. Correlations between single variable were weak in general and did not gain statistical significance in any case. The blocking response had similar extent on both hemispheres. There was no sign of direct dependence from extra- or intracerebral localisation of the causal process. However, the biplot method of computing the results discovered several constellations of variables responsible for blocking and activation. Two of them are reported here. 1) Both responses were more prominent at frontal than at occipital areas. 2) A prodominant slowing localized in mainly frontal regions on the left hemisphere and caused by a process extending from extracerebral or cortical to subcortical showed the greatest tendency to be blocked by opening the eyes while the cause of the focus had no influence.

[Effect of sulfinpyrazone on enhanced platelet aggregation in vitro (author's transl)].

Adding sulfinpyrazone in a concentration of 500 and 250 microgram/ml to blood of patients with enhanced platelet stickiness a significant decrease (p less than 0.01) of the maximum amplitude and the aggregation velocity could be demonstrated with the spontaneous platelet aggregation test, PAT III, of Breddin, as well as with the collagen and the ristocetin induced aggregation. The lower concentration of 50 microgram/ml of sulfinpyrazone yielded the same results when aggregation was induced with the higher concentrations of 1.5 microgram/ml ristocetin and 5 microgram/ml collagen. Only the maximum aggregation velocity of the PAT III was significantly reduced after addition of 50 microgram sulfinpyrazone/ml blood. With sufficient concentrations of sulfinpyrazone the anti-aggregating action of this substance can very well be demonstrated with the aggregation induced by ristocetin or collagen and the PAT III. The effect is dose dependent.

3H-Cymarol in man. Excretion pathways and serum protein binding.

In 10 patients, 5 having received 3H-cymarol i.v., 5 orally, the radioactivity in plasma, urine and in the feces of some patients also was determined. After oral administration the plasma levels rose rapidly reaching maximum levels 1--2 h after administration. After i.v. injection about 30% of the given radioactivity were excreted in the urine. The remaining radioactivity was found in the feces suggesting a high biliary excretion. Only 10% of the radioactivity excreted in the first 24 h were chloroform-extractable. The radioactivity found in the urine after oral administration of the drug amounted to 17.6%. Between 51.1 and 58.5% of the drug were bound to serum proteins.

[The Bifosolar Ideal light-protection spectacles (author's transl)].

Bifosolar glass is made without heating, therefore the phototropic material keeps unchanged its original light sensitivity. A further development of the Biofosolar glass leads to the "Bifosolar Ideal dual strenght glass". This can be used in reduced visual acuity by combining with additional corrections up to 8.0 dioptres. The author describes the manufacture and the technical data of the various Bifosolar glasses.

A simple technique for supporting single cells in electron microscopic immunocytochemistry and embedding.

Cell suspensions of rat anterior pituitaries were filtered with a polycarbonate filter (pore size 3 micron) and fixed on the filter. After fixation the cells were adherent to the filter and immunocytochemical staining could be accomplished by simply dipping the filter into the different incubation media. The cells could be dehydrated and embedded in Epon 812 on the filter. After polymerization the embedded filter was sawn into small blocks and the cell layer was sectioned tangentially on an ultramicrotome. This method also seems to be applicable to other histochemical studies on single cells.
