Immunogenicity of tumor cells modified by trinitrobenzene suflonic acid (TNBS).

The ability of trinitrophenylated tumor cells to stimulate syngeneic antitumor response has been tested in 3 different tumor-host systems: A. Trinitrophenylated and inactivated Moloney induced YAC tumor cells (YAC-TNP) were able to induce the production of cytotoxic antibodies in low responding A/J mice, while inactivated YAC tumor cells (YAC-In) failed to induce such a response. Furthermore, A mice which were injected with YAC-TNP rejected 103 viable YAC tumor cells at a higher frequency than those injected with YAC-In. B. Trinitrophenylated and inactivated Gross virus induced G-35 tumor cells or Monoloney induced LSTRA cells (both syngeneic in BALB/c mice) were as immunogenic as nonmodified inactivated tumor cells. About 50% of the immunized mice survived indefinitely after injection of 103 viable tumors. Fruthermore, spleen cells from mice primed with either modified or nonomodified G-35 cells responded in vitro to G-35 in a mixed leukocyte tumor interaction and generated specific cell-mediated cytotoxic activity to 51Cr-G-35 syngeneic tumors. However, the donors of the primed cells did not produce detectable cytotoxic antibodies to G-35. C. In vitro sensitization of C57B1 spleen cells by trinitrophenylated Mitomycin C treated syngeneic EL-4 generated a stronger cytotoxic response to EL-4 cells than obtained by sensitization with Mitomycin C treated EL-4 cells alone, The superiority of the sensitizing capacity of trinitrophenylated EL-4 was readily demonstrated in conditions which were suboptimal for nonmodified Mitomycin C treated tumor. Both theoretical and practical implications of these results are discussed.

[Inhibition and stimulation of ATPase activity and the nonenzymatic hydrolysis of ATP in electron histochemistry (author's transl)].

In rat liver, homogenized and fractionated, ATPase activity was studied both histochemically and biochemically in the presence of lead ions and with addition of cysteine, EDTA, 2,4-DNP, sodium cyanide, sodium fluoride and p-chloromercuribenzoate. None of the inhibitors concerned permitted to obtain a complete inhibition of ATP hydrolysis. A practically complete inhibition of ATPase was possible only through heat inactivation or after double prefixation with formaldehyde and ethanol. Nonenzymatic ATP hydrolysis plays a negligible role in ATPase histochemistry since phosphate yield taking place due to lead action is insignificant against enzymatic hydrolysis of ATP.

Ultrastructural demonstration of NAD-pyrophosphorylase activity in mouse liver nuclei.

Ultrastructural demonstration of NAD-pyrophosphorylase activity (E.C.2.7.7.1) in isolated mouse liver nuclei was investigated with the use of an electronhistochemical procedure based on the precipitation of pyrophosphate ions with lead ions under conditions permitting simultaneous ATPase inhibition by formaldehyde/ethanol prefixation. In isolated mouse liver nuclei activity of NAD-pyrophosphorylase was found in nucleoli, in interchromatin granules, coiled bodies and strand-like structures in nucleoplasm.

Histological studies after experimental grid irradiation vi. Findings on Guérin-tumours irradiated by high doses.

Author demonstrated subsequent to high-dose grid irradiation in Guérin-tumours of the rat the histological grid pattern corresponding to inhomogeneous dose distribution. Two to four days after irradiation through the grid of narrow openings in some sites certain phenomena could be observed, which seem to refer to the severe damage acquiring a diffuse character, but after 7 days the histological grid pattern has been clearly defined and the severely damaged stripes were about 50 percent narrower than the relatively intact ones. On the basis of the results on the actual tumour and among the applied experimental conditions the existence of a specific grid-tumour effect could not be justified by histological methods.

Histological studies after experimental grid irradiation. IV. Findings in the skin.

In the skin of the rat with longer focus-to-grid distance and wider slot openings the grid pattern corresponding to the inhomogeneous dose distribution may be demonstrated by histological means; after slot irradiation the damaged skin portion corresponding to the slot may be recognised. The shorter length of this skin portion compared with the width of the slot as well as the differences after grid irradiation of the average length of the severely damaged and relatively intact portions is in accordance with the authors conception, justified already by previous experiments, namely that the tissue parts covered by the bars of the grid exert a protective influence on the adjacent directly irradiated tissues.
