Influence of diet composition on the weight gains of Wistar rat.

The composition of diets used for breeding and experimentation on laboratory animals is one of the important factors affecting the quality of the animals and the comparison of experimental data between different laboratories. Six diets used either at the Institute of Physiology, Czechoslovak Academy of Sciences in Prague, or in the Research Centre of Molecular Biology and Medicine, Academy of Sciences GDR in Berlin were analysed for moisture, crude protein, fat, fibre, ash, sodium, potassium, calcium, magnesium, phosphorus and chloride and fed to Wistar rats for 70 and 112 days. Weight gains of the animals were compared and statistically evaluated. The greatest weight gains were achieved with animals fed on the DOS 2b diet.

The influence of atherogenic diet on electromechanical coupling in rabbit myocardium.

To characterize the role of the SR in the electromechanical coupling process in rabbit myocardium after a 12-week-long atherogenic diet containing cholesterol and oil, the influence of changes in Ca2e+ on isometric force, relaxation rate, and resting potentiation of isolated trabeculae were compared with 45Ca2+ binding and uptake by the SR. The diet produces a more strongly expressed resting potentiation of force at 1.25 and 2.5 mM Ca2e+ and an increase in the Vmax equivalent, the contractility index [(dF/dt)max/F'] and the relaxation index [-(dF/dt)max/F]. A higher Ca2e+-accumulating activity of heart microsomes from animals was observed after the diet. It is concluded that changes of SR function are involved in the mechanism responsible for modification of electromechanical coupling induced by cholesterol- and oil-containing diet.

Studies on several 7-substituted N,N-dimethyltryptamines.

Several 7-substituted derivatives of N,N-dimethyltryptamine (DMT) were prepared and evaluated in the rat fundus serotonin receptor assay and in a behavioral (discriminative stimulus) assay in rats. Both 7-Me- and 5-OMe-7-Me-DMT possess a higher pA2, and 5,7-(OMe)2-DMT a lower pA2, than that of DMT itself. Like DMT, all three of these compounds produce behavioral effects in rats which are similar to those of the hallucinogen 5-OMe-DMT. Although 7-ET- and 7-Br-DMT possess a higher serotonin receptor affinity than DMT, neither produce behavioral effects which parallel those of 5-OMe-DMT. In contrast, 6-Me-DMT and its 5-OMe derivative do not interact with the serotonin receptors in a competitive manner and are inactive in the discriminative stimulus assay.

[Application of the serological clostridium assay in tumour diagnostics (author's transl)].

The suitability for serologic tumour diagnostics of the non-oncolysing strain Clostridium butyricum CNRZ 528 has been investigated. By introducing the transplantable Brown Pearce tumour of the rabbit and spontaneous tumours of the dog as test models the antibody production against clostridial rods--indicating malignant growth as had been proved with the Möse strain Cl. oncolyticum M55--has been evaluated. It could be established--utilizing the complement fixation test--that one serological method alone is not sufficient for obtaining clear-cut test results in anyone case. It is to be recommended that the indirect hemagglutination and the complement fixation tests ought to be combined. Our studies on specific clostridial antigens and on optimization of techniques will be continued.

Protocol for determining the auditory percepts of electrical stimulation of the cochlea.

The percepts of electrical stimulation of the auditory nerve can best be determined in man. An eight channel bipolar electrode array has been developed which allows discreet stimulation of segments of auditory nerve via the scala tympani. In addition, a programmable stimulator has been developed which allows the delivery of different waveforms and stimulus patterns. Hopefully this will begin to allow the generation of the elements of speech.

[The virostatic effect of adenine-arabinoside monophosphate. Experimental findings and preliminary clinical experiences].

Adenine arabinoside (Ara-A), a synthetic nucleoside, is an antiviral agent, which is effective against poxviruses and viruses of the herpes group. The monophosphate (Ara-AMP) has the great advantage of better solubility, compared to the parent compound. Experimental studies show the outstanding effect of Ara-AMP on the encephalitis by vaccinia-virus in white mice. Even after immunesuppression by total body irradiation, the survival rate of the Ara-AMP treated animals was significantly higher compared to the untreated controls. In experimental vaccinia-virus infections in rabbits, who had undergone total body irradiation. Ara-AMP has a favourable influence upon the course of the disease and the prognosis. Some clinical observations in children with immunesuppression, who suffered from generalized zoster of varicella-infections, are reported. Furthermore in cases of viral encephalitis Ara-AMP was successful. Controlled studies of a greater number of patients on the therapeutic value of Ara-AMP in infections with DNS-viruses are needed.

[Mechanical characteristics of isotonic work and high-energy phosphates in frog ventricle after 1-fluoro-2,4-dinitrobenzene].

The blocking of the creatinphosphokinase by 1-fluoro-2,4-dinitrobenzene (FDNB) allows to investigate the relationship between ATP-supply, contractility and relaxability of the frog's myocardium. In isotonically working isolated ventricles of frogs the time of work, systolic and diastolic volume, velocity of contraction and relaxation as well as the levels of CP, ATP, ADP and AMP were measured at different intervals until termination of each experiment. CP shows a small variation, ATP decreases to 60% and ADP + AMP increase for the same amount under FDNB during the development of a slight inhibition of contractility and a continuously growing inhibition and retardation of relaxation until systolic arrest. ATP content and volume of relaxation correlated strictly. The contracture and the diminished contractility are caused by the decrease of ATP, producing a lack of substrate for Ca transport and actin-myosin-ATPase. This models the course of events during an insufficiency like in angina pectoris and in myocardial infarction.

Immediate X-radiation induced contractions of the isolated guinea pig terminal ileum: the localization of the effect by drugs.

Tone and motility of the isolated guinea pig ileum were increased by irradiation with a dose of 10 krd. The maximal effect corresponds to that induced by 0.001 microng/ml acetylcholine or 0.3 microng/ml nicotine. The pharmacological analysis of this effect performed with acetylcholine and nicotine and several blocking agents including hexamethonium, atropine, tetrodotoxin, diphenhydramine, and verapamil suggests that radiation acts on the postganglionic parasympathetic neuron and the neuromuscular synapse. The mechanism of radiation is likely to consist of both an increased release of acetylcholine from the postganglionic neuron and a sensibilization of the cholinergic receptor site at the smooth muscle cell. The latter effect is thought to result from an increased contractile action induced by acetylcholine or nicotine in the irradiated ileal smooth muscle.

Contrast media with and without calcium for cardioangiography in children.

The possible beneficial effects of added calcium ions to contrast media to diminish myocardial toxicity was evaluated in 93 cardiac catheterizations with cardioangiography, performed in 50 children and adolescents. Two contrast media with equal iodine content (370 mg/ml) and similar physical properties were used: Isopawue-370, containing 0.34 mg calcium/ml, and Renografin-76, not containing calcium. It is concluded that addition of calcium to contrast media for cardioangiography in children and adolescents does not alter their myocardial, fluid or electrolyte effects when injected selectively into the cardiac chambers or great vessels.

[Effect of anoxia on energy supply and isotonic work performance in the myocardium of the frog].

To investigate correlations between energy supply and mechanical work in the frog's myocardium in true anoxia, the stroke volume, systolic and diastolic volumes and the parameters of velocity of contraction and relaxation of frog hearts were compared to the levels of high energy phosphates and the delivery of lactate. During perfusion with N2 saturated Ringer solution, stroke volume, systolic contractility and diastolic relaxation decrease till a contracture. High preload produces a dilatation growing up to the contracture after retarded and weakened relaxation. The ATP-content decreases during the first quarter of the experiment to 60%. CP decreases continuously to 15%, ADP and AMP remain constant. There is a production of lactate increasing considerably with the onset of contracture. The measured glycolysis is not sufficient for production of mechanical work. The effect of anoxia on the action potential and the reduction of the sequestration of Ca++ and of the break of actomyosin bridges following the decrease of ATP are considered as causing the series of the mechanical events.

[Perioral dermatitis].

Intensive investigations between 1969-1974 on 245 patients with perioral dermatitis are reported. Results are discussed with special regard to clinical behaviour, histology, differential diagnosis and etiology of the disease. Eczematous sources, bacterial and mycotic infections can be ruled out as etiological factors. Therapeutical considerations are discussed.
