Apolar ecdysteroid esters in adult female crickets, Gryllus bimaculatus.

Six hours after injection of 0.5 microCi 3H-ecdysone into the hemocoele of adult female crickets, several labelled compounds could be separated from hemolymph and tissues by silicic acid column chromatography, TLC, and HPLC. The amount of conjugated, polar ecdysteroids was low in all tissues, whereas apolar metabolites were predominant in all tissues. The apolar compound A2, which is the most abundant in quantity, could be hydrolyzed by porcine liver esterase, yielding ecdysone and various long chain fatty acids. This represents a new class of apolar ecdysteroid conjugates not yet found in other insects.

Cytogenetic effects in lymphocytes of formaldehyde workers of a paper factory.

Chromosome analyses were carried out in lymphocytes from 20 male papermakers exposed to formaldehyde (FA) for 2-30 years. 20 male workers from the same factory but without FA exposure served as controls. In the exposed group a significantly increased incidence of dicentrics or dicentrics and ring chromosomes was observed for 11 workers currently employed as supervisors. In contrast to 9 operators their total mean exposure time to FA was about 2.5 times longer. No significantly higher SCE values were found for smoking or for non-smoking FA workers compared with the corresponding control subjects.

Changes of subgingival total colony forming units and black pigmented bacteroides after a single irrigation of periodontal pockets with 1.64% SnF2.

Periodontal pockets of 11 subjects were irrigated once with either 1.64% SnF2 or saline applied by syringe. Changes on the total anaerobic colony forming units (CFU) and black pigmented bacteroides (BPBs) of the irrigated pockets were assessed for 31 days. The results showed little change of the CFUs by either single application of SnF2 or saline. However, SnF2 irrigation produced a decrease in the BPBs for 1 week with the greatest effect (1/2 log reduction) being seen at 3 days after the irrigation. Some subjects had more impressive reductions in BPBs, yet four of the 11 subjects showed no reductions in BPBs in those sites irrigated with SnF2. Further studies with frequent irrigation intervals are necessary to evaluate the potential usefulness of SnF2 for subgingival irrigation.

Changes of the tear film in dry eyes.

This paper describes the alterations in the structure of the tear film and its composition, which can lead to the dry eye syndrome. An attempt has been made to differentiate the individual components of these syndromes and Vogt's method is described as a further aid to diagnosis.

Primary metastasizing aortic endothelioma.

An instance of malignant endothelioma, primary in the aorta, metastasizing to intestine and bone, is reported. The aortic tumor was successfully resected. The unexpected finding of a large hepatic growth at autopsy raised the possibility that the aortic neoplasm was a metastasis. Histochemical, immunologic, and ultrastructural studies supported the diagnosis of an endothelial neoplasm. Although a number of malignant aortic and large vessel tumors have been reported, only four previous instances appear to be endotheliomatous.

[Inoculation hepatitis caused by acupuncture. Clinical cases studied over a 9-year period].

In the period between 1976 and 1983, at a hospital in the south of Germany, 17 cases of hepatitis (16 hepatitis B, 1 NANB) were probably, and 4 further cases possibly, traceable to prior acupuncture performed by a non-medical practitioner. In recent years, the incidence of hepatitis B has, in general, decreased, probably through the elimination of post-infusion type B hepatitis. Acupuncture hepatitis has now attained such a degree of importance that in any case of type B or NANB hepatitis, the patient should be questioned as to possible previous acupuncture treatment. Imperfect disinfection and sterilization in the offices of those practising acupuncture must be disclosed and remedied.

Integration of different keratins into the same filament system after microinjection of mRNA for epidermal keratins into kidney epithelial cells.

We have isolated poly (A)+ RNA, highly enriched in keratin mRNA from bovine muzzle epidermis, and injected it into epithelial cells of a different type, i.e., cultured kidney epithelial cells of the same (MDBK) or taxonomically distant (PtK2) species. Both recipient cell lines contain keratin polypeptides that are different from those present in epidermal cells. Using keratin subtype-specific antibodies in immunofluorescence and immunoelectron microscopy, we show that foreign keratin mRNAs when injected into a different type of epithelial cell can recruit polyribosomes and are translated together with the keratin mRNAs of the host cell. Foreign epidermal keratins are excluded from vimentin filaments and other structures but readily coassemble with the endogenous keratins and appear to be integrated into the meshwork of the preexisting kidney-type keratin filaments. Our observations indicate that different sets of keratin polypeptides from the same or different species can coassemble in the living cell into a common filament system. Thus we have developed a procedure that allows experimental alteration of the intermediate filament cytoskeleton within living epithelial cells.

Chromosome aberrations in human lymphocytes induced by fission neutrons.

The dose-response relationships of dicentrics and excess acentrics were analysed after exposure of human lymphocytes to a mixed fission neutron-gamma-ray beam. From the analysis of exclusively first division cells a linear-quadratic relation was obtained for dicentrics with the ratio of linear and quadratic components, zeta, equal to 2.76 Gy. Over the range of doses studied (0.04-1.97 Gy) intratrack events therefore predominated. This also applied to acentrics which were linearly related to dose. At the lowest level of observed effect and dose, r.b.e. values with respect to 60Co gamma-rays of up to about 11 were derived for dicentrics and acentrics. With increasing neutron dose the r.b.e. decreased.

[Correlation between endoscopic biopsy findings and sonograms of the abdomen in stomach cancer].

35 out of 57 patients with gastric carcinoma presented with a so called "target" pattern. In cases with distal cancer the more advanced cases with endoscopic types Borrmann III/IV showed this sign more frequently than less advanced forms, i.e. suspected early cancer, Borrmann I or Borrmann II. On the other hand in endoscopically advanced cancers Borrmann III/IV a thickening of the gastric wall was more frequent in distal than in proximal localization of carcinoma. The presence of target sign in abdominal ultrasound did, statistically, not influence gastric resection. Other findings, which were observed exclusively in gastric cancer type Borrmann III/IV, like infiltration to the surrounding (pancreas, liver), liver metastasis, and/or ascites, were decisive. Additional sonography of the abdomen is, therefore, a valuable preoperative diagnostic procedure in gastric cancer beside upper GI endoscopy and biopsy.

[Specificity and sensitivity of the solid phase hemadsorption test: a study of treated and untreated syphilitics].

SPHA-test was performed on 317 treated and 78 untreated patients with syphilis and partly compared to 19S-IgM-FTA-ABS- as well as VDRL-test. Equivalent results were obtained in a reference laboratory, the correspondence being 94.8 percent regarding SPHA and 95.6 percent concerning 19S-IgM-FTA-ABS. In comparison to 19S-IgM-FTA-ABS, increased numbers of weakly reactive values occurred in SPHA correlating with the Latex-test. Because of its specificity and sensitiveness, SPHA turned out to be superior to VDRL with regard to the control of treatment. The biological significance of weakly reactive SPHA results is still unclear and does not indicate a necessity for therapy.
