Abnormal chromosome 9 in a neonate program. Report of three cases.

We describe three cases with abnormal chromosome 9. Patient 1 shows translocation in a homologous chromosome, with a karyotype of 46,XX,t(9;9)(9pter----cen----9pter; 9qter----cen::9q13----9qter), 1qh+. This case has a variety of anomalies, including brain anomalies. Patient 2 shows a partial trisomy 9p with a karyotype of 47,XY,+del(9)(pter----q11:). The patient has the typical clinical features of 9p trisomy syndrome. Patient 3 is unique because of partial 9p tetrasomy mosaicism without phenotypic abnormalities; the karyotype is mos 46,XY/47,XY,+dic(9)(pter----cen----q21::q21----cen----pter).

[Trigeminal neuralgia associated with primitive trigeminal artery: report of two cases].

Two cases of trigeminal neuralgia associated with the primitive trigeminal artery are reported. From 1981, the authors have treated 131 trigeminal neuralgia patients with microvascular decompression. Among them, we encountered two rare cases of trigeminal neuralgia associated with the primitive trigeminal artery (PTA) and its variant (PTAV). Case 1 is a 74-year-old woman who was admitted to our hospital due to pain of maxilla and mandible. We diagnosed her pain as trigeminal neuralgia. Preoperative angiogram showed the primitive trigeminal artery arising from the cavernous portion of the right internal carotid artery (ICA). She underwent a microvascular decompression operation. We found that her right trigeminal nerve was compressed by the right superior cerebellar artery (SCA) and the right anterior inferior cerebellar artery (AICA). We transferred the offending arteries, and her pain disappeared. Case 2 is a 48-year-old man who was admitted to our hospital due to severe mandibular pain. We diagnosed his pain as trigeminal neuralgia, and he underwent a microvascular decompression operation. His left trigeminal nerve was found compressed by the left SCA and the AICA, and the AICA was arising from the direction of Meckel's cave. His severe pain disappeared completely after operation. Postoperative angiogram of his left ICA showed an aberrant artery arising from the cavernous portion of the ICA, to the region of the left AICA. This aberrant artery is a variant of PTA (PTAV). PTA and PTAV, the so called persistent congenital arteries, are said to accompany aneurysms and other vascular lesions, and affect hemodynamic stress.(ABSTRACT TRUNCATED AT 250 WORDS)

Epithelial dysplasia produced by carcinogen pretreatment and subsequent wounding.

Golden hamsters in which the tongues were pretreated with DMBA for 4 weeks, subsequently excised, and either received no treatment or received additional postexcisional applications of acetone or DMBA for 9 to 13 days exhibited histologic evidence of epithelial dysplasia; these dysplastic lesions stained positive for papillomavirus genus-specific antigens. Conversely, animals in which the tongues received the same pretreatment yet had no excision failed to show any dysplastic changes, even though the tongues received additional applications of DMBA. Furthermore, papillomavirus genus-specific antigens were not demonstrable. The animals in which the tongues received no pretreatment with DMBA for 4 weeks followed by excisional wounding showed normal healing, even though the tongues were post-treated with DMBA. Papillomavirus genus-specific antigens could not be demonstrated in this last group.

Influence of food on bioavailability from two controlled-release granules of theophylline.

The bioavailability and absorption pattern of theophylline from Theolong granule and Theodur-G were studied in five healthy volunteers both in fasting conditions and after taking two different meals. In all the subjects, no dose-dumping effect was observed with a standard- or a fat-rich breakfast on these preparations. Postprandial administration delayed the absorption of theophylline and a fat-rich breakfast significantly reduced the area under the curve (AUC0-26) for both preparations. The mean AUC decreased by 14.0% for the former and 19.7% for the latter when the fat-rich breakfast was administered. Intake of a standard breakfast resulted in a decrease in the AUC0-26 by 10.6% for Theolong granules but there was little effect observed with the Theodur-G granules.

Preparation of zein microspheres conjugated with antitumor drugs available for selective cancer chemotherapy and development of a simple colorimetric determination of drugs in microspheres.

Zein microspheres conjugated with antitumor drugs (mitomycinc (MMC), daunomycin hydrochloride (DM), peplomycin sulfate (PEP] were prepared by using a dimethyl sulfoxide (DMSO)-H2O system. MMC with low solubility in H2O was easily entrapped by the standard procedure, whereas some modifications were required for moderately and highly soluble drugs such as DM and PEP. Colorimetric determination of the drugs in microspheres was easily achieved by use of the phenol-sulfuric acid method for drugs with sugar moieties in their molecules, such as DM and PEP, while a simple treatment of the microspheres with concentrated sulfuric acid was applied in the case of drugs having a chromophore in their molecules, such as DM and MMC.

Novel preparation of zein microspheres conjugated with PS-K available for cancer immunotherapy.

Microspheres which entrapped PS-K were prepared using Zein as a carrier matrix by a one-step or two-step process in dimethyl sulfoxide-H2O media. Microspheres prepared by the latter process provided satisfactory recovery and uptake of PS-K. Use of a catalytic amount of dl-camphorsulfonic acid, glutaraldehyde and rapid addition of aqueous solution of polyvinylpyrrolidone into the reaction media are crucial for the preparation of fine and mono-dispersed microspheres. The release rate of PS-K could be controlled by altering the ratio of PS-K to Zein and the conditions of the medium. In the presence of actinase E, drug release was considerably increased to 70-80% after 24 h incubation. Sonication of the aggregated microspheres readily yielded a mono-dispersed preparation with a particle diameter of less than 1 micron, which would be a suitable size for phagocytosis by macrophages.

[Morphological changes of pancreatic B cells in ventromedial hypothalamic (VMH)-lesioned obese rats].

Morphological changes of pancreatic B cells were investigated in ventromedial hypothalamic (VMH)-lesioned obese rats. An increase in body weight, a decrease in body length, and a marked increase in fat-pad were observed in VMH-lesioned obese rats. The volume density of pancreatic islets and that of B cells in VMH-lesioned obese rats were more increased than those in sham-operated control rats. The B cells of VMH-lesioned obese rats often showed a slight reaction for aldehyde fuchsin or anti-pig insulin serum. By the electron microscopy, the degranulated B cells were found to contain well developed Golgi apparatus and rough endoplasmic reticulum. These observations indicate increased activities of synthesis and release of insulin. We concluded that VMH-lesioned obesity was caused by accelerated lipogenesis with hypersecretion of insulin.

Endothelium and thrombomodulin: expression and distribution of thrombomodulin on altered endothelium and neoplastic syncytiotrophoblast.

We investigated the expression of thrombomodulin (TM) on endothelium in some pathologic states. We used the cultured endothelial cells treated with interleukin-1 (IL-1) and propagated cells by serial subculture for extended periods of time and assessed cell-surface TM molecules. We also studied the distribution of TM on surgical specimens of chorionic diseases, angiosarcoma and on several established cell lines of human choricarcinoma. Subculture of human umbilical vein endothelial cells (HUVE) up to 2 months (approximately 16 subcultures) decreased the number of cell-surface TM molecules by approximately 20% compared to the primary culture. The number of TM molecules also decreased on HUVE treated by IL-1. The treatment of the cells with IL-1 also induced change of shape. TM was found not only normal syncytiotrophoblast but also on neoplastic syncytiotrophoblast of choriocarcinoma and hydatidiform mole. However, TM was not expressed on the three established cell lines. TM was found on various types of vascular tumors, including angiosarcoma.

[Morphological analysis of resumption of meiosis observed in oocytes of mouse vesicular follicles].

Resumption of meiosis observed in vesicular follicles of immature 32-34 day-old mice was classified morphologically, and the morphological mechanism to induce resumption of meiosis was examined. Resumption of meiosis was observed in 22.1% of vesicular follicles, and the dominant stage of the meiotic division was the first or second metaphase. Cumulus-enclosed and cumulus-free oocytes were separated from ovaries by dissection of ovaries. All of the cumulus-enclosed oocytes contained germinal vesicle, however 57.9% of cumulus-free oocytes did not, suggesting that the disappearance of cumulus cells around oocytes precedes the induction of resumption of meiosis. About 80-90% of the oocytes with germinal vesicle formed first polar body after 15 hrs of incubation. After incubation of oocytes showing resumption of meiosis, but without the first polar body, 36% of the oocytes extruded the first polar body indicating that resumption of meiosis is associated with progression to the second metaphase. Morphological characteristics of the surrounding structures of oocytes showing resumption of meiosis was classified into three categories; extensive proliferation of granulosa cells, degeneration of cumulus cells and their disappearance around oocytes, and detachment of cumulus cell-oocyte complexes from granulosa cell layer. It was suggested that cell-to-cell interaction among cumulus cells, oocytes and granulosa cells was impaired at the time of resumption of meiosis. Granulosa cells secrete an inhibitor of the resumption of meiosis. The resumption of meiosis after the surge of luteinizing hormone follows cumulus dispersion which make oocytes independent from the influence of granulosa cells by the disruption of communication.(ABSTRACT TRUNCATED AT 250 WORDS)

[Basic analysis of extracorporeal shock wave lithotripsy of gallstones].

Influence of extracorporeal shock wave lithotripsy (ESWL) on human gallstones placed in dog gallbladders was investigated. After irradiation of shock wave more than 600 times, maximal diameter of residual stones of 10 mm before irradiation was destroyed to 2 mm or less. Levels of serum transaminase (GOT.GPT) were elevated, in parallel with times of shock waves. These results suggested that 600 times of shock waves were enough to destroy one cholesterol stone, diameter of which was about 1 cm. Data of elevation of serum transaminase showed that more than 600 times of shock waves were possible to induce the damage of liver.

[Histological pattern of the tongue in the Japanese weasels, Mustela itatsi, with special reference to the morphology and distribution of papillae, taste buds and lingual glands].

Light and scanning electron microscopical demonstrations were carried out on the tongues of adult Japanese weasels (Mustela itatsi). Four types of papillae are present on the mucous membrane of the tongue; filiform, fungiform, vallate and foliate papillae. The vallate and foliate papillae are furnished with taste buds. Three types of lingual glands are present in the tongue; mucous (Weber's), serous (Ebner's) and mixed glands. Weber's glands are compound tubular glands which are well developed near the radix. Ebner's glands are compound tubular glands connected with the vallate papillae. Mixed glands are compound tubulo-alveolar glands and present in the lower half of the tongue, near the apex.

Immunohistochemical localization of thrombomodulin in chorionic diseases of the uterus and choriocarcinoma of the stomach. A comparative study with the distribution of human chorionic gonadotropin.

Thrombomodulin (TM) is a newly described protein that functions as a potent natural anticoagulant by converting thrombin from a procoagulant protease to an anticoagulant. TM is found on endothelium of the blood and lymph vessels and on syncytiotrophoblast of the placenta in humans. In the current study, we localized TM by immunohistochemistry in chorionic diseases of the uterus and choriocarcinomas of the stomach. The syncytiotrophoblast of hydatidiform mole contained TM. Uterine choriocarcinoma also had TM in the neoplastic syncytiotrophoblastic cells, although some of the neoplastic syncytiotrophoblastic cells showed no TM expression. Gastric choriocarcinoma never showed TM expression in the neoplastic syncytiotrophoblastic cells. However, all of the syncytiotrophoblastic cells had human chorionic gonadotropin (hCG) in hydatidiform mole, uterine choriocarcinoma, and gastric choriocarcinoma.
