The short-term metabolic effects of cloprednol, a new short-acting synthetic corticosteroid, were evaluated in four normal subjects. Cloprednol, 12.5 mg/day, in one subject had no appreciable effect on urinary excretion of sodium, potassium, nitrogen, or calcium. Cloprednol, 20 mg/day, in three subjects had no significant effect on mean daily urinary sodium and potassium excretion, although mild, transient sodium retention and kaliuresis were observed. One subject had increased nitrogen excretion and all three had a small increase in calcium excretion. Prednisolone, 40 mg/day, a dose with antiinflammatory potency equivalent to 20 mg/day cloprednol, given subsequently to two subjects under identical conditions resulted in sodium and potassium excretion results similar to those of cloprednol, 20 mg/day, but produced a much greater increase in nitrogen and calcium excretion. These results suggest that, like prednisolone, cloprednol lacks the sodium-retaining properties of hydrocortisone and raise the possibility that cloprednol has less of a deleterious effect on nitrogen and calcium excretion than prednisolone.
The effects of the administration of testosterone on plasma testosterone (T), estradiol (E), FSH, LH and prolactin (PRL) were tested in a group of men, aged over sixty. Blood samples were collected on the 2nd, 4th, 7th and 15th day afterr the administration of testosterone propionate (TP) (n = 28 men) and testosterone propionate with testosterone enantate (TP + TE) (n = 21 men). After the administration of TP + TE plasma T increased on days 2-4, returned to the normal basal level on the 7th day, and rose again on the 15th day. With the TP preparation the levels of testosterone remained elevated during the first seven days and returned to the basal values by the 15th day. The FSH and LH were suppressed during the first seven days after the injection of both TP and TP + TE preparations. On the 15th day gonadotropins were suppressed only by the administration of TP. The plasma concentration of E behaved like that of T. We found a significant increase in the PRL levels on the 4th day after treatment with 100 mg of the two preparations. Moreover, we found that PRL basal values were significantly higher in those subjects whose T basal values were low.
The different stages of the phagocytic process by splenic granulocytes of Tinca tinca were studied. Adherence capacity to both endothelium and tissue substrate, mobility rate, the phagocytosis capacity for both cells (Candida albicans) and inert particles (latex beads), candidicide power, and capacity of digestion measured by nitroblue tetrazolium (NBT) reduction were evaluated in splenic granulocytes of healthy adult tench. The capacity of adherence to nylon fiber was possessed by 51% of the granulocytes. The percentage capable of adherence to smooth plastic surfaces rose with incubation time. Casein, an effective chemoattractant, increased the random mobility of the granulocytes. Phagocytosis was greater for opsonized C. albicans than for nonopsonized. However, the number of phagocytosed yeast cells destroyed by the granulocytes did not depend on whether or not the C. albicans had been previously opsonized. The phagocytosis indices and the percent phagocytosis of latex beads were greater than those obtained for the phagocytosis of C. albicans in the absence of serum. Finally, the metabolic activity in these cells following the digestion of ingested material showed a 148 +/- 31% stimulation. The results show that splenic cells of tench have the capacity to make a phagocytic response against both cells (C. albicans) and inert particles (latex beads).
The influence of physical activity stress (swimming until exhaustion) and age on the antibody-dependent cellular cytotoxicity (ADCC) of lymphocytes from spleen, thymus and axillary lymph nodes in BALB/c mice was studied. The results indicate that the ADCC activity decreases after the acute stress both in young and in old mice. However, when swimming until exhaustion is performed after 1 month training, the ADCC activity increases except in the case of spleen lymphocytes from old mice. The basal ADCC capacity of spleen lymphocytes from old controls is higher (P<0.05) than from young controls, but there are no statistically significant differences in this respect between young and old animals in lymphocytes from thymus and axillary lymph nodes. No correlations between the increase in serum corticosterone levels and ADCC response are found.
It is well known that a bidirectional relation exists between the immune and the classic animal regulation systems, the nervous and endocrine systems. It has also been reported that ageing has a negative effect on neuro-immuno-endocrine integration. Since it has been shown that norepinephrine (NE) differently modulates the phagocytic process of macrophages from young or old mice, and considering that NE has a short lifetime, the aim of this study is to evaluate the possible immunomodulatory role of the NE end-metabolite, 4-hydroxy-3-methoxyphenyl-glycol (HMP-glycol), on the superoxide anion levels of peritoneal macrophages from young-adult (22+/-2 weeks) and old (72+/-2 weeks) BALB/c mice. Macrophages were incubated with HMP-glycol in a range of concentrations between 10(-12) and 10(-3)M, and superoxide anion levels (both extracellular and intracellular) were evaluated by using the NBT reduction test. The response of macrophages to HMP-glycol was different between young-adult and old animals. While all concentrations (10(-12), 10(-9), 10(-7), 10(-5), 10(-3)M) of HMP-glycol led to lower extracellular O(-)(2) levels (p<0.05) in macrophages from young-adult mice, only the 10(-3)M concentration of HMP-glycol led to a significant decrease in the extracellular O(-)(2) production in macrophages from old mice. No significant changes were found in the intracellular O(2)(-) levels when macrophages were incubated with the different concentrations of HMP-glycol in either young-adult or old mice. Thus, the modulation of macrophage function by NE not only depends on their concentration, but also on the products of NE catabolism. The effect of NE metabolite will also depend on the age of the animals as there is an age-dependent decreased capacity to respond to low concentrations of HMP-glycol.
To study the possible role of sexual hormones, Testosterone (T), Estradiol (E2), follicle-stimulating hormone (FSH), and luteinizing hormone (LH) on the growth axis, we examined the correlations between the sex, growth hormone (GH), insulin-like growth factor-1 (IGF-1), insulin-like growth factor binding protein 3 (IGFBP3), FSH, LH, T and E2, in growth retarded children ranging in age from 7 to 13 yr. All hormones were measured by Radioimmunoassay (RIA) in a pool of aliquots of samples obtained every 20 min over 12 h (overnight) in each child. Multivariate statistical analysis was performed. We have found: a) Thai only FSH concentrations were significantly higher in girls than in boys; b) A positive correlation between T-IGF-1; T-IGFBP3; FSH-LH; FSH-IGF-1; FSH-IGFBP3, LH-IGF-1, LH-IGFBP3 the sex-FSH; and the sex-IGFBP3 c) A high positive correlation between plasmatic E2 and IGF-1/IGFBP3 ratio (an index of free, active IGF-1). We concluded that the sex, FSH, LH, T and E2 influence the growth axis. The sex through IGFBP3; LH, FSH, and T through IGF1 and IGFBP3; E2 through the IGF-1/IGFBP3 ratio, an index of active IGF-1.
Previous investigations in our laboratory have shown that testosterone implanted into the lateral septum in male rats increases LH and FSH secretion. However, it was unclear whether the effect of testosterone was direct via androgen receptor, or indirect via the estrogen receptor after conversion by aromatization to estradiol. To answer this question, we implanted either testosterone or the non-aromatizable androgen 5 alpha-dihydrotestosterone (DHT), into the lateral septum of adult male rats and measured plasma levels of LH and FSH by radioimmunoassay 2 days after implantation. Both testosterone and DHT significantly increased the plasma LH and FSH concentrations. Mean concentration of LH in control animals was 0.21 +/- 0.06 ng/ml, a figure that increased to 0.7 +/- 0.12 and 0.55 +/- 0.1 ng/ml after DHT or testosterone implantation respectively. Mean concentration of FSH in control animals was 1.5 +/- 0.3 ng/ml; this figure increased to 3 +/- 0.3 and 2.9 +/- 0.3 ng/ml after DHT or testosterone implantation. Neither plasma DHT (64.0 +/- 5.6 vs. 52 +/- 5 ng/100ml) nor plasma testosterone levels (4.1 +/- 0.38 vs. 3.3 +/- 0.18 ng/ml) were significantly affected by the implants. We conclude that androgens independently of conversion to estrogen acting in the lateral septum facilitates the release of LH and FSH.
- The effects of acute alcohol intoxication (AAI) on the pituitary-gonadal axis hormones, and the possible contribution of pituitary-adrenal axis hormones, beta-endorphin and prolactin to alcohol-induced dysfunction of pituitary-gonadal axis hormones were studied in adult men and women. Blood samples were drawn from adults of both sexes who arrived at the emergency department with evident behavioural symptoms of drunkenness (AAI) or from adult volunteers with nil consumption of alcohol (controls). Our results demonstrated that AAI produces a high increase in plasma prolactin, corticotropin (adrenocorticotropic hormone, ACTH), and cortisol in adults of both sexes, a decrease in luteinizing hormone levels only in men, an increase in dehydroepiandrosterone-sulphate (DHEAS) and a contradictory behaviour of testosterone according to gender, with increased plasma testosterone in women and a decrease in men. ACTH and prolactin correlated positively with cortisol, DHEAS and testosterone in women, which suggests that prolactin and ACTH could contribute to stimulated adrenal androgen production. In contrast, the decrease in testosterone and increase in beta-endorphin in men suggests that AAI could have an inhibitory effect on testicular testosterone, perhaps mediated by beta-endorphin. Our results suggest that the effect of alcohol on pituitary-gonadal axis hormones in humans could depend on the gender and degree of sexual maturity of the individual.
In seven patients with active fascioliasis of the liver, CT revealed nodular intrahepatic lesions of diminished attenuation, as well as peripheral branching formations. Computed tomography was negative in a single patient with quiescent disease. Computed tomography can be a useful tool for the diagnosis of this disease during the invasive period and also to evaluate response of patients to medical treatment.
A clinical case of cerebral infection by parenchymal racemose cysticercus, diagnosed by means of morphological characteristics in a patient of Valdivia city is described. The rare frequency of parenchymal location of racemose cysticercus as well as its differential diagnosis with other larval stages of cestodes that develop in the brain and its treatment are discussed.
It is generally assumed that unsaturated fatty acids are important biological mediators that may be applied in the treatment of patients suffering from autoimmune disorders, in the modulation of tumorigenic processes or in the induction or inhibition of apoptosis. In this study, mice divided into three groups were fed dietary lipids; olive oil (20% w/w), fish oil (20% w/w) or hydrogenated coconut oil (20% w/w) for four weeks. Then, survival percentage, natural killer (NK) cell activity and lipid peroxidation were measured after murine lymphoma (LSTRA) transplantation. Survival percentage showed a reduced resistance to LSTRA tumor in mice fed fish oil diet and generaly loss of NK cell activity was observed after lymphoma transplantation. Lipid peroxidation from mice fed dietary lipids was measured in the presence of both phospholipase and cyclooxygenase inhibitors. In general, the data contained in this paper point to a detrimental effect caused by dietary lipids in mice transplanted with a lymphoma.

Nasal mucociliary transport was studied in 33 HIV-positive patients with different stages of infection. Ciliary ultrastructure was studied in 8 of these cases. At the time of study, 24 patients had respiratory infections. Disturbances in mucociliary transport were found in 25 cases. In 6 of the 8 studies of ciliary ultrastructure, changes in the ciliary axonema were found, particularly in the central and peripheral microtubules.
