Urinary glycolipids of YC8 lymphoma bearing mice: effectors of glycosylation process.

Glycolipids from YC8 lymphoma bearing mice urines have peen obtained by the method of Folch and fractionated on silicic column and on silica gel plates. One fraction, among the five obtained, may be used by sera galactosyltransferase as an acceptor for galactose, to form a new glycolipid compound. Furthermore this fraction has shown an inhibitory effect on phosphatase activity, in agreement with hypothesis which relates relates glycosyltransferase and phosphatase activities during tumorous process.

[Determination of anti-acetylcholine receptor antibodies in myasthenia and its treatment by plasma exchange and immunosuppressants].

Recent works have confirmed the auto immune mechanism in myasthenia gravis, that Simpson had hypothetized early as 1960. A post synaptic blockage, of which Ach-R antibodies seem to be chiefly responsible, is now accepted though the exact pathogenesis remains unknown. The immuno assay of these antibodies is still difficult; their blood level evaluates the course of the disease and the effectiveness of the treatment. The withdrawal of the antibodies by plasma exchange (P.E.) and the inhibition of their secretion by immunosuppression (I.S.) lead to remission in many cases. Principle of the immuno assay and results of P.E. and I.S. are reported in 21 cases.

[Demonstration of N-acetylgalactosaminyltransferase activity in the murine lymphoma YC8].

The N-acetyl-galactosaminyltransferase activity has been studied in the biological fluids of YC8-lymphoma bearing Balb/c mice. It is enhanced during tumor development from 1 to 30 times in peritoneal fluids and from 1 to 10 times in sera. This activity is nil in urines. Optimal requirements for activity have been determined. Results suggest the existence, during tumor process not only of an enhancement of enzymatic activity, but also of a new molecule synthesis, molecules which are endogenous acceptors for the enzyme.

Galactose transfer and UDP-galactose hydrolysis in urine from normal Balb/c and Balb/c YC8 mice.

By chromatographic method we have shown the existence of a complex system for galactose transfer from UDP-galactose and for nucleotide hydrolysis in urines from Balb/c YC8 and normal Balb/c mice. By action of sera from normal and ascitic mice as source of enzyme, we have been able to detect transfer for galactose in urines from ascitic mice and an important inhibitory effect of the nucleotide sugar hydrolysis by the sera with urines from normal mice.

[Demonstration of nucleotide pyrophosphatase and phosphohydrolase activities in normal lymphocyte of Balb/c mice and absence of these activities in YC8 tumor cells and in lymphocytes of animals with YC8 lymphoma].

Saline extraction of tumor cells from YC8 lymphoma transplanted in Balb/c Mice, of lymphocytes from animals bearing this tumor and of lymphocytes from control animals, allowed us to show important hydrolytic activities towards UDP-[(14C)]-galactose and galactose-[(14C)]-1-phosphate in normal lymphocytes. These activities disappear in lymphocytes from Mice bearing this tumor and in tumor cells themselves.

[Role of central catecholamines in the psychostimulant activity of pyrovalerone].

A comparative study of the effects of alpha methylparatyrosine (alpha MPT) and/or reserpine pretreatments on mice motorhyperactivity and rat stereotyped behaviour induced by pyrovalerone and amphetamine suggests a different mechanism for these two substances. Both behavioral effects were abolished by alpha MPT but not altered by reserpine in the case of amphetamine which presumably acts through a selective release of "newly synthesized" catecholamines from a "functional" pool. In contrast to this, pyrovalerone would increase spontaneous motor activity through a preferential release of norepinephrine from a "storage" pool since motorhyperactivity was not altered by alpha MPT especially during the first phase, whereas it was abolished by reserpine. Stereotyped behaviour induced by pyrovalerone, was still present after pretreatment with alpha MPT or reserpine; these data suggest an action through a release of both "newly synthesized" and "stored" dopamine. On the other hand, a direct action on dopamine receptors might be involved after high doses of pyrovalerone and amphetamine since stereotyped behaviour was found to be present after a combined pretreatment with alpha MPT + reserpine.
