Spleen samples from idiopathic thrombocytopenic purpura (ITP) patients were investigated for ultrastructure and peroxidase activity. The white pulp was characterized by the presence of many large secondary lymphatic follicles and periarteriolar sheaths formed from lymphocytes and plasma cells. Their ultrastructural details substantiate the process of lymphocyte differentiation and the immunopathologic mechanism of the disease. The vascular lesions proved to be subendothelial hyaline deposits. The platelet destruction process and phagocytosis resulted in the occurrence of fibrin clusters. Macrophages were also described as stainable body macrophages, foamy macrophages and a third type dealing with fibrin phagocytosis.
The ultrastructural pattern of endogenous peroxidase of monocytes has been investigated in bone marrow samples as well as in the blood of patients labelled as myelomonocytic leukaemia (MML) after clinical and laboratory investigation. The predominance or the exclusive presence of a granular pattern in the bone marrow proved to be consistent with chronic MML cases. On the other hand, the presence of the peroxidase in RER, nuclear envelope, Golgi complex and granules in the bone marrow and even in the peripheral blood occurred in acute MML patients.
The authors have carried out a populational study by performing psychomorphological and blood determinations, and have detected the presence of a chronic hyperactivity syndrome of the adrenals in 12,5 percent of the healthy young subjects engaged in various activities that they have investigated. In an experimental study the oxydative processes in the enzymatic cycle of Krebs were cut off by injecting the adrenal medullary area with a 40% solution of sodium salicylate, while the secretion of the adrenal cortex was maintained, as was demonstrated by the unaffected activity of the hexose monophosphate shunt in this tissue. The experimental results demonstrated that the adrenal medullary tissue represents a major pathogenetic link in the complex etiopathogeny of essential arterial hypertension, and this invalidates the role attributed by Volhard (in 1931) to the renal tissue, explaining the failure of haemodialysis, bilateral nephrectomy and kidney transplantation in preventing the recurrence of essential arterial hypertension.
Multivalent hybrid antibody (MHA) complexes with double specificity were prepared by combining two different antibodies, one specific against ferritin (Fer), the other against horseradish peroxidase using protein A from Staphylococcus aureus (SpA). Electron microscopy of mouse spleen lymphocytes and thymocytes (the latter coated with mouse anti-Thy-1 antibody) reacted with anti-HRP/SpA/anti-mIg complex and HRP showed the pattern of surface Ig receptors visualized by the specific peroxidase labelling. When IgG anti-Fer/SpA/IgG-anti-mIg complex was applied to the same cellular stuff, better resolution and higher accuracy were obtained although the distribution was similar. Surface Thy-1 alloantigen and Fe receptors (charged with hIgG) were simultaneously detected on the thymocyte surface by using in the same way a mixture of anti-Fer/SpA/anti-Thy-1 and anti-HRP/SpA/anti-Fab. The concomitant ultrastructural visualization of Thy-1 (with Fer) and Fc (with HRP) on mouse thymocytes clearly showed that their distribution was mainly independent and that the amount of Thy-1 antigen prevailed. These data show that electron microscopy with a mixture of MHA may be a useful technique for the concomitant location of two antigenic determinants on the cell surface.
The selective inhibitory effect of sodium salicylate (2.5 M) on the adrenal medulla was investigated histopathologically, histoenzymatically, electron microscopically and biochemically. It was proved that the different behaviour of nicotinamide-adenine dinucleotide (NAD) and nicotinamide-adenine dinucleotide phosphate (NADP) linked dehydrogenases under the action of sodium salicylate can account for the survival of cortical cells, while the medullar ones are necrosed.
The syndrome of constitutional chronic hyperadrenalism was detected biochemically by blood cathecholamine assays as well as psychically and morphologically in 12.5% of the young apparently healthy subjects from Bucharest, investigated. Experimentally, the enzymatic cellular oxidative process in the adrenal medulla was blocked by chemical sclerosis of the adrenal medulla with a 40% solution of sodium salicylate (2.5 M), without influencing cortical secretion ensured bythe hexose monophosphat shunt. Chemical medullosclerosis associated with splanchnicosympathectomy and with or without suprarenalectomy on the opposite side performed in 60 patients with severe essential arterial hypertension resistant to drug therapy gave good results depending on the anatomical and clinical evolutive stage of disease.

Two patients with congenital dyserythropoietic anemia (CDA) type II were investigated in bone marrow as well as in blood cells. The majority of red cell precursors showed typical morphological abnormalities: high incidence of binucleated erythroblasts, normoblastic hyperplasia, nucleo-cytoplasmic dissociations, karyorrhexis of some nuclei, presence of synartesis phenomenon, invagination of cytoplasmic portions into the nuclear area and presence of marginal cisterna. The modified erythroblasts were present in the peripheral blood, too. The presence of Gaucher-like cells suggests an abnormal catabolism of the red cell precursors membranes.
Six cases of essential thrombocythemia (ET) have been investigated in bone marrow as well as in blood cells. In majority of the cases, the bone marrow aspirate was hypercellular and presented an increased number of megakaryocytes, some of them with displastic appearance. The ultrastructural pattern of platelet peroxidase permitted us to identify atypic megakaryocytes and megakaryoblasts, particularly when present in pheripheral blood.
The study presents 6 cases of leukemia/lymphoma (with mature T-cells) corresponding to the diagnostic criteria of adult T-cell leukemia (ATL): adult age onset of leukemia/lymphoma, organomegaly but normal mediastinum, leukemic cells with typical morphology and phenotype hypercalcemia. The evolution of disease was severe of subacute with resistance or partial response to therapy. The virologic assays were positive according to the ELISA test in four cases of which two presented, according to the Western blot assay, HTLV-I infection. The epidemic aspect of this infection is discussed as well as the possibility of contamination and the geographic spread of the places of origin of the patients. Emphasis is laid on the young age (22-26 years) of three of the patients infected, as a peculiar feature of the disease in Romania.
This review tried to offer a basis for properly integrating genetic, biochemical, and cellular indicators of apoptosis in order to facilitate a better understanding of the control and mechanisms of apoptosis.
This review presents the implications of abnormal regulation of apoptosis in the pathogenesis of a variety of diseases, some of them characterized by a failure of cells to undergo apoptosis, and the others characterized by cell loss. The new perspectives in the therapy of such diseases by developing therapeutic agents that increase or decrease the susceptibility of particular cells to apoptosis are also reviewed.
We established the cell line of bone marrow and blood cells provided by 19 cases of cMPD by ultrastructural studies of their enzymatic content. Myeloblasts, megakaryoblasts and lymphoblasts were identified by the presence of myeloperoxidase, platelet peroxidase and acid phosphatase, respectively.
