Prediction of pulmonary function after resection of primary lung cancer. Utility of inhalation-perfusion SPECT imaging.

To help determine whether preoperative perfusion and inhalation SPECT imagings are useful in predicting postoperative lung function, Tc-99m MAA perfusion SPECT imaging, CT scans, and pulmonary function tests were prospectively performed in 33 patients with primary lung cancer before and after lobectomy or pneumonectomy. Tc-99m Technegas inhalation SPECT imaging was performed in 6 of 33 patients as well. The authors also studied changes in radioactivity on the operated and nonoperated sides before and after surgery, examined the lowest limit value for adaptability to the operation, and made a comparison of both perfusion and inhalation SPECT imaging. The predicted postoperative values obtained from the preoperative Tc-99m MAA SPECT images correlated more closely with the measured 6-month postoperative values than with the measured 3-month postoperative values. The highest correlation coefficient (r = 0.86) was observed between the predicted forced vital capacity (FVC) value and the measured 6-month postoperative FVC value. In many cases, there was not a great difference between the 6-month and 3-month radioactivity on the operated side obtained from Tc-99m MAA SPECT images. This appears to indicate that pulmonary blood flow on the operated side has completely recovered by 3 months after surgery. However, radioactivity in both the upper and lower lobes of the nonoperated side increased soon after surgery compared with that before the operation, and had not returned to preoperative levels 6 months after surgery. Genomic organization of 57 ribosomal protein genes in rice (Oryza sativa L.) through RFLP mapping.

Four hundred cDNA clones from rice (Oryza sativa L.) callus and root cDNA libraries, with a high similarity to about 70 kinds of ribosomal proteins (r-protein) in eukaryotic as well as procaryotic organisms, were identified by their deduced amino acid sequences. Southern hybridization of 114 independent cDNA clones with total rice genomic DNA showed 77 distinct and specific hybridization patterns. Of the 77 clones representing the above hybridization patterns, copies of 67 clones corresponding to 57 r-proteins could be estimated and, among these, only 6 clones were single copy, indicating that almost 90% of these r-proteins in rice were encoded by small multigene families. Loci of 36 r-protein genes could be mapped on the rice linkage map by using 30 full-length cDNA clone sequences from specific RELP bands. Another 21 expressed gene loci were mapped using 3' untranslated region specific cDNA probes amplified from the multicopy cDNA clones representing 17 of the r-protein multicopy gene families. The above 57 loci were mapped from 51 cDNA clones and 41 of these r-protein genes mapped to regions that did not show any clustering, while in 5 cases, pairs of r-protein genes cosegregated or linked closely. The r-protein genes in rice were located throughout the 12 chromosomes and it was found that more than one copy within a multigene family may be expressed simultaneously.

Toward cataloguing all rice genes: large-scale sequencing of randomly chosen rice cDNAs from a callus cDNA library.

A large-scale sequence analysis of rice cDNA was performed for a library from rice callus cultured in a medium containing 1 p.p.m. of 2,4-dichlorophenoxyacetic acid. Random sequencing of 2778 cDNA clones generated 2259 non-redundant expressed sequence tags (ESTs). The strategy of sequencing cDNAs can yield quickly a large number of novel genes. After translation, 690 sequences showed a significant amino acid sequence similarity to sequences already known from PIR. The source of known proteins ranged from bacteria to human. In this report, the non-redundant set of 280 identified ESTs is analyzed in detail.

[A case of clinical Reye syndrome with symmetrical abnormal signal areas in the pons and thalami by MRI].

We report a two-year-old boy with clinical Reye syndrome. Brain CT disclosed symmetrical low-density areas in the pons and thalami, a part of which was recognized as being of high-intensity on both T1 and T2 weighted MRI. These lesions were suspected of being caused by vascular involvement, since the methemoglobin induced by bleeding may have contributed to the high-intensity regions. The lesions gradually disappeared within four months after the onset, and clinical symptoms also improved markedly without serious neurologic impairment. This case was characterized by the marked improvement in neurological and radiological findings in spite of the serious neurological involvement at onset.

[Superselective bronchial arterial infusion therapy with cisplatin and epirubicin hydrochloride, mitomycin C-iohexol-Lipiodol emulsion (EMILE) for hilar lung adenocarcinoma: preliminary clinical experience].

A case of hilar lung adenocarcinoma was treated by superselective bronchial arterial infusion therapy with cisplatin and epirubicin hydrochloride, mitomycin C-iohexol-Lipiodol emulsion (EMILE) using Tracker -18 infusion catheter. The tumor size was reduced on follow-up CT scans. However, EMILE was also distributed to nontumorous lung tissues around the tumor, and a shrinkage of the right upper lobe and elevations of the right hilus and diaphragm followed. No major complaints and clinical complications during and after the treatment occurred. This therapy was safe and effective for local tumor reduction in a case of hilar lung adenocarcinoma.

Antitumor activity of oenothein B, a unique macrocyclic ellagitannin.

The antitumor effect of oenothein B, a macrocyclic ellagitannin from Oenothera erythrosepala Bordas, on rodent tumors was studied. Oenothein B exhibited a strong antitumor activity against MM2 ascites tumors upon intraperitoneal administration to the mice before or after the tumor inoculation. The tannin also inhibited the growth of Meth-A solid type tumor in mice. This antitumor effect of the tannin could not be attributed to its direct cytotoxic action on tumor cells, because the cytotoxicity was very weak in the presence of serum protein. When oenothein B was injected into the peritoneal cavity of mice, peritoneal exudate cells, including cytostatic macrophages, were induced. Furthermore, in the in vitro treatment of macrophages from mice and humans, the tannin stimulated release of an interleukin 1 (IL-1)-like activity and IL-1 beta from the cells. These results suggest that oenothein B exerts its antitumor effect through potentiation of the host-immune defense via activation of macrophages.

Subsite structure of Saccharomycopsis alpha-amylase secreted from Saccharomyces cerevisiae.

The kinetic parameters (kcat/Km) and the cleaved-bond distributions for the hydrolysis of linear maltooligosaccharides Gn (3 less than or equal to n less than or equal to 9) by Saccharomycopsis alpha-amylase (Sfamy) secreted from Saccharomyces cerevisiae were determined at pH 5.25 and 25 degrees C. The subsite affinities of Sfamy were also evaluated from these data. The subsite structure of Sfamy is characteristic of the active site of an endo-cleavage type enzyme, consisting of internal repulsive sites with the catalytic residues and external attractive sites. Moreover, the pKa values of the catalytic residues were calculated from the pH dependence plot of the kinetic parameter (kcat/Km). The amino acid residues which contribute to the subsite affinities and the catalytic activity of Sfamy are proposed and compared with those of Taka-amylase A.

The enzymatic and molecular characteristics of Saccharomycopsis alpha-amylase secreted from Saccharomyces cerevisiae.

The Saccharomycopsis fibuligera alpha-amylase (Sfamy) gene was expressed in Saccharomyces cerevisiae. The highest productivity of Sfamy was 70 mg per liter of culture broth. We purified Sfamy from the culture broth and identified the NH2 terminal primary sequence. This sequence suggests that the Sfamy gene product is synthesized as a pre-pro-precursor, and the pro-sequence is cleaved after a Lys-Arg sequence with the calpain-like endopeptidase encode by the KEX2 gene, resulting in mature Sfamy protein composed of 468 amino acids. Furthermore, the enzyme Sfamy is a glycoprotein in which one N-linked sugar chain containing mannose residues is attached to the Asn residue at the 198 position. The Km and kcat values were 1.1 x 10(-4) M and 1.4 x 10(2) sec-1, respectively, using amylose (the degree of polymerization n = 18) as a substrate. Moreover, the secondary structure, the location of the secondary elements including alpha-helix, beta-sheet, and loop, and tertiary structure were predicted theoretically on the basis of the molecular structure of Aspergillus oryzae alpha-amylase. Taka-amylase A (TAA). These results indicate that Sfamy protein is composed of main (M) and C-terminal (C) domains. The molecular structure of M domain closely resembles that of TAA, but the C domain appears to be more compact than that of TAA because of deletions at three regions forming turns and one region forming alpha-helix.

Liver and kidney nitrogen uptake in rats fed beans enriched with 15nitrogen through (15NH4)2SO4 used as soil fertilizer.

This study was conducted to determine the incorporation of 15nitrogen (15N) into liver and kidney of a group of rats (N = 17) fed a mixture of cooked rice and beans for 4 weeks. The beans (Phaseolus vulgaris), grown in soil cultivated with (15NH4)2SO4, had 1.5% 15N enrichment. Three or four rats were sacrificed weekly and a control group (N = 13) received an isonitrogen and isocaloric reference casein diet. The amount of nitrogen transferred from the beans to the liver at the end of the first, second, third and fourth weeks was 17, 11, 27 and 29% of the total nitrogen, respectively. For the kidney, the respective values were 10, 9, 25 and 27% of the total nitrogen. The total nitrogen content of the liver and kidney of rats receiving the reference casein control diet was similar to that of the rice/beans animals. Weight gain was similar for the two groups, indicating the well-balanced biological value of the experimental diet. The present study shows that it is possible to monitor 15N incorporation into beans and subsequently into rat liver and kidney, in contrast to classical methods for protein metabolism evaluation which measure only total body nitrogen intake and excretion or weight gain.

Glycolipids of the bovine pineal organ and retina.

Neutral and acidic glycolipids from the bovine pineal organ and neutral glycolipids from the bovine retina were characterized. The chemical structures of the isolated glycolipids were determined by means of carbohydrate analysis, methylation analysis, enzyme treatment, fatty acid analysis, long chain base analysis, mass spectrometry, NMR spectroscopy, and IR spectroscopy. GM3, GD3, and GT1 were the major bovine pineal organ gangliosides, GD3 accounting for 75% of the total gangliosides. Galactosylceramide, glucosylceramide, and lactosylceramide were found in both the bovine pineal organ and retina. Sulfatide was also present in both tissues. It had already been reported that the major bovine retina ganglioside was GD3 (Handa, S. & Burton, R.M. (1969) Lipids 4, 205-208). The glycolipid patterns of the two tissues were very similar to each other and quite different from those of other tissues.

[A case of prostate cancer with multiple pulmonary metastases].

A 65-year-old man was hospitalized with bloody sputum. His chest X-ray showed multiple nodules in both lung fields. Transbronchial lung biopsy demonstrated a poorly differentiated adenocarcinoma, which suggested that respiratory abnormalities might be metastatic cancer. Because he had noticed pollakisuria and dysuria, urologic consultation was sought. The findings of digital examination, urethrography, and ultrasonotomography suggested that he had an advanced prostate cancer. In addition, tumor markers of prostatic acid phosphatase (PAP), acid phosphatase (ACP), and prostate antigen (PA) showed abnormal titers; 120 ng/dl, 166 IU/l, and 15.4 ng/ml, respectively. The prostate tissue obtained by transperineal biopsy revealed histopathologically adenocarcinoma and positive findings in immunohistochemical staining for PAP and PA as well as the specimens from the lung. Bilateral orchiectomy and medication of 250 mg of DESD daily as an antiandrogen therapy improved respiratory symptoms. One week after the operation, the multiple shadows on the chest X-ray diminished dramatically. Moreover, serum values of PAP and PA also decreased to the normal range. He is alive in a stable condition 6 months after the operation.

[Formation of experimental obesity by Crisco].

Randomized male litter S.D. rats (weight: about 60 g, 1 group: 10 rats) were used. By using Crisco (hydrogenated cotton seed oil) within the limits of 10 approximately 60%, the intake of the 30% group was high and a good incidence rate of obesity (about 100%) was obtained in the 6 month experiment. In the 30% group, increases of wet weight in perirenal retroperitoneal and subcutaneous fat depots, increases of the number and diameter of fat cells (perirenal retroperitoneal fat depot), elevations of liver fat % and serum insulin level, and a fall of liver phosphoenolpyruvate carboxykinase activity were observed in the 3 month experiment.

[Keypoints and compositions of total parenteral nutrition for patients with low glucose tolerance levels].

Management of total parenteral nutrition (TPN) in depressed glucose metabolism was investigated clinically and experimentally in view of insulin control and/or new component of carbohydrate solution. Fifty TPN cases out of 837 for 9 years were successfully performed insulin control, while 17 patients were unable to get sufficient calory in spite of insulin administration. Cumulative expired CO2 after injection of radioactive carbohydrate in rats showed that each carbohydrate was utilized in the order of glucose, fructose, maltose, sorbitol and xylitol even in depressed glucose metabolism and that depressed carbohydrate metabolism was improved by adequate insulin injection. Combined use of glucose, fructose and xylitol at 4:2:1 (GFX) was was experimentally revealed to be superior to glucose alone as carbohydrate source of TPN in depressed glucose metabolism. Compared with conventional TPN (C-TPN), GFX-TPN showed lower blood glucose and insulin level in rabbits of sepsis and rats of streptozotocin diabetes. Contents of fructose 2,6 bisphosphate and triglyceride and activities of fructose 6 phosphate 2 kinase, acetyl CoA carboxylase and fatty acid synthetase in liver of these animals supported that GFX had favourable effects on glucose and fat utilization in depressed glucose Blood glucose of early postoperative patients was lower in GFX-TPN than in C-TPN.
