This introductory paper outlines the problems and concepts dealt with in a workshop organized by the German Histochemical Society on developmental aspects of the nervous system. Because this workshop also included demonstrations of techniques, it also includes a discussion of some practical problems in the study of the development of the nervous system.
A displacement of structures in the hypothalamic region of the rat has been reported in previous papers. 1 micron granules located in neuroblast like cells containing catalase activity (which were detected by enzyme histochemical methods) undergo a shift from the median eminence (at day 22 after birth) towards the ventromedial part of the hypothalamus (at day 35 after birth). This displacement terminates in the medio-basal part of the arcuate nucleus (at day 55 after birth). A similar displacement of dopaminergic fluorescent perikarya has been described following the same route, with the same time sequence. Neonatal application of monosodium glutamate (MSG) leads to an acceleration of the displacement of both the catecholaminergic and the catalase containing perikarya. However after day 25 (after birth) all fluorescent perikarya disappear whereas the catalase containing cell survive when they finally reach the arcuate area.

In order to get more insight into the mechanism by which the onset of puberty is controlled, a developmental study on the displacement of catalase- and dopamine-containing cells in the hypothalamic region was done in rats which received a neonatal lesion of the suprachiasmatic nucleus. The displacement of cells is delayed after these lesions. However, the time lost at the beginning of the displacement is made up at the end of the migration.
Liposomes are combined with Lucifer Yellow in order to evoke neuronal uptake of this fluorescent dye in the medial basal hypothalamus. Lucifer Yellow liposomes are preferentially taken up by hypothalamic neurons. The dye spreads into the dendrites and axons, producing Golgi-revealing views of several neurons near the injection site in which both untrapped and liposome-entrapped Lucifer Yellow has been microiontophoretically administered. The capriciousness of the labeled neurons makes it possible to study the dendritic arborization and spines, while the axon with its collaterals can be followed to their targets. Moreover, the dye is also taken up retrogradely. When retrograde uptake over great distances occurs, the Golgi-like appearance is missing, because only then is perikaryal labeling found.
After sectioning of the goldfish optic nerve a number of enzyme histochemical changes are observed in the hypertrophied retinal ganglion cells and in the optic nerve. Between one and eighteen days postoperatively an increase in the amount of acid phosphatase reaction product is noted. The enhanced activity decreased to normal first in the optic nerve, followed by the optic tract and tectum. Four days postoperatively higher levels of activity were noted in the hypertrophic retinal ganglion cells for the enzymes NADH tetrazolium reductase, cytochrome oxidase, glutamate dehydrogenase and lactate dehydrogenase. The same enzymes also showed an activity increase in the lesioned optic nerve after four to ten days postoperatively, beginning at the cut and gradually spreading towards the optic tectum. Between fifteen and eighteen days the activity dropped to normal in the hypertrophic retinal ganglion cells, while in the lesioned nerve raised levels of reaction products could be seen till days thirty-five and/or forty-five. It was concluded that the degeneration of the optic pathway is marked by the increase of acid phosphatase activity, whereas the process of regeneration is characterized by an increase of NADH tetrazolium reductase, cytochrome oxidase, glutamate dehydrogenase and lactate dehydrogenase activities. The possible functional implications of these enzymes in the regenerative phenomena are discussed.
The monoclonal human granulocyte antibody B4,3 shows reactivity with structures in the external granular layer of the developing rabbit cerebellum. This reactivity is first observed at post-conceptional day 19-20 and disappears at postnatal day 5-6. The reactivity in the immature rabbit cerebellum is distributed in a longitudinal pattern of bands positive and negative for this monoclonal antibody.

Two histochemical different populations of neurons migrate from the median eminence (ME) into the arcuate nucleus (ARC) at puberty in rats. Therefore the cellular composition of the ARC was studied in normal and monosodium glutamate (MSG) treated female rats at 22, 35, 55 days after birth and in mature ones. Golgi-Cox and mainly Rio-Hortega techniques with Nissl counterstaining were applied. In the mature arcuate nucleus several types of Golgi impregnated cells can be discerned. Neonatal MSG-treatment exclusively destructs the arcuate dopaminergic cells. From the Golgi-preparations it appears that in these animals one type is indeed damaged in or at the ARC.
The administration of monosodium glutamate (MSG) to newborn rats accelerates by two weeks the migration of fluorescent dopaminergic perikarya from the median eminence towards the arcuate nucleus at the time of puberty, which normally occurs from day 22 till 55 after birth. It is hypothesized that MSG affects the trigger mechanism for this migration. The disappearance of fluorescent dopaminergic cells that occurs after MSG administration is considered as a secondary effect.
Enzyme histological changes have been studied in several optic projection areas after right optic nerve lesion in goldfish. An increase in acid phosphatase activity was found in the optic tectum, nucleus rotundus, nucleus geniculatus lateralis and area pretectalis between 2 and 15 days postoperatively. The enzymes glutamate dehydrogenase, lactate dehydrogenase, NADH tetrazolium reductase, cytochrome oxidase, succinate dehydrogenase and beta-hydroxybutyrate dehydrogenase showed a decrease in activity in all or some of these projection areas. No changes were found in acetylcholinesterase activity after optic nerve lesions. Three weeks postoperatively, all enzyme activities returned to the same level as on the normal side. The results are discussed in relation to possible neurotransmitters in goldfish optic terminals.
The transition from infancy to adulthood is characterized by a sequence of events which are not yet fully understood. Hormonal changes influencing neuronal circuits in various hypothalamic area are--at least in part--held responsible. Using catecholamine fluorescence techniques in rats it has been found that a displacement of dopaminergic cells can be seen; the fluorescence activity shifting from the median eminence at day 20 after birth to the dorsolateral part of the arcuate nucleus around day 45 after birth, as is the case for catalase positive cells in these areas. It is concluded that the displacement of at least two populations of cells from the median eminence to the arcuate nucleus runs parallel to the onset of puberty. The relation of this shift in respect to the suprachiasmatic-preoptic control will be discussed.

In this description of the morphology of the mouse cerebellum, the main landmarks for subdivision were the discontinuities of the cortical sheet. In certain cerebellar sulci in the posterior lobe, the cortex is absent. Within the paramedian sulcus the cortex is interrupted rostral and caudal to the copula pyramidis. Rostrally, this discontinuity between vermis and hemisphere occupies a position corresponding to the intercrural sulcus of the ansiform lobule of other mammals. A subdivision into the simple lobule (VI and HVI), ansiform and paramedian lobule (HVII) and vermal lobule VII is proposed for this region, which accounts for the position of this cortexless area in the intercrural sulcus. Caudal to the copula pyramidis is the cortex is interrupted at the bottom of the paramedian sulcus between lobules X, IX and caudal VIII medially, and the paraflocculus and flocculus laterally. In most mammals, this area, devoid of cortex, extends much farther in the interparafloccular sulcus between the dorsal paraflocculus and the flocculus with the ventral paraflocculus. From the medullary surface of the cerebellar peduncles an area without cortex extends in between the ansiform lobule and the paraflocculus onto the copula pyramidis and the paramedian lobule. The nomenclature of the distal, foliated part of the paraflocculus in mouse, rat and rabbit is discussed.
